本発明者は、組み換えHAPLN1(hyaluronan and proteoglycan link protein 1)蛋白質を生産し、その肺胞損傷改善効果、及び慢性閉塞性肺疾患(COPD:chronic obstructive pulmonary diseases)動物モデルでの治療効果を確認することにより、本発明を完成した。
1.組み換えヒトHAPLN1蛋白質のアミノ酸配列
組み換えHAPLN1蛋白質を構成するアミノ酸配列は、次の通りである。

組み換えヒトHAPLN1(rhHAPLN1:gene number 2678736)の製造のために、組み換えヒトHAPLN1アミノ酸配列が符号化されたDNAベクターを、宿主細胞であるExpiTM293細胞(Thermo Fisher Scientific Co.、Waltham、MA,米国)に感染させた。精製のために、アミノ末端に、分泌信号ペプチド、及び10個のヒスチジン(H、His、histidine)、そしてTEV蛋白質分解酵素(TEV protease)認知配列(TEV recognition site)を挿入した状態で発現させた。ベクター感染3日後、培養液を収集し、HisTrap column(GE Healthcare、IL、米国)を介して精製した後、TEVタンパク質分解酵素で、TEVタンパク質分解酵素認知配列を切断し、ヒスチジンを含む分子をDynaBeads(Thermo Fisher Scientific)を使用して除去した。そのように得られた溶液を、40mM Tris-HCl、1M NaCl、pH8.0に対し、16時間透析を行った。最終精製品の蛋白質濃度は、0.11mg/mlであり、溶媒は、20mM Tris-HCl、0.5M NaCl、pH8.0、50%グリセロールにし、1回投与量として小分した後、-20℃冷蔵庫に保管して使用した。
1.実験動物の準備及び飼育
実験動物は、2ヵ月齢の雄C57BL/6J((株)Young Bio、大韓民国)マウスを若年(young)マウスに設定し、20ヵ月齢の雄C57BL/6Jマウスを、老化(old)マウスに設定した。水と飼料は、自由に摂取することができるようにし、飼育室内の温度は、21~24℃、湿度は、40%~60%に維持し、昼と夜との周期は、それぞれ12時間にした。
実験終了時、心臓灌流を施した後、左肺を上下部に二等分し、中性緩衝10%ホルマリン(NBF:neutral buffered 10% formalin)に固定し、上側切片の切断面が見えるように、パラフィン組織切片を作製した。該組織切片を、ヘマトキシリン&エオシン(H&E:hematoxylin & eosin)法で染色し、それを、Ni-U(Nikon)顕微鏡及びDS-Ri1(Nikon)デジタルカメラを利用して撮影し、その結果を図1(A)に示した(縮尺バー=100μm)。
1.実験動物の準備及び飼育
実験動物は、体重20~25gの6週齢から10週齢のめすC57BL6/N((株)Young Bio、大韓民国)マウスを使用し、該マウスは、各群当たり5匹に分けた。水と飼料は、自由に摂取することができるようににし、飼育室内の温度は、21~24℃、湿度は、40%~60%に維持し、昼と夜との周期は、それぞれ12時間にした。
慢性閉塞性肺疾患(COPD)が誘発されたマウスモデルを製造するために、Suki et al (2017, Methods Mol. Biol. 1639: 67-75)及びWright et al (2008, Am J Physiol Lung Mol Physiol 295: L1-L15)を参照した。
エアロゾル吸入群(AH群)は、さらにAH-1:PPE未処理(正常)群、AH-2:PPE+食塩水群、AH-3:PPE+rhHAPLN1群、AH-4:PPE+ヒアルロン酸(HA)群に分け、気管内点滴吸入群(TI群)は、さらにTI-1:PPE未処理(正常)群、TI-2:PPE+食塩水群、TI-3:PPE+rhHAPLN1群、TI-4:PPE+HA群に分けた。ここで、ヒアルロン酸(HA)は、慢性閉塞性肺疾患(COPD)に効能があると知られており、陽性対照群として使用した。ヒアルロン酸(HA)の投与根拠と濃度決定とのために、Cantor et al (2005, Experimental Lung Research, 31: 417-430)を参照し、それにより、ストレブトコックスエキ(Streptococcus equi)由来のヒアルロン酸ナトリウム塩(hyaluronic acid sodium salt)(Sigma-Aldrich; St. Louis, MO、米国;Cat. 73641-10MG)形態で購入して使用した。
Claims (7)
- HAPLN1(hyaluronan and proteoglycan link protein 1)タンパク質を有効成分として含む、老化またはエラスチン減少による肺胞損傷により発病する肺疾患の予防用または治療用の薬学的組成物。
- 前記HAPLN1タンパク質は、配列番号1と、少なくとも95%の配列同一性を有するアミノ酸配列を含む、請求項1に記載の薬学的組成物。
- 前記肺疾患は、
慢性気管支炎、喘息、肺気腫及び慢性閉塞性肺疾患(COPD)からなる群のうちから選択される1以上であることを特徴とする請求項1に記載の薬学的組成物。 - HAPLN1(hyaluronan and proteoglycan link protein 1)タンパク質を有効成分として含む、老化またはエラスチン減少による肺胞損傷により発病する肺疾患の予防用または治療用の健康機能食品組成物。
- 前記肺疾患は、
慢性気管支炎、喘息、肺気腫及び慢性閉塞性肺疾患(COPD)からなる群のうちから選択される1以上であることを特徴とする請求項4に記載の健康機能食品組成物。 - 請求項1に記載の薬学的組成物を個体に投与する段階を含む、それを必要とする個体における、老化またはエラスチン減少による肺胞損傷により発病する、人間以外の動物の肺疾患を予防したり治療したりする方法。
- 前記肺疾患は、
慢性気管支炎、喘息、肺気腫及び慢性閉鎖性肺疾患(COPD)からなる群のうちから選択される1以上である、請求項6に記載の肺疾患を予防したり治療したりする方法。
