Description
(i)抗-HGFR抗体フラグメントは、ヒトHGFRの細胞外部分のエピトープに結合することが可能なパラトープを1つのみ有し、HGFRへ向けたアンタゴニスト活性を有し
(ii)ヒトHGFRの細胞外部分は、安定した様式で肝細胞増殖因子(HGF)に結合することが可能であり、抗-HGFR抗体フラグメントによって認識されるエピトープ内に少なくとも1つのアミノ酸突然変異を含み、抗-HGFR抗体フラグメントがそれに結合するのを防ぎ、および
(iii)抗-HGFR抗体フラグメントおよびヒトHGFRの細胞外部分は、(a)タンパク質形態で、または(b)核酸形態で、患者へ投与するのに適切である。
(i)抗-HGFR抗体フラグメントは、ヒトHGFRの細胞外部分のエピトープに結合することが可能なパラトープを1つのみ有し、HGFRへ向けたアンタゴニスト活性を有し、

decoyMETにおけるDN30-結合エピトープの部位特異的突然変異誘発
MvDN30抗体およびdecoyMETを組み合わせた活性を利用するために、相互中和をもたらす2つの分子間の相互作用を防ぐことが必須である。MvDN30は、IPT-3ドメインとの境界におけるMET細胞外領域のIPT-4ドメイン内のエピトープを認識することが以前に示されている28。以前の研究は、ピコモルの親和性でヒト受容体に結合する親DN30抗体が、イヌおよびラットMETとも相互作用するが11、23、マウスとは交差反応しない21ことを示した。上述の哺乳類種のIPT-3およびIPT-4アミノ酸配列のアライメントの際に、マウスにおいて選択的に変化しているいくつかの残基を同定した(図1A)。単一アミノ酸残基のヒトからマウスへのスワッピングが抗体結合を損なわせ得るかどうか試験するために、IPT3-4ドメイン内に点突然変異を保有する可溶性受容体を生産してDN30抗体に対して試験した。グルタミン酸によるリジン842の置換は、decoyMETK842E(抗体によってもはや認識されない改変された可溶性受容体)を生じさせたが、他の突然変異は全て、相互作用に影響しなかった(図1B)。抗体がdecoyMETK842Eと相互作用することができないことは、アフィニティ精製されたdecoys(固相中)およびDN-30抗体(液相中)を用いて行われたELISAアッセイにおいて確認された(図1C)。それ故に、酸性残基による842位の塩基性アミノ酸の置換は、結合部位の重大な摂動(critical perturbation)をもたらし、decoyMETK842E-抗体の相互作用を妨害した。
本発明者らはそれから、K842Eアミノ酸置換がHGFの結合を妨害するかどうか研究した。ELISA結合アッセイでは、HGFに関するdecoyMETK842Eの親和定数(Kd=1.04±0.05nM)は、野生型decoyMETに関して測定されたKd=1.44±0.07nMと重ねることができた(superimposible)(図2A)。最後に、HGFによって誘導されるMETリン酸化アッセイにおいて、decoyMETK842Eの阻害活性を試験した。図2Bに示されるように、野生型decoyMETおよびdecoyMETK842Eは、A549肺がん細胞におけるHGF-依存性METリン酸化を同等の能力で阻害した。したがって、K842E置換は、HGFとの安定した複合体の形成を妨げず、decoyの阻害活性は影響を受けないままである。
リガンドおよび受容体の同時性の標的化によって引き起こされるMETシグナル伝達に対する阻害活性を評価するために、MvDN30およびdecoyMETK842Eの単独または組み合わせのいずれかの存在下でMETリン酸化を試験した。この目的を達成するために、野生型MET受容体を発現するA549細胞をナノモル濃度のHGFで刺激して、ホスホMET抗体によってMET活性化を測定した。両方の分子が阻害活性を示し、2つの組み合わせがより効率的であった(図3A)。下流のシグナリング伝達因子(signaling transducer)の分析は、組み合わせが、ERKおよびAKT活性化の最も効果的な阻害を達成することを確認した(図3B)。
リガンド主導のMET刺激のマウスモデルにおいて、MvDN30およびdecoyMETK842Eを組み合わせた阻害活性をインビボで評価した。
抗-HGFR抗体フラグメントおよびヒトHGFRの細胞外部分を含む融合タンパク質をコードする単一cDNAを産生するために、本発明者らは、単鎖MvDN30(ScMvDN30)を最初に設計して、軽鎖(VL-CL)および重鎖(VH-CH1)の間にリンカーを導入した。リンカーはフレキシブルであり、グリシン/セリン残基が豊富であり、そして、単量体分子(二量体および/または多量体を生じさせるように、それらは同一ポリペプチドに属する抗体軽鎖および重鎖の関連性によって構成され、別個のポリペプチド(polypepdites)由来ではない)の産生を優先的に可能にする長さを有していた。リンカーの好ましいアミノ酸配列は、配列番号36に報告されて;対応するヌクレオチド配列は配列番号39に報告される。それから、ScMvDN30 cDNAをdecoy METK842Eとインフレームで融合した。2つのグループの融合タンパク質が生産された:i)decoyMETK842EがN末に位置してScMvDN30がC末に位置する、およびii)scMvDN30がN末に位置してdecoyMETK842EがC末に位置する。構造全体に対して高い自由度を保証するために、本発明者らは、2つの部分の間に第2のリンカーを導入した。アミノ酸組成および/または長さが改変された3つの異なるリンカー:i)L45:アラニンおよび荷電残基が豊富な配列の2回繰り返しで構成された45アミノ酸のリジッドリンカー(配列番号35、対応するヌクレオチド配列は配列番号38に報告される);ii)L60,60アミノ酸のグリシン/セリン残基が豊富なフレキシブルリンカー(配列番号36、対応するヌクレオチド配列は配列番号39に報告される;iii)L134,134アミノ酸のフレキシブルおよびリジッド領域の組み合わせ(配列番号37、対応するヌクレオチド配列は配列番号40に報告される)を用いた。融合タンパク質の略図を図9に報告する。
本発明者らは、新たに生産された融合タンパク質が、HGFおよびMETの両方に結合する能力を維持するかどうかをELISAアッセイによって研究した。全ての融合タンパク質は、decoyMETK842Eのものと重ねることができる(superimposable)親和性でHGFと相互作用したが(図10A)、scMvDN30をC末に含む融合タンパク質は、N末に位置する抗体との融合と比較してより低い親和性でMETを認識した。この後者のグループの分子に関して測定されたKdは、scMvDN30単独に関して測定されたものと同等であった(図10B)。
本発明者らは、decoyMETK842Eと等モル比で組み合わせたscMvDN30と比較して、処置の3日後の細胞増殖障害を測定して、融合タンパク質の阻害特性を評価した。全ての融合タンパク質は、用量依存的な様式で阻害特性を示した。報告された実験では、L60リンカーを用いた融合は他の分子よりも強力でなかったが、L45およびL134タンパク質は、scMvDN30-decoyMETK842Eの組み合わせと同様の活性を示した(図11)。
HGF主導の細胞運動の阻害における融合タンパク質の効果を評価するために、本発明者らは、HGFを用いて処理されたHPAF-IIヒト膵臓腺がん細胞をモデルとして用いて、X-CELLigence Real Time Cell Analyzerを用いて定量的な運動性アッセイを行なった。全ての融合は、HGF-依存性の細胞分散を減少させることができた(図12)。
融合タンパク質の阻害活性を、hHGF-Ki SCIDマウスモデルにおいてインビボで評価した。ヒト膵臓腺がん細胞(Capan-I)を、ルシフェラーゼ遺伝子を用いた形質導入によって標識化して、マウスの膵臓内へ同所性に注入した。全身発光の分析によって生着を確認した;生物発光の値に基づいてマウスを同質なグループに階層化して、リンカーL60またはL134を含む融合タンパク質によって処置した。参考文献のように、MvDN30およびdecoyMETK842Eの1:1組み合わせおよびビヒクルを実験グループに含んだ。屠殺の際、細胞注入の5週間後に、肝臓を切除して、IVIS Spectrumによって分析して転移をスコア化した。MvDN30およびdecoyMETK842Eの同時性の投与は、肝臓への、MET主導の転移伝播を有意に阻害した;MvDN30投与が毎日された場合は高い有効性がスコアされたが(COMBO 1グループ)、より少ない頻度の投与が適用された場合は、治療的応答は有意に減少した(COMBO 2グループ)。この制限は、融合タンパク質を用いて処置されたマウスではスコア化されなかった。実際に、それらの全ては、1週間に2回の送達であっても肝臓への転移伝播を高い有効性で減少させた(図13)。
細胞培養
A549ヒト肺腺がん細胞、HPAF-IIおよびCapan-1ヒト膵臓腺がん細胞、およびU87-MGヒト膠芽腫細胞は、ATCC/LGC Standards S.r.l.(Sesto San Giovanni,Italy)から得て;EBC-1ヒト肺癌腫細胞は、Japanese Collection of Research Bioresourcesから得た。全ての細胞は、供給元によって示唆されるとおりに培養した。全ての細胞培養をマイコプラズマ汚染に関して試験した。
HGFRの細胞外部分のエピトープを認識することが可能な抗-HGFR抗体フラグメントの軽鎖(VL-CL)および重鎖(VH-CH1)をコードするcDNAを、配列番号7および配列番号8に報告される配列に従って、Invitrogen GeneArt Gene Synthesis(ThermoFisher)により外部委託で行なわれる遺伝子合成によって生産した。抗体フラグメントを、軽鎖(VL-CL)および重鎖(VH-CH1)をコードするcDNAを発現するpcDNA3.1プラスミド(カタログ番号V79020 Invitrogen Corporation,Camarillo,CA)を用いたHEK-293T細胞の一過的トランスフェクションによって生産した。トランスフェクトされた細胞を3日間飢餓させて、可溶性受容体を含む細胞培養上清を回収した。組み換えタンパク質の精製を、製造元の説明書に従ってHisTrap HPカラム(カタログ番号17524701 GE Healthcare,Freiburg,Germany)を用いたアフィニティクロマトグラフィーによって行なった。大スケールのタンパク質生産および精製を、U-Protein Express BV(Utrecht,The Netherlands)によって行なった。
単一アミノ酸置換を保有するヒトMET外部ドメイン(decoyMET)のcDNA配列を、製造元の説明書に従ってQuickChange II部位特異的突然変異誘発キット(カタログ番号200524 Agilent Technologies,Santa Clara,CA)を用いて生産した。手順は、所望の点突然変異を含むセンスおよびアンチセンスのオリゴヌクレオチドの設計を要する。以下のオリゴを用いた:

血清飢餓A549を、125nMのMvDN30または2μMのdecoyMETK842Eを単独または組み合わせで用いて24時間インキュベートして、それから、50ng/mlのHGF(カタログ番号294-HG-025 R&D Systems)を用いて4℃で2時間刺激した。全細胞溶解物を、以下の一次抗体:抗-METホスホ-Tyr1234/1235(D26,カタログ番号3077 Cell Signaling Technology,Beverly,MA);抗-MET(3D4,カタログ番号08-1366 Invitrogen Corporation);抗-ビンキュリン(クローンhVIN-1,カタログ番号V9131 Sigma Life Science,Saint Louis,MO)を用いてウエスタンブロットによって分析した。抗‐マウスIgG1(カタログ番号JI115035003)および抗‐ウサギIgG(カタログ番号JI111035003)HRP-コンジュゲート二次抗体はJackson ImmunoResearch.(West Grove,PA)由来であった。
decoyMETおよびDN30 mAbの間の相互作用の分析のために、アフィニティ精製された可溶性受容体(野生型decoyMETまたはdecoyMETK842E,100ng/well)をELISAプレート上に固定化して、増加する濃度の抗体(0~100nM)を液相内に添加した。HRP-コンジュゲート抗-マウス抗体(カタログ番号NA931 GE Healthcare)を用いて結合を明らかにした。 および融合タンパク質の間の相互作用の分析のために、アフィニティ精製されたdecoyMET野生型(100ng/well)をELISAプレート上に固定化して、増加する濃度の融合タンパク質またはscMvDN30(0~1000nM)を液相内に添加した。HRP-コンジュゲート抗-ヒトk鎖抗体(カタログ番号A7164 Sigma-Aldrich)を用いて結合を明らかにした。HGFに対するdecoyMETまたは融合タンパク質の結合の分析のために、可溶性受容体(100ng/ウェル)をELISAプレート上に固定化して、溶液中で、増加する濃度のHGF(0~11nM)とともにインキュベートした。結合を、抗-HGFビオチン化抗体(カタログ番号BAF294 R&D Systems)を用いて検出して、HRP-コンジュゲートストレプトアビジン(カタログ番号RPN 1231 GE Healthcare)を用いて明らかにした。
足場非依存性の増殖アッセイのために、2% FBSおよび0.5% Seaplaqueアガロース(カタログ番号50100 BMA,Rockland,ME)で補充された適切な培養培地中に細胞を懸濁して、1%アガロースの上部へ48ウェルプレート(500細胞/ウェル)内に蒔いた。処理を含む新鮮な培地を週に2回供給した。A549細胞を、1μMのMvDN30または1μMのdecoyMETK842Eを単独または組み合わせて用いて、30ng/mlのHGF存在下で処理した。U87-MG細胞を、0.5μMのMvDN30または1μMのdecoyMETK842Eを単独または組み合わせて用いて処理した。培養の12日後にテトラゾリウム塩(カタログ番号T01380 Sigma-Aldrich)を用いてコロニーを染色した。Metamorphソフトウェア(Molecular Devices,Sunnyvale,CA)を用いてコロニー増殖を判定した。細胞浸潤アッセイのために、HPAF-II細胞(1.5×105/ウェル)を、0.5μMのMvDN30または1μMのdecoyMETK842Eの単独または組み合わせの存在下で、血清フリーの培養培地中に懸濁して、30μg/ウェルのマトリゲルマトリックス(カタログ番号354234 Corning Incorporated,NY)で事前にコーティングされたトランスウェルチャンバーの上コンパートメントに蒔いた。2% FBSおよび12.5ng/ml HGFで補充された培養培地を、チャンバーの下コンパートメントに添加した。24時間後、トランスウェルフィルターの上側にある細胞を機械的に除去して、一方で、膜を通って遊走した細胞を、11%グルタルアルデヒド(カタログ番号340855 Sigma-Aldrich)を用いて固定して、0.1%クリスタルバイオレット(カタログ番号C3886 Sigma-Aldrich)を用いて染色した。Image-Jソフトウェアを用いて細胞浸潤を定量化した。細胞分散アッセイのために、HPAF-II細胞(8000/ウェル)を完全培養培地内の96ウェルプレート内に蒔いた。6時間後、増加する濃度(0~4μM)のMvDN30またはdecoyMETK842Eを、単独または1:1の組み合わせで添加した。さらに24時間後、6.25ng/mlのHGFを用いて20時間、細胞を刺激した。細胞を、11%グルタルアルデヒドを用いて固定して、0.1%クリスタルバイオレットを用いて染色した)。リアルタイムの細胞運動アッセイのために、HPAF-II細胞をE-プレート(8000/ウェル;Roche Diagnostics,Mannheim,Germany)内に蒔いて、上記のとおりに処理して;適用される場合(when applied)、融合タンパク質を3μMの濃度で試験した。電気インピーダンスを、X-Celligence RTCA装置(Roche Diagnostic)を用いてデータを10分ごとに記録しながら48時間連続的に監視した。値は、HGF添加の瞬間に標準化された細胞指標として表される。
腫瘍細胞および組織に対する免疫蛍光分析を、記載16、25のとおりに行なった。染色を、抗-METホスホ-Tyr1234/1235一次抗体(D26)を用いて行ない、Alexa Fluor 555-コンジュゲート二次抗体(カタログ番号A31570 Thermo Fisher Scientific)によって明らかにした。488-コンジュゲートファロイジン(カタログ番号A12379 Thermo Fisher Scientific)を用いて細胞を対比染色した。全ての画像を、Leica TCS SP5 AOBS共焦点レーザ走査顕微鏡(Leica Microsystems)を用いてキャプチャーした。免疫蛍光取得設定は、各細胞株または腫瘍組織内で一定に維持した。平均蛍光強度(MFI)を、ImageJソフトウェアを用いて評価して、各チャンネルにおける平均ピクセル強度を測定して、バックグラウンドを差し引いた。細胞株に関してはホスホMET MFIはファロイジンに対して標準化されたが、腫瘍に関しては全てのシグナルはDAPIに対して標準化された。
全ての動物手順は、Fondazione Piemontese per la Ricerca sul Cancroの動物実験に関する倫理委員会およびItalian Ministry of Healthによって承認されたプロトコルに従って行われた。NOD-SCIDマウスは、Charles River(Calco,Italy)から購入し;hHGF-Ki SCIDマウス)は、AVEO Pharmaceuticals,Cambridge,MAから取得した。
平均、標準偏差(SD)および平均の標準誤差(SEM)は、Microsoft Office Excel 2010ソフトウェア(Microsoft Corporation,Redmond,Washington)を用いて計算した。Kd値を計算するために、ELISAアッセイによるデータを分析して、GraphPad Prismソフトウェア(GraphPad Software,San Diego,California)を用いて、非線形回帰、1サイト結合・双曲線カーブ(one site binding hyperbola curve)に従ってフィットさせた。IC50値を計算するために、増殖アッセイによるデータを分析して、GraphPad Prismソフトウェアを用いて、非線形回帰、シグモイド型用量反応曲線(sigmoidal dose-response curve)に従ってフィットさせた。統計的有意性を、両側スチューデントt検定を用いて判定した。全ての実験は少なくとも3回繰り返した(生物学的レプリケート)。インビボ実験は2回繰り返した。図は1つの代表的な実験を示す。
