Description
(i)一般式(I)
mは、3〜5;好ましくは3または5であり;
nは、2〜5;好ましくは3であり;
oは、0または1であり;かつ
pは、3〜17;好ましくは5〜15;より好ましくは11または13であり;
ただし、m+n+o+pの合計が、14以上であり;かつ
Rは、その炭素原子の一つを介して結合性酸素に結合されている炭水化物残基である];
で表される少なくとも1種の抗菌性グリコリピドを含むグリコリピド成分、
または、開鎖の形態および/またはラクトンの形態にあるそれらのエステル、および/または生理学的に許容されるそれらの塩;ならびに
(ii)少なくとも1種の配合安定剤、好ましくは1種または複数のポリソルベートおよび/または1種または複数のシクロデキストリン、好ましくはアルファ−シクロデキストリンを含む配合成分;
を含み、
ここで、前記配合成分の前記グリコリピド成分に対する相対重量比が、その組成物中の一般式(I)で表される全ての抗菌性グリコリピドの全重量を基準にし、かつその組成物中の全ての配合安定剤の全重量を基準にして、100:1から1:2までの範囲内である

− そのグリコリピド成分の含量が、経口的に消費可能な水ベースの製品の全重量を基準にして、0.0005〜1重量%;より好ましくは0.0005〜0.1重量%;さらにより好ましくは0.001〜0.1重量%または0.0005〜0.01重量%;さらにより好ましくは0.005〜0.05重量%の範囲内であるか、および/または
− その配合成分の含量が、経口的に消費可能な水ベースの製品の全重量を基準にして、0.0005〜1重量%の範囲内;より好ましくは0.001〜0.1重量%の範囲、さらにより好ましくは0.005〜0.05重量%;さらにより好ましくは0.0012〜0.5重量%、もしくは0.0012〜0.25重量%、もしくは0.0025〜0.5重量%;または0.0025〜0.5重量%、もしくは0.0025〜0.25重量%、もしくは0.005〜0.5重量%;または0.0012〜0.05重量%、もしくは0.0012〜0.025重量%、もしくは0.0025〜0.05重量%;または0.012〜0.25重量%、もしくは0.012〜0.12重量%、もしくは0.025〜0.25重量%の範囲である。
− そのグリコリピド成分の含量が、経口的に消費可能な水ベースの製品の全重量を基準にして、3〜100ppmw(重量で百万部の中の部数)の範囲;より好ましくは、3〜25ppmw、25〜50ppmw、または50〜75ppmw、または75〜100ppmwの範囲;さらにより好ましくは、3〜10ppmw、または10〜20ppmw、または20〜30ppmw、または30〜40ppmw、または40〜50ppmw、または50〜60ppmw、または60〜70ppmw、または70〜80ppmw、または80〜90ppmw、または90〜100ppmwの範囲であり、および/または
− 好ましくはアルファ−シクロデキストリンを含むか、またはそれからなる、その配合成分の グリコリピド成分に対する相対重量比が、好ましくは20:1から1:1までの範囲内;より好ましくは10:1から2.5:1までの範囲内;さらにより好ましくは7.5:1から2.5:1まで、または10:1から5:1まで;なおより好ましくは7.5:1から5:1まで、あるいは5:1から2.5:1までである。
− 透明な飲料(濁度、0〜10NTU) : 3〜25mg/L
− 曇った飲料(濁度、>10NTU) : 10〜100mg/L
− ジュースおよび果汁含量が50%を超えるフルーツ飲料 : 50〜200mg/L。
− 透明な飲料(濁度、0〜10NTU) : 10〜200mg/L
− 曇った飲料(濁度、>10NTU) : 50〜1000mg/L
− ジュースおよび果汁含量が50%を超えるフルーツ飲料 : 250〜2000mg/L。
次の重量分布を有するグリコリピド混合物を使用した。
アルファ−シクロデキストリン(a−CD)と一般式(I)で表される抗菌性グリコリピドの混合物とを、固体物質としてガラス容器の中に入れ、下の表に示したような相対量を有する容積3mLのストック組成物を生成させた。
1gのアルファ−シクロデキストリン粉体を、二つの60mLガラス容器それぞれに入れた。次いで、一般式(I)で表される抗菌性グリコリピドの混合物(メッシュサイズ0.5mmで粉砕したもの)を、それぞれ1gおよび0.4gの量で加えて、下の表に示した質量分布を得た。
4g(4mL)のポリソルベート80(PS80)を、三つの60mLガラス容器それぞれに入れ、40mLの脱イオン水と混合した。マグネチックスターラーを用いて、室温で30分間、その組成物を激しく撹拌した。三つの分離容器(separate container)の中に、0.4g、1g、および1.6gの、一般式(I)で表される抗菌性グリコリピドの混合物を入れ、次いで、それぞれ三つのポリソルベート80溶液を得た。そのストック組成物は、次の濃度を有していた。
実施例4:抗菌性グリコリピドの混合物と組み合わせて使用される、適切な配合向上剤のスクリーニング
配合安定剤を決めるために、スクリーニングを実施したが、そこでは、水ベースの配合物の中で、食品、化粧品または医療用途で典型的に使用される添加物を抗菌性グリコリピドの混合物と組み合わせた。潜在的な効果を見逃さないように、配合向上剤としてはむしろ高めの濃度の1000mg/Lを使用したが、それに対して、抗菌性グリコリピドの混合物には、典型的な使用濃度、すなわち、5、10および25μg/mLを適用した。
水ベースの製品、特に飲料において抗菌性グリコリピドの混合物を使用するための、アルファ−シクロデキストリンの配合安定剤性能を確認するために、抗菌性グリコリピドの混合物の単独使用では、すなわちアルファ−シクロデキストリンを存在させないと、相溶性に欠ける、市販されている二つの飲料について、濃度を変えて二つの成分を組み合わせて検討した。
実施例5で述べたのと同様のプロトコールを適用して、透明な飲料であるリンゴジュースに対する、アルファ−シクロデキストリンを抗菌性グリコリピドの混合物と組み合わせたものについての、相溶性に関する効果の温度依存性を観察した。しかしながら、10μg/mLの濃度の抗菌性グリコリピドの混合物と、アルファ−シクロデキストリンとの3種の組合せ、25μg/mL(6−1、6−2、6−9、6−10、6−17、6−18)、75μg/mL(6−3、6−4、6−11、6−12、6−19、6−20)および100μg/mL(6−5、6−6、6−13、6−14、6−21、6−22)、ならびに唯一の25μg/mLの濃度の抗菌性グリコリピドの混合物と、100μg/mLのアルファ−シクロデキストリンとの組合せ(6−7、6−8、6−15、6,−16、6−23、6−24)を使用して、それぞれ、二つの試験ポイントで全て使用した。室温の他にも、6℃と40℃も適用し、さらには、観察期間を14日まで延長することにより、次の結果が得られた。
水ベースの製品、特に飲料において抗菌性グリコリピドの混合物を使用するための、ベータ−シクロデキストリンの相溶性改良性能を確認するために、抗菌性グリコリピドの混合物の単独使用では、すなわちベータ−シクロデキストリンを存在させないと、相溶性に欠ける、市販されている二つの飲料について、濃度を変えて二つの成分を組み合わせて検討した。
2種の飲料、「Gerolsteiner Orangenlimonade」(オレンジレモネード、炭酸抜き、曇った飲料)および「Kastell Zironensprudel」(カンキツ類レモネード、滅菌濾過済み、透明な飲料)の2×400mLをそれぞれ、500mLのErlenmeyerフラスコの中に充填した。次いで、下の表に示したような、所定の容積の実施例3で作成したストック溶液を、その飲料の中に注入し、室温で5分間撹拌した。

比較のために、実施例4−8において使用した飲料に対して、配合安定剤、すなわち、ポリソルベートおよびシクロデキストリンを一切加えずに、抗菌性グリコリピドの混合物を適用した。
2種の腐敗菌:サッカロミセス・セレビシアエ(Saccharomyces cerevisiae)MUCL53497およびアスペルギルス・ニガー(Aspergillus niger)ATCC16404に対して、抗菌性グリコリピドの混合物単独の場合、または配合安定剤と組み合わせた場合で求めたMICの値を、下の表に列記する。
水ベースの製品、特に飲料において抗菌性グリコリピドの混合物を使用するための、メチル−ベータ−シクロデキストリンの配合安定化性能を確認するために、抗菌性グリコリピドの混合物の単独使用では、すなわちメチル−ベータ−シクロデキストリンを存在させないと、相溶性に欠ける、市販されている二つの飲料について、濃度を変えて二つの成分を組み合わせて検討した。
水ベースの製品、特に飲料において抗菌性グリコリピドの混合物を使用するための、ヒドロキシプロピル−ベータ−シクロデキストリンの相溶性改良性能を確認するために、抗菌性グリコリピドの混合物の単独使用では、すなわちヒドロキシプロピル−ベータ−シクロデキストリンを存在させないと、相溶性に欠ける、市販されている二つの飲料について、濃度を変えて二つの成分を組み合わせて検討した。

- (i)一般式(II)
sは、1または2であり;
tは、6または7であり;
R 1 は、−Hまたは−OHを意味し;
R 2 は、−Hまたは−C 1 〜C 6 −アルキルを意味し;かつ
R 3 、R 4 、R 5 、R 6 、およびR 7 は、互いに独立して、−Hまたは−C(=O)C 1 〜C 6 −アルキルを意味している]
で表される少なくとも1種の抗菌性グリコリピドからなるグリコリピド成分、
および/または、生理学的に許容されるその塩、ならびに
(ii)シクロデキストリン、ポリソルベート、又はシクロデキストリンとポリソルベートの組み合わせである少なくとも1種の配合安定剤を含む配合成分;
を含む、経口的に消費可能な水ベースの製品であって、
ここで、グリコリピド成分の含量が、経口的に消費可能な水ベースの製品の全重量を基準にして、3〜50ppmw(重量で百万部の中の部数)であり、かつ、
前記配合成分の前記グリコリピド成分に対する相対重量比が、10:1〜2.5:1である、経口的に消費可能な水ベースの製品。 - 前記少なくとも1種の抗菌性グリコリピドが、化合物(II−A)〜(II−D)
から選択される、請求項1に記載の経口的に消費可能な水ベースの製品。 - 前記シクロデキストリンが、アルファ−シクロデキストリン、ベータ−シクロデキストリン、ヒドロキシプロピル−ベータ−シクロデキストリン、およびメチル−ベータ−シクロデキストリンから選択される、請求項1又は2に記載の経口的に消費可能な水ベースの製品。
- 前記配合安定剤がシクロデキストリンを含み、前記シクロデキストリンの、前記グリコリピド成分に対する相対重量比が、5:1から2.5:1までの範囲内である、請求項1〜3のいずれか一項に記載の経口的に消費可能な水ベースの製品。
- 前記配合安定剤がポリソルベートを含み、前記ポリソルベートが、ポリソルベート20、ポリソルベート21、ポリソルベート40、ポリソルベート60、ポリソルベート61、ポリソルベート65、ポリソルベート80、ポリソルベート81、ポリソルベート85、ポリソルベート120、および前述のもののいくつかの混合物からなる群から選択される、請求項1又は2に記載の経口的に消費可能な水ベースの製品。
- 請求項1に記載の経口的に消費可能な水ベースの製品であって、第一から第三の抗菌性グリコリピドを含み、
グリコリピド成分の中に含まれる全ての抗菌性グリコリピドの全重量を基準にして、
− 一般式(II)で表される前記第一の抗菌性グリコリピドの相対重量含量が、30〜50重量%の範囲内であり;
− 一般式(II)で表される前記第二の抗菌性グリコリピドの相対重量含量が、20〜50重量%の範囲内であり;
− 一般式(II)で表される前記第三の抗菌性グリコリピドの相対重量含量が、5〜10重量%の範囲内である、
経口的に消費可能な水ベースの製品。
