Description
(a)溶液のpHの上昇または維持が可能な化合物、および

(ii)pHを約6.5以下に維持しながら第一スズ化合物を混合するステップと
を含む方法によって製造することができる。
(i)CPP−ACFPを含む溶液を得るステップと;
(ii)pHを約6.5以下に維持しながら第一スズ化合物を混合するステップと
を含む方法によって製造することができる。
(i)少なくとも1種類のホスホペプチドを含む溶液を得るステップと;
(ii)pHを約7.0以下に維持しながら、カルシウムイオン、ホスフェートイオン、および水酸化物イオンを含む溶液を混合するステップと

(i)少なくとも1種類のホスホペプチドを含む溶液を得るステップと;

a)約5〜7未満のpHで、カルシウムイオンを含む第1の溶液、ホスフェートイオンを含む第2の溶液、および任意選択によりフッ化物イオンを含む第3の溶液を、ホスホペプチドおよび溶媒を含む溶液と混合するステップと;
b)水酸化物イオンを加えることによって、混合中に溶液のpHを約5.0〜7.0未満に維持するステップと

カゼインホスホペプチド−非晶質リン酸カルシウム(CPP−ACP)を、商標名Recaldent(商標)でCadbury Enterprises Pte Ltdより入手した。CPP−ACP、SnF2、およびNaFを用いて溶液を調製し、0.4%w/vのCPP−ACP、SnF2としての220ppmのF、およびNaFとしての70ppmのFにおいてpH5.6を得た。具体的には、CPP−ACPを蒸留水/脱イオン水に加え、次にpHを4.0〜6.5の間に維持するために1MのHClを加えながら、SnF2(固体)およびNaFを加えることによって、CPP−ACP/SnF2複合体を調製した。pHが6.5を超えないようにした。加えた酸の全体積はCPP−ACP/SnF2溶液の体積の1%未満であった。この溶液を安定化SnF2/ACPと名付けた。NaFをこの方法で加えたが、これは少量成分であり、フッ化物の大部分はSnF2に由来する。この方法は、SnF2のみを用いて(NaFなしで)行うことが可能である。CPP−ACPおよびNaFを用いて別の溶液を調製し、0.4%w/vのCPP−ACP、およびNaFとしての290ppmのFをpH5.6で得た。この溶液を安定化NaF/ACPと名付けた。
この実施例では、溶液中のCPP結合(強く結合した)イオン、緩く結合したイオン、および遊離のイオンを測定するための実験プロトコルを説明する。

以下は、CPP安定化ACP/SnF2溶液のイオン分析のためのプロトコルである。全体(強く結合および緩く結合)および緩く結合した試料を以下のように調製した:
全体(強くおよび緩く結合):0.4%のCPP−ACP/220ppmのF(SnF2として)/70ppmのF(NaFとして)の溶液1mlを採取し、19mlの1MのHNO3中に入れ、一定低速で24時間回転混合(20rpm)しながら室温でインキュベートした。この混合物を約1000gにおいて室温で15分間遠心分離した。上澄みのカルシウム、第一スズ、ホスフェート、およびフッ化物を分析した。
このin situ試験は、5つの溶液:
1)0.4%(w/v)のカルシウムホスホペプチド安定化非晶質リン酸カルシウム(CPP−ACP)、220ppmのフッ化第一スズ(SnF2)および70ppmのフッ化ナトリウム(NaF)(図7中に2%CPP−ACP+SnF2として示される);
2)0.4%(w/v)のCPP−ACPおよび290ppmのNaF;
3)0.4%(w/v)のCPP−ACP;
4)220ppmのSnF2および70ppmのNaF、CPP−ACPは含有せず(図7中にSnF2として示される);および
5)290ppmのNaF
によって促進される確立された口腔内再石灰化モデルを用いて再石灰化を比較するために計画した。
この実施例では、より一般的に前述した異なる目的の組成物のために本発明の複合体を配合できる方法を例示するために、多数の配合物を提供する。これらは、本発明の種々の実施形態を使用して提供できる種類の配合物の単なる例である。

Claims (21)
- ホスホペプチド1モル当たり少なくとも1モルの第一スズの第一スズイオン含有量を有する、第一スズが会合したホスホペプチド安定化非晶質リン酸カルシウム(ACP)および/または非晶質フッ化リン酸カルシウム(ACFP)複合体。
- 前記複合体が、ホスホペプチド1モル当たり少なくとも5モルまたは6モルの第一スズの第一スズイオン含有量を有する、請求項1に記載の第一スズが会合したホスホペプチド安定化非晶質リン酸カルシウム(ACP)および/または非晶質フッ化リン酸カルシウム(ACFP)複合体。
- 前記複合体が、ホスホペプチド1モル当たり約7モルまたは8モルの第一スズの第一スズイオン含有量を有する、請求項2に記載の第一スズが会合したホスホペプチド安定化非晶質リン酸カルシウム(ACP)および/または非晶質フッ化リン酸カルシウム(ACFP)複合体。
- 前記第一スズが、フッ化第一スズ、塩化第一スズ、フッ化第一スズカリウム、フルオロジルコン酸第一スズナトリウム、塩化フッ化第一スズ、酢酸第一スズ、フッ化第一スズナトリウム、ヘキサフルオロジルコン酸第一スズ、硫酸第一スズ、酒石酸第一スズ、グルコン酸第一スズ、クエン酸一第一スズ二ナトリウムからなる群から選択される化合物によって供給される、請求項1〜3のいずれか一項に記載の第一スズが会合したホスホペプチド安定化非晶質リン酸カルシウム(ACP)および/または非晶質フッ化リン酸カルシウム(ACFP)複合体。
- 前記化合物がフッ化第一スズである、請求項4に記載の第一スズが会合したホスホペプチド安定化非晶質リン酸カルシウム(ACP)および/または非晶質フッ化リン酸カルシウム(ACFP)複合体。
- 第一スズが会合した安定化非晶質リン酸カルシウム(ACP)複合体の製造方法であって、pHを約6.5以下に維持しながら、CPP−ACPと第一スズ化合物とを水溶液中で混合するステップを含む、方法。
- 第一スズが会合した安定化非晶質フッ化リン酸カルシウム(ACFP)複合体の製造方法であって、pHを約6.5以下に維持しながら、CPP−ACFPと第一スズ化合物とを水溶液中で混合するステップを含む、方法。
- 請求項1〜5のいずれか一項に記載の第一スズが会合したホスホペプチド安定化非晶質リン酸カルシウム(ACP)および/または非晶質フッ化リン酸カルシウム(ACFP)複合体を含む組成物。
- 前記組成物が、練り歯磨き、歯磨き粉、および液体歯磨きなどの歯磨き剤、マウスウォッシュ、マウスリンス、マウススプレー、バーニッシュ、歯科用セメント、トローチ、チューインガム、歯科用軟膏、歯肉マッサージクリーム、うがい用錠剤、乳製品、およびその他の食品からなる群から選択される口に使用可能な形態である、請求項8に記載の組成物。
- 請求項1〜5のいずれか一項に記載の第一スズが会合したホスホペプチド安定化非晶質リン酸カルシウム(ACP)および/または非晶質フッ化リン酸カルシウム(ACFP)複合体を含有する口腔ケア配合物。
- 歯磨き剤として配合された、請求項10に記載の口腔ケア配合物。
- 歯の表面または表面下を石灰化するための、請求項1〜5のいずれか一項に記載の第一スズが会合したホスホペプチド安定化ACPおよび/またはACFP複合体。
- 前記歯の表面が歯のエナメル質または象牙質である、請求項12に記載の第一スズが会合したホスホペプチド安定化ACPおよび/またはACFP複合体。
- 前記歯の表面が、齲蝕、歯牙酸蝕症、またはフッ素症によって生じたエナメル質中の病変である、請求項12または13に記載の第一スズが会合したホスホペプチド安定化ACPおよび/またはACFP複合体。
- 前記歯の表面が、象牙質知覚過敏を引き起こす露出象牙質である、請求項12〜14のいずれか一項に記載の第一スズが会合したホスホペプチド安定化ACPおよび/またはACFP複合体。
- 保護層がカルシウムおよびホスフェートを含む、歯の表面に結合した前記保護層を形成するための、請求項1〜5のいずれか一項に記載の第一スズが会合したホスホペプチド安定化ACPおよび/またはACFP複合体。
- 前記層が、通常のアパタイトと同等のカルシウム:ホスフェート比を有する、請求項16に記載の第一スズが会合したホスホペプチド安定化ACPおよび/またはACFP複合体。
- 前記層が、前記ACPおよび/またはACFPを安定化させるカゼインホスホペプチドに由来するペプチドを含有する、請求項16または17に記載の第一スズが会合したホスホペプチド安定化ACPおよび/またはACFP複合体。
- 前記層が、炭素、酸素、フッ化物、ホスフェート、カルシウム、および第一スズを含有する、請求項16〜18のいずれか一項に記載の第一スズが会合したホスホペプチド安定化ACPおよび/またはACFP複合体。
- 前記層が、20〜30重量%の炭素、35〜45重量%の酸素、0.1〜1重量%のフッ化物、8〜12重量%のホスフェート、16〜24重量%のカルシウム、および0.5〜2重量%の第一スズを含有する、請求項19に記載の第一スズが会合したホスホペプチド安定化ACPおよび/またはACFP複合体。
- 前記歯の表面または表面下が、ウシ、ヒツジ、ウマ、および家禽などの家畜、ネコおよびイヌなどのコンパニオンアニマル、または動物園の動物の歯の表面または表面下である、請求項12〜20のいずれか一項に記載の第一スズが会合したホスホペプチド安定化ACPおよび/またはACFP複合体。
