(57)【要約】 【構成】本発明はラクトフェリンを有効成分とする抗リ
ウマチ剤に関する。 【効果】ラクトフェリンは優れたコラゲナーゼ阻害作用
を示し、リウマチの治療剤として有用である。

【発明の詳細な説明】
【０００１】
【産業上の利用分野】本発明はラクトフェリンを有効成
分とする抗リウマチ剤に関する。
【０００２】
【従来の技術】ラクトフェリンはヒトや牛などの乳や涙
液に含まれている蛋白質で、その薬理作用としては抗菌
作用やリンパ球の増殖作用等が知られている（特開平２
−４８５３４号公報等参照）。
【０００３】
【発明が解決しようとする課題】しかしながら、天然物
であるラクトフェリンの適用範囲を拡大するため、潜在
する効果をさらに研究する事が望まれていた。
【０００４】
【課題を解決するための手段】そこで、本発明者等はラ
クトフェリンの新しい薬理作用を見つけるべく種々検討
を行なった。その結果、ラクトフェリンが抗リウマチ剤
として有用であることを見い出した。
【０００５】
【発明の構成】本発明はラクトフェリンを有効成分とす
る抗リウマチ剤に関する。
【０００６】天然の蛋白質であるラクトフェリンは、医
薬として種々の応用可能性が期待されている。しかしな
がら、既に報告されているのは抗菌作用やリンパ球の増
殖作用等にすぎない。そこで、本発明者等はラクトフェ
リンの効果をさらに検討したところ、抗リウマチ剤とし
て有用であることを見い出した。
【０００７】抗リウマチ剤としての有用性を検討する指
標として、コラゲナーゼ阻害活性がある。コラゲナーゼ
阻害活性とリウマチとの関連は既に文献で報告されてい
る(W. H. Johnson et. al., J. Enzyme Inhibition, 2,
1 (1987) ; Woolley D.E.et. al., Arthritis Rheum,
20, 1231 (1977) ）。そこで、本発明者らはラクトフェ
リンのコラゲナーゼ阻害活性を調べた。
【０００８】詳細なデータは薬理試験の項で示すが、ラ
クトフェリンは有意にコラゲナーゼによるコラーゲンの
分解を抑制しており、優れたコラゲナーゼ阻害作用を有
することが明らかとなった。
【０００９】ラクトフェリンの投与は通常膝等の関節に
注射する方法を用いるが、経口でも投与することができ
る。投与量は症状や剤型等によって決められるが、注射
剤であれば０．１〜１０重量％、好ましくは０．５〜５
重量％のものを患部の必要量に合わせて用いればよく、
また経口剤であれば通常１日１〜５０００ｍｇ、好まし
くは５０〜１０００ｍｇを１回または数回に分けて投与
すればよい。
【００１０】ラクトフェリンの製剤は常法により調製す
ることができる。注射剤の場合であれば、ラクトフェリ
ンに必要に応じて、浸透圧調節剤、ｐＨ調整剤、増粘剤
等の通常用いられる賦形剤を加え、注射用の蒸留水に溶
解して調製する。錠剤の場合は、ラクトフェリンに必要
に応じて結合剤、滑沢剤等の通常用いられる賦形剤を加
えて調製する。以下に処方例を示す。
【００１１】「処方例」 「薬理試験」抗リウマチ剤としての有用性を評価するた
め、永井らの方法（炎症，４，１２３（１９８４））に
準じて、ラクトフェリンのコラゲナーゼ阻害作用を測定
した。
【００１２】Ｉ．細菌由来コラゲナーゼに対する作用 （実験方法）フルオレセイン イソチオシアネートで標
識したコラーゲンと細菌（Clostridium histolyticum)
由来コラゲナーゼをトリス−塩酸緩衝液（０．０５Ｍ、
ｐＨ７．５；０．２Ｍ塩化ナトリウム、５ｍＭ塩化カル
シウム、０．０２重量％アジ化ナトリウム（ＮａＮ3 ）
を含む）に加えて溶解し、各々の濃度が０．０２５重量
％および１２．５ユニット／ｍｌとなるようにする。
【００１３】この溶液０．４ｍｌにラクトフェリンを加
え、褐色試験管中、３５℃で６０分間インキュベートす
る。これに８０ｍＭの濃度でｏ−フェナンソロリンを溶
解した水−エタノール（１：１Ｖ／Ｖ）溶液１０μｌを
加えて反応を停止し、さらに３５℃で６０分間インキュ
ベートする。その後、トリス−塩酸緩衝液（０．０５
Ｍ、ｐＨ９．５；０．２Ｍ塩化ナトリウムを含む）とエ
タノールの混液（３：７Ｖ／Ｖ）４００μｌを加えて撹
拌し、遠沈した。上清の蛍光強度を励起波長４９５ｎ
ｍ、測定波長５２０ｎｍで測定することにより分解した
コラーゲンの量を求めた。コントロールとしてはラクト
フェリンを加えないものを用いた。
【００１４】（実験結果）ラクトフェリン添加によるコ
ラーゲン分解抑制率を表１に示した。

【表１】 II．組織コラゲナーゼに対する作用 （実験方法）Burns らの方法（ Invest. Ophthalmol. V
is. Sci., 30, 1569 (1989) ）に従ってウサギ角膜より
採取した組織コラゲナーゼを用い、Ｉと同様の方法で実
験を行なった。尚、添加したコラゲナーゼの量は２．５
ｍｇタンパク質／ｍｌとした。
【００１６】（実験結果）結果を表２に示した。

【表２】 表１および２に示すように、細菌由来および組織由来の
コラゲナーゼのどちらを用いても、ラクトフェリンはコ
ントロールと比較して濃度依存的に分解物の生成を抑制
しており、ラクトフェリンがコラゲナーゼ阻害作用を有
することが立証されている。
【００１８】
【発明の効果】薬理試験の結果に示すように、ラクトフ
ェリンは優れたコラゲナーゼ阻害作用を有しており、抗
リウマチ剤として有用である。
