Description
本実施形態では、分析用基板1を用いて検出対象物質11を検出する(図3参照)。図1〜図3を用いて本実施形態に用いられる分析用基板1について説明する。
次に、図11のフローチャートを用いて、本実施形態の分析方法について説明する。なお、試料液によっては検出対象物質11が含まれていない場合もある。この場合、分析用基板1の反応領域10には検出対象物質11、第1粒子20及び第2粒子30が捕捉されない。そこで、説明を分かりやすくするために、反応領域10に検出対象物質11、第1粒子20及び第2粒子30が捕捉されている場合について説明する。
まず、エクソソームに特異的な抗原タンパク質であるCD9を認識する抗体を、光ディスク基板の反応領域に固定した。そして、光ディスク基板を洗浄液で洗浄した。
光ディスク基板に第2粒子を捕捉させない以外は、実施例1と同様にして分析用基板を作製した。
CD63を発現しているエクソソームは、CEAを発現しているエクソソーム比較して多量に存在する場合が多いため、従来法でも十分に検出可能である。しかしながら、CEAを発現しているエクソソーム数は、CD63を発現しているエクソソームと比較して1%以下と非常に微量であるため、このように微量な場合であってもエクソソームが検出可能であるか評価した。具体的な評価方法は以下の通りである。

Claims (6)
- 反応領域を有する分析用基板の前記反応領域に検出対象物質が結合され、前記検出対象物質に結合する抗体が設けられた第1粒子が前記検出対象物質に結合され、前記抗体に結合する抗原が設けられて金属により形成されかつ前記第1粒子と異なる複素屈折率を有する第2粒子が前記第1粒子に結合された前記分析用基板に光ピックアップがレーザ光を照射し、
前記光ピックアップが前記反応領域からの反射光を受光して受光レベル信号を生成し、
判定回路が前記反応領域において所定の信号レベルよりも高い信号レベルの受光レベル信号を粒子検出信号として抽出し、
計数回路が前記抽出された粒子検出信号に基づいて検出対象物質を検出する分析方法であって、
前記第2粒子が結合した前記第1粒子に起因する受光レベル信号は、前記所定の信号レベルよりも高いことを特徴とする分析方法。 - 前記所定の信号レベルは、検出対象物質が存在しない領域からの反射光を受光して生成された信号レベルである請求項1に記載の分析方法。
- 前記第2粒子の前記複素屈折率をn−ki(nは前記第2粒子の屈折率を表し、iは虚数単位を表し、kは前記第2粒子の消衰係数を表す。)で表した場合に、(k−0.23)2/1.22+(n−1.36)2/0.942>1を満たす、請求項1又は2に記載の分析方法。
- 前記第2粒子は金、銀、白金及び銅からなる群より選択される少なくとも1種の金属により形成される請求項1〜3のいずれか1項に記載の分析方法。
- 前記抗原は、タンパク質及びタンパク質断片の少なくともいずれか一方である請求項1〜4のいずれか1項に記載の分析方法。
- 反応領域を有する分析用基板の前記反応領域に検出対象物質が結合され、前記検出対象物質に結合する抗体が設けられた第1粒子が前記検出対象物質に結合され、前記抗体に結合する抗原が設けられて金属により形成されかつ前記第1粒子と異なる複素屈折率を有する第2粒子が前記第1粒子に結合された前記分析用基板にレーザ光を照射し、前記反応領域からの反射光の受光レベルを検出して受光レベル信号を生成する光ピックアップと、
前記反応領域において所定の信号レベルよりも高い信号レベルの受光レベル信号を粒子検出信号として抽出する判定回路と、
前記粒子検出信号に基づいて検出対象物質を検出する計数回路と、
を備え、
前記第2粒子が結合した前記第1粒子に起因する受光レベル信号は、前記所定の信号レベルよりも高いことを特徴とする分析装置。
