Claims (1)
- 【特許請求の範囲】 1.(a)酵素の水性混液をフェニル−セファロース樹脂に、キモシンが樹脂と 結合するのに充分な時間接触させる段階と、 (b)樹脂と待合キモシンとを、水性混液から分離する段階と、 (c)キモシンを樹脂から溶離する段階とから成る、酵素の水性混液からキモシ ンを分離する方法。 2.前記混液を樹脂と接触させる前に、酵素の水性混液に塩を添加することを特 徴とする請求の範囲第1項記載の方法。 3.前記添加塩が、少なくとも約1Mの濃度を有する無機塩水から構成されるこ とを特徴とする請求の範囲第2項記載の方法。 4.前記溶離段階が、約0.5M未満の濃度を有する希塩水の使用を包含するこ とを特徴とする請求の範囲第1項記載の方法。 5.前記最初の酵素水性混液が、発酵段階で作られることを特徴とする請求の範 囲第1項記載の方法。 6.前記最初の酵素水性混液が、発酵段階で作られることを特徴とする請求の範 囲第2項記載の方法。 7(a)水性混液のpHを約3未満に調節する段階と、(b)pH調節水性混液 を、フェニル−セファロース樹脂に、キモジンが樹脂と結合するのに充分な時間 、接触させる段階と、 (c)樹脂と結合キモシンとを、水性混液より分離する段階と、 (d)キモシンを樹脂より溶離する段階とから成る、キモシンを酵素の水性混液 から分離する方法。 8.前記樹脂と結合キモシンとを、少なくとも約1Mの濃度を有する塩水で溶離 する段階の前に、洗浄することを特徴とする請求の範囲第7項記載の方法。 9.前記溶離段階が、約0.5M未満の濃度を有する希塩水の使用を包含するこ とを特徴とする請求の範囲第7項記載の方法。 10.最初の酵素水性混液が、発酵段階からであることを特徴とする請求の範囲 第7項記載の方法。 11.最初の酵素水性混液が、発酵段階からであることを特徴とする請求の範囲 第8項記載の方法。
