ホルモン分泌を変化させる組換え体融合タンパク質発明の分野
本発明は組換えDNA技術によって製造されそして本来的に抗原でないペプチド
に対して能動免疫原として使用される融合タンパク質の製造に関するものである
。肝炎表面抗原(BBsAg)のような抗原性の高いタンパク質の遺伝子と、本
来は抗原性でないタンパク質、タンパク質の1部またはペプチドをコードするc
DN^とを組合わせることによって、融合タンパク質が発現される。この抗原性
の高い融合タンパク質の注入後に、抗原性の高いタンパク質および問題のタンパ
ク質の双方に向けられたポリクローナル抗体が製造される。かくして、この技術
は生体内で産生されたホルモンを免疫的に無効化させる方法を提供し、そして生
殖力のない状態を誘導するがまたは急速に増殖する組織のホルモン依存性増殖を
阻止するために使用することができよう。
本発明の背景
1960年代後期中のギレメン(Guille+m1n)、 シャリ−(Sch
ally)および共同研究書違の開拓的な研究結果として、視床下部デカペプチ
ド、即ち黄体形成ホルモン放出ホルモン(LHR)りが全ての哺乳動物の生殖能
力の開始および維持に深く係わっていることが明らかになってきた。
LHRHの合成または、視床下部と下垂体間の体液連絡をもたらす毛細管網への
LBRHの移送に係わる神経経路の崩壊または破壊によってゴナドトロピンの合
成および放出の減少並びに不妊症がもたらされる( KalraおよびKalr

同様に、LHRH−下垂体相互作用の低下はLHHの長時間作用性競合的拮抗剤
の投与によって誘導することができる。これらの合成ペプチドは下垂体Ll(R
Eレセプターに結合するが、レセプター後細胞内活性は刺激しない。上記ペプチ
ドは、タンパク質分解作用の不活性化に対する抵抗性によって異常に長時間レセ

LER[+=下垂体相互作用の破壊に使用されるもう1つの方法はLBRHこ対
する抗体の産生であった。LHRH能動免疫1973) 、雌ヒツジ(C1ar

能動免疫法によるLBRHこ対する抗体産生は、痛みはないにしても、動物にと
って不快なものである。この方法は不妊状態を誘導し、そして発情行動を減少さ
せる(性腺ステロイドのwI環低下のため)か、ベット避妊剤として使用するこ
とはできないであろう。
米国ではネコやイヌの数の深刻な過剰があり、これは一般大衆、動物愛護協会、
獣医および保健省役人の重大関心事である。ネコの集団においては、繁殖速度は
これら動物に住居が利用できるようになるほぼ2倍の速度であると報告されてい

せるために、繁殖力のあるネコの約8ε%の卵巣を除去すべきである( Nas

その内約80%が殺される。不要のネコやイヌの収容および屠殺費用は 1年当

妊娠制限の効果的で、安全で、利用し易くそして回復可能な手段を提供する組換
え体融合タンパク質またはワクチンを提供することか本願発明の目的である。投
与の容易なワクチンは外科的不妊状態に取って替わり、イヌ、ネコおよび他の動
物に効果的な避妊を提供するように思われる。
妊娠した雌ウシは典型的には、発育する胎児に多くの栄養が必要であるので、飼
料から肉への魅力ある転換を示さない。その結果、飼育項生は一般的には、妊娠
していることがわかっていないかまたは確認されていない雌ウシに支出する金顕
はかなり少ない、成る割合の雌ウシが飼育場で妊娠し、胎児が浪費を示す。かく
して、積み出しの6〜10ケ月前に不妊状機を誘導する方法は、成長性のある、
価値ある製品であると考えられる。 ウシが「妊娠していないJと農場主が保証
出来る場合には、彼はその動物により高い値段を要求できるであろう。
それ故、飼育場の未経産雌ウシの排卵周期の阻害を生じさせる注入可能な組換え
体製品を提供することがもう1つの目的である。
ヒトのホルモンが介在した組lai!J形成に関する例は多数ある。男性では、
前立腺癌(BPH)が増殖継続を前立腺テストステロンに依存していることは良
く知られている。精巣ステロイド分泌を阻止するためにゴナドトロピンの血清値
を減少させ、その結果過形成組織の退化を誘導することは有益であろう。長期に
作用するLHRH拮抗剤が前立腺過形成の治療に使用できることが最近報告され
ている( PetersおよびWalshSNew England Jo
urnalMedicine 317−1599.1987)。 この治療の作
用メカニズムは、下垂体LHRHレセプターの調節低下によるLHおよびFS)
!分泌の阻害である。 その結果、テストステロン産生の低下を生じさせるゴナ
ドトロピン分泌の阻害がもたらされる。血清テストステロンの減少は前立腺過形
成の退行をもたらす。しかし乍ら、LIIRH拮抗剤の注入により誘導されるL
EI分泌阻害には長時間が必要であり、それ故、反復して投与する必要があると
著者は述べている。
ゴナドトロピン分泌を阻害するため、即ち、内因性LHRHの免疫無効化のため
にLHR■拮抗剤の複数回の注入を避けることが本願発明の更にもう1つの目的
である。
浄書(白参に変更なし) −実質的に非抗原性ペプチドを、好ましく
はフロイントの完全アジュバントのようなアジュバントを使用することを必要と
しないで、高度に免疫原性にする方法を提供することが本願発明の更にもう1つ
の目的である。

本願発明の原理および目的に従って、潜在的な免疫原として使用可能な融合体タ
ンパク質を創製する方法が提供される。理想的には、本願発明の上記方法はcD
NA (複数)を−緒に融合し、そして独特な融合タンパク質を産生させるため
それらを真核細胞発現ベクター中にいれる合タンパク質の大部分は非常に抗原性
であり且つ潜在的な免疫原である。これらは実質的に親水性でなければならない
、このような免疫原の良好な例はB型肝炎表面抗原のような被覆タンパク質であ
る。黄体形成ホルモン放出ホルモン分子のような、抗体を生じさせることが望ま
れる問題のペプチドをコードするオリゴヌクレオチドを1つまたはそれ以上のタ

本願発明の融合タンパク質は、アジュバントまたは多数回の注入の使用(これら

原性である。

ルモン配列の挿入を示す。上部はnBsAgタンパク質の略図であり、 その際
、LFIRHデカペプチドを有する部位が点描されている。その下にはBBsA
gをコードするcDNAに沿ってJVa 11部位があり、その際、LFIRB
のcDNAが理想的に挿入されている。下の図は被覆タンパク質配列間に挿入さ
れたLHRHを有するウィルス被覆粒子を示す。
図2は、BBsAgのAVa n部位(図1参照)に挿入される合成りNAリン
カ−を示す。このリンカ−はHBsAHのアミノ酸119(グリシン)の後のオ
リゴヌクレオチド配列用に フレキシブルな挿入部位を提供する。 これによっ
て、flBsAg配列を元に戻す舶に プロリン残基(120位)に3個のアミ
ノ酸が挿入される。好ましくは、LHRIIIフラグメントはリンカ−の5−a
lおよび適当な読み取りフレームが保持される(図3参照)。
図3は、BBsAg cDNA内の合成リンカ−に挿入されるLHRIIペプチ
ドのヌクレオチド配列を示す。適当な読み取りフレームを保持するために、挿入
されたLHRIIペプチドのすぐ後にグリシン(GGG)を付加し適当な読み取
りフレームを保持するオリゴヌクレオチドの3“末端に2個のヌクレオチド(G
G)が付加されている。
合成りNAリンカ−(図2)に挿入されたとき、適当な読み取りフレームを保持
するために3゛末端に付加されたGOIご注意すること。本発明者は、切断リン
カ−に適切に挿入するためにも1工Hのオーバーハングも付加した。
図4は、マウスのメタロチオネインプロモーター(CL28ベクター)にHBs
Ag −LHRH融合体を挿入しそしてその後BPVゲノムを挿入するために使
用される概略図である。 プラスミドBPV−BLはC127細胞を形質転換す
るために好都合に使用される。

リA付加シグナルは挿入物の下流に存在する。表面抗原タンパク質は培地中に分
泌され、その際、該タンパク質は粒子の形態で見い出される。組換え体産生物(
即ち、核物質を有していない表面抗原)はモルモットで抗原性の高いことが示さ
れている。
DNA配列から、肝炎抗原のアミノ酸配列が決定される。
この配列は、最も親水性の特性を有すると思われる分子の領域について分析した
。該領域(図1のAva Uで同定される)に、独特のクローニング部位を提供
する短いオリゴヌクレオチド配列(図2)をクローニングした。この領域に、特
定のタンパク質をコードするcDNA配列を、固有の違す、1部位かまたはBg
l工■部位(図2参照)のどちらかでのプラント末端連結によって挿入すること
ができる。
視床下部遺伝子の公表された構造(Seeburgおよび^del*an、
Nature 311.666.1984: 図3)を使用するが、上記のよ
うに修正して、LHRHをコードするオリゴヌクレオチドは、カラサーズ(Ca
ruthers)等(GeneticEngineering 4.1〜17.
1982)が開発した固相合成法を使用してアプライドバイオシステムズ(Ap
plied Blo−5ystess) 380−A DNA自動合成器で合
成した。オリゴヌクレオチドは濃縮水酸化アンモニウムを使用して固体支持体か
ら切り離した。精製は分離用ゲル電気泳動を使用して達成した。精製したオリゴ
ヌクレオチドはポリヌクレオチドキナーゼであり、大きさおよび純度はゲル電気
泳動で分析した。
LIIRHオリゴヌクレオチドは 下記の方法でSmalおよびBglIIクロ
ーニング部位に挿入した: LHRHオリゴヌクレオチドは3°プラント末端
および5°CTAGオーバーハングを有するように(図3に示されるように)合
成した。垣1およびジ1工■でHBsAg DNA (合成挿入物を有する)を
消化した後、Ll’lRHDNAを挿入した。得られた構造はHBsAg −L
HR)Iと命名した。 合成11旧リンカ−を HBgAg −LHR)1cD
NAの各末端に付加し、 そして適当な配向でベクターCL2g/Ban中に挿
入した。ディーンハーv −(Dean Ba5er)博士(NIHの)から入
手したCL211/Bagは、全メタロチオネイン遺伝子を含有する 3.8k
bpのマウスDNAフラグメントを有しており、細菌ベクターpBR322のE
coR1部位に挿入された上流の制御エレメントを有している。CL2g/Ba
gはpJYMMT(E)の誘導体であって(Dean [1aIler等、J、
Mol。
Appl、 Genet、 1. 4.273〜288(1982) ) 、S
V40配列が除去されている以外はPJYMMT(E)と同一である。マウス遺
伝子の転写開始領域の直後に位置する固有の醜±■部位挿入可能になる。HBg
Ag −LHRH融合遺伝子をCL2g/Bagに挿入しそして適当な制限パタ
ーン(ネズミのプロモーターに関して)による配向をエンドヌクレアーゼで証明
した後、 ウシ乳頭腫ウィルス(BPV)の全ゲノムは7.8kbpのBag旧
−3allフラグメントとして挿入した。最終的な構造は、アガロースゲルおよ
びポリアクリルアミドゲルの両方での診断制限酵素消化パターンの試験によって
証明された。本方法の図示は図4で提供する。
かくして、得られたB型肝炎表面タンパク質は該分子の親水性領域内にLHRI
I配列を含有していた。
融合HBsAg/LHRHタンパク質を発現する細胞株を作成するために、精製
したBPv−HBsAg−Ll(RHcDNA 108gを、担体としてサケ精
子DNA LOagを含有する250IIIM CaCL溶液0.5mlに加え
た。この混合物を28+mM NaCl、50℃M EEPESおよび1.5m
Mりん酸ナトリウムの0.5 ml中に吹き込んで加えた。室温で30〜40分
間置いて装ん酸カルシウムの沈澱物を形成させた。
形質転換の24時間府に、 5×10′1個のマウスC127細胞を1.Ohm
のベトリ皿に入れた。外来DNAを加える直前に、細胞に新鮮な培地を与えた(
ダルベツコの修正イーグル培地+10%のウシ胎児血清)。りん酸カルシウム沈
澱物1■Lを各皿(lfil、)に加え、細胞は37℃で6〜8時間時間インベ
コベート。
培地を吸引器で吸い取り、そしてりん酸緩衝生理食塩液(plに7.0)中20
%のグリセロ−・・ル 5mlを用いて室温で2分間置き換えた。細胞はりん酸
緩衝生理食塩液で洗浄し、fowlの培地を与えそして37℃でインキュベート
した。20〜24時間後、培地を交換し、細胞は3〜4日毎に再度加えた。
14〜21日後、形質転換した細胞の集まり(形態の変化が特徴である)を同定
し、モしてT−25フラスコに移して株膨張を開始させた。細胞株から得た培地
はHBsAgの存在およびLHRH免疫活性を試験した。組織培地は低速遠心機
で澄明にし、モしてHBsAg粒子は42.00Orpm(200,000Xg
)で4時間沈澱させた。ベレットは少量のりん酸緩衝生理食塩液中に再懸濁させ
、そして出発物質および上澄液のHBsAg活性は、 クリニカルアッセイズイ
ンコーボレーテッド(C1inical As5ayS、 Inc、)から購入
した市販のラジオイムノアッセイ(RIA)で分析した。培地上澄液および再懸
濁ベレットのL!’IR)l免疫活性は、アルネルアンタイボディーズインコー
ボレーテッド(Arnel Anti−bodies、 Inc、)から購入し
たLHR[(抗血清を使用するラジオイムノアッセイで分析した。ヨード化用!
、11 RHおよび参照調製物はシグマ ファインケミカルズインコーボレーテ
ッド(Sigma Fine Che+miea1g、 Inc、)から購入し
そして不溶性のタンパク質A(ベレット抗原抗体コンプレックスへの)はシグマ
ファインケミカルズインコーポレーテッドから入手した。
これらの方法に続いて、融合タンパク質を発現する細胞株を作成し、そしてこれ
は標準的スクリーニング法で容易に確認された。2.3ミリグラムの精製材料で
は、雄ウサギまたは他の動物の不妊状態を誘導するのに適す−る投与量は滴定タ
イプの方法で決定することができる。
例えば、血液試料採集と組み合わせてタンパク質0.5、lまたは511gを雄
ウサギに注入して血清ゴナドトロピン値および精巣ステロイド値並びに抗LHR
H抗体価を決定することができる。同様に、適当な追加免疫間隔も決定すること
ができる。
上記から、当該技術分野の熟練者は、本願発明の精神または範囲のいずれかから
離れることなく、融合遺伝子および遺伝子産生物に免疫学的に活性が低下しない
抗原性遺伝子またはタンパク質遺伝子の選択のような多くの修正が可能であるこ
とを容易に決定できよう。例えば、他の親水性の被覆タンパク質またはB型肝炎
コア抗原のような、HBsAg以外の免疫原性の高いタンパク質を使用すること
ができる。更に、当該技術分野の熟練者は、LHRH以外のペプチドが使用でき
ることを理解しよう。免疫系に抗原性エピトープを与えるために、オーストラリ
ア特許69792/17で同様な方法が報告されているが、ウィルス、細菌およ
び原生動物エピトープのような抗原性エピトープが常に使用されている。先行技
術のハプテン化技術の不利益を有することなく、非抗原性ペプチドに対する抗体
の産生誘導を可能にするために、このような融合タンパク質中で非抗原性ペプチ
ドを使用することはこれまで示唆されていない。
実質的に非抗原性ペプチドの例は、種によって構造が実質的に異なっていないペ
プチドまたは、異なる動物に注入するとき、生じる抗体価が実質的に大きくなら
ない任意の他のペプチドまたはタンパク質である。LHRHの他に、成長ホルモ
ン放出ホルモン、副腎皮質ホルモン、副甲状腺ホルモン、インヒビンおよびゴナ
ドトロピンのサブユニットを挙げることができる。このようなペプチドの一部ま
たは僅かに修正したものも使用することができる。

Claims (10)
- 1.1つのプロモーターで制御される2つの遺伝子の発現で得られる組換え体融 合タンパク質であって、その際、第1の遺伝子は高い抗原性で高い親水性のタン パク質の産生をコードしており、そして第2の遺伝子は、それ単独では実質的に 抗原性でないペプチドまたはこのようなペプチドの1部をコードしている。
- 2.融合タンパク質が高度に抗原性でありそして上記の実質的に非抗原性ペプチ ドに対する抗体を有する抗体を産生させる請求の範囲第1項に記載の融合タンパ ク質。
- 3.上記ペプチドがホルモンからなりそして産生された抗体が内在的に産生され る同じホルモンを免疫無効化することができる請求の範囲第2項に記載の融合タ ンパク質。
- 4.上記の高い抗原性で高い親水性のタンパク質が肝炎表面抗原である請求の範 囲第1項に記載の融合タンパク質。
- 5.第2の遺伝子が、内在的に産生されるホルモンの活性と実質的に同じホルモ ン活性を有するタンパク質をコードしている請求の範囲第4項に記載の融合タン パク質。
- 6.上記第2の遺伝子が黄体形成ホルモン放出ホルモンをコードしている請求の 範囲第5項に記載の融合タンパク質。
- 7.上記第2の遺伝子が上記第1の遺伝子の1つまたはそれ以上の領域に挿入さ れている請求の範囲第1項に記載の融合タンパク質であって、その際、上記領域 の各々は上記タンパク質の高親水性領域をコードしている。
- 8.上記2つの遺伝子、転写開始領域、プロモーターおよび発現ベクターからな る請求の範囲第1項に記載の融合タンパク質をコードするDNAであって、その 際該DNAは哺乳動物の細胞を形質転換して融合タンパク質を発現させることが できる。
- 9.マウスのメタロチオネインプロモーターとウシ乳頭種ウイルスゲノムとが適 当な配列で融合したHBsAg−LHRHタンパク質および読み取りフレームを コードしているDNAからなる請求の範囲第8項に記載のDNAであって、これ にようて黄体形成ホルモン放出ホルモンタンパク質が抗原性特性を有して発現さ れる。
- 10.上記第2の遺伝子が上記第1の遺伝子の1つまたはそれ以上の領域に挿入 されている請求の範囲第8項に記載のDNAであって、その際上記領域の各々は 上記タンパク質の高親水性領域をコードしている。
