Description
本発明は、T細胞リンパ腫または白血病の処置において有用な細胞および作用因子に関する。
リンパ系悪性腫瘍は、T細胞に由来するものまたはB細胞に由来するもののいずれかに大きく分けることができる。T細胞悪性腫瘍は、臨床的かつ生物学的に不均一な障害の群であり、合わせて非ホジキンリンパ腫の10~20%および急性白血病の20%を占める。最も一般的に同定される組織学的サブタイプは、特定不能末梢性T細胞リンパ腫(PTCL-NOS);血管免疫芽球性T細胞性リンパ腫(AITL)および未分化大細胞型リンパ腫(ALCL)である。全ての急性リンパ芽球性白血病(ALL)のうちの一部20%がT細胞表現型である。

T細胞受容体(TCR)は、Tリンパ球の表面上に発現され、主要組織適合性遺伝子複合体(MHC)分子に結合した抗原の認識に関与する。TCRが抗原性ペプチドおよびMHC(ペプチド/MHC)と会合すると、関連する酵素、補助受容体、特殊アダプター分子、および活性化または放出された転写因子によって媒介される一連の生化学的事象を通じてTリンパ球が活性化される。
TCRはまた、最大5つの不変鎖γ、δ、ε(集合的にCD3と称される)およびζも含む。CD3およびζサブユニットは、αβまたはγδによる抗原の認識後に第2のメッセンジャーおよびアダプター分子と相互作用する特異的な細胞質ドメインを通じてTCRシグナル伝達を媒介する。TCR複合体の細胞表面発現の前に、サブユニットが対で集合し、そこではTCRαおよびβならびにCD3γおよびδの膜貫通ドメインおよび細胞外ドメインの両方が役割を果たす。
「悪性」という用語は、本明細書では、その標準の意味に従って使用され、その成長が自己限定されず、隣接組織内に侵入することができ、かつ、遠位組織に拡散することができる細胞を指す。そのように、「悪性T細胞」という用語は、本明細書では、リンパ腫または白血病の状況でクローン的に拡大したT細胞を指すために使用される。
選択的作用因子は、TRBC1またはTRBC2のいずれかに相互排他的に結合する。
(i)TRBCの3位におけるNをKから識別;
(ii)TRBCの3位におけるKをNから識別;
(iii)TRBCの4位におけるKをNから識別;
(iv)TRBCの4位におけるNをKから識別;
(v)TRBCの36位におけるFをYから識別;
(vi)TRBCの36位におけるYをFから識別;
(vii)TRBCの135位におけるVをEから識別;および/または
(viii)TRBCの135位におけるEをVから識別。
本発明の方法で使用する作用因子は、枯渇性モノクローナル抗体(mAb)もしくはその機能性断片、または抗体模倣物であってよい。

実施例12に記載の通り、ファージディスプレイを使用して、TRBC1よりもTRBC2との結合に対して高度に選択的ないくつかの抗体結合ドメインが同定されている。
配列番号23~32で示されるscFvのうちの1つからの
(i)重鎖CDR3および/もしくは軽鎖CDR3;
(ii)重鎖CDR1、CDR2およびCDR3ならびに/もしくは軽鎖CDR1、CDR2およびCDR3;または

抗体の調製は、標準の実験室技法を使用して実施することができる。抗体は、動物血清から得ることができ、あるいはモノクローナル抗体またはその断片の場合には、細胞培養物中で産生することができる。細菌または哺乳動物細胞培養物において、確立された手順に従って抗体を産生するために組換えDNA技術を使用することができる。
TRBC1またはTRBC2のいずれかに対して選択的な抗体は、当技術分野における標準の方法を使用して同定することができる。抗体の結合特異性を決定するための方法としては、これだけに限定されないが、ELISA、ウエスタンブロット、免疫組織化学的検査、フローサイトメトリー、フェルスター共鳴エネルギー移動(FRET)、ファージディスプレイライブラリー、酵母ツーハイブリッド法、免疫共沈降、二分子蛍光補完法およびタンデムアフィニティー精製が挙げられる。
あるいは、作用因子は、免疫グロブリンに由来するものではないまたは免疫グロブリンに基づくものではない分子であってよい。非抗体ポリペプチドの結合能力を活用するためにいくつもの「抗体模倣」設計反復タンパク質(DRP)が開発されてきた。
抗体または模倣物は、抗体または模倣物と、別の作用因子または抗体とのコンジュゲートであってよく、例えば、コンジュゲートは、検出可能な実体または化学療法用実体であってよい。
抗体様結合ドメインを2つ有するという基本概念に基づく多種多様な分子が開発されてきた。
(i)TRBC1またはTRBC2のいずれかに結合する第1のドメイン;および
(ii)T細胞を活性化することができる第2のドメイン
を含んでよい。
キメラ抗原受容体(CAR)は、キメラT細胞受容体、人工T細胞受容体およびキメラ免疫受容体としても公知であり、任意の特異性を免疫エフェクター細胞に移植する、工学的に作製された受容体である。古典的CARでは、モノクローナル抗体の特異性がT細胞に移植されている。CARをコードする核酸を、例えば、レトロウイルスベクターを使用してT細胞に移入することができる。この方法では、養子細胞移入のための多数のがん特異的T細胞を生成することができる。この手法の第I相臨床試験では有効性が示されている。
本発明は、BiTEまたは本発明の第1の態様のCARなどの作用因子をコードする核酸をさらに提供する。
本発明は、本発明の1つまたは複数の核酸配列を含むベクター、またはベクターのキットも提供する。そのようなベクターを使用して核酸配列を宿主細胞に導入し、そうすることで宿主細胞に本発明の第1の態様に記載のCARを発現させることができる。
本発明は、本発明の第1の態様にしたがうCARを含む免疫細胞などの細胞にも関する。
(i)対象または上に列挙されている他の供給源由来のT細胞またはNK細胞を含有する試料の単離;および
(ii)T細胞またはNK細胞へに本発明のCARをコードする核酸配列の形質導入またはトランスフェクト
によって作製することができる。
本発明は、本発明の第1の態様のCARを発現する複数の細胞を含有する医薬組成物にも関する。医薬組成物は、薬学的に許容される担体、希釈剤または賦形剤をさらに含んでよい。医薬組成物は、任意選択で、1つまたは複数のさらなる薬学的に活性なポリペプチドおよび/または化合物を含んでよい。そのような製剤は、例えば、静脈内注入に適した形態であってよい。
本発明は、T細胞リンパ腫および/または白血病を処置するための作用因子、細胞および方法に関する。
末梢性T細胞リンパ腫は、比較的稀なリンパ腫であり、全ての非ホジキンリンパ腫(NHL)の10%未満を占める。しかし、それらは侵攻性の臨床経過を伴い、大多数のT細胞リンパ腫の原因および正確な細胞起源は未だ十分に定義されていない。
PTCL-NOSは、全ての末梢性T細胞リンパ腫およびNK/T細胞リンパ腫の25%超を占め、最も一般的な亜型である。これは、除外診断によって決定され、現行のWHO 2008に列挙されている特定の成熟T細胞リンパ腫実体のいずれにも対応しない。そのため、これは、特定不能びまん性大細胞型B細胞性リンパ腫(DLBCL-NOS)と類似している。
AITLは、リンパ節が関与する多形浸潤、顕著な高内皮細静脈(HEV)および濾胞樹状細胞(FDC)網目構造の血管周囲増大を特徴とする全身性疾患である。AITLは、通常は胚中心において見いだされる、濾胞ヘルパー型のαβT細胞(TFH)に由来するデノボのT細胞リンパ腫と考えられる。
ALCLは、ALCL-「未分化リンパ腫キナーゼ」(ALK)+またはALCL-ALK-に細分され得る。
EATLは、腸の上皮内T細胞に由来すると考えられる侵攻性新生物である。2008
WHO分類ではEATLの形態学的、免疫組織化学的かつ遺伝学的に別個の型が2種類認識されている:I型(大多数のEATLを表す)およびII型(症例の10~20%を占める)。
HSTLは、一般に自然免疫系のγδ細胞傷害性T細胞に由来する侵攻性の全身性新生物であるが、稀な症例ではαβT細胞に由来することもあり得る。これは最も稀なT細胞リンパ腫の1つであり、一般には、青年および若年成人(年齢の中央値、35歳)に影響を及ぼし、強力に男性優性である。
節外性NK/T細胞リンパ腫、鼻型は、侵攻性疾患であり、多くの場合、破壊的な正中病変および壊死を伴う。大多数の症例はNK細胞に由来するが、いくつかの症例は細胞傷害性T細胞に由来する。これは、普遍的にエプスタイン・バーウイルス(EBV)に関連する。
本発明の方法は、皮膚T細胞リンパ腫を処置するためにも使用することができる。
斑(萎縮性または非萎縮性):非特異的皮膚炎、体幹下部および臀部の斑;そう痒が最小/存在しない;
プラーク:強いそう痒性プラーク、リンパ節腫脹;および
腫瘍:潰瘍形成易発性
に分けられる。
C-ALCLは、多くの場合、形態ではALC-ALK-と区別できない。これは、細胞の75%超がCD30を発現する未分化形態、多形性形態または免疫芽球性形態の大細胞の皮膚腫瘍と定義される。リンパ腫様丘疹症(LyP)と共に、C-ALCLは一次皮膚CD30陽性T細胞リンパ球増殖性障害のスペクトルに属し、これは群として、菌状息肉腫の次に2番目に多い皮膚T細胞リンパ球増殖の群を含む。
T細胞急性リンパ芽球性白血病(T-ALL)は、小児コホートのALLの約15%を占め、成人コホートのALLの約25%を占める。患者は、通常、高白血球数を有し、そして臓器肥大、特に縦隔肥大およびCNSの関与を示す可能性がある。
T細胞前リンパ球性白血病(T-PLL)は、侵攻性の挙動ならびに血液、骨髄、リンパ節、肝臓、脾臓、および皮膚の関与への偏好を伴う成熟T細胞白血病である。T-PLLは、主に30歳を超える成人に影響を及ぼす。他の名称として、T細胞慢性リンパ球性白血病、「こぶ状」型T細胞白血病、およびT前リンパ球性白血病/T細胞リンパ球性白血病が挙げられる。
本発明の方法は、当該作用因子を医薬組成物の形態で投与するステップを含み得る。
当該作用因子の投与は、活性成分が生物により利用可能になるような種々の経路のいずれかを使用して実現することができる。例えば、当該作用因子は、経口経路および非経口経路によって、腹腔内に、静脈内に、皮下に、経皮的に、筋肉内に、局所送達によって、例えばカテーテルまたはステントによって投与することができる。
本発明は、第1の態様の方法に従ったT細胞リンパ腫の処置において使用するための作用因子も提供する。作用因子は上で定義された任意の作用因子であってよい。
本発明は、第1の態様の方法に従ったT細胞リンパ腫の処置において使用するための、上で定義された作用因子を含むキットをさらに提供する。
本発明は、さらに、対象におけるT細胞リンパ腫または白血病の存在を決定するための方法であって、対象由来の試料中のTRBC1陽性またはTRBC2陽性のいずれかであるT細胞の割合を決定するステップを含む方法に関する。
TRBC1発現細胞およびTRBC2発現細胞の識別
JOVI-1抗体はVineyら(Hybridoma;1992年;11巻(6号);701~713頁)によって以前に開示されており、市販されている(Abcam、ab5465)。本発明者らは、JOVI-1により、TRBC1またはTRBC2の特異的な発現に基づいて細胞を識別することができることを決定した。
正常なドナーCD4+T細胞およびCD8+T細胞は、別々のTRBC1陽性集団およびTRBC1陰性集団を含有する
本発明者らは、JOVI-1抗体を正常ドナーの初代ヒトT細胞に対して試験した。これらの分析により、全てのドナーが、ある割合のTRBC1を発現するCD4+T細胞およびCD8+T細胞の両方とある割合のTRBC1を発現しないそれぞれを有することが明らかになった。正常なCD4+T細胞およびCD8+T細胞のおよそ20~50%がTRBC1+veである(図6および7)。
TCRを発現するクローン性T細胞株は、TRBC1陽性または陰性である
細胞株は、患者における元のクローン性腫瘍集団に由来するものである。TCRを発現するT細胞株の染色により、T細胞がTRBC1またはTRBC2のいずれかを発現することが示され、これがクローン性のマーカーとして確認される。試験した3つのT細胞株のうち、ジャーカット細胞(TRBC1+であることが分かっている)はJOVI-1で染色され、HPB-ALL細胞またはHD-Mar-2細胞(TRBC2+であることが分かっている)はJOVI-1で染色されず、TRBC1またはTRBC2のいずれかが排他的に発現されることが支持される(図8)。
T前リンパ球性白血病の患者における初代クローン性T細胞はTRBC1陽性または陰性である
T前リンパ球性白血病(T-PLL)の患者の末梢血から抽出したクローン性T細胞は、均一にTRBC1陽性またはTRBC1陰性のいずれかである。
TCBC1に独特の残基の変異の影響
TCRβ鎖定常領域におけるハイブリッドTRBC1/2変異以外は同一である、TCRをコードするプラスミドベクターを生成した。分析から、JOVI-1がTCRβ定常鎖の3位および4位の残基の差異を認識することが示され、これらの残基が抗体認識に利用可能なものであり、TRBC1をTRBC2から、またはTRBC2をTRBC1から識別する作用因子を生成するための最良の標的であり得ることが示された(図5)。
TRBC1 TCR発現T細胞は特異的に溶解されるがTRBC2 TCR発現T細胞は特異的に溶解されない。
野生型ジャーカットT細胞(CD34-、TRBC1+)を、CD34マーカー遺伝子と同時発現させたTRBC2を用いて形質導入したTCRαβノックアウトジャーカットT細胞(CD34+TRBC2+)と混合した。これらの細胞を、1時間にわたり、JOVI-1のみと一緒に、またはJOVI-1および補体と一緒にインキュベートした。細胞を洗浄し、CD34、アネキシンVおよび7-AADについて染色した。細胞をフローサイトメトリーによって分析した。
mAbへの長期曝露後のTCR内部移行に起因するTRBC1 TCRの検出失敗を回避した。細胞を洗浄し、CD34、アネキシンVおよび7-AADについて染色した。細胞をフローサイトメトリーによって分析した。生細胞(すなわち、アネキシンV陰性および7-AAD暗である細胞)に関してゲーティングすることにより、本発明者らは、TRBC1 T細胞が補体の存在下でJOVI-1によって選択的に死滅することを決定することができた(図9)。
ポリクローナルエプスタイン・バーウイルス(EBV)特異的T細胞は2つのほぼ同等のTRBC1/2集団に分割することができる。
正常血液ドナーから末梢血T細胞を採取した。単核細胞を単離し、細胞の大部分を凍結保存した。少数の細胞にEBVの実験室株(B95-8)を感染させた。数週間を越えて、リンパ芽球様細胞株(LCL)として公知の不死化EBV感染細胞株が現れた。そのような細胞株は、異なるEBV抗原の大きな集合を示すことが公知である。予め凍結保存しておいた単核細胞を解凍し、このLCL株を用い、IL2の存在下で週に1回、4週間にわたって繰り返し刺激した。このプロセスにより、末梢血単核集団からEBV特異的T細胞が選択的に増大する。そのようなプロセスにより、T細胞の90%超がEBV特異的であり、ドナーのEBV免疫系を表すポリクローナル株がもたらされることも公知である。この株の特異性は、自己由来LCLは高程度に死滅するが同種異系LCLまたはK562細胞は死滅しないことが示されることによって確認される(図10a)。次いで、この細胞株をJOVI-1で染色し、TRBC1 T細胞およびTRBC2 T細胞のほぼ同等の混合物を含有することが示された(図10b)。
循環末梢性T細胞リンパ腫のJOV1染色。
仮説は、クローン性であるT細胞リンパ腫は、TRB1 T細胞受容体またはTRBC2 T細胞受容体のいずれかを発現するが、ポリクローナルである正常T細胞は、TRBC1を有するものとTRBC2を有するものの混合物であるT細胞集団で構成されるというものであった。これを実証するために、リンパ腫が末梢血中を循環している患者からT細胞リンパ腫の血液試料を得た。末梢血単核細胞を単離し、CD5およびJOVI1を含む抗体のパネルを用いて染色した。T細胞の総集団(リンパ腫および正常T細胞の両方を含有する)を最初に同定した。この集団は、正常(明るい)CD5発現を有するT細胞および中間/暗いCD5発現を有するT細胞で構成されていた。前者は正常T細胞を代表し、後者はリンパ腫を代表する。次にJOVI-1結合を調査し、図12に結果が示されている。
JOVI-1のVH/VL配列の解明
マウスIgG CH1の定常領域およびマウスカッパの定常領域にアニーリングするプライマーを用いた5’RACEを使用して、本発明者らは、ハイブリドーマJOVI-1から単一の機能性VH配列および単一の機能性VL配列を単離した。VHおよびVLの配列はそれぞれ配列番号1および2である(上記を参照されたい)。VHおよびVLのアノテートされた配列が図11に示されている。
JOVI-1に基づくCARの機能
JOVI-1 scFvをCAR構成にクローニングした。いずれのスペーサー長により最適なJOVI-1に基づくCARがもたらされるかを解明するために、ヒトFcスペーサー、ヒトCD8ストークスペーサーまたはIgG1ヒンジに由来するスペーサーのいずれかを用いて第3世代CARを生成した(図14)。正常ドナー由来の初代ヒトT細胞にこれらのCARを形質導入し、ジャーカットおよびTCRがノックアウトされたジャーカットの死滅を比較した。IgG1ヒンジスペーサーまたはCD8ストークスペーサーのいずれかを有するJOVI-1 scFvを有するCARにより、ジャーカットは死滅したが、TCRがノックアウトされたジャーカットは死滅せず(図15)、これにより、予測された特異性が実証された。CARを形質導入した正常ドナーT細胞はTRB1/2 T細胞の混合物を有するはずであるので、培養物が「自己パージ」することが予測された。実際に、これが観察された。100%TRBC2陰性になったCAR T細胞培養物のJOVI-1染色が図16に示されている。
JOVI-1の特異性の実証
よく特徴付けられたヒトTCRならびに都合のよいマーカー遺伝子をコードするトリシストロン性レトロウイルスカセットを生成した。TCRα鎖およびβ鎖のコード配列を、デノボ遺伝子合成を使用してオーバーラッピングオリゴヌクレオチドから生成した。これらの鎖を口蹄疫2Aペプチドとインフレームで接続して共発現を可能にした。切断型CD34マーカー遺伝子をPCRによってcDNAからクローニングし、内部リボソーム進入配列(internal ribosome entry sequence)(IRES)を使用してTCR鎖と共発現させた。このカセットをレトロウイルスベクターに導入した。この構築物のバリアントを、所望の変異を導入するプライマーを用いたオーバーラップPCRによってスプライシングすることによって生成した。構築物の正確さを、サンガー配列決定によって確認した。ジャーカット76株は、TCRα鎖およびβ鎖の両方がノックアウトされた、ジャーカットT細胞株のよく特徴付けられた誘導体である。このジャーカット株に上記のレトロウイルスベクターを用いて、標準の技法を使用して形質導入した。
ジャーカットをECACCから得、上記の通り操作した。他のT細胞株もECACCから得た。正常ドナーから静脈穿刺によって末梢血を採取した。血液をフィコール処理して単核細胞を単離した。細胞をJOVI-1ならびに全てのTCRおよびCD3を認識する市販のモノクローナル抗体で染色した。操作したT細胞の場合には、細胞を、CD34を認識する抗体で染色した。末梢血単核細胞の場合には、細胞を、CD4およびCD8を認識する抗体で染色した。抗体は、適切なフルオロフォアとコンジュゲートした状態で購入し、そうすることで、フローサイトメーターを用いた細胞の分析中に独立した蛍光シグナルを得ることができた。
野生型ジャーカットT細胞(TRBC1-TCR)、およびTRBC2 TCRを導入したジャーカットT細胞TCR KOを1:1の比率で混合した。次いで、このジャーカットの混合物を1ug/mlのJOVI-1モノクローナル抗体と一緒に、補体の不在下または存在下でインキュベートした。4時間後、細胞をアネキシン-Vおよび7AADおよびCD34で染色した。都合よく、マーカー遺伝子CD34により、野生型(TRBC1)ジャーカットとトランスジェニック(TRBC2)ジャーカットとを区別することができる。細胞集団をフローサイトメトリーによって分析した。生細胞を、アネキシン-V陰性かつ7AAD暗であるフローサイトメトリー事象に関してゲーティングすることによって選択的に試験した。このように、トランスジェニック(TRBC2)T細胞の生存と野生型(TRBC1)T細胞の生存を試験した。
T細胞リンパ球増殖性障害のクローン性の調査
悪性細胞が均一にTRBC1陽性または陰性のいずれかであることを確認するために、4名の患者:T細胞大型顆粒リンパ球増殖性障害(T-LGL)3名;および末梢性T細胞リンパ腫(PCTL)1名を試験した。
ファージディスプレイを使用した、T細胞受容体β鎖定常ドメインの2つのアイソフォームを区別するモノクローナルヒト抗体の生成
TRBC2ペプチドとTRBC1ペプチドとを区別する抗体を生成するために、2つのTRBCアイソフォームの間の領域の差異を包含する断片を合成した。TRBC2とTRBC1との間の4つのアミノ酸の差異のうち、2つは定常ドメインの始めに見いだされる。これらの領域を表すペプチド(以下を参照されたい)を合成し、抗体生成のために使用した。
抗体ファージディスプレイ選択
ヒトファージディスプレイライブラリーを構築し、(Schofieldら、2007年、Genome Biol 8巻、R254頁)に記載の通りファージ選択を行った。抗体ライブラリーからTRBC1特異的抗体およびTRBC2特異的抗体を同定するために、複数のラウンドのファージディスプレイ選択を行った。2つのファージ選択戦略を並行して使用して、特異的な結合物質の大きなパネルが生成する機会を最大限にした。これらの戦略は、固相選択および液相選択として公知である(図21)。固相選択では、ファージ抗体を、固体表面に固定化した標的抗原に結合させる(Schofieldら、2007年、上記の通り)。液相選択では、ファージ抗体を溶液中のビオチン化した抗原に結合させ、次いで、ファージ抗体-抗原複合体をストレプトアビジンまたはニュートラアビジンでコーティングした常磁性ビーズによって捕捉する。固相選択戦略の中で、2つの異なる固定化または抗原提示手法を用いた。第1の手法を使用する場合、ウシ血清アルブミン(BSA)またはオボアルブミン(OA)とコンジュゲートしたTRBCペプチドをMaxisorp(商標)immunotube上に直接吸着によって固定化した。第2の手法を使用する場合、ビオチン化TRBCペプチドを、ストレプトアビジンまたはニュートラアビジンで予めコーティングしたMaxisorp(商標)immunotubeチューブ上に間接的に固定化した。
ラウンド2およびラウンド3選択アウトプットからのscFv集団をpSANG10-3F発現ベクターにサブクローニングし、E.coli BL21(DE3)細胞へと形質転換した。1128種の個々の形質転換体(564クローン/TRBCペプチド)を選び取って12×96ウェル培養プレート(94クローン/プレート)に入れ、自己誘導培地を使用して抗体発現を誘導した。一晩の誘導後に培養上清中に分泌された組換えモノクローナル抗体を、ニュートラアビジンでコーティングされたNunc Maxisorp(商標)96ウェルプレートに固定化したビオチン化TRBC1およびTRBC2への結合について試験した。TRBC1選択からスクリーニングされた564クローンのうち、255クローンが、TRBC1に特異的であることが見いだされた(TRBC1に対して>10000TRF単位およびTRBC2に対して<1000TRF単位)。TRBC2選択からスクリーニングされた564クローンから、138種のTRBC2特異的結合物質(TRBC2に対して>10000TRF単位およびTRBC1に対して<2000TRF単位)が同定された。図24に、TRBC1(図24A)またはTRBC2(図24B)のいずれかに対する選択から生じた単一の96ウェルプレートからの代表的な結合プロファイルが示されている。異なる選択条件を使用して生成した特異的な結合物質の詳細が表5に要約されている。
CDR3配列およびVL CDR3配列が上の表1に要約されている。TRBC2特異的クローンおよびそれらのVH CDR3配列およびVL CDR3配列が上の表2に要約されている。
ウサギのペプチド免疫化によるTRBCポリクローナル抗体の産生
TRBC2とTRBC1とを区別する抗体を生成するために、2つのTRBCアイソフォーム間を区別する原理領域を包含する2つのペプチドを合成し、ウサギを免疫化するために使用した。以下のペプチド配列を使用した:

を含む二重特異性T細胞エンゲージャー。 - 前記第1および第2のドメインが、単鎖可変断片(scFv)である、請求項1に記載の二重特異性T細胞エンゲージャー。
- 前記第1のドメインが、TRBC1に選択的に結合する、請求項1または2に記載の二重特異性T細胞エンゲージャー。
