Description
精子無力症における精子運動能の回復のためのTreamidの有効性についての研究
精子無力症は、顕著なプロオキシダント特性を呈するエトポシドを使用することによってシミュレーションした。フリーラジカルのレベルの上昇を誘導する薬物を使用したのは、近代思想により、精子無力症の直接的原因が酸化ストレスであるという事実によるものであった(Kao S. H.ら、Increase of oxidativestress in human sperm with lower motility. Fertil. and Steril. 2008;89:5:1183〜1190頁)。
低ゴナドトロピン性性腺機能低下症における精子運動能の回復のためのTreamidの有効性についての研究
低ゴナドトロピン性性腺機能低下症は、Synestrolの投与によってシミュレーションした。エストロゲンは、LH(黄体ホルモン)及びFSH(卵胞刺激ホルモン)の分泌を阻害することができる。雄の体におけるエストロゲン濃度の増加は、遊離テストステロンの濃度の減少及び精子無力症の出現を導く性ステロイド結合グロブリンの含有量の増加を伴う。
交尾活動の回復のためのTreamidの有効性についての研究
アンドロゲン欠乏によって引き起こされる交尾活動の低減は、雄のラットへのSynestrolの投与によってシミュレーションした。
雄のラットにおいて、加齢変化により低減した交尾機能及び性的動機付けの回復のためのTreamidの有効性についての研究
勃起不全は、後期及び老齢の生殖可能年齢(14〜19カ月)の動物を使用してシミュレーションした。性的動機付けの抑制は、後期生殖可能年齢(14カ月)の動物を使用してシミュレーションした。
交尾活動の季節による抑制状態でのインタクト動物の雄のラットの交尾活動の回復のためのTreamidの有効性についての研究
生殖活動の季節による抑制は、冬期間(12月の終わり)に実験を行うことによってシミュレーションした。バックグラウンド群として、インタクト動物(3カ月齢)を6月に試験した。
慢性非細菌性前立腺炎(CAP)又は良性前立腺肥大症(BPH)における前立腺組織の構造の再生及び正常化のためのTreamidの有効性についての研究
これは、これらの疾患に典型的な重度の形態的変化の減少において現れる。CAPは、腹側葉の腺を絹糸で縫合することによってシミュレーションし、BPHは、Sulpirideの投与によってシミュレーションした。基準薬は、Prostamol Unoであった。
40頭の雄のWistarラット(3カ月齢)について実験を行った。慢性非細菌性前立腺炎をシミュレーションするために、前立腺の腹側葉を絹糸で縫合した(麻酔下)。良性前立腺肥大症(BPH)のモデルは、雄のラットに40mg/kgの用量のSulpirideを2カ月間毎日腹腔内注射して(Eglonil、Sanofi Winthrop Industry社、France)、高プロラクチン血漿を誘導することによって、ラットにおいてシミュレーションした。
前立腺の構造を正常化するためのTreamidの能力を、Sulpiride投与の存在下で研究した。この目的のために、Treamidを、Sulpirideと同時に、0.5mg/kgの用量で2カ月間胃内投与した。対照動物は、等量の溶媒(2%のデンプン粘液)を受けた。比較群の動物を、50mg/kgの用量のProstamol Unoの溶液で胃内注射した。投与過程の終了時に、動物を秤量し、CO2チャンバー内で安楽死させた。Sulpirideは、ラットの腹側葉の腺におけるBHPの発症を引き起こすことが知られている[Van Coppenolle Fabien、Christian Slomianny、Francoise Carpentier Effects of hyperprolactinemia on rat prostate growth:evidence of androgeno - dependence.//Physiol. Endocrinol. 精巣組織の修復再生のプロセス及び精子形成の増殖プールの枯渇によって引き起こされる精巣機能不全の処置の強化のためのTreamidの有効性についての研究
後者は、雄のラット(3カ月齢)への細胞分裂阻害薬パクリタキセル(Mitotax、Dr. Reddy's社、India)の単回注射によってシミュレーションした。薬物の選択は、これが、精子形成-幹細胞の前駆体に損傷を与える医薬の1つであるという事実による。
ストレプトゾトシンに誘導される膵臓病変のモデルの膵臓細胞の再生に関するTreamidの効果についての研究
研究は、4週齢の雄のC57BL/6マウスについて行った。膵臓の損傷は、0.25mlのリン酸緩衝液中40mg/kgの用量のストレプトゾトシンを5日間毎日腹腔内注射することによって誘導した[Szkudelski T. The Mechanism of Alloxan and Streptozotocin Action in B Cells of the Rat Pancreas//Physiological Research. - 2001. - 50巻 - 536〜546頁]。
メタボリックシンドロームモデルの雄のマウスにおける膵臓細胞の再生及び精巣組織の機能回復に関するTreamidの効果についての研究
代謝障害は次のようにシミュレーションした:誕生の2日後に、雄のC57Bl/6マウスに、甲領域において、200mg/kgの用量のストレプトゾトシンで1回皮下注射し、注射した溶液の体積は30μlであった。誕生の4週間後から、動物を高脂肪食に移した。この目的のために、使用した食餌は、重い飽和脂肪が豊富であった(30%の脂肪)(30%の脂肪を有するSiff EF R/M Cat No. E15116-34、Germany)。高脂肪食を用いる給餌期間は42日であった(実験の28日目から70日目まで)。Treamidを、実験の49日目から70日目まで毎日、1日1回、10mg/kgの用量で胃内投与した。対照動物は、試験化合物の代わりに等量の溶媒(2%デンプン粘液)を受けた。

メタボリックシンドロームを、動物を高脂肪食(「カフェテリアダイエット」)で12週間維持することによって、雄のWistarラット(260〜280g)においてシミュレーションした。「カフェテリアダイエット」については、Current Protocols in Pharmacologyに従って、バランスをとった[Current Protocols in Pharmacology (2005) Animal Models of Disease//Contributed by Petter Hedlund、Kenshi Matsumoto、及びKarl-Erik Andersson]。食餌は、げっ歯類の標準の食餌に加えて、25〜35%の脂肪及び25〜30%の可消化炭水化物を含有した。Treamidは、食餌の7週目〜11週目に毎日、1日1回、0.5mg/kgの用量で胃内投与した。基準薬メトフォルミンを、同じレジメンに従って、200mg/kgの用量で胃内投与した。

慢性中毒性肝炎は、雄のCBA/CaLacマウスにおいて、研究の0、3、7、10、14、及び17日目にマウス1頭当たり0.2mlの体積で、オリーブ油中20%の四塩化炭素(CCl4)溶液を胃内投与することにより誘導した。Treamidは、研究の14日目から27日目までに、毎日、1日1回、10mg/kgの用量で胃内投与した。28日目に、動物をCO2チャンバー内で安楽死させ、肝生検試料を組織解析のために採取した。
肺気腫モデルの肺組織の回復に関するTreamidの効果についての研究
肺気腫は、0.9%のNaCl 30μl中マウス1頭当たり0.6uの用量でエラスターゼ(Sigma社、USA)を単回気管内投与することによって、雌のC57Bl/6マウスにおいて誘導された[Luthje L.、Raupach T.、Michels H.ら//Respiratory Research 2009、10:7]。操作日を実験0日目とした。Treamidは、研究の7日目から60日目までに、毎日、1日1回、10mg/kgの用量で胃内投与した。7、14、30及び60日目に、動物をCO2チャンバー内で安楽死させ、それらの肺を単離した。
肺線維症の発症に関するTreamidの効果についての研究
肺線維症を引き起こす肺胞上皮の毒性損傷を、30μlの生理食塩水中マウス1頭当たり80μgの用量で、1回、細胞分裂阻害薬であるブレオマイシンを気管内投与することによってシミュレーションした。操作中の麻酔は、吸入エーテルを使用することによって実現させた。術野は、70%エタノール溶液で消毒し、術野の体毛は除去した。皮膚、皮下組織及び首の筋膜を、首の中心線に沿って解剖した。筋肉は、組織の鈍分離の方法によって、器官の腹側で取り外した。ブレオマイシン注射を、Hamiltonシリンジによって、エアフローに沿った吸入で行った。傷を縫合した後、術野を消毒薬で処置した。ブレオマイシン投与を実験0日目とした。
腫瘍を有する動物のカヘキシーの発症に関するTreamidの効果についての研究
血行性転移するルイス肺癌(LLC)をシミュレーションする場合、雄のC57Bl/6マウスにおいて、カヘキシーが発症した。腫瘍細胞を、0.1mlの生理食塩水中5×106の濃度で、1回、マウスに皮下注射した(Yu-Chou T.、Samuel K.K.、I-Lu L.ら、Preclinical Investigation of the Novel Histone Deacetylase Inhibitor AR-42 in the Treatment of Cancer-Induced Cachexia//JNCI J Natl Cancer Inst (2015) 107(12))。
