Description
かかる問題を解決するため、生体関連物質を、糖鎖、ポリエチレングリコールなどの親水性高分子、アルブミンなどによって化学修飾する試みが行われている。その結果、分子量の増大や水和層の形成などにより、生体関連物質の血中半減期を延長することが可能となる。
また、ポリエチレングリコールで修飾することで、生体関連物質の毒性や抗原性の低下、難水溶性薬物の溶解性向上などの効果が得られることも良く知られている。
ポリエチレングリコールは、生体関連物質の血中半減期を延長させ得る優れた素材であり、ポリエチレングリコールで修飾した生体関連物質の研究が多数行なわれ、それらの血中半減期を有意に延長できるという結果が得られている。
しかしながら、さらに血中半減期を延長できる新たな素材が求められている。
非特許文献1では、酵素であるペルオキシダーゼに負電荷を付与したところ、腎臓からの排出が有意に抑制されたことが報告されている。また非特許文献2では、負電荷を付与したデキストランポリマーにおいて、腎臓からの排出が抑制されたことが確認されている。
ングリコール類とポリグルタミン酸とのブロックポリマーに、抗癌剤を結合させた水溶性高分子抗癌剤が記載されている。特許文献3には、ポリエチレングリコールとポリアスパラギン酸またはポリグルタミン酸とのブロックポリマーからなる薬物の担体が記載されている。
上記特許文献に記載されたブロックポリマーは、総じて自己会合により高分子ミセルを形成し、ミセル内部に取り込まれた薬物の送達に用いられる。ブロックポリマーのポリアミノ酸部分の側鎖に存在するカルボキシル基は、薬物を共有結合させるのに利用され、高分子ミセルが形成された際には、ミセルの内部に取り込まれることになる。それゆえ、これらポリアミノ酸部分におけるカルボキシル基が負電荷を有するとしても、腎臓からの排出は抑制されない。また、これらブロックポリマーは、ポリアミノ酸部分のカルボキシル基以外に官能基を有しないことから、生体関連物質を修飾することができない。さらに、前記ブロックポリマーは、水溶液中ではミセルを形成してしまうため、生体関連物質の修飾には適していない。

上記の修飾された生体関連物質は、ブロックポリマーを構成するポリエチレングリコールによる分子サイズの増大効果と、アニオン性のポリアミノ酸による負電荷の付与効果により、血中半減期が延長され、生体の血中での安定性が向上される。
本発明は、アニオン性ブロックポリマーと、生体関連物質との結合体(以下、本明細書にて「本発明の結合体」ともいう)を提供する。
本発明の結合体は、ポリエチレングリコールとポリアミノ酸とが直接またはリンカーを介して結合されたブロックポリマー誘導体と、生体関連物質とが結合されて形成され、下記の式(1)で示される。
式(1)中、nはアミノ酸の重合度を示し、通常2~100であり、好ましくは5~50であり、さらに好ましくは5~30である。
式(1)中のエチレングリコールの重合度と、アミノ酸の重合度との比(m/n)は、通常1~500であり、好ましくは2~240であり、さらに好ましくは5~200である。
m/n値が1よりも小さい、つまりエチレングリコールの重合度がアミノ酸の重合度よりも小さいと、ポリエチレングリコールによる分子サイズの増大効果が十分に得られない可能性がある。また、m/n値が500よりも大きい、つまりアミノ酸の重合度が小さい場合、アニオン性のポリアミノ酸による負電荷の付与効果が十分に得られない可能性がある。
ポリアミノ酸部を構成するアミノ酸としては、グルタミン酸(式(1)中、x=2)がより好ましい。
また、ポリアミノ酸部を構成するアミノ酸としては、L体、D体、DL体のいずれでもよいが、より好ましくはL体である。
アシル基としては、炭素数2~4の飽和アシル基が好ましく、具体的な例としてはアセチル基、プロピオニル基などが挙げられ、好ましくはアセチル基である。
従って、式(1)中、Qは、2個または3個の活性水素を有する化合物の残基を示す。
ここで、「活性水素」とは、有機化合物の分子内水素のうち、反応性の高い水素をいい、たとえば、水酸基、カルボキシル基、アミノ基、チオール基等の水素が挙げられる。なお、本発明においては、第1級アミノ基の場合は、活性水素は1個と数える。
本発明において、2個または3個の活性水素を有する化合物としては、好ましくはエチレングリコール、グリセリン、リシン、アスパラギン酸、グルタミン酸等が挙げられ、より好ましくはエチレングリコールおよびグリセリンが挙げられる。
それゆえ、「2個の活性水素を有する化合物の残基」としては、エチレングリコールなどの残基が挙げられ、「3個の活性水素を有する化合物の残基」としては、グリセリン、リシン、アスパラギン酸、グルタミン酸などの残基が挙げられる。
なお、酵素などの生体関連物質においては、複数のポリマー誘導体を結合させても活性が低下しないことが知られている。
より具体的には、サイトカインとしては、免疫を調節するインターフェロンタイプI、タイプII、タイプIII、インターロイキン、リンフォカイン;造血因子であるエリスロポエチン、顆粒球コロニー刺激因子(GCSF);細胞増殖因子である上皮成長因子(EGF)、線維芽細胞成長因子(FGF)、血小板由来成長因子(PDGF)、肝細胞成長因子(HGF)、トランスフォーミング成長因子(TGF);細胞傷害因子である腫瘍壊死因子(TNF-α)、リンフォトキシン(TNF-β);アディポカイン;神経栄養
因子である神経成長因子(NGF);それらの受容体アンタゴニストなどが挙げられる。
成長因子としては、インスリン様成長因子(IGF)、トランスフォーミング成長因子(TGF)、トロンボポエチン、骨形成タンパク質(BMP)などが挙げられる。
血液凝固因子としては、フィブリノーゲン、フィブリン、血液凝固第V因子、第VII因子、第VIII因子、第IX因子、第X因子、第XII因子などが挙げられる。
ホルモンとしては、カルシトニン、インスリン、そのアナログ、グルカゴン様ペプチド(GLP-1)、エクセナチド(GLP-1受容体アゴニスト)、ソマトスタチン、ヒト成長ホルモンなどが挙げられる。
抗体としては、完全長抗体、また抗体フラグメントとして、FabやsvFV、ナノボディなどが挙げられる。
酵素としては、アスパラギナーゼ、スーパーオキシドディスムターゼ、尿酸オキシダーゼなどが挙げられる。
これらタンパク質は、血中での安定性が低く、本発明の結合体として修飾することにより、血中半減期の延長が期待される。
本発明において、タンパク質としては、好ましくはインターフェロン、インターロイキン、エリスロポエチン、GCSF、血液凝固第VIII因子、血液凝固第IX因子、ヒト成長ホルモン、抗体フラグメントが用いられ、より好ましくは、ヒト成長ホルモン、インターフェロン、GCSF、エリスロポエチン、抗体フラグメント(特にFab)が用いられ、さらに好ましくは、ヒト成長ホルモン、GCSFが用いられる。
本発明において、ペプチドとしては、好ましくはインスリン、ビバリルジン、テリパラチド、エクセナチド、エンフビルチド、デガレリクス、ミファムルチド、ネシリチド、ゴセレリン、グラチラマー、オクトレオチド、ランレオチド、イカチバント、ジコチニド、プラムリンチド、ロミプロスチム、カルシトニン、オキシトシン、リュープロレリン、グルカゴンが用いられ、より好ましくは、インスリン、エクセナチド、カルシトニンが用いられる。
また、核酸は1~3本鎖のいずれも用いることができるが、好ましくは1本鎖又は2本鎖である。
さらに、本発明において用いられる核酸は、他のタイプのヌクレオチド(たとえば、プリンまたはピリミジン塩基と、リボース、デオキシリボース以外の糖とのN-グリコシド等)、あるいは非ヌクレオチド骨格を有するオリゴマー(たとえば、市販のペプチド核酸(PNA)等)などであってもよい。さらにまた、該核酸は、たとえば、公知の修飾の付加された核酸、当該分野で知られた標識のされた核酸、キャップの付加された核酸、メチル化された核酸、1個以上の天然ヌクレオチドを類縁物で置換した核酸等であってもよい。
より好適な核酸としては、血中で作用するアプタマーが挙げられる。
かかる結合体は、下記式(1’)で表され、上述したように、ブロックポリマー誘導体分子の結合数の増大により、活性が低下する可能性のある生体関連物質の修飾体として、好ましい。
本発明の中間体は、下記式(2)で示されるブロックポリマー誘導体である。

従って、本発明は、生体関連物質の修飾方法(以下、本明細書にて「本発明の修飾方法」ともいう)をも提供する。
本発明の修飾方法において、本発明の中間体は、式(2)中、Xで示される官能基を介して生体関連物質と結合される。それゆえ、生体関連物質に結合したブロックポリマー誘導体のポリアミノ酸部分の側鎖に存在するカルボキシル基は遊離の状態で存在するため、ポリアミノ酸部分による負電荷を生体関連物質に十分に付与することができる。
生体関連物質の有する官能基と、本発明の中間体におけるXで示される官能基との反応は、後述する通り、一般的な反応条件および方法にて、たとえば縮合剤や還元剤を加えて反応させる等により、行うことができる。
ここで「保護基」とは、ある反応条件下で分子中の特定の化学反応可能な官能基の反応を防止または阻止する基である。保護基は、保護される化学反応可能な官能基の種類、使用される条件および分子中の他の官能基もしくは保護基の存在により変化する。保護基の具体的な例は多くの一般的な成書に見出すことができるが、例えば「Wuts, P. G. M.; Greene, T. W. Protective Groups in Organic Synthesis, 4th ed.; Wiley-Interscience: New York, 2007」に記載されている。
また、保護基で保護された官能基は、それぞれの保護基に適した反応条件にて脱保護、すなわち化学反応させることで、元の官能基を再生させることができる。保護基の代表的な脱保護条件は前述の文献に記載されている。

得られたブロックポリマー誘導体(23)と、GCSFとの結合体(28)の同定は、上述したようにMALDI-TOF-MSにより分子量を測定し、また、下記に示す通り、SDS-PAGE、円偏光二色性測定およびサイズ排除クロマトグラフィーによる分析にて行い、目的物が得られたことを確認した。
測定結果を図4に示した。図4より、MALDI-TOF-MSにより測定される分子量は24,438Daであり、結合体(28)の生成が示唆された。
SDS-PAGEキットとして「4-15%ミニプロティアン(登録商標)TGX(商標)プレキャストゲル(バイオラッド(BIO-RAD)社」を用い、キットの推奨測定条件に従い、結合体(28)の評価を行った。
染色液としては、クマシーブリリアントブルー溶液(CBB溶液)およびヨウ素染色溶
液(塩化バリウム溶液+ヨウ素溶液)を用いた。
SDS-PAGEの結果を図5に示す。図5中、レーンMは分子量マーカーを、レーン1はGCSFを、レーン2は結合体(28)を示す。左図はクマシーブリリアントブルー(CBB)染色によるタンパク質の検出結果を示し、右図は、ヨウ素染色によるPEGの検出結果を示す。
結合体(28)の分析結果(レーン2)においては、タンパク質やペプチドを選択的に染色するCBB染色でバンドが検出され、さらに、ポリエチレングリコールを染色するヨウ素染色でもバンドが検出された。両方の染色でバンドが検出されたことから、ブロックポリマー誘導体(23)とGCSFとが結合した目的物が得られたことが確認された。
円偏光二色性の測定は、タンパク質の変性状態を調べるのに有効な手段として知られている。下記の装置を用い、下記波長にて、円偏光二色性を測定した。

測定により得られたGCSFのスペクトルと、結合体(28)のスペクトルとを重ね合わせたところ、形状が一致していたことから、ブロックポリマー誘導体(23)による修飾により、GCSFが変性していないことが確認された。

GCSFおよび結合体(28)について得られたクロマトグラムを比較したところ、結合体(28)のピークが、GCSFのピークと比べて、高分子量側にシフトしていること、つまりブロックポリマー誘導体(23)がGCSFに結合し、分子量が増大したことが確認された。
原料としてブロックポリマー誘導体(24)を使用し、陽イオン交換クロマトグラフィーによる精製の際のグラジエント条件を表3に示す条件とした他は、実施例9(1)と同様に、ブロックポリマー誘導体(24)とGCSFとの結合体(29)を合成した。
原料としてブロックポリマー誘導体(25)を使用し、陽イオン交換クロマトグラフィーによる精製の際のグラジエント条件を表4に示す条件とした他は、実施例9(1)と同様にして、ブロックポリマー誘導体(25)とGCSFとの結合体(30)を合成した。
原料としてブロックポリマー誘導体(26)を使用し、実施例9(1)と同様に、ブロックポリマー誘導体(26)とGCSFとの結合体(31)を合成した。RP-HPLCを用いて分析を行い、図6に示すように、反応が進行したことを確認した。
次いで、陽イオン交換クロマトグラフィーを用い、下記精製条件にて精製した。

原料としてブロックポリマー誘導体(27)を使用し、実施例9(4)と同様に、ブロックポリマー誘導体(27)とGCSFとの結合体(32)を合成した。
得られたブロックポリマー誘導体(27)とGCSFとの結合体(32)の同定は、実施例9(1)の場合と同様に、MALDI-TOF-MSによる分子量の測定、SDS-PAGE、円偏光二色性測定、サイズ排除クロマトグラフィーによる分析にて行い、目的物が得られたことを確認した。
GCSFを3mg含有する10mMリン酸ナトリウム緩衝液(pH=8.0、5(w/v)%ソルビトール含有)に、実施例1で得たアミノ基を有するブロックポリマー誘導体(19)を3当量添加し、GCSF濃度を2.0mg/mLに調整した。次いで、微生物由来のトランスグルタミナーゼ(TGage、「Activa(登録商標)WM」(味の素株式会社製))を0.06mg(2重量%/GCSF)加え、25℃にて4時間撹拌した。さらに、N-エチルマレイミドをTGageに対して0.8当量添加し、25℃にて1時間撹拌することで反応を停止した。
RP-HPLCを用いて分析を行い、図8に示すように、反応が進行したことを確認した。RP-HPLCの分析条件は、上記実施例9(1)における分析条件と同じである。
RP-HPLC分析の結果、反応4時間後の反応率はおよそ84%であった。

原料としてブロックポリマー誘導体(20)を使用し、陽イオン交換クロマトグラフィーによる精製の際のグラジエント条件を表7に示す条件とした他は、実施例10(1)と同様に、ブロックポリマー誘導体(20)とGCSFとの結合体(34)を合成した。
原料としてブロックポリマー誘導体(21)を使用し、陽イオン交換クロマトグラフィーによる精製の際のグラジエント条件を表8に示す条件とした他は、上記(1)と同様に、ブロックポリマー誘導体(21)とGCSFとの結合体(35)を合成した。
GCSFを3mg含有する10mM酢酸ナトリウム緩衝液(pH=4.6、5(w/v)%ソルビトール含有)にメトキシPEGアルデヒド(日油株式会社製「SUNBRIGHT ME-200AL」、数平均分子量=約20kDa)を3当量添加し、GCSF濃度を3.0mg/m
Lに調整し、4℃にて1時間撹拌した。次いで、還元剤であるシアノ水素化ホウ素ナトリウム100当量を添加し、4℃にて24時間撹拌した。さらに、グリシルグリシンを100当量添加し、4℃にて1時間撹拌することで反応を停止した。RP-HPLCを用いて分析を行い、反応が進行したことを確認した。RP-HPLCの分析条件は、実施例9(1)における分析条件と同じである。
RP-HPLC分析の結果、反応24時間後の反応率はおよそ78%であった。

グラジエント条件:表9に示す通り。
=4.6、5(w/v)%ソルビトール含有))。得られた溶液のGCSF濃度は、280nmにおける吸光度を測定することで算出した。
得られたメトキシPEG-GCSF結合体(36)の同定は、実施例9(1)の場合と同様に、MALDI-TOF-MSによる分子量の測定(図9)、SDS-PAGE、円偏光二色性測定、サイズ排除クロマトグラフィーによる分析にて行い、目的物が得られたことを確認した。
GCSF、実施例9(4)で得たブロックポリマー誘導体(26)とGCSFとの結合体(31)、および比較例1で得たメトキシPEG-GCSF結合体(36)を使用し、Sprague-Dawleyラット(体重200g~270g)を用いて、薬物動態試験を行った(3匹/グループ)。
GCSFサンプルは、200μLのリン酸緩衝生理食塩水(PBS)(pH=7.4)を用いて調製した後、0.22μmのフィルターにてろ過した。調製したサンプル溶液を尾静脈注射(GCSFとして100μg/kg)した後、投与前(t=0)および投与後の任意の時間に血液サンプルを採取した。動物実験の全ての作業はイソフルランによる麻酔下で行った。
血液サンプルを遠心分離(1500×g、20分)し、得られた血清に含まれるGCSF量をELISAキット(Invitrogen社製)により測定した。結果はprogram 2.0 PK solverシステムにより、2コンパートメントモデルを用いて
評価し、血中濃度曲線下面積(AUC)を比較した(表10、図10)。
また、表10および図10に示されるように、比較例1で得たメトキシPEG-GCSF結合体(36)と比べても、有意に血中滞留性を改善できることが確認できた。
ブロックポリマー誘導体(26)とGCSFとの結合体(31)と、メトキシPEG-GCSF結合体(36)のPEG部分は、同分子量(20kDa)であることから、ブロックポリマー誘導体(26)とGCSFとの結合体(31)にて観察される血中滞留性の向上は、PEG鎖の末端にポリアニオンであるポリグルタミン酸(平均重合度=9)を導入したことに起因するものであることが示唆された。
hGH(アミノ酸配列:下記の通り)(シェナンドー(Shenandoah)社)を2mg含有する100mM酢酸ナトリウム緩衝液(pH=5.5)に実施例4で得たアルデヒド基を有するブロックポリマー誘導体(26)を5当量添加し、hGH濃度を1.0mg/mLに調整し、4℃にて1時間撹拌した。次いで、還元剤であるシアノ水素化ホウ素ナトリウム50当量を添加し、25℃にて24時間撹拌した。さらに、グリシルグリシンを100当量添加し、25℃にて1時間撹拌することで反応を停止した。逆相液体クロマトグラフィー(RP-HPLC)を用いて、下記に示す条件にて分析を行い、図11に示すように、反応が進行したことを確認した。

e社製(分画分子量=3.5kDa)、外液:10mM酢酸ナトリウム水溶液(pH=4
.7))、その後、陽イオン交換クロマトグラフィーにより、下記精製条件にて精製し、図12に示す結合体(37)の画分を分取した。

得られたブロックポリマー誘導体(26)とhGHとの結合体(37)の同定は、実施例9(1)の場合と同様に、MALDI-TOF-MSによる分子量の測定、SDS-PAGE、円偏光二色性測定、サイズ排除クロマトグラフィーによる分析にて行い、目的物が得られたことを確認した。
なお、SDS-PAGEによる分析結果を図13に示した。図13中、レーンMは分子量マーカーを示し、レーン1はブロックポリマー誘導体(26)とhGHとの結合体(37)を示し、レーン2はhGHを示す。
また、左図はクマシーブリリアントブルー染色によるタンパク質の検出結果を示し、右図はヨウ素染色によるPEGの検出結果を示す。
図13において、ブロックポリマー誘導体(26)とhGHとの結合体(37)については、タンパク質とPEGの双方のバンドが検出された(レーン1)。
原料としてブロックポリマー誘導体(27)を使用した他は、実施例11(1)と同様にして、ブロックポリマー誘導体(27)とhGHとの結合体(38)を合成した。
得られたブロックポリマー誘導体(27)とhGHとの結合体(38)の同定は、実施例11(1)の場合と同様に、MALDI-TOF-MSによる分子量測定、SDS-PAGE、円偏光二色性測定、サイズ排除クロマトグラフィーによる分析にて行い、目的物が得られたことを確認した。
原料として、メトキシPEGアルデヒド(日油株式会社製「SUNBRIGHT ME-200AL」、数平均分子量約=20kDa) を使用した他は、実施例11(1)と同様にして、メトキシPE
G-hGH結合体(39)を合成した。RP-HPLC分析の結果、反応24時間後の反
応率はおよそ77%であった(図14)。
得られたメトキシPEG-hGH結合体(39)の同定は、実施例11(1)の場合と同様に、MALDI-TOF-MSによる分子量の測定、SDS-PAGE、円偏光二色性測定、サイズ排除クロマトグラフィーによる分析にて行い、目的物が得られたことを確認した。
下垂体を摘出したOVAラット(オス、体重=約90g)はCharles Rive
r Laboratories (Lecco, Italy)より購入した。ラットを無作為に6グループ(4匹/グループ)に分け、実験前に動物施設にて2週間飼育した。
PBSコントロールグループには、PBS溶液200μLを尾静脈より投与した。hGH投与グループには、ネイティブのhGHを6日間尾静脈より投与した(0.3mg/kg/日、約27μg/ラット/日、合計約162μg/ラット)。実施例11(1)で得たブロックポリマー誘導体(26)とhGHとの結合体(37)を投与するグループ、実施例11(2)で得たブロックポリマー誘導体(27)とhGHとの結合体(38)を投与するグループ、および比較例2で得たメトキシPEG-hGH結合体(39)を投与するグループには、各結合体をそれぞれPBSに溶解し、単回で尾静脈より投与した(1.8mg/kg、約162μg/ラット)。ラットは11日間、同時刻にモニターおよび体重測定を行い、結果を図15に示した。また、各ラットの頚骨長を11日間測定し、ラットの初期の体重で補正した頚骨長の増加量を図16に示した。
Claims (10)
- 式(1)において、yが1である、請求項1に記載の結合体。
- 式(1)において、mが40~1200であり、nが5~50である、請求項1または2に記載の結合体。
- 式(1)において、Qが、エチレングリコール、グリセリン、リシン、アスパラギン酸およびグルタミン酸からなる群より選択される化合物の残基である、請求項1~3のいずれか1項に記載の結合体。
- 式(1)において、L1、L2、L3が、それぞれ独立して、単結合、フェニレン基、アミド結合、エーテル結合、チオエーテル結合、ウレタン結合、第2級アミノ基、カルボニル基、ウレア結合、トリアジン基、チオールの付加されたマレイミド基、オキシム結合、ならびに、単結合、フェニレン基、アミド結合、エーテル結合、チオエーテル結合、ウレタン結合、第2級アミノ基、カルボニル基、ウレア結合、トリアジン基、チオールの付加されたマレイミド基およびオキシム結合からなる群より選択される1種以上を含んでいてもよいアルキレン基からなる群より選択される、請求項1~4のいずれか1項に記載の結合体。
- 式(1)において、Aが、サイトカイン、ホルモン、酵素、抗体および核酸からなる群より選択される生体関連物質である、請求項1~5のいずれか1項に記載の結合体。
- 式(2)において、Qが、エチレングリコール、グリセリン、リシン、アスパラギン酸およびグルタミン酸からなる群より選択される化合物の残基である、請求項7に記載のブロックポリマー誘導体。
- 式(2)において、L2、L3、L4が、それぞれ独立して、単結合、フェニレン基、アミド結合、エーテル結合、チオエーテル結合、ウレタン結合、第2級アミノ基、カルボニル基、ウレア結合、ならびに、単結合、フェニレン基、アミド結合、エーテル結合、チオエーテル結合、ウレタン結合、第2級アミノ基、カルボニル基およびウレア結合からなる群より選択される1種以上を含んでいてもよいアルキレン基からなる群より選択される、請求項7または8に記載のブロックポリマー誘導体。
- 式(2)において、Xが、活性エステル基、活性カーボネート基、アルデヒド基、イソシアネート基、イソチオシアネート基、エポキシ基、カルボキシル基、チオール基、マレイミド基、置換マレイミド基、ヒドラジド基、ピリジルジチオ基、置換スルホ基、アミノ基、オキシアミノ基、ヨードアセトアミド基、アルキルカルボニル基、アルケニル基、アルキニル基、アジド基、アクリロイル基、スルホニルオキシ基、α-ハロアセチル基、アリル基およびビニル基からなる群より選択される反応性官能基である、請求項7~9のいずれか1項に記載のブロックポリマー誘導体。
