る赤血球のより長期の保存のための浮遊培養液。 2 約80〜150mMのグルコース、30〜80mMマ
ンニトール、6〜90mMクエン酸カリウム、1〜
5mMアデニンおよび10〜200mMのアンモニウム
塩を必須成分とし、PH約6.8から7.2、オスモル濃
度約180〜220ミリオスモルである、赤血球を生き
たまた保存可能な水溶液における、約4℃での赤

化合物である請求の範囲第1項記載の浮遊培養
液。 4 グルコース約110mM、マンニトール55mM、

塩化アンモニウムまたは酢酸アンモニウム50mM
を含む請求の範囲第3項記載の浮遊培養液。 5 適量の一または二塩基性燐酸カリウムを用い
て、PH値を約7.0に維持する請求の範囲第4項記
載の浮遊培養液。 6 オスモル濃度が約200mOsmである請求の範
囲第5項記載の浮遊培養液。 7 上記アンモニウム塩が塩化アンモニウムと酢
酸アンモニウムから成る群より選ばれる化合物で

範囲第9項記載の方法。 発明の背景 技術分野 本発明は、生理学的に十分に自然な状態での赤
血球の保存に関する。特に本発明は、輸血に適す
る状態での約4℃における、人間の赤血球の保存
用の新しい浮遊培養液に関する。本発明の新規な
浮遊培養液により、赤血球の保存期間をこれまで
可能であつたよりも延長することができる。 従来の技術 標準的な血液銀行の処方に従つて約4℃の浮遊
培養液中に赤血球を保存および貯蔵することは衆
知である。出願人の知る限りでは、赤血球の品質
の維持に関して、低張圧の浮遊培養液または透過
性の塩、たとえばアンモニウム塩の添加による影
響を報告した参考文献はない。 しかし、連続的冷蔵保存の間に、人間の赤血球
は、最終的に小胞へと分離する針状物の発生を伴
う形状変化を起すことが観察された。これにより
赤血球の表面積は減少し、恐らく赤血球は弾力性
を減じ、脾臓のろ過機構を通過しにくくなる。こ
のことは更に輸血後数分のうちに赤血球が血液の
循環から除去されてしまうことに繋がる。輸血可
能な赤血球に対する一般に認められた基準は、少
くとも70%の赤血球が輸血後24時間循環している
ことを必要とする。 本発明により、従来の方法に伴う問題点と限界
とを克服できる。浸透膨脹による赤血球の表面張
力の増大が針状物の発生を妨げることが理論的に
考えられる。従つて、赤血球の輸血用にすでに確
立された基準によつて、輸血に適する状態で4℃
で赤血球より長期間保存することができる、浸透
圧性と膜透過性をもつ浮遊培養液が開発された。
浮遊培養液中にアンモニウム塩、好ましくは
NH4ClおよびNH4OOCH3のような特定の透過性
化合物を含有することは、低イオン濃度の有害な
影響を防ぐのみならず、以下に詳細を記す有益な
結果をもたらすことが発見された。 発明の概要 本発明の一つの目的は、保存赤血球の輸血用品
質を保ちながら、約4℃における浮遊赤血球の保
存期間を十分に増加させる赤血球用の浮遊培養液
を提供することにある。 また本発明のもう一つの目的は、人体への輸血
用の保存赤血球としての適合性を保ちながら、従
来の方法で可能である以上に、4℃における保存
期間を伸ばすことができる赤血球の保存方法の提
供にある。 本発明の記述が進むに従つて、他の目的および
利点は明らかになる。

前述またはその他の本発明の目的、特徴および
付随する利点の多くは、次の詳細を別添の図とと
もに一読することで明らかになる。 第1図は、人間の赤血球のATP量(本来の値
に対するパーセント)と、種々の浮遊培養液中に
おける4℃での保存時間との関係を示した。 第2図に、異なる浮遊培養液中において4℃で
の種々の時間間隔で保存した後の人間の赤血球の
生体内生存率をパーセントで示した。 特定の理論または説明とは別に、従来の浮遊培
養液中において冷蔵保存の間に、赤血球中に好ま
しくない針状物が形成されるのを防ぐために、赤
血球細胞の表面張力を浸透膨脹により増大するこ
とが必要と考えられる。従つて、赤血球は、約
180〜220mOsm(ミリオスモル)、特に約
200mOsmの培養液中に保存される。浮遊培養液
のPHは、約6.8から7.2の範囲に、好ましくは7.0付
近に保たれる。このことは、膜透過性の増加に繋
がるイオン濃度の低下を伴うので、赤血球細胞の
正常なイオン濃度を、一定の透過性を有し、イオ
ン化可能な塩、このましくは濃度約10〜200mM、
好ましくは50mMの酢酸アンモニウムおよび塩化
アンモニウムからなるユニークな成分を浮遊培養
液中に持つことによつて保つ。当然のことなが
ら、アンモニウム以外の適当な陽イオン、および
酢酸塩、塩化物以外の適当な陰イオンもまた、そ
れらの塩が本発明の目的と基準にかなう限り、用
いることができる。その塩の一例は、メチルアミ
ン塩化水素である。浮遊培養液の他の成分は、従
来用いられてきた適当な濃度の、燐酸カリ、マン
ニトール、アデニン、牛リンパ液タンパク質、イ
ノシン、ピルベート(pyruvate)、クエン酸カリ
ウムおよびグリコースである。例えばグルコース
の濃度は約80〜150mM、マンニトール約30〜
80mM、クエン酸カリウム約6〜90mM、アデニ
ン約1〜5mMである。一および二塩基性の燐酸
塩の濃度は約180〜220mOsmの浮遊培養液のPHを
約6.8から7.2に保つように調整される。 「相当延長された」または「より長期の」保存
等の語は、単に従前より知られている血液銀行の
処理方法および浮遊培養液を用いて可能であるよ
りも多くの期間、生きた状態で赤血球を保存また
は貯蔵できることを意味する。 実施例の説明 本発明による典型的な浮遊培養液は、次の物質
からなる水溶液である。 グルコース 110mM アデニン2mM マンニトール 55mM 塩化アンモニウム
50mM クエン酸カリウム79mM 二塩基性燐酸カリウム 25.8mM 一塩基性燐酸カリウム 14.7mM 同様なものも、PHが約7.0であり、オスモル濃
度が約200mOsmである限り簡単に調製できる。 成分調製の通常の工程において、血漿から分離
された赤血球は、ほぼ等容積の浮遊培養液中に再
浮遊される。浮遊培養液は、参考文献として用い
た“Clinical Practice of Blood Transfusion”
editors:Petz & Swisher,Churchill−
Livingston Publishers,N.Y.,1981に記載され
ている標準的血液銀行処方に従つて、約4±2℃
に保存される。この文献の第11章第281頁には、
従来の手法による赤血球の生存率に関するデータ
が示されている。 本発明による浮遊培養液中にて保存された場
合、初めの4から5週目にかけての保存期間中に
アデノシン三燐酸(ATP)の進行性の増加がみ
られる。ATPの水準は、輸血後の赤血球の生存
と相関関係にある。4週後のATPの水準は、初
期値の165%を平均に、115%から250%にわたる
値を示す。その後ATPの水準は、さらに保存を
続けると着実に下がり、約11週後には初期値に達
し、およそ16週後には初期値の50%にまで下が
る。 第1表に赤血球を16週間まで保存した浮遊培養
液中の種々の成分と溶質の濃度を示す。本発明に
よる優れた浮遊培養液である番号20の溶液が、赤
血球のATPを第1図に示されている様に、テス
トされた他の溶液に比べて、より高いレベルでよ
り長期間保つていることに注意されたい。
付随する利点の多くは、次の詳細を別添の図とと
もに一読することで明らかになる。 第1図は、人間の赤血球のATP量(本来の値
に対するパーセント)と、種々の浮遊培養液中に
おける4℃での保存時間との関係を示した。 第2図に、異なる浮遊培養液中において4℃で
の種々の時間間隔で保存した後の人間の赤血球の
生体内生存率をパーセントで示した。 特定の理論または説明とは別に、従来の浮遊培
養液中において冷蔵保存の間に、赤血球中に好ま
しくない針状物が形成されるのを防ぐために、赤
血球細胞の表面張力を浸透膨脹により増大するこ
とが必要と考えられる。従つて、赤血球は、約
180〜220mOsm(ミリオスモル)、特に約
200mOsmの培養液中に保存される。浮遊培養液
のPHは、約6.8から7.2の範囲に、好ましくは7.0付
近に保たれる。このことは、膜透過性の増加に繋
がるイオン濃度の低下を伴うので、赤血球細胞の
正常なイオン濃度を、一定の透過性を有し、イオ
ン化可能な塩、このましくは濃度約10〜200mM、
好ましくは50mMの酢酸アンモニウムおよび塩化
アンモニウムからなるユニークな成分を浮遊培養
液中に持つことによつて保つ。当然のことなが
ら、アンモニウム以外の適当な陽イオン、および
酢酸塩、塩化物以外の適当な陰イオンもまた、そ
れらの塩が本発明の目的と基準にかなう限り、用
いることができる。その塩の一例は、メチルアミ
ン塩化水素である。浮遊培養液の他の成分は、従
来用いられてきた適当な濃度の、燐酸カリ、マン
ニトール、アデニン、牛リンパ液タンパク質、イ
ノシン、ピルベート(pyruvate)、クエン酸カリ
ウムおよびグリコースである。例えばグルコース
の濃度は約80〜150mM、マンニトール約30〜
80mM、クエン酸カリウム約6〜90mM、アデニ
ン約1〜5mMである。一および二塩基性の燐酸
塩の濃度は約180〜220mOsmの浮遊培養液のPHを
約6.8から7.2に保つように調整される。 「相当延長された」または「より長期の」保存
等の語は、単に従前より知られている血液銀行の
処理方法および浮遊培養液を用いて可能であるよ
りも多くの期間、生きた状態で赤血球を保存また
は貯蔵できることを意味する。 実施例の説明 本発明による典型的な浮遊培養液は、次の物質
からなる水溶液である。 グルコース 110mM アデニン2mM マンニトール 55mM 塩化アンモニウム
50mM クエン酸カリウム79mM 二塩基性燐酸カリウム 25.8mM 一塩基性燐酸カリウム 14.7mM 同様なものも、PHが約7.0であり、オスモル濃
度が約200mOsmである限り簡単に調製できる。 成分調製の通常の工程において、血漿から分離
された赤血球は、ほぼ等容積の浮遊培養液中に再
浮遊される。浮遊培養液は、参考文献として用い
た“Clinical Practice of Blood Transfusion”
editors:Petz & Swisher,Churchill−
Livingston Publishers,N.Y.,1981に記載され
ている標準的血液銀行処方に従つて、約4±2℃
に保存される。この文献の第11章第281頁には、
従来の手法による赤血球の生存率に関するデータ
が示されている。 本発明による浮遊培養液中にて保存された場
合、初めの4から5週目にかけての保存期間中に
アデノシン三燐酸(ATP)の進行性の増加がみ
られる。ATPの水準は、輸血後の赤血球の生存
と相関関係にある。4週後のATPの水準は、初
期値の165%を平均に、115%から250%にわたる
値を示す。その後ATPの水準は、さらに保存を
続けると着実に下がり、約11週後には初期値に達
し、およそ16週後には初期値の50%にまで下が
る。 第1表に赤血球を16週間まで保存した浮遊培養
液中の種々の成分と溶質の濃度を示す。本発明に
よる優れた浮遊培養液である番号20の溶液が、赤
血球のATPを第1図に示されている様に、テス
トされた他の溶液に比べて、より高いレベルでよ
り長期間保つていることに注意されたい。

第1図に、4,8,10,12,14および16週目に
おける下記の溶液中に浮遊させた赤血球のATP
値を示す。すなわち(a)第1表の番号7に対応する
等浸透圧のクエン酸カリウム、燐酸、マンニトー
ル、グルコースおよびアデニン溶液、(b)第1表の
番号14に対応する(a)溶液に酢酸アンモニウムを加
えた溶液、(c)第1表の番号12に対応する(a)溶液か
ら低張性にするためクエン酸カリウムを除いた溶
液、(d)第1表の番号13に対応する、(c)溶液に酢酸
アンモニウムを加えた低張溶液、(e)第1表の番号
20に対応する(c)溶液に塩化アンモニウムを加えた
低張溶液。この結果は、低張性とアンモニウム塩
の添加の両方から得られる効果を示している。低
張性による効果は、針状物の発生が遅れることと
関連があるであろう。しかし、ATP水準の増大
の理由は明らかでない。浮遊培養液に加えられた
場合に、本発明のアンモニウム塩がATPを増加
させるメカニズムもまた不明である。 第2図に、125日後までの種々の期間、(c)溶液
中に保存された赤血球の24時間生体内生存率を示
す。 アンモニウム塩を含む低張性細胞浮遊液が輸血
に適することは考えにくいことは注意に価する。
従つて、輸血に先立ち遠心分離により赤血球細胞
は沈澱処理され、輸血可能な溶液に移されたのう
ち輸血される。 本発明によつて保存される赤血球の溶血は約
0.3から3.8%である。 これらの例及び実施例は、例示のためにのみ記
述されており、本発明に基づく種々の変更、改変
は、この技術の習熟者に示唆されるとともに、出
願の要旨と添付される請求の範囲に含まれる。

20に対応する(c)溶液に塩化アンモニウムを加えた
低張溶液。この結果は、低張性とアンモニウム塩
