本発明は概して、開発及び製品化のための治療用タンパク質の選択に関し、より具体的には、高濃度タンパク質を容易に実現できる可能性が高く、それに付随する凝集体が形成される可能性が低いタンパク質の選択に関する。本発明は特に、タンパク質のコロイド相互作用の評価、及び高濃度で製剤化するためのタンパク質の選択に関する。
大きな生物学的分子、すなわち「バイオ治療薬」は重要な薬物種である。例えばモノクローナル抗体は、優れた治療特異性、長い生物学的半減期、及び高い安全性プロファイルを示す。

タンパク質の動的ビリアル範囲を決定するための簡易なSINS系アッセイを提供する。タンパク質は複雑な挙動を示すが、これはタンパク質が化学的物理的環境の影響を極めて受けやすいためである。したがって、ある特定された環境状況下でタンパク質のビリアル係数を評価しても、他の環境におけるそのタンパク質のビリアル係数を決定するには不十分である。無数の条件下で所与のタンパク質に関する多数のビリアルを得るのは労力と時間を要する。本発明者らは本明細書で、タンパク質の動的コロイド安定性を決定するハイスループットアッセイを開示している。これは、凝集に付随する問題を伴わずに、または少なくとも凝集の潜在的な問題を最小限にして、高濃度での製剤化またはそれ以外の高濃度での維持が可能なタンパク質を選択する情報を提供する。
好ましくない動的コロイド相互作用プロファイルを有するタンパク質、すなわち自己会合する傾向のあるタンパク質は一般に、高濃度では粘度が高くなる可能性がある。そのような高濃度での粘度の高さは、タンパク質を非経口注射に望ましくないものにする可能性がある。本明細書に記載のCD-SINSアッセイは、粘度降下賦形剤の選別に有用である。一態様では、減粘性タンパク質製剤の製造方法を提供する。一実施形態は、粘度降下能のある賦形剤である。
本明細書で使用される場合、用語「コロイド安定性」は、「自己会合する傾向」または「凝集する傾向」と同義に使用することができ、タンパク質の引力性、中性、または斥力性の相互作用の能力をもたらし得るスルースペース分子力、例えば、静電気力、ファンデルワールス力などの正味(全体的)効果を意味する。
(a)タンパク質、ナノ粒子、及び緩衝塩を組み合わせて試料を作製する工程;
(b)前記試料を光で励起する工程;
(c)前記試料を透過した光を測定する工程;
(d)前記試料の吸収強度比を算出する工程
を含み、
前記吸収強度比が閾値を超える場合、前記タンパク質が高濃度で安定である、
タンパク質が自己会合する可能性を決定するための方法。
(e)高濃度の高可溶性タンパク質を賦形剤と組み合わせて、製剤化した原薬を作製する工程
をさらに含む、先行条項のいずれか1項に記載の方法。
(a)少なくとも2種のナノ粒子;
(b)少なくとも2相のタンパク質;及び
(c)塩または緩衝液
を含む、生物分析用混合物。
モノクローナル抗体(mAb)をEESYR(登録商標)細胞によって産生し、プロテインAクロマトグラフィー及び陰イオン交換または疎水性相互作用クロマトグラフィーの研磨ステップによって精製した。緩衝液成分は、Sigma-Aldrich、VWR、またはJT-Bakerから入手したものであり、入手可能な最高等級のものであった。Illustra NAPカラム(カタログ番号17-0853-02)及びXK26/100 Superdex 200pgカラム(カタログ番号90-1002-73)を、GE Healthcare Life Sciencesから購入した。Amicon遠心フィルターユニット(カタログ番号UFC905024)は、EMD Milliporeから購入した。Slide-A-Lyzer(商標)G2透析カセット(カタログ番号87732)は、ThermoFisher Scientificから購入した。20nm金ナノ粒子(カタログ番号HD.GC20)は、BBI solutionsから購入した。吸収スペクトルを取得する際の反応容器として、Thermo Scientific(商標)Nunc(商標)Microwell(商標)96ウェルマイクロプレート(カタログ番号12-565-66)を使用した。
濃縮した(100g/L)モノクローナル抗体(mAb1、mAb2、mAb3、mAb4、mAb5、及びmAb6)を、Akta avant(GE Healthcare Life Sciences)を用い、10mM MES pH6.0、50mM塩化ナトリウムでXK26/100 Superdex 200pgカラムを通して、それぞれ精製した。単量体mAb画分を回収し、30,000MWCO HY膜搭載のVivaFlow 200と直列に連結したEasy-Load MAsterFlex L/S(Cole Parmer)ポンプを使用して濃縮した。150mLの各濃縮抗体を3つの画分に分割し、10,000MWCOの透析カセットに装填し、2Lの10mM MES pH6.0、250mM塩化ナトリウム;10mM MES pH6.0、50mM塩化ナトリウム;及び10mM MES pH6.0、10mM塩化ナトリウムと交換した。50,000MWCO搭載の遠心フィルターユニットを使用して、それぞれの対応する緩衝液中で最終容量約15mLまで溶液を濃縮した。SoloVPE(C Technologies,Inc.)を使用して濃度を測定した。10mM MES pH6.0、10mM塩化ナトリウム中では、抗体は公称60g/Lを上回って濃縮することはできなかった。他の2つの塩濃度(250mM及び50mM塩化ナトリウム)を公称80g/Lに調整した。3つの条件から得たアリコートを採取し、10g/Lに希釈した。80g/L、10g/Lの試料及び緩衝液をCG-MALS装置(Wyatt Technology)に加え、光散乱シグナルをmAb濃度の関数として測定した。
異なる塩条件下の各抗体(mAb1、mAb2、mAb3、mAb4、mAb5、及びmAb6)のサブアリコートを、別々の15mLファルコンチューブに加えた。光学密度1(1OD)の20nm金ナノ粒子溶液5mLを溶液に添加し、得られる最終タンパク質濃度を50μg/mLにした。30分待機した後、吸収スペクトル及びλ最大をSPECTRAmax 340PC(Molecular Devices)で記録した。
100mM緩衝溶液を適切なpHに調製した。Illustra NAPカラムを、2.4mLの100mM緩衝溶液(例えば、目標pHに応じてMESまたはリン酸ナトリウム)で平衡化した。濃縮された(50~75g/L)mAb原液を、平衡化した脱塩カラムを使用して緩衝液交換した。得られた濃度を、SoloVPEを使用して測定した。続いて各抗体を100mM緩衝液で5.12mg/mLに希釈した。次いで、mAbをマイクロウェルプレートの列1に添加し、100mM緩衝液で0.04mg/mLまで順次希釈した。80μLの金ナノ粒子を96マイクロウェルプレートの列2、3、及び4に添加した。列1の順次希釈を続行しながら、順次希釈したmAb40μLを、マルチチャネルピペットを使用して、金ナノ粒子を入れた各列に添加した。続いて、357mM、71mM、及び14mMの塩化ナトリウム塩原液280mLを、マルチチャネルピペットを使用してそれぞれ列2、3、及び4に添加した。得られた溶液を30分間平衡化した後、吸収スペクトルをSPECTRAmax 340PCで記録した。最大吸収強度は、450nmでの吸光度に対して正規化した。この比を抗体の最終濃度の関数としてプロットする。
1ミリリットルあたり約6.3×1011粒子の20nm金ナノ粒子を、2mM NaCl、20mM NaCl、または200mM NaCl、pH6の10mM MES緩衝液中で様々な濃度のヒト血清アルブミン(HAS)と結合させた。3.125μg/mL、6.25μg/mL、12.5μg/mL、25μg/mL、50μg/mL、100μg/mL、200μg/mL、及び400μg/mLを含んだ個々の試料に吸光度分光法を行った。これらのスペクトログラムをプロットし(図4、200mM NaClの試料に関するスペクトルを示す)、450nmでの初期吸光度に対する最大吸光度(λ最大)の吸収強度の比を各タンパク質濃度について算出し、プロットした(図5)。

製剤化条件によっては好ましくない動的コロイド相互作用プロファイルを有する高粘度の製剤化抗体(mAb7)を使用して、追加賦形剤が抗体のCG-SINSプロファイルを改善できるかをスクリーニングした。異なる緩衝液及びイオン強度(図13)、ならびに糖及び/またはアミノ酸賦形剤(図14)を含んだ様々な溶液で製剤化したモノクローナル抗体7(mAb7)のCG-SINSプロファイルは、高濃度での調剤に好ましい吸光度最大/吸光度0の閾値を下回ったままであった(例えば1.7未満)。約100mg/mLの製剤化したmAb7の粘度の実測は約100cPであった。
Claims (20)
- (a)タンパク質、ナノ粒子、及び緩衝塩を組み合わせて試料を作製する工程であって、前記緩衝塩は、緩衝液と塩、または緩衝能を有する塩である、工程;
(b)前記試料を光で励起する工程;
(c)前記試料を透過した光を450nm~750nmの範囲の複数の波長で測定する工程;
(d)前記試料の吸収強度比を算出する工程であって、前記吸収強度比は、450nmでの初期吸光度に対する最大吸光度(λ最大)の吸収強度の比である、工程;および
(e)前記吸収強度比が1.7を超える場合、前記タンパク質が高濃度でその天然構造の90%以上を保持すると決定する工程;
を含み、
異なるイオン強度を用いて工程(a)~(e)を少なくとももう一回繰り返す工程をさらに含む、
タンパク質が自己会合する可能性を決定するための方法。 - (i)前記タンパク質が、抗原結合タンパク質である、
(ii)前記タンパク質が、抗原結合タンパク質であり、前記抗原結合タンパク質が、抗体、抗体断片、または受容体-Fc融合タンパク質である、
(iii)前記タンパク質が、抗原結合タンパク質であり、前記抗原結合タンパク質が、ヒトモノクローナル抗体である、
(iv)前記タンパク質が、前記試料中に約2μg/mL~約512μg/mLの濃度で存在する、
(v)前記ナノ粒子が金ナノ粒子である、
(vi)前記ナノ粒子が、金ナノ粒子であり、前記金ナノ粒子が、直径約20nm~約100nmを有する、
(vii)前記ナノ粒子が、金ナノ粒子であり、前記金ナノ粒子の直径が約20nmである、
(viii)前記試料が、約5×1011~約8×1011ナノ粒子/mLを含む、
(ix)前記試料が、約6~6.5×1011ナノ粒子/mLを含む、
(x)前記緩衝塩の塩が、前記試料中に約2mM~約250mMの濃度で存在する、
(xi)前記緩衝塩の塩が、塩化ナトリウムである、
(xii)前記緩衝塩の塩濃度が、約2mM、約20mM、または約200mMである、
(xiii)前記透過光が、約450nm~約750nmの範囲の複数の波長で測定される、
(xiv)前記吸収強度比が、最大吸光度と初期吸光度の比である、または
(xv)前記吸収強度比の閾値が、約1.7である、
請求項1に記載の方法。 - (i)前記試料において異なる濃度のタンパク質を用いて工程(a)~(e)を繰り返す工程をさらに含む、
(ii)前記試料において異なる濃度の塩を用いて工程(a)~(e)を繰り返す工程をさらに含む、および/または
(iii)異なるpHの前記試料を用いて工程(a)~(e)を繰り返す工程をさらに含む、
請求項1または2に記載の方法。 - (f)高濃度の高可溶性タンパク質を賦形剤と組み合わせて、製剤化した原薬を作製する工程
をさらに含む、請求項1~3のいずれか1項に記載の方法。 - 前記タンパク質の濃度が約50mg/mL~約500mg/mLである、請求項4に記載の方法。
- (i)前記賦形剤が、等張化剤、緩衝液、界面活性剤、安定剤、及びそれらの組み合わせからなる群から選択される、
(ii)前記賦形剤が、等張化剤であり、前記等張化剤が、塩である、または

- (a)請求項1~9のいずれか1項に記載の方法に従って、タンパク質が自己会合する可能性を決定する工程;
(b)粘度降下賦形剤を前記タンパク質と組み合わせる工程;
(c)前記粘度降下賦形剤の存在下で、請求項1~9のいずれか1項に記載の方法に従って、前記タンパク質が自己会合する可能性を決定する工程;及び
(d)前記粘度降下賦形剤を含まない場合よりも前記タンパク質溶液の粘度を少なくとも50%低下させるレベルで前記粘度降下賦形剤を加えて前記タンパク質を製剤化する工程
を含む、タンパク質を含む低粘度医薬製剤を製造する方法。 - 前記粘度降下賦形剤がパラアミノ安息香酸(PABA)である、請求項18に記載の方法。
