Description
本出願は、2015年5月11日に出願された米国特許仮出願第62/159,733号、及び2015年7月8日に出願された同第62/189,991号に基づく優先権を主張するものであり、これらの開示は、参照によりその全体が本明細書に組み込まれる。
病原体及びその他の微生物の検出のための従来法は、培養法をベースとしたものであるが、それらには時間と手間がかかり、迅速に結果を提供するという品質管理機関及び臨床検査機関のニーズをもはや満たしていない。
本開示は、核酸増幅反応におけるサンプル阻害(sample inhibition)を排除するための組成物、及びこの組成物を使用する核酸増幅方法を提供する。

実施形態Aは組成物であり、かかる組成物は、複数の酸化ジルコニウム粒子と、非イオン性界面活性剤と、有機鉄キレート剤と、を含有し、組成物は約8.45〜8.85のpHを有し、有機鉄キレート剤は、第二鉄に対し104.2以上の第1のアフィニティ定数と、マグネシウムに対し103.8未満の第2のアフィニティ定数と、を有し、第1のアフィニティ定数及び第2のアフィニティ定数は、20℃にてpH8.45の脱イオン水中で測定される。
a) 複数の酸化ジルコニウム粒子と、非イオン性界面活性剤と、エチレングリコール四酢酸(EGTA)の一価の塩と、を含む組成物を、対象とするサンプルと接触させて、水性混合物を形成する工程であって、組成物が約8.45〜8.85のpHを有する工程と、
b) 工程a)の水性混合物を熱による溶菌プロセスにかける工程と、
c) 工程b)の後で、水性混合物の一部を核酸増幅プロセスにかける工程と、を含む。
複数の酸化ジルコニウム粒子と、
非イオン性界面活性剤と、
有機鉄キレート剤と、を含有する、キットであり、
有機鉄キレート剤は、第二鉄に対し104.2以上の第1のアフィニティ定数と、マグネシウムに対し103.8未満の第2のアフィニティ定数と、を有し、第1のアフィニティ定数及び第2のアフィニティ定数は、20℃にてpH8.45の脱イオン水中で測定される。
Claims (10)
- 核酸増幅反応におけるサンプル阻害を排除するための水性組成物であって、
複数の酸化ジルコニウム粒子と、
0.005%(質量/体積)以上の濃度の非イオン性界面活性剤と、
有機鉄キレート剤と、を含有し、
前記組成物は、8.45〜8.85のpHを有し、
前記有機鉄キレート剤は、第二鉄に対し104.2以上の第1のアフィニティ定数と、マグネシウムに対し103.8未満の第2のアフィニティ定数と、を有し、前記第1のアフィニティ定数及び前記第2のアフィニティ定数は、20°CにてpH8.45の脱イオン水中で測定される、水性組成物。 - 前記複数の粒子が、100nm以下の平均粒子径を有する、請求項1に記載の水性組成物。
- 前記有機鉄キレート剤が複数のカルボキシレート基を含む、請求項1又は2に記載の水性組成物。
- 第二鉄を更に含有する、請求項1〜3のいずれか一項に記載の水性組成物。
- ナノ粒子分散安定剤を更に含有する、請求項1〜4のいずれか一項に記載の水性組成物。
- 前記非イオン性界面活性剤が、11〜16の親水性−親油性バランスを有する、請求項1〜5のいずれか一項に記載の水性組成物。
- ポリビニルピロリドンを更に含有する、請求項1〜6のいずれか一項に記載の水性組成物。
- 指示染料を更に含有する、請求項1〜7のいずれか一項に記載の水性組成物。
- a)複数の酸化ジルコニウム粒子と、非イオン性界面活性剤と、エチレングリコール四酢酸(EGTA)の一価の塩と、を含み、8.45〜8.85のpHを有する組成物を、対象とするサンプルと接触させて、水性混合物を形成する工程と、
b)工程a)の水性混合物を熱による溶菌プロセスにかける工程と、
c)工程b)の後で、前記水性混合物の一部を核酸増幅プロセスにかける工程と、を含む、核酸増幅方法。 - 複数の酸化ジルコニウム粒子と、
