Description
本発明は、細胞免疫療法の分野、より具体的には、新世代のキメラ抗原受容体(chimeric antigen receptor: CAR)に関し、これは、小分子との相互作用を介して、そのようなCARを付与された免疫細胞の制御を可能にする。より詳細には、本発明は、少なくとも1つのエクトドメインに、該受容体の抗原結合機能を制御する分子スイッチを含む、キメラ抗原受容体に関する。したがって、本発明は、より制御された、かつ潜在的により安全な、遺伝子改変されたCAR付与免疫細胞、例えばTリンパ球、を提供する。
養子免疫療法は、エクスビボで作製された自己抗原特異的T細胞の移入を含む療法であって、ウイルス感染症およびがんを治療するための有望な戦略である。養子免疫療法に使用されるT細胞は、抗原特異的T細胞の増殖、または遺伝子操作によるT細胞の向け直しのいずれかによって作製され得る(Park, Rosenberg et al. 2011)。ウイルス抗原特異的T細胞の移入は、移植関連ウイルス感染症および希少ウイルス関連悪性腫瘍の治療に使用される確立した手法である。同様に、腫瘍特異的T細胞の分離および移入は、メラノーマの治療に成功したことが示されている。

付着標的細胞に関して:2×104個の特異的標的抗原(STA)陽性細胞またはSTA陰性細胞を、96ウェルプレートでウェルあたり0.1ml中に播種する。プレーティングの翌日に、STA陽性細胞とSTA陰性細胞をCellTrace CFSEで標識し、4×105個のT細胞と4時間共培養する。次いで、細胞を回収し、固定可能な生存率解析用色素fixable viability dye (eBioscience社)で染色し、MACSQuantフローサイトメーター(Miltenyi社)を用いて解析する。

-「エクトドメイン」は、細胞外空間(細胞の外側の空間)へと延びる本発明のキメラ抗原受容体の一部を指す。
構築物およびmRNA調製
全ての構築物はpCLS24707 (SEQ ID NO:38)が起源であったが、これは多重鎖CAR(mcCAR)のα鎖(SEQ ID NO:39)、β鎖(SEQ ID NO:40)およびγ鎖(SEQ ID NO:41)をコードする。FRBドメイン(SEQ ID NO: 2)およびFKBPドメイン(SEQ ID NO: 1)をコードする配列は、デノボ合成された(GeneCust社)。scFv、ヒンジ-膜貫通-細胞質内α鎖ドメイン、FRBおよびFKBPをPCRによりさらに増幅して、ゴールデンゲートアセンブリ(golden gate assembly)適合性の断片(SEQ ID NO:42〜45)を作製した。さらに、FRB-FKBPおよびFKBP-FRB断片を、4-EAAAR-リンカー(SEQ ID NO:19)または−GS-4x-EAAAR-リンカー(SEQ ID NO:20)を用いて標準的な分子生物学手法により作製した(SEQ ID NO:46および47)。次いで、これらの断片を一連の制限およびライゲーションによりアセンブルして、pCLS26563、pCLS26564、pCLS26881、およびpCLS27123 (SEQ ID NO:48〜51)を生じさせた。これらの構築物によってコードされるそれぞれのアミノ酸配列をSEQ ID NO:52〜55に示す。
活性化の3〜6日後、Tリンパ球に、AgilePulse MAXシステム(Harvard Apparatus社)を用いてメッセンジャーRNAのエレクトロトランスファー(electrotransfer)によりトランスフェクトした。活性化ビーズの除去後、細胞をペレット化し、サイトポレーション媒体T中に>28×106細胞/mlで再懸濁した。5×106個の細胞を、0.4cmキュベット中で6.9μgの全RNA (2.5μgのα鎖、1.9μgのβ鎖および2.5μgのγ鎖)と、または8.4μgの全RNA (4μgの改変α鎖、1.9μgのβ鎖および2.5μgのγ鎖)と混合した。エレクトロポレーションは、2回の1200Vでの0.1msパルス、続いて4回の130Vでの0.2msパルスからなっていた。エレクトロポレーション後、細胞を2mLの培地中に希釈し、37℃/5%CO2でインキュベートした。mRNAエレクトロトランスファーの2時間後、ビヒクル(DMSO)またはラパマイシン(100nM)を19時間添加した。
α鎖の検出のための1回目の標識化は、2%PBS FBS, 2mM EDTA, 0.1%アジド中の抗Fab'2-ビオチン(ヤギ抗マウスIgG、Fab'2フラグメント特異的、115-066-072、Jackson Immunoresearch社)を用いて4℃で20分間行い、続いて2%PBS FBS, 2mM EDTA, 0.1%アジドを用いて2回の洗浄工程を行った。2回目の標識化は、2%PBS FBS, 2mM EDTA, 0.1%アジド中のストレプトアビジン-APCを用いて4℃で20分間行い、続いて2%PBS FBS, 2mM EDTA, 0.1%アジド中での洗浄工程とPBS中での洗浄工程を行った。細胞の生存を、PBS中のefluor450 (ebioscience社 65-0863-14)を用いて4℃で20分間モニターし、続いて2%PBS FBS, 2mM EDTA, 0.1%アジドによる洗浄工程を行い、2%PFAで固定した。フローサイトメトリーをMACSQUANT (Miltenyi Biotec社)を用いて行い、データ解析をFlowJoソフトウェアで行った。
構築物、mRNA調製およびフローサイトメトリー
scFv/抗原相互作用をオン/オフ状態間で切り替える統合システムを設計するために、FRB、FKBP12、またはFRBとFKBP12の融合物を、CD8aヒンジドメインとscFvドメインの間に挿入した(図1B)。開始実験として、初代T細胞に、多重鎖CAR (mcCAR)の各鎖をコードするmRNAをトランスフェクトした。ラパマイシンの添加後に、細胞外ヒンジドメインの検出の変化を、CD19標的化scFvのFab'2ドメイン(100nM、20時間)を追跡することによってモニターした。小分子(ラパマイシン)の非存在下では、高レベルの表面検出は、図3Aに示されるように、野生型mcCARおよびFKBP-mcCARについてのみ達成され、この場合、陽性細胞が90%超で、全体的に高いMFIを有することが見出された。ストーク(stalk)領域におけるFKBPとFRBの両方の存在は、それらの相互位置とは無関係に、CD19 scFvの表面検出を事実上消滅させた(陽性細胞が40%未満で、該mcCARと比較してMFIの最大40倍減少を有する)。興味深いことに、陽性細胞の割合またはMFIを考慮するとき、ラパマイシンの添加は、mcCAR、FRB-mcCARおよびFKB-mcCAR構築物にほとんど影響を与えなかった(図3B)が、それは、FKBP/FRB-mcCARおよびFRB/FKBP-mcCAR構築物の表面検出を強く改善し(MFIを考慮する場合には最大15倍、陽性細胞の割合を考慮する場合には3倍)、該システムをオフからオンの状態に変えた。ラパマイシン添加時のこの検出の変化は、スイッチオン成分を含有しているCAR鎖の安定化を含めて、様々な要因に起因し得る。しかし、この小分子は、T細胞の表面で該CARのFKBP/FRB-CARの検出を効率的にオンにするために、常にいつでも必要とされた、ことに留意しなければならない。
実施例2からのFKBP-FRB* mcCAR CD19を付与された遺伝子改変T細胞の細胞溶解活性は、フローサイトメトリーベースの細胞傷害アッセイを用いて評価した。このアッセイでは、CAR標的抗原を提示する標的細胞(Daudi CD19陽性)がCellTrace(商標)CFSEで標識され、対照細胞がCellTrace(商標)バイオレットで標識される。混合した標的細胞集団(1:1比)は、X-Vivo-15培地100μLの最終容量中の様々な比率の遺伝子改変エフェクターCAR T細胞(エフェクター/標的比20:1)と共に、ビヒクル(エタノール)またはAP21967 (100nM)の存在下に一定時間(5時間)にわたり37℃で共インキュベートした。
mRNA調製、トランスフェクションおよびフローサイトメトリーによる表面提示の測定は、実施例2に記載のように実施し、トランスフェクトされたT細胞を10nMのAP21967および漸増量のタクロリムスと共に同時にインキュベートした。
実施例2に示したCAR細胞外ドメイン(α鎖)を、一本鎖CAR (scCAR)をコードするプラスミドDNAを作製するための鋳型として使用した。CD8α膜貫通ドメイン(SEQ ID NO:32)、CD3-T細胞受容体のζ鎖の細胞質内シグナル伝達領域(SEQ ID NO:28)、および共刺激4-1BB (CD137)由来のシグナル伝達ドメイン(SEQ ID NO:31)を用いてCARを完成させた。標準的な分子生物学的手法に従って、一連の制限およびライゲーションを用いたゴールデンゲートクローニングによりscCARをアセンブルして、pCLS27572 (FKBP-FRB*)(SEQ ID NO:65)、pCLS27603 (FKBP)(SEQ ID NO:66)、pCLS27604 (FRB*)(SEQ ID NO:67)を生じさせた。これらの構築物によりコードされるそれぞれのアミノ酸配列をSEQ ID NO:68〜70に示す。

この研究の目的は、ヒトCD123抗原に対する誘導性キメラ抗原受容体(CAR)を発現するようにCellectis社によって遺伝子改変された、ヒトT細胞のインビボでの抗腫瘍活性を実証することであった。この誘導性システムは、非免疫抑制性のラパマイシン合成アナログ(ARIAD社が開発したAP21967, # 635055)により作動する。

第4群(CAR T細胞/AP21967)は、D0に、X個のCAR陽性T細胞を含むx個の改変CAR T細胞(RPMI 1640中200μL)の単回IV注射を受け、続いてAP21967の1日2回のIP注射を10日間連続して受ける(2Q1D×10)。
Claims (15)
- キメラ抗原受容体(CAR)であって、
(a)(i) 細胞外抗原結合性ドメインと、
(ii) 少なくとも第1の多量体化リガンド結合性ドメインおよび第2の多量体化リガンド結合性ドメインを含むスイッチドメインであって、該2つの結合性ドメインが所定の多価リガンドに結合して、該2つの結合性ドメインとそれらに結合可能な多価リガンドとを含む多量体を形成することができる、スイッチドメインと、
を含む少なくとも1つのエクトドメイン;
(b) 少なくとも1つの膜貫通ドメイン;ならびに
(c) シグナル伝達ドメイン、および任意で共刺激ドメインを含む少なくとも1つのエンドドメイン
を含み、該スイッチドメインが細胞外抗原結合性ドメインと膜貫通ドメインとの間に位置しており、かつ分子スイッチを構成する該第1の多量体化リガンド結合性ドメインおよび該第2の多量体化リガンド結合性ドメインが、同じポリペプチド分子上に存在することを特徴とする、キメラ抗原受容体。 - 第1の多量体化リガンド結合性ドメインおよび第2の多量体化リガンド結合性ドメインが、化学物質誘導二量体化(CID)システムから誘導される、請求項1記載のキメラ抗原受容体。
- 第1の多量体化リガンド結合性ドメインおよび第2の多量体化リガンド結合性ドメインが同じであるか、または異なっている、請求項1または2記載のキメラ抗原受容体。
- 第1の多量体化リガンド結合性ドメインおよび第2の多量体化リガンド結合性ドメインが、FK506結合タンパク質(FKBP12):mTORのFKBP-ラパマイシン結合ドメイン(FRB)、FKBP12:FKBP12、FKBP(F36V):FKBP(F36V)、FKBP12:FRB(T2098L)、FKBP12:カルシニューリンA(CnA)、FKBP12:シクロフィリン(CyP)、GyrB:GyrB、GAI:GID1A、GAI:GID1B、GAI:GID1C、Snap-tag:Halo-tag、Snap-tag:CLIP-tag、DHFR:DHFR、グルココルチコイド受容体リガンド結合性ドメイン:DHFR、およびPYL1:ABI1からなる多量体化リガンド結合性ドメインの対から選択される、請求項1〜3のいずれか一項記載のキメラ抗原受容体。
- 第1の多量体化リガンド結合性ドメインが、FKBP12 (SEQ ID NO: 1)、もしくはFKBP12 (SEQ ID NO: 1)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体であり、かつ第2の多量体化リガンド結合性ドメインが、FRB (SEQ ID NO: 2)、もしくはFRB (SEQ ID NO: 2)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体である、または、
第1の多量体化リガンド結合性ドメインが、FKBP12 (SEQ ID NO: 1)、もしくはFKBP12 (SEQ ID NO: 1)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体であり、かつ第2の多量体化リガンド結合性ドメインが、カルシニューリンA(CnA)(SEQ ID NO: 5)、もしくはCnA (SEQ ID NO: 5)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体である、または、
第1の多量体化リガンド結合性ドメインが、FKBP12 (SEQ ID NO: 1)、もしくはFKBP12 (SEQ ID NO: 1)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体であり、かつ第2の多量体化リガンド結合性ドメインが、シクロフィリン(CyP)(SEQ ID NO: 6)、もしくはCyP (SEQ ID NO: 6)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体である、または、
第1の多量体化リガンド結合性ドメインおよび第2の多量体化リガンド結合性ドメインが、GyrB (SEQ ID NO: 7)、そのクメルマイシン結合性断片、もしくはGyrB (SEQ ID NO: 7)もしくはそのクメルマイシン結合性断片に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体から選択される、または、
第1の多量体化リガンド結合性ドメインが、GAI (SEQ ID NO: 8)、もしくはGAI (SEQ ID NO: 8)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体であり、かつ第2の多量体化リガンド結合性ドメインが、GID1A (SEQ ID NO: 9)、もしくはGID1A (SEQ ID NO: 9)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体である、または、
第1の多量体化リガンド結合性ドメインが、GAI (SEQ ID NO: 8)、もしくはGAI (SEQ ID NO: 8)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体であり、かつ第2の多量体化リガンド結合性ドメインが、GID1B (SEQ ID NO:10)、もしくはGID1B (SEQ ID NO:10)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体である、または、
第1の多量体化リガンド結合性ドメインが、GAI (SEQ ID NO: 8)、もしくはGAI (SEQ ID NO: 8)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体であり、かつ第2の多量体化リガンド結合性ドメインが、GID1C (SEQ ID NO:11)、もしくはGID1C (SEQ ID NO:11)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体である、または、
第1の多量体化リガンド結合性ドメインが、Snap-tag(SEQ ID NO:12)、もしくはSnap-tag(SEQ ID NO:12)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体であり、かつ第2の多量体化リガンド結合性ドメインが、HaloTag (SEQ ID NO:13)、もしくはHaloTag (SEQ ID NO:13)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体である、または、
第1の多量体化リガンド結合性ドメインが、Snap-tag(SEQ ID NO:12)、もしくはSnap-tag(SEQ ID NO:12)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体であり、かつ第2の多量体化リガンド結合性ドメインが、CLIP-tag(SEQ ID NO:14)、もしくはCLIP-tag(SEQ ID NO:14)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体である、または、
第1の多量体化リガンド結合性ドメインが、DHFR (SEQ ID NO:15)、もしくはDHFR (SEQ ID NO:15)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体であり、かつ第2の多量体化リガンド結合性ドメインが、DHFR (SEQ ID NO:15)、もしくはDHFR (SEQ ID NO:15)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体である、または、
第1の多量体化リガンド結合性ドメインが、グルココルチコイド受容体リガンド結合性ドメイン(SEQ ID NO:16)、もしくはグルココルチコイド受容体リガンド結合性ドメイン(SEQ ID NO:16)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体であり、かつ第2の多量体化リガンド結合性ドメインが、DHFR (SEQ ID NO:15)、もしくはDHFR (SEQ ID NO:15)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体である、または、
第1の多量体化リガンド結合性ドメインが、PYL1 (SEQ ID NO:17)、もしくはPYL1 (SEQ ID NO:17)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体であり、かつ第2の多量体化リガンド結合性ドメインが、ABI1 (SEQ ID NO:18)、もしくはABI1 (SEQ ID NO:18)に対して少なくとも90%、例えば、少なくとも95%、少なくとも98%もしくは少なくとも99%、の配列同一性を有するその変異体である、請求項1〜3のいずれか一項記載のキメラ抗原受容体。 - 前記少なくとも1つのエクトドメインが、
(iii)スイッチドメインと膜貫通ドメインとの間に位置する、ヒンジ
を含む、請求項1〜5のいずれか一項記載のキメラ抗原受容体。 - 請求項1〜6のいずれか一項記載のキメラ抗原受容体をコードするヌクレオチド配列を含む、ポリヌクレオチド。
- (i) 免疫細胞を提供する段階;および
(ii) 該免疫細胞の表面に、請求項1〜6のいずれか一項記載の少なくとも1つのキメラ抗原受容体を発現させる段階
を含む、免疫細胞を遺伝子操作するための方法。 - (iii)前記免疫細胞の表面に、少なくとも1つの共刺激受容体を発現させる段階
をさらに含む、請求項8記載の方法。 - 前記共刺激受容体がNKG2DおよびDAP10からなる群より選択される、請求項9記載の方法。
- 請求項1〜6のいずれか一項記載の少なくとも1つのキメラ抗原受容体を含む、免疫細胞。
- 前記免疫細胞が、内因性mTORタンパク質のアミノ酸配列内に、1つまたは複数のアミノ酸置換を含み、かつ該1つまたは複数のアミノ酸置換が、2035位にてセリンが別のアミノ酸で置き換えられるアミノ酸置換を含む、請求項11記載の免疫細胞。
- 内因性FKBP12遺伝子が不活性化されている、請求項11または12記載の免疫細胞。
- がんまたはウイルス感染の治療のための医薬の調製における、請求項11〜13のいずれか一項記載の免疫細胞の使用。
- B細胞悪性腫瘍の治療のための医薬の調製における、請求項11〜13のいずれか一項記載の免疫細胞の使用。
