本発明の分野は、広域中和抗HIV-1抗体(bNab)およびその使用方法である。

本発明は、広域中和抗HIV-1抗体(bNab)およびその抗原結合部分に関する。特に、本発明は、いくつかの異なるモノクローナル抗体、BG18、NC37、およびBG1、限定するものではないがそれらの相補性決定領域(CDR)を含むそれらの抗原結合部分、ならびにそれらの誘導体および変異体に関し、これらはインビボでHIV-1の広域かつ強力な中和を示す(下記実施例1参照)。したがって、本発明の抗HIV-1抗体は、本明細書中で考察されるように、単独で、または互いに組み合わせて、もしくは他のbNabと、もしくは他のHIV治療と組み合わせて、強力な治療的有用性を有する。
本発明の一実施形態は、本発明の少なくとも1つの抗HIV-1抗体またはその抗原結合部分を含む医薬組成物、ならびにそれを必要とする患者の治療でのそれらの使用方法に関する。患者は、HIV-1による潜在感染または活性感染を有し得る。これらの組成物および方法に利用される抗HIV-1抗体またはその抗原結合部分は、本発明の任意の抗HIV-1抗体またはその抗原結合部分であり得るが、特に有用なものは、組換え作製されたその変異体および/または操作された誘導体を含むBG18またはその変異体を含む抗HIV抗体またはその抗原結合部分である。
本発明の一実施形態は、試料中に存在するHIV-1を検出するためのキットに関する。これらのキットは、本発明の抗HIV-1抗体またはその抗原結合部分および様々な試薬、例えば、抗HIV-1抗体とHIV-1上に存在するエピトープ、例えばgp120および/またはgp140、またはそれらの抗原性断片との間の結合の検出を助ける試薬を含み得る。
範囲の値が提供される場合、文脈上明らかに別段の指示がない限り下限の単位の10分の1まで、その範囲の上限と下限との間にある介在値、およびその記載された範囲内の任意の他の記載された値または介在値のそれぞれが、本発明内に包含されると理解される。これらの小さい方の範囲の上限および下限は、独立して小さい方の範囲に含まれ得、同様に本発明内に包含され、記載された範囲内の任意の具体的に除外される限界に従属する。記載された範囲が限界のうち一方または両方を含む場合、それらの含まれる限界の両方を除く範囲も本発明に含まれる。
1.ウイルス血症コントローラー(Viremic Controller)に見られる強力なHIV-1広域中和抗体および抗体感受性ウイルスの共存
A.方法
HIV-1感染者EB354
HIV-1感染長期非進行者394例のコホート内で、中和の広域性および効力について上位1%に入ったドナーEB354から得られたIgGを精製した。ドナーEB354は1986年にHIV-1クレードBと診断された。ドナーEB354は1995年から1998年の間にジダノシンおよびスタブジンによる治療を受けたが、それ以来抗レトロウイルス療法は受けていない。ドナーEB354は、2002年以降臨床的に頻繁に追跡されてきた(表3:HLA A*01:01、24:02、B*27:05、57:01、C*01:02、06:02)。ほとんどの個体では、感染直後に株特異的抗体が急速に発現する。この応答は、場合によってはbNAbを誘発する耐性ウイルス変異体の選択と関連している。ただし、bNAbの発現が、共存するbNAbに対して耐性である自己由来のプラズマウイルスの迅速な選択と関連しているという点で、詳細に試験された個体とEB354とは異なる。感受性かつ耐性のウイルス株がbNAbと共存しているためEB354は重要であり、耐性株は、何らかの方法で部分的にしか有効でないか、CD8+T細胞によって抑制されているため、高レベルのウイルス血症を引き起こすことができない。したがって、bNAb感受性プラズマウイルスはこの個体の免疫圧力を逃れられず、ウイルスが持続するbNAb:ウイルスの平衡状態をもたらすが、高レベルのウイルス血症を引き起こすことはできない。EB354は、エリートコントローラーおよびエリートニュートラライザーの両方であるという点で独特である。HIV-1エリートコントローラーとは、長年にわたってウイルス量を低く維持する感染者である。これらの個体はウイルスを伝播する可能性が低く、AIDSを発症せず長期間生存を維持する。HIV-1エリートコントローラーの85%には、HLA対立遺伝子B57*01および/またはB27*05が見られる。これらの対立遺伝子は、CD8T細胞の細胞傷害活性の増強と関連している。ウイルス血症進行者(すなわち、高いウイルス量を有する患者)と比較して、エリートコントローラーはbNAbを発現する可能性が低い。EB354では、感染を部分的に制御している可能性がある堅牢なCD8+T細胞応答に関係なく、bNAbの発現および親和性成熟を補助するのに十分なHIV-1複製が見られた。本明細書中で考察されるように、ドナーEB354の血清学的中和活性を説明するbNAbは、いくつかの重複しないエピトープを認識する。
J.F.Scheidら、A method for identification of HIV gp140 binding memory B cells in human blood.Journal of immunological methods 343, 65-67(2009)に記載されているように、ドナーEB354 PBMC由来のベイト+CD19+IgG+B細胞の単一細胞選別を行った。抗CD19磁気ビーズ(MACS)を用いてメモリーB細胞を予め濃縮し、以下の4種類のベイトを用いて染色した:ベイトとしてのgp120 2CCコアタンパク質、gp140YU2、gp14092UG37.8(クレードA)+gp140CZA79012(クレードC)の1:1混合物およびBG505 SOSIP.664。レスキュープライマーを使用して重鎖およびIgλ遺伝子の両方を増幅し、通常のプライマーをIgK鎖に使用した。PCR産物をいずれも配列決定し、Ig遺伝子使用、CDR3、およびVH/VL体細胞超変異の数(IgBLASTおよびIMGT)について分析した。T.Tillerら、Efficient generation of monoclonal antibodies from single human B cells by single cell RT-PCR and expression vector cloning.Journal of immunological methods 329, 112-124(2008)に以前に記載されているように、精製し消化したPCR産物をヒトIgγ1、IgκまたはIgλ発現ベクターにクローニングし、F.Kleinら、Enhanced HIV-1 immunotherapy by commonly arising antibodies that target virus escape variants.The Journal of experimental medicine 211, 2361-2372(2014)に記載されているように、指数関数的に増殖するHEK 293-6E細胞へのIgH、IgKおよびIgL発現プラスミドの一過性トランスフェクションによって作製した。
ルシフェラーゼベースのTZM.bl細胞アッセイを用いて、HIV-1中和を評価した。要約すると、エンベロープシュードウイルスを単一抗体の5倍連続希釈物とともにインキュベートし、ルシフェラーゼレポーター遺伝子を保有するTZM.bl細胞に適用した。48時間後、細胞を溶解し、発光を測定した。IC50およびIC80は、相対発光単位(RLU)をそれぞれ50%および80%減少させる単一抗体濃度を反映する。抗体データベース計算ツールを使用して、範囲曲線を構築した。
Asn26HCをGlnに変異させることによって重鎖位置26にある潜在的なN結合型グリコシル化部位が除去されたBG18 Fabの結晶が、野生型BG18 Fabの結晶と比較して優れたサイズおよび形態を示したため、BG18N26Q Fabを用いて構造決定を行った。BG18N26Q Fabの結晶(空間群P21;a=46.12Å、b=71.04Å、c=69.54Å;β=98.48°;非対称単位当たり1分子)は、0.2μLの18mg/mLタンパク質溶液と、0.2mLの0.1M酢酸ナトリウムpH4.5、26.8%(v/v)ポリエチレングリコール(PEG)400および13.4%(w/v)PEG8,000とを20℃で混合し、20%(v/v)エチレングリコールを補充した母液中で凍結保護し、液体窒素中で急速冷却することによって得られた。1.5:1のタンパク質:リザーバー比で250nLの液滴を用いる0.1M酢酸ナトリウム三水和物pH4.6および2.0M硫酸アンモニウムのリザーバーを用いたシッティングドロップでの蒸気拡散により、NC102Fab(空間群P212121;a=60.2Å、b=82.5Å、c=117.2Å;非対称単位当たり1分子)の結晶を作製した。25%(v/v)エチレングリコールを補充した母液中で結晶を凍結保護し、液体窒素中で急速冷却した。1.5:1タンパク質:リザーバー比でタンパク質とリザーバー(0.1M HEPES pH7.5および20%(w/v)PEG10,000を含む)とをインキュベートすることによる250nLのシッティングドロップでの蒸気拡散により、NC37-93TH057複合体(空間群P212121;a=63.6Å、b=67.4Å、c=210.4Å;非対称単位当たり1個の複合体)の結晶を作製した。20%(w/v)PEG400を補充した母液中で結晶を凍結保護し、液体窒素中で急速冷却した。
USCFキメラを用いてEM構造を視覚化した。以下の選択肢を有するUCSFキメラ内のフィット・イン・マップ・ユーティリティ(fit in map utility)を使用して、結晶構造からの座標をサブトモグラム平均化またはネガティブ染色単一粒子EM構造に適合させた:リアルタイム相関/平均更新、原子からシミュレートしたマップの使用、分解能25Å。BG505 SOSIP.664構造(PDB 4TVP)を最初に密度に適合させ、次いでBG18 FabおよびCH103 Fab(PDB 4JAM;179NC75 Fabのモデルとして)について個々に対応する密度に適合させた。FabのCH1-CLドメイン領域は低い密度を示した。
一過性トランスフェクションによって6×-Hisタグ付きBG18、BG18N26Q、NC102およびNC37 FabをHEK293-6E細胞に発現させ、Ni2+-NTAアフィニティークロマトグラフィー(GE Healthcare)およびSuperdex 200 16/60サイズ排除クロマトグラフィー(SEC)(GE Healthcare)を用いて、トランスフェクトした細胞上清から精製した。切断されたHisタグ付き93TH057 gp120コアタンパク質をバキュロウイルス感染昆虫細胞内で産生させ、Ni2+アフィニティークロマトグラフィー(GE Healthcare)を用いて、記載されているように精製した(Diskinら、Science 2011;Diskinら、NSMB 2010)。結晶化試験の前に、精製NC37および93TH057タンパク質を同時インキュベートし(2:1モル比のFab対gp120コア)、gp120タンパク質1mg当たり5kUのエンドグリコシダーゼHを用いて25℃で3時間処理した。Superdex 200 10/300 GLカラム(GE Healthcare)を用いたサイズ排除クロマトグラフィーによって、エンドグリコシダーゼH処理複合体を精製した。
製造業者の指示に従ってQiagen MinElute Virus Spinキットを用いて、患者の血漿からHIV-1 env遺伝子HIV-1-RNAの単一ゲノム配列決定(SGS)を抽出した。抽出したRNAを、SuperScript III Reverse TranscriptaseおよびHIV-1特異的プライマーenvB3outを用いてEnv特異的cDNA合成に供した。

Li, M.ら、Human immunodeficiency virus type 1 env clones from acute and early subtype B infections for standardized assessments of vaccine-elicited neutralizing antibodies.J Virol, 2005.79(16):p.10108-25に記載されているように、293T細胞(ATCC)にHIV-1 Env発現プラスミドと、pSG3ΔEnvと呼ばれるEnv欠損ゲノムバックボーンプラスミド(Env-deficient genomic backbone plasmid)とをトランスフェクトすることによってシュードウイルスを作製した。中和アッセイで使用するために、シュードウイルスをトランスフェクションの72時間後に回収した。上記のLiらに記載されているように、単一ラウンドの複製シュードウイルスアッセイとTZM-Bl標的細胞とを使用して中和活性を評価した。要約すると、TZM-bl細胞を96ウェル平底プレートに播種した。このプレートにシュードウイルスを加え、これを抗体の連続希釈物とともに37℃で1時間プレインキュベートした。感染の72時間後に、溶解およびBright-Glo TMルシフェラーゼ基質(Promega)の添加時にルシフェラーゼレポーター遺伝子発現を定量した。IC50値を決定するために、非線形回帰により用量反応曲線を適合させた。
Amin, M.N.ら、Synthetic glycopeptides reveal the glycan specificity of HIV-neutralizing antibodies.Nat Chem Biol, 2013.9(8):p.521-6に記載の手順に従って、標的糖ペプチドを作製するために、GlcNAc-ペプチド前駆体の自動固相ペプチド合成およびその後のGlcNAc-ペプチドの酵素的糖転移反応からなる化学酵素的方法を用いて、いくつかのHIV-1株に由来するV3糖ペプチドの合成を達成した。25℃でBIAcore T200システム(GE Healthcare)を用いるSPRにより、ビオチン化合成糖ペプチドとBG18およびBG8 IgG抗体との相互作用を評価した。200応答単位(RU)が達成されるまで、HBS-P緩衝液(10mM HEPES、150mM NaCl、P20界面活性剤0.05%v/v、pH7.4)中のニュートラアビジン被覆CM5センサーチップ上にビオチン化糖ペプチドを固定化した。BG18(またはBG8)を、BS-P緩衝液中で4μMから始めて2倍の系列希釈の濃度で流速40μL/分で180秒間注入した。次いで、流速40μL/分でBS-P緩衝液を1210秒間注入して解離させた。流速50μL/分で3M MgCl2を3分間注入してから、流速50μL/分でHBS-P緩衝液を5分間注入することによって再生を行った。
週2回、3週間にわたって各抗体1mgを皮下(s.c.)投与することにより、ヒト化NOD Rag1-/-Il2rgnull(NOD.Cg-Rag1tm1Mom Il2rgtm1Wjl/SzJ)マウス(The Jackson Laboratory)を治療し、合計6回の抗体注射を実施した。同じドナー由来のヒト細胞により対照huマウスを再構成し、HIV-1YU2に感染させたが、抗体により治療はしなかった。プラズマウイルス量を毎週測定した。F.Kleinら、HIV therapy by a combination of broadly neutralizing antibodies in humanized mice.Nature 492, 118-122(2012)に記載されているように、リバウンドウイルスを有するマウス由来のgp120配列を得た。
PBS中の5μg/mLの精製2CCコア、gp120.YU2(野生型または変異体)またはgp140.YU2フォールドオン三量体により、高結合96ウェルELISAプレート(Costar)を一晩コーティングした。PBS+0.05%Tween20で6回洗浄した後、2%BSA、1μM EDTAおよび0.05%Tween-PBS(「ブロッキング緩衝液」)を用いてプレートを2時間ブロックし、次いで、初期濃度4μg/mLからPBS中7連続1:4希釈物として加えたIgGとともに1時間インキュベートした。さらに洗浄した後、ヤギHRPコンジュゲート抗ヒトIgG抗体(Jackson ImmunoResearch)(ブロッキング緩衝液中0.8μg/mL)とともに1時間インキュベートし、続いて西洋ワサビペルオキシダーゼ(HRP)発色基質(ABTS溶液;Invitrogen)とともにインキュベートすることによってプレートを発色させた。競合ELISAでは、0.5μg/mLのBG505.SOSIP.664によりプレートをコーティングし、洗浄し、ブロッキング緩衝液を用いて2時間ブロッキングし、次いで、初期濃度32μg/mLからPBS中7連続1:4希釈物として加えたIgGとともに、4μg/mLの一定濃度のビオチン化抗体の存在下で1時間インキュベートした。次いで、HRPコンジュゲートストレプトアビジン(Jackson ImmunoReseach)(ブロッキング緩衝液中1μg/mL)を用いてプレートを発色させた。
確立されたプロトコルに従って、CMV-env発現カセットを作製した(52)。要約すると、以下のプライマーを用いてpcDNA 3.1D/V5-His-TOPO発現ベクターからCMVプロモーターを増幅した:

Rev19 5’-ACTTTTTGACCACTTGCCACCCAT-3’(配列番号104)(1×High Fidelity緩衝液、2mM MgSO4、0.2mM dNTP、0.5単位のHigh Fidelity Platinum Taqおよび0.2μMの各プライマーを含有する20μL容量中)。サイクル条件は、94℃、2分;(94℃、15秒;55℃ 30秒; 68℃、4分)×35;68℃、10分であった。0.7%アガロースゲルを用いた分析によりenvの存在を確認し、Macherey-Nagel GelおよびPCR Purificationキットを用いて産物を精製した。次いで、プライマーCMVenvおよびRev19を用いて、10ngのエンベロープおよび0.5ngのCMVを3連でオーバーラップPCRに供した。全反応容量6は、1×High Fidelity緩衝液、0.2μM MgSO4、0.2mM dNTP、1単位のHigh Fidelity Platinum Taqおよび0.4μMの各プライマーを含有する50μLであった。PCRは、94℃、2分間;(94℃、30秒;60℃ 30秒;68℃、4分)×25;68℃、10分で行った。6ウェルプレート中で500ngのCMV-envをpSG3Δenvとともに293T細胞に同時トランスフェクトし、48時間後に上清を回収した。上記のように、TZM-blによる中和試験に上清を供した。
van ’t Woutら、Isolation and propagation of HIV-1 on peripheral blood mononuclear cells.Nat Protoc, 2008. 3(3):p.363-70に記載されているように、患者の末梢血単核細胞(PBMC)と健常ドナーPBMCとの共培養によって、ドナーEB354由来のウイルスを得た。ロックフェラー大学の試験プロトコルMNU-0628の下で、白血球除去によって患者から健常ドナーPBMCを得た。10%FBS、1%ペニシリン-ストレプトマイシンおよびPHAを1μg/mL含有するIMDM中、1mL当たり5×106細胞の密度で37℃および5%CO2で健常ドナーPBMCを2~3日間予備刺激した。次いで、刺激したドナーPBMC 6×106個を、10%FBS、1%ペニシリン-ストレプトマイシン、10IU/mLのIL-2および5μg/mLのポリブレンを含有するIMDMに移し、5~10×106個のEB354 PBMCと37℃および5%CO2で共培養した。培地を毎週交換し、Lenti-X p24 Rapid Titerキット(Clontech)により培養上清中のp24の存在を定量した。上清1mL当たり1ngのp24を超える培養物を凍結し、-80℃で保存した。上記のプロトコルに従ってTZM-bl中和アッセイを用いて、組織培養感染用量50(TCID50)の決定と、その後の様々な広域中和抗体および自己由来血清IgGに対する自己由来ウイルスの感受性に関する試験とを行った。中和アッセイはいずれも2連で行った。
製造業者の仕様書(Agilent Technologies)に従って、QuikChange(multi-)site-directed mutagenesisキットを用いて、野生型HIV-1YU2エンベロープに一重、二重および三重の突然変異を導入した。
ClustalW(バージョン2.11)を使用して、またはGeneious(バージョン8.1.6)配列分析ソフトウェアを使用する手動アラインメントによって、envヌクレオチド配列のアラインメントを生成した。明確にアラインさせることができなかった領域は、系統発生分析および多様性計算のために除去した。jModelTestを用いて、最尤系統発生分析のための進化モデルクラスを選択した。モデルパラメータ値および系統発生の結合推定とともにPhyML(バージョン3)を使用して、最尤分子系統樹を生成した。DIVEIN webtoolの2標本U統計量検定を使用して、標本間のペアワイズ遺伝的多様性を比較した。
Cutadapt v1.8.3を用いて配列アダプターを除去した。各ウイルスに対するリードアセンブリは3つの工程で行った。まず、Spades v3.6.1を使用してデノボアセンブリを実行し、長いコンティグファイルを作成した。続いて、255bpよりも長いコンティグをHIVエンベロープ参照配列にアラインし、Geneious 8を用いてコンセンサス配列を生成した。最後に、ギャップを埋めるためにコンセンサス配列にリードを再アラインし、最終コンセンサスを生成した。二重ピークを有する配列(任意の残基に対するカットオフコンセンサス同一性<75%)は、下流分析から省略した。
マン-ホイットニー検定によって統計的差異を分析した。GraphPad Prismソフトウェアを分析に使用し、*p≦0.05、**p≦0.01および***p≦0.001でデータを有意と見なした。
複数のbNAb
HIV-1感染長期非進行者394例のコホート内で、中和の広域性および効力について上位1%に入ったドナーEB354から得られたIgGを精製した。このドナーは1986年にHIV-1クレードBと診断された。EB354は1995年から1998年の間にジダノシンおよびスタブジンによる治療を受けたが、それ以来抗レトロウイルス療法は受けていない。2002年に、EB354のウイルス量は<400コピー/mLであり、CD4およびCD8の数はそれぞれ954および1046細胞/mm3であった。EB354は、2002年から2006年の間にウイルス血症に3つの実証されたピークを有した(図1A)。ウイルス量が<400コピー/mlであった2006年に、血清学的中和効力および広域性が最初に評価された(図1Aおよび図1Bならびに表3)。中和活性は2010年まで増大し、その時以来広域かつ強力なままである(図1B)。HLAタイピングでは、HLA A*01:01、24:02、B*27:05、57:01、C*01:02、06:02が明らかにされた(表3)。
アミノ酸レベルでは、BG18は、ともにVH4-59生殖系列前駆体から生じる10-1074およびPGT121の重鎖とそれぞれ63%および62%同一である。BG18は、密接に関連した(90.8%の同一性)生殖系列遺伝子VH4-4から生じる(図6A)。PGT121、10-1074およびBG18ならびにそのクローン変異体の重鎖の相補性決定領域(CDR)のアラインメントは、CDRH1とCDRH2との間に高度な類似性を示す。BG18のCDRH3は、PGT121/10-1074よりも3残基短い(21対24残基;図2Aおよび図6B)。さらに、BG18は、PGT121ファミリーに共通のフレームワーク領域内に7つの突然変異のうち6つを共有するが(図2A)、PGT121/10-1074のメンバーとは異なり、いかなる挿入または欠失も含有しない。
gp120コアに結合したNC37 Fabの2.7Å分解能の結晶構造(図1C)は、NC37がCD4bsを認識することを証明した(図8Aおよび図8B)。NC37ファミリーの一部のメンバーはVH1-2に整列し、他のメンバーはVH1-46に整列したため、配列のみに基づいて、NC37のVHがVH1-2またはVH1-46生殖系列遺伝子に由来していたかどうかを決定することは当初困難であった(表13)。しかし、構造比較から、NC37がVH1-46由来抗体8ANC131/134と同様の配向および結合角度をとることが明らかにされた(図8Aおよび図8B)。結果として、NC37がその軽鎖を用いてループD残基Asn280gp120と接触するのに対して、VH1-2由来抗体、例えばNIH45-46はそれらの重鎖を用いてAsn280gp120と接触する。NC37のCDRH3は、gp120内部ドメインに向かって伸長し、CDRH3残基Tyr100GHCとgp120内部ドメイン残基Lys97gp120およびGlu102gp120との間に接触を形成する。NC37 Fab-gp120複合体をSOSIP三量体構造に重ね合わせると、NC37 CDRH3がEnv三量体内の隣接プロトマーと接触することが示唆される(図8C)。
自己由来ウイルスに対する3つのbNAbの効果を検討するために、2006年~2015年の間の5つの異なる時点(2006年、2010年、2013年、2014年および2015年)に得られた単一ゲノム配列決定(SGS)によって、循環プラズマウイルス由来の個々のenv遺伝子をクローニングした。37個のみの機能的インフレーム転写物(functional in-frame transcript)が回収されたが、これは一つには、全時点での400コピー/mL未満のウイルス量の値によるものである。さらに、2015年から回収された配列はいずれも非機能的であった(図10)。37個の機能的配列は、以下の3つのクラスターを形成した-残りの配列と約15%異なる2006年に得られた1つの配列を含有するクラスターA、4つの時点すべてからの配列を含有する最大のクラスターであるクラスターB、およびいずれも2014年に得られた3つの配列を含有するクラスターC(図3A)。2014年に出現した配列の新しいクラスターと一致して、2013年から2014年の間にウイルス多様性の増加が見られた(図3B)。
BG18またはNC37が独立してインビボでHIV-1に選択的圧力をかけることができるかどうかを決定するために、ヒト化マウス(huマウス)をHIV-1YU2に感染させ、確立された感染を治療した。BG1は、HIV-1YU2に対して低い活性を示したため(IC50=15.8μg/mL)、その後の併用治療実験にのみ使用し、単独療法実験には使用しなかった。10-1074と同様に、それほど強力ではないが依然として有効なクローン変異体であるBG18またはBG8の投与は、ウイルス量の急速な減少(平均1.5log10)、その後のリバウンドウイルス血症と関連していた(図4A、図11および図12)。リバウンドウイルス配列はいずれも、Asn332gp120のN結合型グリコシル化部位を変化させた変異を含有していた(図4Bおよび図4C)。NC37による単独療法もウイルス血症を一過性に抑制したが、ウイルス量の減少の大きさは、BG18を用いた場合よりも顕著ではなく、平均0.5log10であった(図4Aおよび図12)。これは、BG18およびNC37がHIVを治療するのに有効であり、BG18が最も顕著な効果を有することを示している。NC37リバウンドウイルスEnv配列の大部分は、CD4bs抗体に対する耐性と関連するR456K突然変異を示した(図4Bおよび図4C)。これはさらに、huマウスでは、BG18およびNC37がインビボでHIV-1YU2抗体耐性変異体を選択することができることを示している。

- 前記抗体抗原またはその結合部分が組換え作製される、請求項1~3のいずれか一項に記載の単離された抗HIV-1抗体またはその抗原結合部分。
- 請求項1~4いずれか一項に記載の単離された抗HIV-1抗体またはその抗原結合部分を含む、二重特異性抗体。
- 請求項1~4のいずれか一項に記載の抗HIV-1抗体もしくはその抗原結合部分、または請求項5に記載の二重特異性抗体をコードする配列を含む、単離された核酸。
- 請求項6に記載の核酸を含む、ベクター。
- 請求項7に記載のベクターを含む、培養細胞。
- (i)請求項1~4のいずれか一項に記載の第1の抗HIV-1抗体もしくはその抗原結合部分、または請求項5に記載の二重特異性抗体と、(ii)薬学的に許容される担体とを含む、医薬組成物。
- 前記重鎖および前記軽鎖が、配列番号9および13のそれぞれの配列を含む、請求項9に記載の医薬組成物。
- 第2の抗HIV-1抗体またはその抗原結合部分をさらに含む、請求項9または10に記載の医薬組成物。
- HIV-1感染を治療するための薬剤の製造における、第1の請求項1~4のいずれか一項に記載の抗HIV-1抗体もしくはその抗原結合部分、もしくは請求項5に記載の二重特異性抗体、または請求項9~11のいずれか一項に記載の医薬組成物の治療有効量を含む第1の治療剤の使用。
- 前記治療が、第2の治療剤を投与することをさらに含む、請求項12に記載の使用。
- 前記第2の治療剤が、非ヌクレオシド逆転写酵素阻害剤、プロテアーゼ阻害剤、エントリー阻害剤または融合阻害剤、インテグラーゼ阻害剤、および第2の抗HIV-1抗体またはその抗原結合部分からなる群から選択される、請求項13に記載の使用。
- 請求項1~4のいずれか一項に記載の抗HIV-1抗体もしくはその抗原結合部分、または請求項5に記載の二重特異性抗体を含む、ワクチン組成物。
- 前記重鎖および前記軽鎖が、配列番号9および13のそれぞれの配列を含む、請求項15に記載のワクチン組成物。
- HIV-1感染を予防するための薬剤の製造における、請求項15または16に記載のワクチン組成物の使用。
