CD47(Cluster of Differentiation 47)は、1980年代にヒト卵巣癌で腫瘍抗原
として最初に同定された。それから、CD47は、急性骨髄性白血病(AML)、慢性骨
髄性白血病、急性リンパ性白血病(ALL)、非ホジキンリンパ腫(NHL)、多発性骨

マットは、1本鎖可変断片(VH+リンカー+VL)である。ライブラリーサイズは1.
1×1010であり、配列多様性を以下のようにして分析した。ライブラリーから選択し
、さらに配列を決定した(sequenced)62クローンについて、16配列がトランケーショ
ン(truncation)、フレームシフトまたはアンバーコドン(amber codon)を有し;46配列
が全HCDR3配列が固有である全長scFvを有する。46の全長scFvにおいて、
13配列がラムダ軽鎖を有し、33配列がカッパ軽鎖を有する。
ヒトCD47-IgVドメインに特異的に結合するファージクローンを得るために、2
つのファージパンニング方法を使用した。
パンニング
本方法において、上記したのと同様にして開発したファージライブラリーを、まず、カ
ゼイン被覆イムノチューブ中で2時間インキュベートした。ヒトCD47-IgV-Fc
融合タンパク質を第1のパンニングラウンドで使用した。PBSTで5~20回洗浄する
ことによって、非結合ファージを除去した。結合ファージを新たに調製した100mMト
リエチルアミン溶液で溶出させ、Tris-HClバッファーを添加することによって中
和して、第1のアウトプットファージプールとした。この第1のアウトプットファージプ
ールを、増幅を目的として大腸菌TG-1(E. Coli TG-1)細胞に感染させることにより回
収した(rescue)後、抗原としてビオチン化ヒトCD47-IgVを用いた第2のパンニン
グラウンドを行った。結合ファージを同様の方法で溶出させ、第2のアウトプットファー
ジプールとし、さらにこれを回収した後、抗原としてヒトCD47-IgV-Fc融合タ
ンパク質を用いた第3のパンニングラウンドを行った。さらに、結合ファージを第3のア
ウトプットファージプールとし、ビオチン化ヒトCD47-IgVを用いた第4のパンニ
ングラウンドを行った。
本第2の方法において、ファージライブラリーを、まず、カゼインブロック(casein-bl
ocked)100μL ストレプトアビジン-磁気ビーズ(streptavdin-magnetic bead)中で
インキュベートして、ストレプトアビジンビーズバインダーを消耗させた。ストレプトア
ビジン-磁気ビーズ及びAG0084-huIgG1/kをネガティブ消耗(negative de
pletion)に使用した。消耗したライブラリーを回収した後、抗原としてビオチン化ヒトC
D47-IgVを用いた第2のパンニングラウンドを行い、さらに、カゼインブロックス
トレプトアビジン-磁気ビーズを用いてネガティブ消耗を行った。PBSTで5~20回
洗浄することによって、非結合ファージを除去した。結合ファージを新たに調製した10
0mMトリエチルアミン溶液で溶出させ、Tris-HClバッファーを添加することに
よって中和した後、回収し、さらにヒトCD47-IgV-Fc融合タンパク質を用いた
第3のパンニングラウンドを行い、AG0084-huIgG1/kを用いて消耗させた
。さらに、結合ファージを第3のアウトプットファージプールとし、ビオチン化ヒトCD
47-IgVを用いた第4のパンニングラウンドおよびカゼインブロックストレプトアビ
ジン-磁気ビーズを用いたネガティブ消耗を行った。

A(Phage single point ELISA)(SPE)のための高力価ファージ(HT)と作製した。
フィルターリフトから選択した陽性シングルクローンをヒトCD47-IgV-Fc融合
タンパク質およびビオチン化ヒトCD47-IgVドメインタンパク質に結合させた。ま
た、これらの陽性シングルクローンのVHおよびVL遺伝子について配列を決定した。特
有のVH及びVL遺伝子を有するすべての陽性ヒットを発現ベクターpFUSE2ss-

ロセスにより、本発明の抗体分子が臨床用途に全体的に最良の特性で構築できる。
によるスクリーニング
組換ヒトCD47-Fc融合タンパク質(Acrobiosystems)を、室温(RT)で2時間、
マイクロタイタープレートにリン酸緩衝食塩水(PBS)における2μg/mLで被覆し
た。抗原を被覆した後、ウェルを、室温(RT)で1時間、1% BSAを含むPBS/
0.05% Tween(PBST)でブロックした。ウェルをPBSTで洗浄した後、
単一のクローンから精製したファージをウェルに加えて、RTで1時間、インキュベート
した。結合ファージクローンを検出するために、M13に対するHRP標識2次抗体(HRP
conjugated secondary antibodies)(Jackson Immuno Research)を添加した後、蛍光発生
基質(Roche)を添加した。すべてのインキュベーション工程間で、プレートのウェルをP
BSTで3回洗浄した。蛍光をTECAN Spectrafluorプレートリーダー

における1% BSAでブロックした後、1μg/mLのSIRPα-Hisタンパク質
を、RTで1時間、CD47抗体(10μg/mL)の不存在下でまたは存在下で添加し
た。次に、プレートを3回洗浄し、RTで1時間、HRP標識抗His2次抗体(HRP-con
jugated anti-His secondary antibody)と共にインキュベートした。洗浄後、TMB溶液
を30分間各ウェルに加え、反応を2.0M H2SO4で停止し、ODを490nmで
測定した。

凝集係数(agglutination index)」で表わされる、赤血球凝集アッセイの滴定結果を示す

AML患者(AML-PB003F)の初期PBMC(Primary PBMC)を1μM CFSEで10分間
標識した後、5:1の腫瘍細胞:食細胞の割合でMDMに添加し、所定のCD47抗体を
様々な濃度で添加した。3時間インキュベート後、取り込まれない(non-phagocytosed)標
的細胞をPBSで洗い流し、残った食細胞をかきおとし、CD47抗体で染色し、フロー
サイトメトリーによって分析した。食作用は、CD14+細胞でゲーティングし(gating)
、さらにCFSE+細胞の割合(%)を評価することによって測定した。食作用を前記と
同様にして測定した。
抗体よりかなり高く、本発明の試験CD47抗体はすべて有意なAML結合能(75%を
超える)および食作用能(phagocytosis capabilities)(少なくとも36%)を示した。
ンビボ有効性
NSGマウスに、尾静脈注射により100万細胞/マウスの濃度でRaji Luc-
EGFPを移植した。マウスをインビボで撮像し、移植してから5日目に移植レベルを測
定した。CD47抗体(即ち、1F8、5F9、及び2A1)の処置を、10mg/kg
の用量で同日に開始した。すべてのマウスに腹腔内注射により1日おきに注射した。マウ
スを、IVIS Lumina IIIイメージングシステム(IVIS Lumina III imaging
system)により以下の時点でインビボで撮像した:抗体処置の0日目、処置後2日目、処
置後6日目、及び処置後9日目。インビボライブイメージングシステム(in vivo live im
aging system)により生物発光輝度(bioluminescent radiance)を分析することによって、
マウスの腫瘍成長を測定した。

54個のPDXサンプル(7種のヒト癌タイプ)を、免疫-組織学染色によってCD4
7の発現について分析した。様々なPDXサンプルにおけるCD47染色のレベルを形状
(geometry)および染色強度によって採点した(score)。図16a、図16bおよび図16
cから、CD47抗体による処置後に様々な発現レベルのCD47が示される。
投薬を受けていない(Naive)カニクイザル(cyno monkey)の静脈内にベヒクル(n=2)
、1F8(n=3、15mg/kg)および5F9(n=3、15mg/kg)を注入し
た。血液採取してから24時間以内に、注射前に2回ならびに抗体投与してから3、6、
10、14及び21日目に、血液学(CBC)を分析した。赤血球数(Erythrocyte count
)(RBC)、ヘモグロビン(HGB)、絶対網状赤血球数(Absolute Reticulocytes Cou
nts)及び血小板数を含むCBCパラメーターを試験した。結果を図17a~17dに示す
が、これから、1F8処置はカニクイザルの血液パラメーターに影響を及ぼさなかったこ
とが示される。

用いて検出した。カニクイザルの薬物動態パラメーターをWinolinによって分析し
、図17e及び下記表に示す。
投薬を受けていない(Naive)カニクイザル(cyno monkey)の静脈内に抗体13H3(20
mg/kg)を単回投与および繰り返し投与(毎週投与)で注入した。抗体投与してから
所定の時期に、赤血球数(Erythrocyte count)(RBC)、ヘモグロビン(HGB)、血
小板数及びリンパ球数を含む血液学(CBC)パラメーターを試験した。

CD47抗体のエピトープビニングを、競合ELISAによって評価した。CD47
ECDタンパク質及び第1抗CD47抗体を予めインキュベートして、ストレプトアビジ
ン-HRP抗体によって検出されるビオチン化第2抗CD47抗体に添加した。第1抗C
D47抗体が第2抗体へのCD47 ECDの結合に対して競合した場合には、双方の抗
体を同じまたは重複するエピトープビン(epitope bins)に入れた。競合しない場合には、
非重複エピトープビン(non-overlapping epitope bin)に入れた。図18a及び図18b
に示される結果から、本発明のCD47抗体である1F8は参考抗体5F9及び2A1と
は異なるエピトープを有することが示される。

のコンプレックス構造とは異なり、CD47-1F8 Fabのコンプレックス構造はよ
りまっすぐのヘッドツゥヘッド配向(straighter head to head orientation)をとる。残
基L3、V25、T26、N27、M28、E29、A30、Q31、T34、E35、
Y37、A53、L54、L74、K75、G76、T99、E100、L101、T1
02及びR103を含み、このうちL3、N27、E29、Q31、T34、E35、Y
37、A53、T99、E100、L101、T102及びR103はSIRPαとの相
互作用にかかわりのあるCD47の1F8エピトープは、不連続でかつ広範囲であり、1
F8のアンタゴニスト特性を説明する。また、このコンプレックス構造から、1F8のV
HドメインがCD47への8個の水素結合及び4個の塩架橋(salt bridge)を形成し、1
F8のVLドメインがCD47への8個の水素結合を形成することが明らかである。

のの、1F8抗体はほとんどが標的の異なるエピトープに結合する。CD47の1F8エ
ピトープは、立体配座的に不連続であり、CD47のTNMEAQループ(残基26~3
1)、T34、E35、L74、およびLTRヒンジ(残基101~103)を有する。
多くの水素結合相互作用が抗体残基の側鎖とCD47の主鎖の酸素原子との間に形成され
る。また、塩架橋は、1F8のR103とCD47のE35との間に形成される。また、
適切な配向を保持するのに重要であるいくつかのファン・デル・ワールス接触(Van der W
aals contacts)が観察される。抗体1F8のVHドメインがCD47のT34、E35お
よびLTR ヒンジ(残基101~103)への結合に主に関与する一方、VKドメイン
がTNMEAQループ(残基26~31)及びL74と相互作用する。CD47のこれら
のエピトープは、5F9抗体及びSIRPαのものとは異なる。構造分析から、1F8抗
体の2つの長いループ(残基26~38および52~59)がほとんど垂直な配向でCD
47に結合するのを助け、これにより付近の細胞のCD47が抗体によって架橋されない
ように抗体を分離し、これによりほとんどの血液細胞の血球凝集を防止することが示唆さ
れる。
構造の重ね合わせから、CD47の結合配向がこれらの2つのコンプレックスで非常に異
なることが明らかである。双方の抗体は共にhead-to-head結合配向を有する
ものの、CD47は約180°水平に回転する。1F8/CD47コンプレックスの構造
は軽鎖ループ残基61~64付近にCD47 N-末端ピログルタメートを有するが、5
F9はW52、N32及びW101の3つの重鎖ループの中にCD47 N末端を有する
。抗体1F8では、重鎖残基Trp33及びArg103が、ファン・デル・ワールス接
触およびそれぞれ、CD47のLeu101及びGlu35との塩架橋を形成する。同じ
位置で、抗体5F9の残基Tyr101はファン・デル・ワールス接触を介してCD47
のN-末端方向に向き、Arg102はCD47のGlu104と水素結合を形成する。
抗体1F8のループ残基Asn31、Trp33、ならびにヒンジ残基Arg53及びA
sp56は、ドメイン間水素結合ネットワークを形成し、さらにAsn31及びArg5
3はCD47のLeu101及びThr34の主鎖と水素結合を形成する。同じ界面で、
5F9は、残基Tyr52がCD47のLeu3とのファン・デル・ワールス接触を形成
する以外は、相互作用を形成しないように見える。ヒンジ(残基52~56)は1F8(
残基52~59)より3残基短い。軽鎖では、Fab 1F8及び5F9は双方とも、ル
ープ(1F8ではV29-Y38および5F9ではV152-Y158)のCD47との
いくつかの重要な水素結合相互作用を有する。1F8の残基Y97及びY98はループ(
残基26~38)を押しのけ(“push” away)、後者は1F8とCD47との間に、即ち
、1F8のArg34とCD47のLeu74の主鎖との間に、および1F8のArg3
6とCD47のThr26の主鎖との間に、2個の水素結合を形成する。しかしながら、
5F9の残基Gly218及びSer219(1F8のTyr97及びTyr98に相当
する)により、5F9のループ(残基149~158)がCD47と3つの水素結合(A
sn157-Lys39、Tyr159-Glu104及びLys177-Thr99で
)を形成する。また、重鎖も同様にして、5F9のループ(残基149~158)が1F
8(残基26~38)より3残基短い。1F8のこれらの関連するより長いループは、C
D47の結合配向に主として寄与する。
抗ヒトCD47抗体を得るために、BALB/C、C57/BL6またはSJLマウス
を含む6~8週齢の異なる種を組換ヒトCD47細胞外ドメインタンパク質で数ラウンド
免疫処置した。免疫処置後、十分な力価の抗CD47 IgGを有するマウスを、同じ抗
原で追加免疫した後、融合した(fusion)。ハイブリドーマ上清について、ELISAスク
リーニングによってヒトCD47 ECDタンパク質との直接結合およびCD47へのS
IRPα結合の競合を試験した。一連のスクリーニングアッセイにより、34C5が上記
したアッセイに従うヒト化およびさらにインビトロでの特性化(characterization)用に選
択された。
