1つの実施形態において、上記ユニット型ポリイオンコンプレックスは、2分子の上記ブロックコポリマーと1分子の上記核酸とから構成される。

1つの実施形態において、上記ポリエチレングリコールセグメントの分子量が、60×103以上である。
1つの実施形態において、上記ポリエチレングリコールセグメントが二鎖分枝型であり、各ポリエチレングリコール鎖の分子量が、20×103以上である。
1つの実施形態において、上記カチオン性ポリアミノ酸が、構成アミノ酸としてオルニチンを含む。
本発明の別の局面によれば、上記ユニット型ポリイオンコンプレックスと、該ユニット型ポリイオンコンプレックスを構成し得るブロックコポリマーであって、遊離のブロックコポリマーと、を含む、核酸送達用組成物が提供される。
1つの実施形態において、上記組成物中における上記ブロックコポリマー由来のアミノ基のモル濃度と上記核酸由来のリン酸基のモル濃度との比(N/P比)が、1〜20である。
本発明によれば、ポリエチレングリコールセグメントとカチオン性ポリアミノ酸セグメントとを有する特定数のブロックコポリマーと、特定数の核酸と、から構成されるユニット型ポリイオンコンプレックスであって、該ユニット型ポリイオンコンプレックスにおけるブロックコポリマーのカチオン性ポリアミノ酸セグメントの側鎖由来の正の電荷量の合計が、核酸由来の負の電荷量の合計で相殺されていないユニット型ポリイオンコンプレックス(結果として、電気的に中和されていないユニット型ポリイオンコンプレックス)が提供される。以下、本発明の実施形態について説明するが、本発明はこれらの実施形態には限定されない。なお、本発明に用いられるブロックコポリマーおよび核酸はそれぞれ、重合反応物として得られる場合に、ある程度の多分散性を示し得る。よって、本明細書において、ブロックコポリマーの特性(分子量、重合度、電荷等)および核酸の特性(塩基長、電荷等)について言及する際は、特段の記載がない限り、多分散性を示す重合反応物全体の平均について言及するものとする。したがって、例えば、ブロックコポリマーの電荷量は、実際に測定して得られるカチオン性アミノ酸残基の重合度を平均重合度とし、該平均重合度に基づいて算出される。また例えば、ポリエチレングリコールセグメントの分子量としては、実際に測定して得られる平均分子量(例えば、数平均分子量)が適用され得る。

[A−1−1.ユニット型ポリイオンコンプレックスの全体構成]
図1(a)〜(d)は、本発明の第1の実施形態におけるuPICの一例を説明する概略図である。具体的には、図1(a)は、本発明で使用され得るブロックコポリマーの概略図であり、図1(b)は、本発明で使用され得る核酸の概略図であり、図1(c)は、図1(a)のブロックコポリマーと図1(b)の核酸とから構成されるuPICの概略図であり、図1(d)は、図1(c)のuPICにおけるPEGセグメントの空間的な広がりを示した概略図である。
本実施形態で使用され得るブロックコポリマーは、PEGセグメントと、カチオン性ポリアミノ酸セグメントと、を有し、送達対象の核酸に対して3.0×105以上、好ましくは5.0×105以上(例えば、8.0×105以上、1.0×106以上、1.5×106以上、または2.0×106以上)の結合定数(Ka)を有する。核酸に対して上記所定の親和性を有するブロックコポリマーを用いることにより、血中においてuPICからブロックコポリマーが解離し難くなる、あるいは、解離したとしてもすぐに再会合することができ、結果として、核酸由来の負電荷とブロックコポリマー由来の正電荷とが相殺されていない状態においても、核酸を良好に保護して高い血中滞留性を得ることができると推測される。一方、結合定数(Ka)の上限値は、特に限定されないが、実現容易性の観点から、例えば、4.0×106以下であり得る。なお、本明細書において、ブロックコポリマーは、該ブロックコポリマーの薬学的に許容可能な塩も包含する。

本明細書において、核酸とは、プリンまたはピリミジン塩基、ペントース、リン酸からなるヌクレオチドを基本単位とするポリもしくはオリゴヌクレオチドを意味する。例えば、オリゴもしくはポリ二本鎖RNA、オリゴもしくはポリ二本鎖DNA、または、同一の鎖にRNAとDNAが混在したオリゴもしくはポリ二本鎖核酸等の二本鎖核酸が好ましく用いられ得る。当該核酸に含有されるヌクレオチドは天然型であっても、化学修飾された非天然型のものであっても良く、またアミノ基、チオール基、蛍光化合物などの分子が付加されたものであっても良い。
本発明の第2の実施形態によれば、ポリエチレングリコールセグメントとカチオン性ポリアミノ酸セグメントとを有する特定数のブロックコポリマーと、特定数の核酸と、から構成されるユニット型ポリイオンコンプレックスであって、
該ブロックコポリマーのカチオン性ポリアミノ酸セグメントの側鎖由来の正の電荷量の合計が、該核酸由来の負の電荷量の合計で相殺されず、
該核酸の鎖長が、10〜50塩基長であり、
該ポリエチレングリコールセグメントの分子量が、40×103以上であり、
該ブロックコポリマーが、配列番号1で示されるセンス鎖および配列番号2で示されるアンチセンス鎖からなる二本鎖核酸(ただし、各鎖の5’末端にリン酸基が付加されていない)に対して所定の結合定数(Ka)を有する、ユニット型ポリイオンコンプレックスが提供される。
本発明のuPICは、例えば、任意の適切なブロックコポリマーと核酸とを、必要により緩衝化された水溶液(例えば、リン酸緩衝生理食塩水、HEPES緩衝液)中で混合することにより調製することができる。
本発明の核酸送達用組成物は、A項に記載のuPICと、該uPICを構成し得るブロックコポリマーであって、遊離状態のブロックコポリマーと、を含む。本発明の核酸送達用組成物によれば、uPICを単独で用いる場合よりも優れた核酸の血中滞留性が得られ得る。このような効果が得られる理由は、例えば以下のように推測できる。すなわち、uPICを含む溶液においては、核酸−ブロックコポリマー間の解離および会合が可逆反応として生じており、その結果、uPICが可逆的に分解および再形成されている。よって、uPICに加えて遊離状態のブロックコポリマーを配合することにより、平衡を会合側にシフトさせることができ、結果として、uPIC単独の場合よりも核酸を好適に保護できると考えられる。
種々の分子量を有する二鎖分枝型のポリ(エチレングリコール)誘導体(NOF社製)を用い、特許文献1の実施例に記載のブロックコポリマーの調製方法と同様の方法で、以下のブロックコポリマー(塩酸塩):PEG−pOrn(37×2−11)、PEG−pOrn(37×2−14)、PEG−pOrn(37×2−20)、PEG−pOrn(37×2−40)、PEG−pLys(37×2−9)、PEG−pLys(37×2−14)、PEG−pLys(37×2−20)、PEG−pLys(37×2−40)の白色粉末を得た。ポリアミノ酸セグメントの重合度は、1H-NMRによって決定した。
PEG−pOrn(37×2−11)とsiRNAとを、10mM HEPES緩衝液(pH7.3)に別々に溶解し、siRNAの終濃度が10μMとなるように、かつ、N/P比が0.25、0.5、1、2、3、5または10となるように混合してPIC溶液を調製した。siRNAとしては、各鎖の5’末端にリン酸基が付加されていないsiLuc(ルシフェラーゼに対するsiRNA)を用い、必要に応じて、5’末端にAlexa647等の標識を付して用いた。

PEG−pOrn(37×2−11)の代わりにPEG−pOrn(37×2−14)を用いたこと以外は実施例1と同様にして、PIC溶液を調製した。
PEG−pOrn(37×2−11)の代わりにPEG−pLys(37×2−14)を用いたこと以外は実施例1と同様にして、PIC溶液を調製した。
PEG−pOrn(37×2−11)の代わりにPEG−pLys(37×2−9)を用いたこと以外は実施例1と同様にして、PIC溶液を調製した。
PEG−pOrn(37×2−11)の代わりにPEG−pLys(37×2−20)を用いたこと以外は実施例1と同様にして、uPIC溶液を調製した。参考例1で得られたuPIC溶液(N/P比=10)は、特許文献1の表2において医薬製剤Bとして開示されたuPIC溶液に略相当し、1分子のsiRNAと2分子のブロックコポリマーとから構成されるuPICを含むものである。
上記実施例1〜3および比較例1で調製したPIC溶液について、沈降平衡法による超遠心分析(SE−AUC分析)および蛍光相関分光解析(FCS解析)を行うことによって、PIC1個あたりの核酸会合数およびブロックコポリマー会合数を求めた。具体的な方法は以下のとおりである。
SE−AUC分析は、吸収光学系を備えた分析用超遠心システムBeckman Optima XL−A(Beckman Coulter社製)を用いて行った。上記実施例および比較例においてN/P比=1で調製したPIC溶液をsiRNA濃度が0.6μMになるように150mM NaClを含む10mM HEPES緩衝液(pH7.4)で希釈して測定試料とした。20℃で72時間遠心後、波長260nmにおける吸収をrの関数として測定した。各試料の測定結果を、ORIGINソフトウェアを用いて解析してPICの分子量(Da)を求めた。
FCS解析は、40×対物レンズ(C−Apochromat、Carl Zeiss社製)およびConfoCor3モジュールを搭載した共焦点レーザスキャン顕微鏡(Carl Zeiss社製、製品名「LSM510」)を用いて行った。上記実施例および比較例においてN/P比=1で調製したPIC溶液をsiRNA濃度が100nMになるように150mM NaClを含む10mM HEPES緩衝液(pH7.4)で希釈して測定試料とした。測定試料を8ウェルチャンバーに移してサンプリング時間15秒、繰り返し回数5回にてFCS測定を行った。測定結果をConfoCor3ソフトウェアで解析することにより測定領域内における平均蛍光粒子数を求めた。次いで、以下の式(a)を用いて計算することによって、PIC1個あたりの核酸会合数を求めた。

(式中、ANcopolymerは、PIC1個あたりのブロックコポリマー会合数であり、MWPICは、上記SE−AUC分析で得たPICの分子量(Da)であり、MWsiRNAおよびMWcopolymerはそれぞれ、siRNAおよびブロックコポリマーの構造式に基づいて算出される分子量(Da)である。)
上記参考例1で得られたuPIC溶液について、上記uPICの構造解析1と同様にFCS解析を行うことによって、PIC1個あたりの核酸会合数およびPICの平均粒子径(流体力学的直径:DH)を求めた。なお、PICの平均粒子径(DH)は、ConfoCor3を用いて、上記FCS測定で得られた自己相関曲線を拡散時間に変換し、次いで、該拡散時間をCy5色素を対照として用いて拡散係数(DC)に変換し、該拡散係数をアインシュタインの関係式(Stokes−Einstein equation)に代入することによって算出した。結果を図2に示す。
上記で得られた各ブロックコポリマーのsiRNAに対する結合定数(Ka)を以下の方法により決定した。結果を表2に示す。
結合定数は、エチジウムブロマイド(EtBr)とブロックコポリマーのsiRNAに対する結合の競合を、EtBrが核酸に結合すると増強する蛍光の強度を定量することにより決定した。上記蛍光強度の定量は、蛍光分光光度計(日本分光社製、FP‐6600)を用いて行った。具体的には、測定対象のブロックコポリマーと上記siRNA(実施例および比較例で用いたsiLuc、ただし、Alexa647で標識されていない)とを、150mM NaClを含む10mM HEPES緩衝液(pH7.3)に別々に溶解し、siRNAの終濃度が1μMとなるように、かつ、N/P比が0.25、0.5または1となるように混合してPIC溶液を調製した。各N/P比のPIC溶液 1mLに対して、約0.5mMのEtBr水溶液を10μLずつ10回滴下し、滴下する度に蛍光強度(ex 525nm,em 600nm)の測定を行った。得られた蛍光強度を、既報の方法(J.Chem.Edu.71,77(1994))に基づいて解析して結合定数を算出した。
実施例、比較例および参考例で得られたuPIC溶液(N/P比=1)を6週齢の雌性BALB/cマウスに尾静脈投与した。このとき、siLucの投与量が2nmol/マウスとなるように投与した。その後、In vivo共焦点イメージングシステム(Nikon A1R、ニコン社製)を用いて、マウスの耳血管内でのsiLuc由来の蛍光強度を経時的に測定し、次式によって相対蛍光強度を求めた(N=1)。投与直後、5分後、10分後、30分後および60分後の相対蛍光強度を表3に示す。

N/P比=10のuPIC溶液を用いたこと以外は、siRNAの血中滞留性評価1と同様にして、6週齢の雌性BALB/cマウスにuPIC溶液を尾静脈投与し、マウスの耳血管内でのsiLuc由来の蛍光強度を経時的に測定して相対蛍光強度を求めた(N=1)。相対蛍光強度の経時的変化を図3に示す。

該ユニット型ポリイオンコンプレックスを構成し得るブロックコポリマーであって、遊離のブロックコポリマーと、を含む、核酸送達用組成物。 - 組成物中における前記ブロックコポリマー由来のアミノ基のモル濃度と前記核酸由来のリン酸基のモル濃度との比(N/P比)が、1〜20である、請求項7に記載の核酸送達用組成物。
