Description
つまり、研究者が、ヒト肝細胞培養と肝臓再生の新しいメカニズムに多くの進歩を遂げたが、現在、ヒト肝細胞を遺伝子編集なしに増殖させ続け、体内で効率的に再増殖の能力を持たせることは不可能だ。
もう一つの好ましい例では、上記のWntシグナル経路アクティベーターは、以下から選べられるアクティベーターを含む:Wnt3a タンパク質、Wnt3a馴化培地、CHIR、又はそれらの組み合わせ。
もう一つの好ましい例では、上記の細胞培地には、以下から選べられる培地を基礎培地とする:DMEM、MEM、RPMI(例えばRPMI1640)、Neuronal basal又はFischers;好ましくに、上記のDMEMは、Advanced DMEM/F12、DMEM/F12から選べられる。
もう一つの好ましい例では、上記のN2添加物の使用量は、1Xである。
もう一つの好ましい例では、上記のB27添加物の使用量は、1Xである。
一つの好ましい例では、上記の肝細胞は、成熟肝細胞および肝前駆細胞との間にある中間状態細胞である。
本発明のもう一つには、薬物組成物を提供し、ただし、上記の肝細胞培養物又は単離・精製された肝細胞;及び薬学的に許容される担体を含む。
本発明のもう一つには、インビトロで肝細胞を増殖すると肝細胞の成熟を誘導するためのキットを提供し、ただし、前記のいずれか一つの培地を含む。
本文に開示された内容に基づき、当業者にとって、本発明の他の面は自明なものである。
特に明記しない限り、本発明に培養されるものは、肝細胞、特に初代肝細胞、より特にヒト肝細胞、ヒト初代肝細胞である。
本発明は、インビトロで肝細胞を増殖する方法を開示する:肝細胞を、本発明の「肝細胞培地(インビトロ増殖培地)」に培養し、多量の成熟肝細胞と肝前駆細胞の間にある中間状態細胞を得る。本発明の一部の実施形態において、それが、肝前駆細胞に関連するマーカーとするタンパク質SOX9、CK19とCK7を発現することは見られ、そのALB、HNF4A、TTR又はCYP3A4発現レベルは、初代肝細胞より低く、そのCYP1A2、CAR、C3又はUGT1A1発現レベルは、初代肝細胞に近い或いはこれより高い。これらの細胞を、さらに肝成熟細胞に誘導することができる。
本発明者が、インビトロで肝細胞を増殖する培地を提供する。上記の培地は、「インビトロ増殖培地」と「肝向け成熟培地」を含む。
本発明の新しい発見によって、本発明に記載された方法で得られる肝細胞培養物、又は当該肝細胞培養物から単離・精製された肝細胞(成熟肝細胞と肝前駆細胞の間にある中間状態細胞を含む)、或さらに誘導してなる成熟肝細胞を提供する。
上記の組成物を投薬に使用される場合、通常に、1×102-1×1010個細胞/kg体重、好ましくは1×103-1×108個細胞/kg体重は適切であり、臨床医の診断および患者の症状の重症度に依存する。
1、実験材料
1.1 肝細胞培地
肝細胞培地(インビトロ増殖培地、HM)の配合及び来源は、表1に示された。ただし、各配合の作業濃度は、培地中の最終濃度である。
肝細胞誘導培地(肝向け成熟培地、HIM)の配合及び来源は、表2に示された。ただし、各配合の作業濃度は、培地中の最終濃度である。
*当該組分は、30% Wnt3a馴化培地又は0.1uM CHIR(CHIR99021)に代わられても良い。ただし、Wnt3a馴化培地は、2日間の培養後のL-Wnt3a細胞株(HansClevers研究室から入手)によって生成された細胞上清から採取された。
異なるロット番号のヒト肝細胞は、米国のCelsis In Vitro Technologies(Blatimore、MD)から購入した。L-Wnt3a細胞株は、Hans Clevers研究室から入手した。Snu-398細胞株は、米国のATCCセルバンクから購入した。
Fah-/- Rag2-/-IL2rg-/-(FRG)マウス:フマリルアセトアセターゼ(FAH)ノックアウトマウス(略語:Fah-/-マウス)は、1993年に確立された。Fah-/-マウスは、完全なチロシン代謝をできないので、肝臓にスクシニルを蓄積し、肝臓実質細胞の死をもたらす。NTBCを添加した飲料水で飼育されたFah-/-マウスは、野生型マウスと表現型に明らかな違いはなく、正常な肝機能を有し、正常に発育および繁殖できるが、NTBCの給餌を停止してから4~6週間以内に肝不全で死亡する。Fah-/-マウスは広範囲かつ継続的な肝障害があり、その肝臓微小環境は移植細胞の増殖に特に適し、野生型肝細胞(正常なFAH遺伝子発現を有する)は、脾臓移植を経った後にFah-/-マウスの脾臓をほぼ完全(90%以上)に再構築でき、アクセプターとするマウスは正常な肝機能に戻った。Fah-/-マウスとRag2-/-IL2rg-/-免疫不全マウスとを交配した後、3つの遺伝子ノックアウトを有するFah-/-Rag2-/-IL2rg-/-マウス(FRGマウス)が得られた。FRGマウスは、成熟したT、B細胞、およびNK細胞を欠いて、ヒト肝細胞の移植後、ヒト肝細胞を高度(>90%)に再増殖することができる。
2.1ヒト肝細胞培養
冷凍ヒト肝細胞は、米国のCelsis In Vitro Technologies(Blatimore、MD)から購入した。凍結したヒト肝細胞を10%血清を含むDMEM培地で蘇生し、2×104細胞/cm2の密度でI型コラーゲンでコーティングされた培養プレートに植え、培養プレートを37℃、5%CO2、5%O2のインキュベーターで培養し、1日培養後に肝細胞培養培地に交換し、3日ごとに細胞培養培地を交換した。細胞培養の6日後、生い茂った細胞をトリプシン処理し、I型コラーゲンでコーティングされた培養プレート上に3×104細胞/cm2の密度で再び植え、培養プレートを37℃、5%CO2、5%O2のインキュベーターで培養した。
増殖されたヒト肝細胞を培養し、いっぱいになった後、細胞をトリプシン処理し、細胞を肝細胞培養培地に再懸濁し、5×105細胞/ウェルの密度で日本のElplasia会社の非付着24ウェルプレートに植え、1日後、細胞培養培地を肝細胞の肝向け誘導培地に交換し、細胞はゆっくりと凝集して細胞球になった。培地を2日ごとに交換し、細胞誘導の10日後に誘導された細胞を収集した。
凍結したヒト肝細胞を10%血清を含むDMEM培地で蘇生し、2×104細胞/cm2の密度でI型コラーゲンをコーティングした6ウェルプレートに植えった。細胞が壁に付着した後、培地を肝細胞培養に変更し、細胞プレートをincuCyte FLR装置の37℃、5%CO2インキュベーターで培養し、細胞を3日間連続して培養し、同じ視野の細胞を30分ごとに写真を撮った。細胞培養・増殖のビデオはソフトウェアによって生成される。
1)Trizol(Invitrogen)またはRNeasy FFPE Kit(Qiagen)試薬を使用して、細胞RNAを抽出した。
2)Trizolで抽出されたRNAから1 ugを取って、RNeasyFFPEキットで抽出されたRNAから、1ugの特定の値またはすべてを取って、M-MLV逆転写酵素(Promega)キットでcDNAを得た。
3)リアルタイム定量PCRにはSYBR Premix Ex Taq(TaKaRa)キットを使用し、遺伝子発現の検出にはABI StepOnePlusリアルタイムPCRシステム(Applied Biosystems)を使用した。すべてのQ-PCRデータには少なくとも2回の繰り返しがあり、プライマー配列は表3に示された。
検出にはALBELISAキット(Bethyl Laboratories)を使用した。
培養された細胞の尿素分泌能を検出するために、細胞の24時間の細胞培養上清を採取し、Abnova会社製の尿素検出キットを用いて、実験操作過程に従って実験を行った。
細胞のPAS(Periodic Acid-Schiff stain、過ヨウ素酸シッフ染色)染色は、Sigma会社の実験キットに従って使用された。細胞をオイルレッド染色し、細胞から細胞培養液を吸引し、PBSで洗浄した。4%パラホルムアルデヒドを加え、細胞を30分間固定し、パラホルムアルデヒドを除去し、PBSで3回洗浄した。オイルレッドO(Sigma-Aldrich)で10分間染色し、70%エタノール溶液で2回洗浄し、顕微鏡で観察して写真を撮った。
細胞老化に関連するβ-ガラクトシダーゼ染色し、細胞プレート内の細胞から細胞培養培地を除去し、PBSで洗浄した。4%パラホルムアルデヒドを加え、細胞を5分間固定し、パラホルムアルデヒドを除去し、PBSで3回洗浄した。β-ガラクトシダーゼ染色キット(Beyotime)の指示に従い、顕微鏡で観察して写真を撮った。
増殖されたヒト肝細胞のCYP2B6代謝能を検出するために、10日間の肝向け誘導後、細胞と新たに蘇生されたヒト肝細胞を、それぞれに100μmのブプロピオンを含む培地にインキュベートし、インキュベーションの異なる時点での培養上清を採取した。細胞上清をRuideLiver Company会社に送って、質量分析を使用してCYP2B6代謝物の生成を検出した。産物は、市販の標準を使用して標準曲線を描いた。
実験手順に従って、細胞RNAをTrizol(Invitrogen)試薬で抽出した。Illumina Traseq RNAサンプル前処理キットを使用して、1マイクログラムの総RNAからシーケンスライブラリーを調製した。Illumina HISEQ2000シーケンサーで、シングルエンドリード長100bpで実行した。Tophatを使用して、読み取り数をヒトリファレンスゲノム(HG19)にマッピングした。Cufflinksを使用して、デフォルトのパラメーターを使用し、UCSC遺伝子のFPKMを計算した(100万分の1のフラグメントの100万分の1のエクソンフラグメントのマッピング)。FPKMは、下流の遺伝子分析のためにLog2に変換された。遺伝子発現については、一元配置分散分析を実行し、差異発現遺伝子(DEGS)を特定した。各分析では、P値と変化の倍数を計算する必要がある。Partek Genomics Suite 6.6に基づく選択されたプローブセットの平均リンケージクラスターのユークリッド距離を使用して、分類または遺伝子全体の無差別クラスタリングとヒートマップ生成を行った。
遺伝子セット濃縮分析(GSEA)は、DEG経路の濃縮に使用された。DEGsリストには、オンラインMSIGDBツールが使用された(http://Studio.Org/gSe/MSigDb/ index.jsp)。GSEA V2デスクトップソフトウェアは、RNASEQの結果から大幅に濃縮された経路を特定することに使用された。
1)細胞移植の6日前に、飲料水中のNTBCの濃度を除去することにより、マウスの肝障害が引き起こされた。
2)マウスの脾臓側を上に向けて、肋骨の1~2cm下の表皮と筋層を切り取った。脾臓に付着している褐色脂肪を見つけ、それを引き出して脾臓を引き出した。脾臓の前端を糸で軽く結紮した。
3)5×105のproliHHまたはヒト初代肝細胞(約100ul)をインスリン針で採取し、結紮位置を超えて脾臓に針を挿入した。注入が完了したら、30~60秒間一時停止し、針を抜いて糸をしっかりと結び、液体が逆流しないようにした。
4)脾臓を腹腔に入れ、筋肉と表皮を縫合し、傷口をアルコール綿で消毒し、ケージに入れた。移植後、体重と死亡状態を毎週記録した。移植細胞がなく、体重が30%減少したマウスを陰性コントロールとして使用した。
5)12週間生存したマウスを処理し、その後の分析のために肝臓と血液のサンプルを収集した。
1)ProliHHと肝癌細胞株Snu398を消化し、2×106個の細胞を採取し、免疫不全NOD-SCIDマウスの鼠径部に直接注入した。
2)2か月後、マウスの腫瘍形成を観察し、記録のために写真を撮った。
全文統計分析にはGraphPadPrism5ソフトウェアを使用した。両側ログランク検定を使用して、初代肝細胞移植後のラットの生存曲線を計算した。サンプルの血清学的データ統計は、対応のないスチューデントのT検定を採用した。図に示されているデータは、平均±標準誤差である。* P <0.05を示すものは、著しい違いである。
インビトロの増殖には、増殖されたヒト肝細胞は、一部の成熟肝臓遺伝子の発現をダウンレギュレートし、肝前駆体関連遺伝子の発現をアップレギュレートした。それがインビトロで成熟肝細胞に誘導され得るかどうかを試験するために、本発明者らは、ProliHHを三次元培養システムに置き、肝向け成熟培地(HIM)を添加し、10日間培養した(図5A)(通常の酸素)。成熟まで誘導した後、ProliHHは成熟肝細胞に特有の典型的な多角形と二重の核を有する(図5B)。細胞形態の変化に加えて、ALB、TTR、HNF4A、AATなどの肝細胞遺伝子も大幅にアップレギュレートされ、SOX9、CK19、CK7などの肝前駆体遺伝子は大幅にダウンレギュレートされた(図5C)。さらに重要なことは、薬物と尿素の代謝に関与する重要な遺伝子も、成熟まで誘導された後に有意にアップレギュレートされ、それに応じて、CYP2B6と尿素の代謝能も有意に増加した(図5D-F)。
増殖されたヒト肝細胞ProliHHは、インビトロでの増殖中に、肝前駆体関連遺伝子の発現をアップレギュレートした。それが体内で成熟肝細胞に誘導され得るかどうかを試験するために、本発明者らは、FRGマウスの肝臓に組み込まれたProliHHを分析・同定した。免疫蛍光染色は、ProliHHが体内で成熟肝細胞マーカータンパク質ALB、FAH、HNF4AとCYP3A4を発現したが、肝前駆細胞関連遺伝子CK19およびCK7を発現しなかったことを示した(図8AとB)。qPCR検出にヒト特異的肝臓遺伝子プライマーを使用すると、体内で、フェーズIとフェーズII代謝酵素及びトランスポータータンパク質などの成熟肝細胞遺伝子のProliHHでの発現が、インビトロで培養されたProliHHよりも有意に高かったことが示された(図8C)。全トランスクリプトーム分析によって、体内でのProliHHが、PHHと非常に高い類似性を持っていることを示し(r2=0.94)、これは、体内での肝細胞には、ProliHHの遺伝子発現がPHHの遺伝子発現に近いことを示している(図8D)。

であることを特徴とする、インビトロで肝細胞を増殖する方法。 - 前記細胞培養培地は、DMEM、MEM、又はRPMIから選ばれる培地を基礎培地とする、ことを特徴とする請求項1に記載の方法。
- 前記DMEMは、Advanced DMEM/F12およびDMEM/F12から選ばれることを特徴とする、請求項2に記載の方法。
- 請求項1で増殖された肝細胞を成熟肝細胞に誘導する方法であって、
前記細胞培養培地にフォルスコリン、デキサメタゾン、オンコスタチンMを
フォルスコリン 3~7 μM、
デキサメタゾン 5~20 μM、
オンコスタチンM 10~30 ng/ml、
の濃度で添加することを特徴とする方法。 - 前記細胞培養培地においてフォルスコリン、デキサメタゾン、オンコスタチンMは、
フォルスコリン 4~6 μM、
デキサメタゾン 7~15 μM、
オンコスタチンM 15~25 ng/ml、
の濃度であることを特徴とする請求項4に記載の方法。 - 前記成分の濃度が、
Wnt3a タンパク質 40~60 ng/ml、
CHIR 0.07~1.5 μM、
N-アセチル-システイン 0.7~1.5 mM、
ニコチンアミド 7~15 mM、
FGF10 2~3 ng/ml、
EGF 40~60 ng/ml、
HGF 20~30 ng/ml、
[Leu15]-ガストリンI 7~15 mM、
A 83-01 4~6 μM、
Y-27632 7~15 μM、
血清 0.7~1.5 %、
であることを特徴とする請求項1に記載の方法。 - 前記方法は、増殖した肝細胞を継代培養する請求項1に記載の方法。
- 前記継代培養は低酸素状態で行われ、前記低酸素状態は、体積比で0~15%の酸素を含む、ことを特徴とする、請求項7に記載の方法。
- 前記低酸素状態は、体積比で10%未満の酸素を含む、ことを特徴とする、請求項8に記載の方法。
- 前記低酸素状態は、体積比で6%未満の酸素を含む、ことを特徴とする、請求項8に記載の方法。
- 前記培養は、3次元培養である、ことを特徴とする、請求項8に記載の方法。
- 上記の肝細胞は、ヒト肝細胞であることを特徴とする請求項1に記載の方法。
- 請求項1~3および6~12のいずれか一つの方法で得られた肝細胞培養物、又は、当該肝細胞培養物から単離・精製された成熟肝細胞と肝前駆細胞の間にある中間状態細胞である肝細胞。
- 請求項13に記載の、肝細胞培養物、又は、単離・精製された成熟肝細胞と肝前駆細胞の間にある中間状態細胞である肝細胞、の使用であって、
肝臓の再生を促進する薬物の調製における使用;または、
肝臓疾患を予防、緩和又は治療する薬物組成物又はキットの調製における使用;または、
インビトロモデルとして、肝臓に関連する疾患又は薬物に対する研究における使用;または、
生体内の細胞移植の組成物の調製における使用;または、
Albuminタンパク質の産生における使用。 - 前記肝臓に関連する疾患又は薬物的研究は、薬物の送達、薬物の代謝、肝臓の形成、肝臓の再生に対する研究;或いは肝毒性試験、肝細胞毒性物質のスクリーニング、肝細胞機能を調節する物質のスクリーニングであることを特徴とする請求項14に記載の使用。
- 請求項13に記載の肝細胞培養物又は単離・精製された肝細胞;及び薬学的に許容される担体;を含むことを特徴とする薬物組成物。
- 請求項13に記載の肝細胞培養物又は単離・精製された肝細胞を含む;又は請求項16に記載された薬物組成物を含む;ことを特徴とするキット。
- 培養培地が、成分として、Wntシグナル経路アクティベーター、N-アセチル-システイン、ニコチンアミド、FGF10、EGF、HGF、[Leu15]-ガストリンI、A 83-01、Y-27632、および血清を含有し、
前記Wntシグナル経路アクティベーターは、Wnt3a タンパク質またはCHIRであり、前記成分の濃度は、
Wnt3a タンパク質 30~70 ng/ml、
CHIR 0.05~2 μM、
N-アセチル-システイン 0.5~2 mM、
ニコチンアミド 5~20 mM、
FGF10 1~4 ng/ml、
EGF 30~70 ng/ml、
HGF 15~40 ng/ml、
[Leu15]-ガストリンI 5~20 mM、
A 83-01 3~7 μM、
Y-27632 5~20 μM、
血清 0.5~2 %、
であることを特徴とする、インビトロで肝細胞を増殖するための培養培地。 - 請求項18に記載のインビトロで肝細胞を増殖する培養培地、及び、フォルスコリン、デキサメタゾン、オンコスタチンMを含む、インビトロでの肝細胞の増殖、および、肝細胞の成熟を誘導する培養培地であって、前記フォルスコリン、デキサメタゾン、オンコスタチンMは、
フォルスコリン 3~7 μM、
デキサメタゾン 5~20 μM、
オンコスタチンM 10~30 ng/ml、
の濃度であることを特徴とする、インビトロで肝細胞を増殖、および、肝細胞の成熟を誘導する培養培地。 - 前記培養培地が含む成分が、基礎培地に存在し、前記基礎培地は、DMEM、MEM、又は、RPMIから選ばれることを特徴とする請求項18に記載の培養培地。
- 前記DMEMは、Advanced DMEM/F12およびDMEM/F12から選ばれることを特徴とする、請求項20に記載の培養培地。
- 前記培養培地における成分の濃度は、
Wnt3a タンパク質 40~60 ng/ml、
CHIR 0.07~1.5 μM、
N-アセチル-システイン 0.7~1.5 mM、
ニコチンアミド 7~15 mM、

であることを特徴とする請求項18に記載された培養培地。 - インビトロで肝細胞を増殖するとともに、肝細胞の成熟を誘導することを特徴とする請求項18~22のいずれか一つの培養培地の使用。
- 請求項18~22のいずれか一つの培養培地を含むことを特徴とするインビトロで肝細胞を増殖するとともに肝細胞の成熟を誘導するキット。
