Abstract

Denne oppfinnelse frembringer en fremgangsmåte ved , utvinnelse og rensing av chymosin fra fermentasjon anvendt for mikrobiologisk produksjon av chymosin. Fermentasjonsblandingen filtreres for å gi et filtrat som inneholder chymosinet. Filtratet prepareres fcrst ved enten å innstille det til en lav pH-verdi, såsom ca. ; 2, og/eller innstilles saltkonsentrasjonen i filtratet ; til minst ca. 2M. Den vandige filtratblanding bringes i ; kontakt med en fenyl-sefarose-harpiks som selektivt binder chymosinet fra filtratet. Filtratet som er igjen etter kontakten med fenyl-sefarose-harpiksen kastes, og : fenyl-sefarose-harpikskolonnen elueres med vann eller i fortynnet saltopplcsning, slik at man oppnår i alt j vesentlig rent chymosin i et eneste elueringstrinn.
