Claims (14)
- 1.膣分泌物の試料中に存在するタンパク質を測定することに基づいた、胎児膜 の破裂を検出する診断方法であって、 検出されるタンパク質がインスリン様増殖因子結合タンパク質すなわちIGFB P−1であり、胎児膜の破裂によってもたらされる IGFBP−1の存在が、 IGFBP−1の少なくとも一つの特異的結合物質によって試料中に検出される ことを特徴とする診断方法。
- 2.試験に用いられる結合物質:試料中のIGFBP−1の濃度の比率が、試料 中のIGFBP−1の濃度が羊水の存在以外の起源由来のいき値以上である場合 のみ、陽性の試験結果が出現するように調節されることを特徴とする請求の範囲 1記載の方法。
- 3.使用される特異的結合物質がIGFBP−1に対する抗体であることを特徴 とする請求の範囲1または2に記載の方法。
- 4.抗体がモノクローナル抗体であることを特徴とする請求の範囲3記載の方法 。
- 5.測定が、IGFBP−1結合物質および標識をつけた別の結合物質でそのシ グナルが検出可能な結合物質に基づいた試薬を用いて行われることを特徴とする 請求の範囲1〜4のいずれか一つに記載の方法。
- 6.膣から採取した試料は、試料中の IGFBP−1の濃度を低下させるため に、試験を行う前に希釈することを特徴とする請求の範囲2,3,4また5に記 載の方法。
- 7.希釈率が少なくとも1:10で、好ましくは1:20であることを特徴とす る請求の範囲3記載の方法。
- 8.標識を付けた試薬を用い、そのシグナルは、測定されるIGFBP−1の濃 度が充分高い場合にのみ陽性の試験結果を与えることを特徴とする請求の範囲2 ,3,4または5に記載の方法。
- 9.胎児膜の破裂によって膣分泌物試料中にもたらされる、インスリン様増殖因 子結合タンパク質1すなわちIGFBP−1の存在を検出するため、IGFBP −1の特異的結合物質を含有する少なくとも一つの試薬を備えていることを特徴 とする胎児膜の破裂を診断するのに用いる試験キット。
- 10.さらに、結合反応を検出する標識を備えていることを特徴とする請求の範 囲9記載の試験キット。
- 11.標識が生成するシグナルの感度の範囲は、羊水の存在で高濃度のIGFB P−1がもたらされる場合にのみ陽性の試験結果が起こるような方式で調節され ることを特徴とする請求の範囲10記載の試験キット。
- 12.試薬が、IGFBP−1の特異的結合物質として、特異的1GFBP−1 抗体を、好ましくはモノクローナル抗体を含有していることを特徴とする請求の 範囲9,10または11に記載の試験キット。
- 13.結合反応を検出する標識が、他のIGFBP−1結合物質に結合されたシ グナル生成物質であることを特徴とする請求の範囲10または11に記載の試験 キット。
- 14.特異的結合物質に基づいた試薬が、固相もしくは不溶性粒子に結合した結 合物質、または結合物質の溶液からなることを特徴とする請求の範囲9に記載の 試験キット。
