Description
a)前記断片の末端へのXi個のヌクレオチド(Xは1〜5とすることができ、好ましくは1〜3である。iはサイクルの数である。)の酵素的付加段階であって、n+Xi個のヌクレオチドを含む断片を得ることを可能する、第1段階である、以下のステージを含む段階:
−伸長の過程で、第1の支持体に、n個のヌクレオチドを含む最初の核酸断片または核酸断片の第1の末端を付着させる第1の付着ステージ;
−前記核酸断片の第2の末端へのXi個のヌクレオチド(Xは1〜5とすることができ、好ましくは1〜3である。iはサイクルの数である。)の酵素的付加ステージの酵素的付加に必要な試薬を添加する添加ステージ;
−反応溶媒から好ましくない試薬を除去する任意の除去ステージ;
−前記第1の支持体から、n+Xi個のヌクレオチドを含む前記断片を分離する分離ステージ;
−n+Xi個のヌクレオチドを含む前記断片を移動させる第1の移動ステージ;
b)n+Xi個のヌクレオチドを含む正しい配列を有する断片の精製段階であって、第2段階である、以下の連続したステージを含む段階:
−第2の支持体に、第1段階の間に付加したXi個のヌクレオチドを運ぶその末端によって、n+Xi個のヌクレオチドを含む前記断片を付着させる第2の付着ステージ;
−付加されていない断片および第2の支持体に付着していない断片の除去ステージ;
−前記第2の支持体から、n+Xi個のヌクレオチドを含む前記断片を分離する分離ステージ;
−反応溶媒から、好ましくない残留試薬を除去する任意の除去ステージ;
c)例えばPCRによる、n+Xi個のヌクレオチドを含む正しい配列を有する断片の増幅、好ましくは酵素的増幅の任意の段階であって、第3段階である、以下の連続したステージを含む段階:
−増幅に必要な試薬を添加する添加ステージ;
−n+Xi個のヌクレオチドを含む断片の増倍率Yiによる増倍ステージ(任意に、可能性のある方法を形成するサブステージから成る)(iはサイクルの数である。Yは1〜4×1010とすることができ、好ましくは1〜109である。);
−n+Xi個のヌクレオチドを含む断片の移動ステージ。
各サイクルは、水性溶媒等の酵素的付加と酵素的増幅に適合する反応溶媒中で実施され、さらに当該合成方法は、iの伸長サイクルの全ての終了時、増倍率Yfによる最終的な増幅ステージを含む。
好ましい実施態様では、増幅に使用されるヌクレオチドは天然型ヌクレオチドである。
本発明は、上記方法によって合成された核酸にも関する。これらの核酸のヌクレオチド配列は、伸長サイクルの実施の間、前記ヌクレオチドの重合の順序を定め、予め決定することができる。しかしながら、ランダムな配列を有する核酸の合成に本発明の方法を使用することが想定され得る。
本発明の構成要件である方法の自動化は、この方法を最適化することができる事前に適合させた任意の装置によって、特に伸長サイクルの持続時間を最小限にすることによって、付加、洗浄操作および流体の流れの正確さを最大化することによって、ならびに、これらのサイクルの様々なステージの間に使用される反応条件を最適化することによって、行うことができる。
本発明はまた、上記の方法を実施するための任意のキットに関し、以下を含むキットである:
−n個のヌクレオチドを含む核酸断片を含む反応媒体;
−ヌクレオチドの付加のための酵素試薬;
−前記酵素的付加試薬によって付加され得るヌクレオチドまたはヌクレオチドの組み合わせ;
−精製ステージのための洗浄および/または緩衝液;
−核酸を増幅するための酵素試薬と、酵素的増幅の試薬に使用できる天然型ヌクレオチドとを含む増幅反応媒体;
−使用説明書。
−合成プライマーとして使用される核酸断片;
−n個のヌクレオチドを含む核酸断片を付着させるための第1の支持体;
−付加された核酸断片を付着させるための第2の支持体;
−ヌクレオチドの付加のための酵素試薬および酵素的付加の試薬によって付加され得るヌクレオチドおよびヌクレオチドの組み合わせを含む付加反応媒体;
−付着、精製および分離ステージのための洗浄および/または緩衝液;
−核酸を増幅するための酵素試薬と、酵素的増幅の試薬に使用できる天然型ヌクレオチドとを含む増幅反応媒体;
−使用説明書。
−第1および/または第2の付着支持体への核酸断片の付着に好ましい緩衝液媒体;および/または
−第1および/または第2の付着支持体からの核酸断片の分離に好ましい緩衝液媒体。
合成方法の特定のステージの間、核酸断片を固定するための付着支持体を用いた本発明の好ましい態様の例示的な実施態様の説明を、図面を参照して以下に記載する。図1〜7は、方法の様々なステージを図式的に表す:
図1:開始断片の固定化;
図2:固定された断片へのヌクレオチドの付加;
図3:洗浄ステージと、付加された核酸断片の分離;
図4:付加された核酸断片の第2の支持体への固定;
図5:付加された核酸断片の脱保護;
図6:増幅ステージ;
図7:前のサイクルからの付加された断片を使用する新しいサイクルの開始;
そして、図8は、酵素的付加段階と、正しい配列の断片の精製段階を実施するための「反応装置」の可能性のある配置図を示す。
そして、合成の終了前または新しい伸長サイクルi+1の実施前には、その後の精製ステージは必要ではない。
これにより合成された核酸は、最後の増幅ステージの後、第2の反応装置の出口17で回収される。
Claims (9)
- 所望の配列の核酸の合成方法であって、以下を含む方法:
a)第1の支持体に、最初の核酸断片または伸長された核酸断片を付着させる工程であって、最初の核酸断片または伸長された核酸断片が3’−OH基を有する工程;
b)ステージ(i)および(ii)のサイクルを実施して、最初の核酸断片または伸長された核酸断片がi番目のサイクル中にX個のヌクレオチドだけ伸長される工程であって、ステージ(i)が、付着された最初の核酸断片または伸長された核酸断片を、修飾されたヌクレオシド三リン酸および鋳型非依存性DNAポリメラーゼに接触させることで伸長させ、それにより最初の核酸断片または伸長された核酸断片が修飾されたヌクレオチドを含むこととなる工程を含み、ここで前記修飾されたヌクレオチドはヌクレオチドの複数の付加を阻止するおよび特異的に第2の支持体に結合する保護基を含み、ならびにステージ(ii)が、伸長された断片を脱保護して当該断片上に3’−OH基を与える工程を含む、工程;
c)前記第1の支持体から、伸長された、最初の核酸断片または伸長された核酸断片を分離する工程;
d)適切に伸長された断片を、前記第2の支持体にそれらの保護基を特異的に結合させることで、精製する工程;
e)保護基を脱保護することで、適切に伸長された断片を遊離する工程;
f)所望の配列の核酸が得られるまでa)からe)を繰り返す工程。 - 前記の付着させる工程を繰り返す前に、遊離された適切に伸長された断片を増幅させる工程をさらに含む、請求項1に記載の方法。
- 前記X個のヌクレオチドが、どのサイクルiにおいても、1〜5個のヌクレオチドである、請求項1または2に記載の方法。
- 前記の付着させる工程が、最初の核酸断片または伸長された核酸断片の区間を、前記第1の支持体に付着された相補的オリゴヌクレオチドと、ハイブリダイズさせる工程を含む、請求項1〜3のいずれか一項に記載の方法。
- 前記鋳型非依存性DNAポリメラーゼが、ターミナルデオキシヌクレオチジルトランスフェラーゼ、テロメラーゼ、ポリメラーゼηもしくはζ、PNPアーゼ、または鋳型非依存性RNAポリメラーゼである、請求項1〜4のいずれか一項に記載の方法。
- 前記鋳型非依存性DNAポリメラーゼが、ターミナルデオキシヌクレオチジルトランスフェラーゼである、請求項5に記載の方法。
- 前記修飾されたヌクレオシド三リン酸が、3’−O−保護ヌクレオシド三リン酸である、請求項1〜6のいずれか一項に記載の方法。
- 前記の精製する工程が、前記第2の支持体に3’−O−保護基を特異的に結合させる工程を含む、請求項1〜6のいずれか一項に記載の方法。
- 前記修飾されたヌクレオシド三リン酸が、3’−O−保護基および修飾された窒素塩基を含む、請求項1〜6のいずれか一項に記載の方法。
