本開示の第1の態様においては、発現系が実施形態において提供される。本開示の実施形態によれば、前記発現系は、細胞特異的プロモーターを組み込む第1の核酸分子と;前記第1の核酸分子に機能可能に結合され、転写アクチベーターをコードする第2の核酸分子と;前記転写アクチベーターの第1の認識配列を組み込む第3の核酸分子と;前記第3の核酸分子に機能可能に結合され、第1のプロモーター及び第1の調節エレメントを組み込む第4の核酸分子と;前記第4の核酸分子に機能可能に結合され、第1の調節タンパク質をコードする第5の核酸分子と;前記転写アクチベーターの第2の認識配列を組み込む第6の核酸分子と;前記第6の核酸分子に機能可能に結合され、第2のプロモーター及び第2の調節エレメントを組み込む第7の核酸分子と;前記第7の核酸分子に機能可能に結合され、第2の調節タンパク質をコードする第8の核酸分子と;前記第5の核酸分子に機能可能に結合され、前記第1の調節タンパク質の発現を条件付きで阻害するように構成される第9の核酸分子、及び前記第8の核酸分子に機能可能に結合され、前記第2の調節タンパク質の発現を条件付きで阻害するように構成される第10の核酸分子からなる群から選択される少なくとも1つと、を含み、前記第1の調節エレメントが、前記第2の調節タンパク質に結合することによって前記第1のプロモーターの機能を阻害するようにされ、前記第2の調節エレメントが、前記第1の調節タンパク質に結合することによって前記第2のプロモーターの機能を阻害するようにされる。強力な制御、高い効率、及び特異性を有する本開示の実施形態に係る前記発現系を用いて、特定の細胞環境下で目的の遺伝子を特異的に発現させながら、相互抑制方式によって調節されることができる。
1)アデノウイルスは、広範囲の宿主に有用であり、ヒトに対する病原性が低く、それによってアデノウイルスベクター系は、ヒト及び非ヒトタンパク質の発現に広く用いられる。更に、アデノウイルスは、様々な哺乳類細胞に感染することができるため、殆どの哺乳類細胞及び組織における組換えタンパク質の発現に有用であることができる。アデノウイルスは、上皮細胞性(epitheliophilic)であり、ヒト腫瘍の殆どは上皮細胞から由来されることに特に留意される。更に、アデノウイルスの複製遺伝子及び病原性遺伝子は、十分に明確であり、アデノウイルスに対する抗体は、ヒト集団において存在する(70〜80%の大人は、アデノウイルスに対する中和抗体を有する)。ヒトは、野生型アデノウイルスに感染した後にのみ僅かな自己制限症状を引き起こし、リバビリンによる処置が有効である。
2)アデノウイルスは、増殖細胞及び非増殖細胞において感染し、遺伝子発現することができる。レトロウイルスは、増殖細胞のみ感染することができ、DNAトランスフェクションは、非増殖細胞で行われることができず、それによって細胞は、インキュベーション中に保持される必要がある。アデノウイルスは、アデノウイルスに対する幾つかのリンパ腫細胞を除き、ほぼ全ての細胞型に感染することができる。アデノウイルスは、非増殖性の初代細胞での遺伝子発現を研究するのに最も良好な系であり、形質転換細胞及び初代細胞から得られた結果間の直接比較を可能にする。
3)アデノウイルスは、効果的に増殖でき、高い力価を有する。アデノウイルス系は、1010〜1011VP/mlLの力価を生じさせることができ、濃縮後に最大1013VP/mLになることができる。したがって、この機能は、遺伝子治療を非常に好適にする。
4)アデノウイルスは、ヒトの遺伝子と相同である。ヒトウイルスは、一般にベクターとして用いられ、ヒト細胞は、アデノウイルスベクター系の宿主として用いられ、これは正確な翻訳後プロセッシング及びヒトタンパク質の適切なフォールディングのための理想的な環境を提供する。殆どのヒトタンパク質は、高い発現レベルを達成でき、完全に機能する。
5)アデノウイルスは、染色体に組み込まれず、挿入後に変異原性を引き起さない。レトロウイルスは、宿主の染色体にランダムに組み込まれ、遺伝子の不活性化又は癌遺伝子の活性化を引き起こす。しかしながら、アデノウイルスは、卵細胞を除く殆ど全ての公知の細胞の染色体に組み込まれないため、宿主における他の遺伝子を妨害しない。アデノウイルスの単一コピーで組み込まれた卵細胞は、特定の特性を有するトランスジェニック動物を作製するための優れた系である。
6)アデノウイルスは、懸濁培養培地で増幅されることができる。293細胞は、懸濁培養によって調整されることができるため、アデノウイルスの大幅の増幅を可能にする。293細胞の懸濁液が、1〜20Lのバイオリアクターにおいて組換えタンパク質を発現することができることが多数の事実によって示されている。
7)アデノウイルスは、複数の遺伝子を同時に発現させる能力を有する。これは、同じ細胞株又は組織において複数の遺伝子を発現するように設計された最初の発現系である。発現系の構築のための最も簡単な方法は、2つの遺伝子を含む二重発現カセットをアデノウイルス導入ベクターに挿入する、又は目的の細胞株を異なる組換えウイルスでそれぞれコトランスフェクトし、タンパク質を発現させることを含む。個々の組換えタンパク質の相対的な同時発現は、異なる組換えウイルスのMOI比を決定することにより正確に推定されることができる。

本開示の第3の態様においては、実施形態において、組換え細胞が提供される。本開示の実施形態によれば、前記組換え細胞は、上記に記載される発現系を含む。本開示の実施形態に係る組換え細胞は、人体の全身性免疫応答を効果的に活性化し、腫瘍細胞などの異種細胞を高い安全性及び特異性で攻撃することができる。
本開示の第4の態様においては、実施形態においては、癌の治療のための医薬の調製における、上記発現系、上記組換えウイルス、及び上記組換え細胞の使用が提供される。本開示に記載される発現系は、目的のタンパク質を腫瘍細胞で特異的に発現させることができる。本開示の実施形態に係る薬剤は、より効果的、特異的、及び安全な方法で、癌を治療することができる。この実施形態においては、本発明者らは、合成生物学の概念に従って、アデノウイルス転写に関連する重要な遺伝子E1Aの発現を調節するために、複数の標的に応答した遺伝子回路を設計した。同時に、そのような同時発現は、全身性免疫応答に関連するサイトカイン又は抗体遺伝子を活性化することができる。第一に、本発明者らによって設計された遺伝子回路は、従来の腫瘍溶解性アデノウイルスとは異なり、アデノウイルスのパッケージングを複数のレベルで調節できる。第1の工程においては、本発明者らによって、腫瘍−特異的プロモーターが用いられ、遺伝子回路においてマスタースイッチGal4VP16の発現を調節する。第2の工程においては、遺伝子回路におけるmicroRNAの標的配列は、異なる細胞株におけるmicroRNAの発現に応じて、非腫瘍細胞から腫瘍細胞を区別することができる。第3の工程においては、安定した相互抑制スイッチが、細胞特異的プロモーター及びmicroRNAシグナルなどの外部入力シグナルに効率的に応答することができる本発明者らによって設計された遺伝子回路で用いられ、前記遺伝子回路は、入力シグナルの差を更に拡大することができ、非腫瘍細胞から腫瘍細胞を区別することにおいてより効率的であることができる。第4の工程においては、本発明者らによって設計されたアデノウイルスベクターにおける発現系は、アデノウイルスE1B遺伝子を含まず、E1B遺伝子は、正常細胞内のP53遺伝子と相互作用し、正常細胞における成功した(successful)アデノウイルスの増殖を保証することができる。E1B遺伝子を有さないアデノウイルスは、P53を発現する正常細胞では増殖できないが、一方P53遺伝子を欠く腫瘍細胞では更に効率的に増殖することができる。したがって、実施形態におけるアデノウイルスベクターは、マルチレベル調節により、従来の腫瘍溶解性アデノウイルスよりも、より特異的で安全である。更に、この操作されたアデノウイルスは、実施形態において様々なサイトカイン及び抗体を発現する遺伝子を有するためのベクターとして用いられる。したがって、アデノウイルスは、腫瘍溶解性効果を示すだけでなく、同時発現因子を使用することにより、全身性免疫応答を活性化することもでき、腫瘍溶解性アデノウイルスの有効性を改善する。
本開示の第5の態様においては、実施形態において、発現系を用いることによって目的のタンパク質を発現させる方法が、提供される。前記発現系は、上記に記載される発現系である。本開示の実施形態によれば、前記方法は、(1)前記目的のタンパク質をコードする核酸配列を含む第5の核酸分子を提供することと;(2)第10の核酸分子によって、第2の調節タンパク質の発現を阻害し、前記目的のタンパク質を発現させることと、を含む。実施形態において発現系を用いて目的のタンパク質を発現させる方法を用いて、前記第1のプロモーターにおける前記第2の調節タンパク質媒介抑制メカニズムが解放され、それによって前記目的のタンパク質は、前記細胞特異的プロモーター及び前記第1のプロモーターの相互作用下で特定の細胞で効果的に発現される。
1.細胞培養及びトランスフェクション
HEK293(293−H)細胞株は、Invitrogen Co.,Ltdから購入した。ヒト肝癌細胞株HepG2、正常肝細胞Chang及びHepa1−6は、ATCCから購入した。ヒト肝癌細胞株Huh7は、BeNa Co.,Ltdから購入した。細胞を、4.5g/Lのグルコース、0.045ユニット/mLのペニシリン、0.045g/mLのストレプトマイシン、及び10%FBS(Invitrogen Co.,Ltd.から購入)を含む高グルコースダルベッコ変法イーグル培地(即ち、DMEM完全培地)で、37℃、湿度100%、5%CO2濃度にて培養した。
この実施例では、2つの初期アデノウイルスベクター配列を用いる。即ち、ViraPowerTM Adenoviral GatewayTM Expression Kit(K49300 Invitrogen)とAdeno−X−Adenoviral System 3(632266 Clontech)である。これらの2つのアデノウイルスベクターの構造をそれぞれ図1と図2に示す。
トランケートされたAFPプロモーター誘導体に関連するプラスミドを構築した。AFPエンハンサー及びAFPプロモーター配列は、PCR増幅を介して、特異的に設計されたPCRプライマーの下で肝癌細胞株のゲノムから直接得た。AFPプロモーター配列の点突然変異は、オーバーラップエクステンションPCR法を使用して取得した。AFPエンハンサー及びAFPプロモーター配列は、エントリークローン中に構築され、エントリークローンの両端に特異的に設計されたLRクロナーゼの認識配列が存在する。特異的プロモーターと活性化遺伝子Gal4VP16は、LR反応により1つの同一ベクター中に構築した。microRNAの標的配列は、プライマーアニーリング、酵素消化、及びライゲーションによりCMV−EYFPベクター(XbaI/SalI)に挿入した。発現系の一次エレメントライブラリーは、ゴールデンゲート法によって構築した。タイプIIs制限酵素(例えば、BsaI、BbsI、BsmBI、SapIなど)は、その認識部位が非パリンドロームであり且つ対称な配列であり、その切断部位が認識部位の外側に位置する特定のクラスの制限酵素である。これは、その認識部位がパリンドロームであり且つ対称な配列であり、その切断部位が認識配列と重複している分子生物学で最も一般的に使用されるタイプII制限酵素とは異なる。したがって、タイプIIs制限酵素によって消化された2つのフラグメントを後続の消化及びライゲーション反応でライゲーションすると、以前あった認識部位が存在しないため再度は切断されることがなく、接続(welding)の効果が得られる。ゴールデンゲートクローニング法では、タイプIIs制限酵素の特性により、異なる切断部位配列が認識配列の外側に人工的に設計される。これにより、1つの同一のタイプIIs制限酵素が異なる付着末端を生成することができるので、複数のフラグメントを一度に組み立てることを確実にし、複数の異なる制限酵素を使用するなどの、従来のマルチフラグメントアセンブリの欠点を克服する。このゴールデンゲートクローニング法の最大の特徴は、従来の消化連結法と比較して、更なる「傷跡(scar)」が導入されず、DNAフラグメントのシームレスな連結が達成されることである。この実施例の一次エレメントライブラリーで使用されるタイプIIs制限酵素は、Esp3Iである。発現系の2番目の論理回路ライブラリーも、IIs型制限酵素BsaIを使用して、ゴールデンゲート法によって構築した。次に、ゲートウェイ(Gateway)法によりViraPowerTM Adenoviral GatewayTM Expression Kitのアデノウイルスベクター配列に2番目の論理回路を挿入した、又はギブソン(Gibson)クローニングによりAdeno−X Adenoviral System 3のアデノウイルスベクター配列に挿入した。ゲートウェイ技術は、BP及びLR反応からなる、十分に研究されたファージλ部位特異的組換え系(attB×attP→attL×attR)に基づく。BP反応は、attB DNAフラグメント又は発現クローンと、attPドナーベクターとの間の組換え反応であり、エントリークローンを作成する。LR反応は、attLエントリークローンとattR宛先ベクターとの間の組換え反応であり、並行反応における1以上の宛先ベクターに目的の配列を挿入するために使用される。本発明者らは、この実施例においてLR反応を使用した。「ギブソンサーモスタットワンステップアセンブリ法」としても知られるギブソンアセンブリ技術は、米国のJ.CraigVenter Instituteでギブソンらによって作成されたDNAアセンブリ法であり、T5エキソヌクレアーゼ、DNAポリメラーゼ、及びリガーゼの相乗効果の下で、重複する末端配列を有する複数のDNAフラグメントがin vitroで組み立てられる。中でも、T5エキソヌクレアーゼは、5’→3’エキソヌクレアーゼ活性を有し、DNAフラグメントの一方の鎖の5’末端から重複末端配列を有するDNAフラグメントを切断し、相補鎖の3’末端に重複配列を有するオーバーハング末端を生成する。この相補鎖は、50℃で特異的にアニーリングされ(この温度で、T5エキソヌクレアーゼは徐々に不活性化される)、最後に、DNAポリメラーゼとTaqリガーゼの存在下で完全なDNA分子が形成され、シームレスなアセンブリが達成される。
細胞RNAはTrizol法により抽出した。1mLのTrizolを遠心分離によりペレット化した5×106個の細胞に添加した後、ピペットで繰り返しピペッティングする又は激しく振とうして細胞を溶解した。15〜30℃の室温で5分間放置した後、0.2mLのクロロホルムを添加し、混合物を15秒間激しく振とうし、室温で更に2〜3分間維持し、12,000×gで4℃にて15分間、遠心分離した。0.5mLのイソプロパノールを、分離した上部の水層に添加し、室温で10分間放置し、12,000×gで4℃にて10分間、遠心分離した後、1mLの75%エタノールを添加し、ボルテックスし、7500×gで4℃にて5分間、遠心分離した。上清を廃棄し、沈殿したRNAを室温で自然乾燥させた。乾燥したRNAをRNaseフリー水に溶解させた。QuantiTect Rev. Transcription Kit(カタログ番号/ID:205310 QIAGEN)を使用して、逆転写実験を行った。定量的PCRプライマーは、qAFP for:CAAAGCTGAAAATGCAGTTGAATG(配列番号15)、qAFP rev:TTCCCCATCCTGCAGACAATCC(配列番号16)、GAPDH for:AGAAGGCTGGGGCTCATTTG(配列番号17)、及びGAPDH rev:AGGGGCCATCCACAGTCTTC(配列番号18)である。蛍光色素は、PowerUpTM SYBR(登録商標)GreenMaster Mix(A25741 Thermo Fisher)である。
高純度のエンドトキシンフリー組換えアデノウイルスプラスミドを抽出した。HEK293細胞に、線状化したアデノウイルスDNAをトランスフェクトし、これを一晩PacIで消化した後、培養培地を完全培地に交換した。培養1週間後に、細胞を培養培地1mLと共に15mL遠心管に回収した(培養中の細胞増殖状態に応じて新鮮な培地を添加)。次いで、これを液体窒素による37℃での3回連続の凍結融解サイクルにより溶解させ、2000rpmで5分間、遠心分離した。上清を一次ウイルス溶液として得た。連続する3世代のトランスフェクション後、得られたアデノウイルスを増幅のために10枚の15cmプレートに播種し、CsCl密度勾配遠心分離と透析との組合せにより更に精製した。CsCl密度勾配は、1.4g/mLの密度のCsCl溶液2mLを添加し、更に1.3g/mLの密度のCsCl溶液3mLをゆっくりと添加した後、5mLのウイルス懸濁液を添加することにより調製した。20000rpm、4℃にて10時間の遠心分離後、1.3g/mL〜1.4g/mLの密度のCsCl溶液に濃縮されたアデノウイルスを透析バッグに回収し、使用前に10分間、10mM EDTA−Na2で煮沸した。透析バッファーは、50gのスクロース、10mLの1M Tris−HCl、及び2mLの1M MgCl2を使用して調製し、水で1Lにし、pH8.0に調整する。4℃で一晩攪拌し、透析バッファーを2回交換した後、ウイルス力価の測定のためにアデノウイルスを回収した。ウイルス力価は、感染細胞から抽出されたウイルスDNAを定量することにより検出した。具体的には、約2×106のHeLa細胞/ウェルを6ウェル組織培養プレートに播種し(インキュベーション1日間後に細胞被覆率が90〜100%に達する)、無血清培地500μLで希釈したウイルスストックの5μLにて2連で感染させた後、37℃で3〜6時間インキュベートし、培養培地を除去し、1mLのPBSで2回洗浄した。500μLの新鮮なNP−40溶解バッファー(0.65%NP−40(Calbiochem、Billerica、MA、USA)、150mMのNaCl、10mMのTris、pH8.0を含む)を添加し、室温で5〜10分間インキュベートすることにより細胞を溶解した。ここで、完全に溶解するために10〜15回ピペッティングした後、ウイルス沈殿のために2000×gで3分間、遠心した。上清を廃棄した後、1mLのNP−40溶解バッファーを添加し、短時間ボルテックスして、2000×gで3分間、遠心分離した。上清を再び廃棄し、沈殿物を200μLのPBSに懸濁した。DNAの単離及び精製中に、DNeasy Blood&Tissue Kit(QIAgen、Valencia、CA、USA)を使用してDNAを抽出し、その後の定量反応に使用した。定量プライマーは、L2フォワードプライマー:TTGTGGTTCTTGCAGATATGGC(配列番号19)、及びL2リバースプライマー:TCGGAATCCCGGCACC(配列番号20)である。
アデノウイルスを、各種細胞に対する認識能力と殺滅能力について検出した。Chang細胞(1×104細胞/ウェル)、HepG2及びHuh7細胞(それぞれ5×104細胞/ウェル)を96ウェルプレートに播種し、102〜10−4に10倍希釈された7種の感染多重度(MOI)勾配でアデノウイルスベクターに感染させた。ここで、6日間の観察後に、MTS法にて細胞生存率を測定した。
細胞数はMTS法で測定した。20μLのMTS/PMS混合物(Promega)を、試験対象細胞を含む96ウェルプレートの各ウェルに添加した後、混合し、37℃で1〜4時間インキュベートした。発色後、プレートを10秒間振とうして均一な色を得た。マイクロプレートリーダーを使用して、490nmの波長での吸光度(OD値)を測定した。標準曲線を確立した。目的細胞の数は、標準曲線を参照して検出した。
サイトカインの生物活性は、サイトカイン依存性細胞株の増殖を介して検出できる。IL−2、mGM−CSF、及びhGM−CSFの生物学的活性は、それぞれCTLL−2、FDC−P1、及びTF−1細胞株を使用して検出した。回収後、1×104個の試験対象のサイトカイン依存性細胞を96ウェルプレートに播種し、試験対象のサイトカインを含むサンプルを感染させ、37℃で24時間又は48時間、恒温培養した。増幅した細胞をMTS法により測定した。用量増殖標準曲線を確立した。測定したサンプル中のサイトカインの生物活性は、標準曲線を使用して推定した。
共免疫沈降は、以下のようにして行った。適切な量のタンパク質溶解バッファー(Biyuntian Co.,Ltd.)を、試験対象の回収した細胞に添加し、遠心分離して細胞片を除去した。後続のウエスタンブロット分析のために、少量のタンパク質溶解バッファーを残した。残りのタンパク質溶解バッファーと、対応するタグ抗体(Thermo Fisher Scientific Co.、Ltd.)をコンジュゲートさせた10〜30μLのプロテインA/G−アガロースビーズとを、得られた細胞溶解物に添加し、軽く振とうしながら4℃で一晩インキュベートした後、遠心分離して上清を除去した。沈殿したビーズを3回洗浄した後、溶離液を使用して抗体及び捕捉されたタンパク質をアガロースビーズから溶出し、次いで、SDS−PAGEローディングバッファー(Shanghai Shenggong Co.,Ltd.)を添加して煮沸した。関連タンパク質の濃縮は、ウエスタンブロット法によって検出した。
実験動物は、HuafuKang Co.,Ltd.又はWeitonglihua Co.,Ltd.から購入した。腫瘍モデルは、SPF環境で飼育された6週齢の雄又は雌のマウスを使用して構築した。ヒト腫瘍モデルでは、6週齢のヌードマウスの右側腹部にマトリゲル(BD Co.,Ltd.)中の1×107個のヒト肝癌細胞HepG2を皮下注射した。マウス腫瘍モデルでは、1×106個のマウス肝癌細胞Hepa1−6を、6週齢の野生型C57BL/6Jマウスの右側腹部に皮下注射した。皮下腫瘍サイズは、3日ごとにノギスで測定し、次の式を使用して計算した:体積=0.5×長さ×幅2。腫瘍体積が100mm3を超えた後、腫瘍溶解性アデノウイルス又は対照処置を行った。動物実験倫理の要求にしたがって、腫瘍サイズが直径15mmに到達後、担腫瘍マウスを安楽死させた。
1.合成生物学の使用によって癌細胞を非癌細胞から区別する各種バイオマーカーに対応する相互抑制スイッチの構築
近年、関連する研究により、腫瘍溶解性ウイルスが癌の治療に非常に有効であり、腫瘍溶解性アデノウイルスに関する多くの臨床研究が開発されていることが示されている。既存の臨床研究結果によると、腫瘍溶解性アデノウイルスのリークの割合が比較的高いので、臨床段階で腫瘍溶解性アデノウイルスの安全性をいかにして改善するかが重要な課題である。既存の腫瘍溶解性アデノウイルスの分析に基づくと、大部分の腫瘍溶解性アデノウイルスは、アデノウイルス複製の鍵となる遺伝子E1の発現を調節する腫瘍特異的プロモーターなど、単一のバイオマーカーに応じて調節される。しかし、1つのバイオマーカーだけが、比較的高いリーク確率を引き起こすことがある。したがって、本発明者らは、この実施例において、各種バイオマーカーに対応する相互抑制スイッチを設計し、様々なレベルでアデノウイルスE1A遺伝子の発現を調節することにより、認識に対する腫瘍溶解性アデノウイルスの特異性を改善し、安全性を向上させた。
2.1肝癌特異的アルファ−フェトプロテイン(AFP)プロモーターの構築
アルファ−フェトプロテイン(αFP又はAFP)は、主に胎児の肝臓で合成され、健常な成人では発現しないが、肝細胞が癌になると再び発現し、肝癌の進行に伴って血清中のAFP量は劇的に増加する。現在、アルファ−フェトプロテインは、原発性肝癌の診断のための特異的臨床指標であるので、本発明者らは、実施例において肝癌細胞を非肝癌細胞と区別するためのプロモーターマーカーとしてアルファ−フェトプロテインプロモーターを選択した。
最近の研究から、腫瘍の形成と成長が、microRNA(miRNA)の発現レベルを大きく変えることが示されている。表1に示すように、各種癌細胞で様々なmicroRNAが高レベルに発現している。本発明者らは、正常な肝細胞の高発現を特徴付けるmicroRNAマーカーとしてmiR199aを選択し、同様に、既知の研究結果の分析に基づき、肝癌細胞の高発現を特徴付けるmicroRNAマーカーとしてmiR21及びmiR122を選択した。まず、本発明者らは、microRNA蛍光プラスミドレポーター系を設計、構築した。図7に示すように、本発明者らは、microRNAの標的配列を、通常発現されるEYFPの3’非翻訳領域に挿入し、別の通常発現されるEBFPを蛍光対照として使用した。幾つかの異なる細胞株に、EYFP及びEBFP蛍光プラスミドをそれぞれトランスフェクトして、microRNA標的配列の発現を検出した。実験結果を図8に示す。これは、miR199aが肝癌細胞株と比較して正常肝細胞で高度に発現している一方で、miR21は、肝癌細胞株で有意に高い発現レベルを示しているので、miR199aとmiR21を、肝癌細胞と正常細胞を区別するために本発明者らが設計した相互抑制スイッチの有効なマーカーとして使用できることを示している。
腫瘍溶解性アデノウイルスの限られたパッケージング能力(最大8Kbpの外来遺伝子しかパッケージできない)のため、本発明者らは、この実施例ではできる限り短く有効な阻害エレメントを選択した。本発明者らは、この実施例でLacI及びtetR−KRABを選択した。図9に示すように、本発明者らは、EYFP遺伝子がtetR−KRAB側で共発現されるかどうかに応じて2つの論理回路、即ち、EYFPを発現しないSwitch I(SI)と、EYFPを発現するSwitch II(SII)を構築した。2つのスイッチを個別にHEK293細胞に一過性トランスフェクトし、それぞれを、shRNA−FF4及びshRNA−FF5のそれぞれで共トランスフェクトした。図10に示すように、これらの2つのスイッチはいずれも、各種入力信号下で効率的に反転できる。SIは、LacI側でより高いレベルで発現するが、tetR−KRAB側ではSIIよりも高いリークレベルを示す。総合的に考慮した後、本発明者らは、後続の実験で異なるエフェクターを有する本腫瘍溶解性アデノウイルスを構築するためのバックボーンとしてSI系を選択した。これは、SIスイッチ系が、各種入力信号に応答し、目的の細胞中でE1遺伝子を効率的に発現することができるからである。実験室での過去の結果に基づいて、本発明者らは、相互抑制スイッチSIに対する各種アクチベーターの調節効果を試験した。図11に示すように、本発明者らは、アクチベーターGal4VP16、Gal4esn、dCas9−VP64、及びrtTAの調節効率を試験した。実験結果から、遺伝子回路がこれらの各種アクチベーターに効果的に応答することができ、そのような遺伝子回路の安定性を証明していることが示されている。
多くの臨床情報は、腫瘍マーカーの発現が個人によって大きく異なることを示す。したがって、本発明者らは、各個人に異なるマーカーを選択することができる。個々の患者の正確な治療を達成するために、本発明者らは、各種バイオマーカーを有する様々な腫瘍溶解性アデノウイルスを調製する必要がある。しかし、これらの腫瘍溶解性アデノウイルスを従来の酵素消化及びライゲーション法により迅速に構築することは非常に困難である。本実施例において、本発明者らは、3段階ライブラリーを設計、構築し、CGGA法(カスケードゴールデンゲートゲートウェイ/ギブソンアセンブル法)により腫瘍溶解性アデノウイルスを迅速に改変した。まず、癌細胞と正常細胞の区別に関連するマーカーエレメント(例えば、腫瘍特異的プロモーター、論理回路関連抑制エレメント、癌細胞を区別するmiRNA認識配列など)を一次プラスミド上に構築することにより、図12に示す一次エレメントライブラリーを作製した。一次エレメントの2つの端部にはそれぞれ、EI(認識部位と切断部位が異なる制限酵素、例えば、Esp3IやBsaIなど)の認識部位が挿入され、ゴールデンゲート法により複数の一次エレメントを1つの二次発現系に構築する。発現系は、以下のように構築される。具体的には、複数の一次エレメントが一次エレメントライブラリーから選択され、ゴールデンゲート法によって3つの発現系に組み立てられる。このような3つの発現系は、腫瘍特異的プロモーター系、第1の調節エレメント媒介抑制系、及び第2の調節エレメント媒介抑制系を含む。次いで、図13に示すように、ゴールデンゲート法によって3つの発現系をランダムに組み立てることにより、完全な論理回路が形成される。更なる実験のために、論理回路の両端に、ギブソン相同配列又はゲートウェイ認識部位(標的配列)が挿入される。最後に、論理回路は、図14に示すように、ギブソン法又はゲートウェイ法によってアデノウイルスベクター(E1及びE3の一部を除去)に構築される。操作されたアデノウイルスベクターは、PacI消化により線状化され、特定の細胞にトランスフェクトされ、アデノウイルス粒子をパッケージングする。したがって、1以上の腫瘍バイオマーカーを、本発明者らによって置き換えることができる。
本発明者らによって構築された腫瘍溶解性アデノウイルスにおける相互抑制スイッチが、肝癌細胞を効果的に区別し、殺滅できるかどうかを検出するために、本発明者らは、腫瘍溶解性アデノウイルスのin vitro細胞殺滅アッセイを設計した。各種細胞株に、様々な感染多重度で腫瘍溶解性アデノウイルスを感染させた。ここで、Chang細胞は正常な肝細胞であり、Huh7及びHepG2細胞はヒト肝癌細胞株である。細胞の生存率は、インキュベーションの6日後にMTS法で測定した。図15に示すように、感染の多重度がそれぞれ100、10、1の場合、肝細胞腫細胞は重篤な細胞毒性により重度の死滅を示し、肝癌細胞は0.1の感染多重度において僅かな細胞死を示すことが分かった。同様に、感染多重度が10以下の場合、Chang細胞は、細胞死を示さずに正常な増殖状態である一方、感染多重度100では僅かな細胞毒性応答を示し、細胞体積が大きくなり、細胞成長が遅くなった。このデータは、特定の感染多重度の範囲内でパッケージされたアデノウイルスが、正常な肝細胞の成長と増殖に干渉することなく、肝癌細胞を効果的に区別して殺滅できることを示している。
現在臨床診療で使用されているアデノウイルスベクターは、自然界に広く存在するヒトAd2又はAd5型アデノウイルスである。更に、これらのアデノウイルスに拮抗する中和抗体は、人体に存在することが知られている。したがって、臨床データは、アデノウイルス単独の腫瘍溶解効果に依存する腫瘍治療の治療効果が低いことを示した。そのため、本発明者らは、実施例において、腫瘍部位にエフェクター遺伝子を運ぶ送達系として腫瘍溶解性アデノウイルスを使用し、全身性免疫応答を引き起こすようにした。図16に示すように、本発明者らは、まず、これらのエフェクター遺伝子を高頻度に発現するプラスミドベクターに構築し、一過性トランスフェクションによりこれらのエフェクターの活性を検出した。結果は、本発明者らによって構築されたエフェクター遺伝子が、生物学的機能を有する免疫因子、及び免疫学的チェックポイント遺伝子の抑制因子を効率的に発現できることを示している。本発明者らは、腫瘍溶解性アデノウイルスベクター上でこれらのエフェクター遺伝子を更に構築して、パッケージング及び精製後にこれらのエフェクターを発現できる腫瘍溶解性アデノウイルスを得た。本発明者らは、肝癌細胞HepG2の感染後、上清を抽出して、上清中の免疫因子の活性を検出した。
図18に示すように、1×107個のHepG2、Huh7、及びHepa1−6肝癌細胞をヌードマウスの右側腹部に皮下移植し、3日ごとに皮下腫瘍体積を測定した。HepG2及びHuh7細胞はヒト肝癌細胞であり、Hepa1−6細胞はマウス肝癌細胞である。腫瘍体積が100mm3を超えた後、マウスをランダムに2つの群に分け、処置群には、1×109個の腫瘍溶解性アデノウイルスを腫瘍内注射し、対照群には、PBSを注射した。処置後の腫瘍体積の変化を継続的に観察した。PBSを注射した対照群の腫瘍は成長を続け、腫瘍体積が増大していくのに対し、処置群の腫瘍は、腫瘍溶解性アデノウイルスの注射後に有意に抑制されており、腫瘍溶解性アデノウイルスが、腫瘍細胞の溶解によってin vivoでの腫瘍成長を抑制でき、形成された腫瘍に対する顕著な治療効果を発揮することを示す。マウス肝癌の処置において、本発明者らは、非腫瘍溶解性ウイルス(第二世代アデノウイルスベクター、GFP遺伝子含有ウイルス、Ad−GFP)を別のウイルス対照として使用した。実験結果から、PBS及びAd−GFP処置群の腫瘍は成長し続け、腫瘍溶解性ウイルス処置群は腫瘍HepG2及びHuh7の成長がある程度抑制され、腫瘍Hepa1−6の成長を僅かに抑制することが分かる。これは恐らく、HepG2及びHuh7細胞よりもHepa1−6細胞の成長速度が速いことと、Hepa1−6細胞でのAFPの発現レベルが比較的低いことに起因する。したがって、本発明者らは、後続の実験において、免疫エフェクター含有腫瘍溶解性アデノウイルスを使用してHepa1−6腫瘍モデルを処置し、非常に効果的な結果を達成した。
C57BL/6マウスの左右側腹部に、マウス肝癌細胞Hepa1−6を皮下注射した。腫瘍塊の形成後、サイトカインhIL−2、mGM−CSF、及び抗PD−1 scFvをそれぞれ発現する腫瘍溶解性アデノウイルスを、右側腹部の腫瘍に注射した。対照としてPBS及びAdGFPを注射した。腫瘍成長を継続的に測定した。処置の14日間後の担癌マウスの左右側腹部の腫瘍における浸潤リンパ球を単離精製し、腫瘍内の浸潤T細胞の表現型をフローサイトメトリー法により検出した。本発明者らは、Ki−67+T細胞又はインターフェロン−γを発現しているT細胞の割合が、腫瘍溶解性アデノウイルスを右側腹部に注射した腫瘍だけではなく、腫瘍溶解性アデノウイルスを注射しなかった左側腹部の腫瘍においても、対照群に比べて高いことを見出した。腫瘍溶解性アデノウイルスの処置によって引き起こされる免疫応答は、注射部位の腫瘍だけでなく、遠位側の腫瘍でも機能できるが示される。これは、腫瘍溶解性アデノウイルスが注射部位の腫瘍に治療効果があるだけでなく、遠位転移の腫瘍にも治療効果を示すことを示唆する。
図22に示すように、腫瘍溶解性アデノウイルスの処置プロセスを、非感染腫瘍細胞、感染腫瘍細胞、遊離ウイルス、腫瘍溶解性アデノウイルス抗原活性化免疫細胞、及び腫瘍抗原活性化免疫細胞(これらの5種の成分をS、I、V、ZV、及びZTでそれぞれ表す)を含む腫瘍ウイルス免疫系に抽象化する。モデルは、非感染腫瘍細胞の成長が一般化されたロジスティック成長モデルに適合することを想定しており、γは成長速度を示し、Kは収容力を示し、εは非線形性を示す。非感染腫瘍細胞に遊離ウイルスを感染させ、κの割合で感染腫瘍細胞に変換することができる。感染腫瘍細胞は速度δで溶解し、速度αで大量の遊離ウイルスを放出する。遊離ウイルスは、環境内で、速度ωで自然減衰する。ZVは、腫瘍溶解性アデノウイルス又はウイルス感染腫瘍細胞の表面抗原の活性化によってそれぞれcV1及びcV2の速度で増加する一方、速度pVで感染腫瘍細胞の生成を阻害する。ZTは、溶解した腫瘍細胞から放出される抗原の活性化により速度cTで増加し、速度pTで感染及び非感染腫瘍細胞の生成を阻害する。ZV及びZTは、環境内で、速度βで自然減衰する。
シミュレーション結果から、系は、完全な腫瘍の除去、飽和に向かう腫瘍成長、安定した腫瘍体積、及び不安定な腫瘍体積に対する傾向を含む幾つかの動的な挙動を示すことが分かる。一般に、腫瘍に対する免疫細胞の阻害効果が強いほど、系により多くの腫瘍が除去され、一方、ウイルスに対する免疫細胞の抑制効果が強いほど、系内のより多くの腫瘍が制御不能となる。しかし、図24に示すように、系は、腫瘍の安定性と不安定性の傾向に非単調な変化を示す。

- 前記免疫エフェクターが、PD−1遺伝子をアンタゴナイズする阻害配列、PD−L1遺伝子をアンタゴナイズする阻害配列、CTLA4遺伝子をアンタゴナイズする阻害配列、Tim−3遺伝子アンタゴナイズする阻害配列、GM−CSF、IL−2、IL−12、又はIL−15からなる群から選択される少なくとも1つの配列を含む請求項11に記載の発現系。
- 前記目的のタンパク質及び前記第1の調節タンパク質が、融合タンパク質の形態で発現され、前記目的のタンパク質及び前記第1の調節タンパク質が、切断可能なリンカーペプチドによって結合される請求項10に記載の発現系。
- 前記第9の核酸分子及び前記第10の核酸分子が、それぞれ独立して、RNA干渉によって、前記第1の調節タンパク質又は前記第2の調節タンパク質の発現を阻害する請求項1に記載の発現系。
- 前記第1のmicroRNAが、miR199a、miR95、miR125、miR25b、Let−7、miR143、miR145、及びmiR200Cからなる群から選択される少なくとも1つを含む請求項1に記載の発現系。
- 前記第2のmicroRNAが、miR21、miR223、miR224、miR221、miR18、miR214、miR146a、及びmiR1792からなる群から選択される少なくとも1つを含む請求項1に記載の発現系。
- 前記第1の核酸分子及び前記第2の核酸分子が、第1の発現ベクターにローディングされ;前記第3の核酸分子、前記第4の核酸分子、前記第5の核酸分子、及び任意に前記第9の核酸分子が、第2の発現ベクターにローディングされ;前記第6の核酸分子、前記第7の核酸分子、前記第8の核酸分子、及び任意に前記第10の核酸分子が、第3の発現ベクターにローディングされる請求項1に記載の発現系。
- 前記第1の発現ベクター、前記第2の発現ベクター、及び前記第3の発現ベクターが、それぞれ独立して、プラスミド、ウイルス、ナノ物質、リポソーム、分子結合ベクター、ネイキッドDNA、染色体ベクター、及びポリマーの少なくとも1つから選択される請求項19に記載の発現系。
- 前記ウイルスが、アデノウイルス、ワクシニアウイルス、ヘルペスウイルス、及びレトロウイルスからなる群から選択される少なくとも1つを含む請求項20に記載の発現系。

任意に、前記第3の発現ベクターが、配列番号8に示されるヌクレオチド配列を有する核酸を有する請求項19に記載の発現系。 - 前記アデノウイルスが、5’末端から3’末端に、第1の末端逆位配列、パッケージングシグナル、AFPIII、Gal4VP16、5×UAS、tetO、miniCMV、tetO、E1A、2A、免疫エフェクター、2A、LacI、microRNA199a特異的認識配列、5×UAS、LacO、miniCMV、LacO、tetR−KRAB、microRNA21特異的認識配列、アデノウイルスE2遺伝子領域、アデノウイルスE3遺伝子領域、アデノウイルスE4遺伝子領域、及び第2の末端逆位配列を含み、
任意に、前記アデノウイルスベクターが、配列番号9〜14からなる群から選択されるヌクレオチド配列を有する核酸を有する請求項23に記載の発現系。 - 前記アデノウイルスが、
アデノウイルスベクターから、アデノウイルス複製及びパッケージングに関連する前記アデノウイルスE1遺伝子及び前記アデノウイルスE3遺伝子の一部を除去する工程と;
段階的ゴールデンゲート(Golden Gate)法によって、遺伝子回路に前記アデノウイルスE1A遺伝子を挿入する工程と;
ゲートウェイ(Gateway)法又はギブソン(Gibson)法によって、前記アデノウイルスベクターに前記遺伝子回路を組み込む工程と、によって得られる請求項27に記載の発現系。 - αフェトプロテイン特異的プロモーターである腫瘍細胞特異的プロモーターを組み込む第1の核酸分子と;
前記第1の核酸分子に機能可能に結合され、Gal4VP16である転写アクチベーターをコードする第2の核酸分子と;
前記転写アクチベーターの5×UASである第1の認識配列を組み込む第3の核酸分子と;
前記第3の核酸分子に機能可能に結合され、第1のプロモーター及び第1の調節エレメントを組み込む第4の核酸分子であって、前記第1のプロモーターが、miniCMVであり、前記第1の調節エレメントが、複数の繰り返されたtetO配列を含み、前記複数の繰り返されたtetO配列の少なくとも1つが、前記第1のプロモーターの下流に設定される第4の核酸分子と;
前記第4の核酸分子に機能可能に結合され、LacIである第1の調節タンパク質をコードする第5の核酸分子であって、
前記第5の核酸分子が、目的のタンパク質をコードする配列を更に含み、前記目的のタンパク質が、ウイルス性複製タンパク質、好ましくはアデノウイルス複製タンパク質及びパッケージングタンパク質、並びに免疫エフェクターを含み、前記免疫エフェクターが、個々のタンパク質又は融合タンパク質の形態で発現され、前記ウイルス性複製タンパク質及び前記免疫エフェクターが、切断可能なリンカーペプチドによって結合され、
前記目的のタンパク質及び前記第1の調節タンパク質が、融合タンパク質の形態で発現され、前記目的のタンパク質及び前記第1の調節タンパク質が、切断可能なリンカーペプチドによって結合され;
前記転写アクチベーターの5×UASである第2の認識配列を組み込む第6の核酸分子と;
前記第6の核酸分子に機能可能に結合され、第2のプロモーター及び第2の調節エレメントを組み込む第7の核酸分子であって、前記第2のプロモーターが、miniCMVであり、前記第2の調節エレメントが、複数の繰り返されたLacO配列を含み、前記複数の繰り返されたLacO配列の少なくとも1つが、前記第2のプロモーターの下流に挿入される第7の核酸分子と;
前記第7の核酸分子に機能可能に結合され、tetR−KRABである第2の調節タンパク質をコードする第8の核酸分子と;
前記第5の核酸分子に機能可能に結合され、前記第1の調節タンパク質の発現を条件付きで阻害する第9の核酸分子であって、前記第9の核酸分子が、正常細胞特異的microRNAである第1のmicroRNAによって特異的に認識される核酸配列を含む第9の核酸分子と;
前記第8の核酸分子に機能可能に結合され、前記第2の調節タンパク質の発現を条件付きで阻害する第10の核酸分子であって、前記第10の核酸分子が、腫瘍細胞特異的microRNAである第2のmicroRNAによって特異的に認識される核酸配列を含む第10の核酸分子と、
を含み、
前記第1の調節エレメントが、前記第2の調節タンパク質に結合することによって前記第1のプロモーターの機能を阻害し、
前記第2の調節エレメントが、前記第1の調節タンパク質に結合することによって前記第2のプロモーターの機能を阻害することを特徴とする組換えウイルス。 - 前記組換えウイルスが、アデノウイルス、ワクシニアウイルス、レトロウイルス、及びヘルペスウイルスからなる群から選択される少なくとも1つを含む請求項29に記載の組換えウイルス。
- 前記組換えウイルスが、アデノウイルスである請求項29に記載の組換えウイルス。
- 前記免疫エフェクターが、PD−1遺伝子をアンタゴナイズする阻害配列、PD−L1遺伝子をアンタゴナイズする阻害配列、CTLA4遺伝子をアンタゴナイズする阻害配列、Tim−3遺伝子をアンタゴナイズする阻害配列、IL−2、IL−12、IL−15、又はGM−CSFからなる群から選択される少なくとも1つの配列を含む請求項29に記載の組換えウイルス。
- 請求項1から28のいずれかに記載の発現系を含むことを特徴とする組換え細胞。
- 前記発現系の少なくとも一部が、前記組換え細胞のゲノムに組み込まれる請求項33に記載の組換え細胞。
- 癌の治療のための医薬の調製における、請求項1から28のいずれかに記載の発現系、請求項29から32のいずれかに記載の組換えウイルス、及び請求項33から34のいずれかに記載の組換え細胞の使用。
- 前記癌が、肝癌、肺癌、結腸直腸癌、メラノーマ、乳癌、又は前立腺癌を含む請求項35に記載の使用。
- 請求項1から28のいずれかに記載の発現系を用いることによって、目的のタンパク質を発現させる方法であって、前記方法が、
(1)前記第5の核酸分子が前記目的のタンパク質をコードする核酸配列を含むような請求項10に記載の発現系を提供する工程と;
(2)前記第10の核酸分子によって、前記第2の調節タンパク質の発現を阻害し、前記目的のタンパク質を発現させる工程と、を含むことを特徴とする方法。 - 前記発現が、細胞内で行われる請求項37に記載の方法。
- 前記方法が、工程(2)において、前記第2のmicroRNAを前記第10の核酸分子に接触させることを更に含む請求項37に記載の方法。
- 請求項1から28のいずれかに記載の発現系を用いることによって、目的のタンパク質を発現させる方法であって、前記方法が、
(1)前記第8の核酸分子が第1の目的のタンパク質をコードする核酸配列を含むような請求項10に記載の発現系を提供し、前記第5の核酸分子が第2の目的のタンパク質をコードする核酸配列を含む工程と;
(2)前記第9の核酸分子によって、前記第1の調節タンパク質の発現を阻害し、前記第1の目的のタンパク質を発現させる工程;又は前記第10の核酸分子によって、前記第2の調節タンパク質の発現を阻害し前記第2の目的のタンパク質を発現させる工程と、
を含むことを特徴とする方法。
