Description

本開示はまた、上述の核酸を含む、発現カセットおよび/またはベクターを提供する。好適には、本開示のキメラタンパク質を符号化する核酸は、プロモータに動作可能に結合される。プロモータは当該技術分野において周知である。宿主細胞で機能する任意のプロモータを、本開示のC1V1および/またはその任意の変異形の発現に使用することができる。特定の動物細胞においてC1V1キメラタンパク質またはその変異形の発現を駆動するために有用である開始制御領域またはプロモータは非常に多く、当業者によく知られている。これらの核酸を駆動することが可能である実質的にあらゆるプロモータを使用することができる。

本開示は、中で発現されたときに、膜電圧を修飾する光活性化キメラオプシンに関する。本開示は、必ずしもこれらの文脈に限定されるわけではなく、本開示の種々の態様は、これらおよび他の文脈を使用した実施例の説明を通じて、理解することができる。
この実施例では、皮質のE/I上昇の駆動および皮質切片でのγ振動の監視、ならびに生きた動物でのインビボ実験を可能にするであろう手段を、1)用量反応の調査を可能にする、さらにより高い力価、2)E/Iバランスの段階様の変化を可能にする低い脱感作、および3)同じ調製物内での異なる集団の相対的な駆動を可能にする、赤方偏移した励起、の3つの主要な特性とともに、模索した。
キメラチャネルロドプシン変異形C1V1を、重複伸長PCRにより、野生型またはヒトコドン最適化のいずれかのチャネルロドプシン−1を、ヒトコドン適応VChR1(GenBankTM受託番号ACD70142.1)と融合させることによって生成した。C1V1スプライス変異形を、重複PCRによって生成した。変異形1は、ChR1の最初の145のアミノ酸およびVChR1のアミノ酸102〜316を含有していた。変異形2は、ChR1の最初の162のアミノ酸およびVChR1のアミノ酸119〜316を含有していた。結果として得られたキメラPCRフラグメントを、CaMKIIαプロモータ下で、pECFP−N1(Clonetech,Mountain View,CA)およびレンチウイルス発現ベクターにクローニングした。膜輸送シグナルは、Kir2.1チャネル1から導出した。突然変異は、コード配列およびスプライス部位のシークエンシングによって確認した。AAV仲介の遺伝子送達については、CaMKIIαプロモータとともに、オプシン−EYFP融合体を、pAAV2−MCSベクターの修飾形にサブクローニングした。Cre依存性オプシンの発現は、オプシン−EYFPカセットを、伸長因子1a(EF−1α)プロモータの制御下で、互換性のないlox部位(loxPおよびlox2722)の対の間に逆向きにクローニングして、二重floxの逆位オープンリーディングフレーム(D10)を生成することによって、達成した。全ての構築物は、Deisseroth Lab(www.optogenetics.org)から入手可能である。
興味深いことに、キメラにおける最も強い改善は、ヘリックス1および2が、ChR1からの相同体で置換された場合に見られた(図1C〜D)。膜標的化および光電流の大きさについて、2つのキメラChR1−VChR1チャネルを、培養HEK293細胞において試験した。第1のものを、ChR1のA1a145後の第2の細胞内ループに結合させ、第2のものを、ChR1のTrp163後のヘリックス3内に結合させた(図1C)。いずれの変異形も、HEK293細胞で同程度に十分に発現したが(図1D)、培養ニューロンにおいては、第2の変異形がより強く発現し(図1E)、VChR1−EYFPと比較して、非常に強化されたピーク光電流(888±128pA、n=11個の細胞、p<0.0005)を示した(図1B)。作用スペクトルピークは、ChR2に対して強く赤方偏移したままであり(表1、図1F)、キメラのイオン選択性は、ChR2およびVChR1について以前に報告されたものと類似していた(図1G)。Kir2.1輸送配列を、このハイブリッドに付加することは、さらに増加した光電流をもたらす傾向を見せた(1104±123pA、n=12個の細胞、VChR1−EYFPと比較してp<0.0005、C1V1−EYFPと比較してp=0.23、図1B、表1〜2)。結果として得られるハイブリッドChR1/VChR1キメラは、ChR2配列を全く含まず、単独では十分に発現しない2つのオプシン遺伝子から顕著に誘導され、本明細書にC1V1と称される(図1A、H)。
この赤方偏移したオプシンの速い非活性化特性28は、可視スペクトルの青色末端の方向に位置する波長によって活性化される、他のオプシンからの最大限の時間的ならびにスペクトル的な分離に必要となるであろう。しかしながら、C1V1−ts−EYFPによって提示される光電流は、ChR2よりも10倍を上回って遅い減衰、およびもともとのVChR1よりもさらに遅い減衰を示し(図2A、Toffが、それぞれ、C1V1−ts−EYFP(n=4)およびVChR1−EYFP(n=5)に対して、156±12msおよび132±12ms、表1)、速い発火レートを要する用途へのC1V1の使用を排除する可能性があることが見出された。C1V1の光電流動態を補正するために、発色団領域に、既知の構造モデル22、28を使用して、より速い光周期動態、減少した不活性化、および減少した青色吸収を有する突然変異を探した(図1H)。次に、この突然変異が不活性化を減少させることを示す、ヘリックス2におけるグルタミン酸塩が豊富なモチーフの研究に基づいて3、グルタミン酸塩−122をスレオニンに突然変異させた。
C1V1ベクターでの全ての点突然変異は、部位特異的な突然変異生成(Agilent Technologies,Palo Alto,CA)によってプラスミドにおいて生成した。膜輸送シグナルは、Kir2.1チャネルから導出した。突然変異は、コード配列およびスプライス部位のシークエンシングによって確認した。
ChETA−相同突然変異E162T28は、光周期を、ほぼ3倍、著しく加速させた(Toff 58±4.7ミリ秒、n=7個の細胞、図2A、表1)。驚くべきことに、ChR2または他の微生物オプシンにおける類似の突然変異は、赤方偏移をもたらし28,29、C1V1においては、この突然変異は、望ましくない方向にある作用スペクトルを、浅色から530nmに偏移させた(図1F、表1)。C1V1−E122Tは、ChR2の46%の非活性化と比較して、26%不活性化しただけであり(図2B、表1)、加えて、スペクトルは、さらに546nmまで赤方偏移され(図1F、表1)、C1V1作用スペクトルの厄介な青色ショルダーの顕著な損失を示した。最終的に、二重変異E122T/E162Tにおいては、作用スペクトルの赤方偏移および青色ショルダーの非存在を保持しながらも、電流の不活性化は、E122T変異のものよりもさらに低くさえあり、光周期は、E162Tと比較してもさらに速かった(Toff34±4.9ミリ秒、n=7個の細胞、図2A、図2C、表1)。さらに、E122変異が、光電流の振幅を大幅に減少させる一方で(図2D)、二重変異は、本来のC1V1−tsの非常に高い電流特性を再現した。したがって、個々の突然変異の複数の驚くべきかつ有用な特性は、二重変異の輸送強化されたC1V1キメラにおいて保存された。
2ミリ秒の光パルスをピーク活性化波長に使用して、HEK細胞におけるピーク活性化波長を記録した。2ミリ秒の光パルスを最大活性化波長に使用して、非活性化動態(τoff)およびピーク光電流を培養海馬ニューロンにおいて記録した。ChR2とのコンビナトリアル活性化に最適な変異形を識別するために、405nmおよび560nmでの反応率をHEK細胞において記録した。光電流の脱感作を、300ミリ秒の光パルスを使用して記録し、ピーク光電流(Imax)の減衰を定常状態に対して定量化した。
これらの新規なC1V1オプシン遺伝子をニューロンにおいて試験するために、C1V1−ts−EYFPを符号化するレンチウイルスベクターおよび点突然変異の組み合わせを生成した。これらのオプシンを、次いで、培養海馬ニューロンに発現させ、同一な刺激条件下(2ミリ秒のパルス、542nmの光、5.5mW mm−2)で全細胞光電流を記録して、光電流振幅における改善が、VChR1と比較して、増加したC1V1の発現から直接的にもたらされたかどうかを判定した。
動物
野生型またはトランスジェニックのパルブアルブミン::Cre C57/BL6J雄性マウスを、3〜5匹の群でケージに収容し、餌および水を自由に摂取させて12時間の逆明/暗周期で保管した。実験プロトコルは、Stanford University IACUCに認可されており、National Institutes of Health基準のCare and Use of Laboratory Animalsについての指針を満たす。
初代海馬培養物を、POスプラーグドーリーラットから単離し、Matrigel(Invitrogen)コーティングしたガラスのカバースリップに蒔き、FUDRで処理して、神経膠の過剰成長を阻害した。内毒素不含プラスミドDNAを、HEPES緩衝化した生理食塩水/CaPO4混合物を使用して、培養ニューロンにトランスフェクトした。蛍光タンパク質発現によって識別される個々のニューロンからの電気生理学的記録を、3〜5MΩのガラスピペット中の標準的な内部溶液([mM]130 Kグルコン酸塩、10 KCl、10 HEPES、10 EGTA、2 MgC12、KOHでpH7.3)を使用して、タイロード培地([mM] 150 NaCl、4 KCl、2 MgC12、2 MgC12、10 D−グルコース、10 HEPES、NaOHでpH7.35)において取得した。皮質切片の生理学については、8〜9週齢の野生型C57BL/6Jまたは事前にウイルスを注入したPV::Creマウスからの300gmの急性冠状切片を、ビブラトーム(Leica)を使用して、氷冷スクロース切断溶液([mM]11 D−グルコース、234スクロース、2.5 KCl、1.25 NaH2PO4、10 MgSO4、0.5 CaC12、26 NaHCO3)に得た。切片を、32℃で1時間、酸素化した人工脳脊髄液(ACSF;[mM] 124 NaC1、3 KC1、1.3 MgC12、2.4 CaC12、1.25 NaH2PO4、26 NaHCO3、10 D−グルコース)中で回復させた。蛍光タンパク質の発現を識別した後、一定なACSF灌流下で、示される前頭前皮質層から個々のニューロンパッチを取得した。広い波長のキセノンランプ源(Sutter Instruments DG−4)からフィルタ処理した光を、顕微鏡(Leica DM−LFSA)の蛍光ポートに結合させた。バンドパスフィルタ(Semrock)は、20nmのバンド幅を有し、追加の減光フィルタ(ThorLabs)で調節して、スペクトル全体にわたって光パワーの出力を均一化した。
レンチウイルス仲介およびAAV仲介の両方の遺伝子送達を、マウスにおけるオプシンの異種発現に使用した。示されるオプシンを、皮質の興奮性ニューロンを標的とするヒトカルモジュリン依存性タンパク質キナーゼIIα(CaMKIIα)プロモータ、またはCre誘発性カセットおよび続くウッドチャック肝炎ウイルス転写後調節要素(WPRE)と併用した伸長因子1a(EF−1a)のいずれかによって、駆動した。Cre誘発性組み換えAAVベクターは、University of North Carolina Vector Core(Chapel Hill,NC,USA)によって生成し、パルブアルブミン::Creトランスジェニックマウスと併せて使用して、パルブアルブミン陽性の介在ニューロンを標的化した。簡単にいうと、University of North Carolinaにおいて、AAV構築物をpAAV2−MCSの修飾形にサブクローニングし、AAV5コーティングしたタンパク質で血清型にして、ウイルスベクターコアによってパッケージ化した。AAVベクターの最終的なウイルス濃度は、1×1012ゲノム複製(gc)/mLであった。レンチウイルス構成物を、報告した通りに生成した。全ての構築物は、Deisseroth Lab(www.optogenetics.org)から入手可能である。定位ウイルス注入を、Stanford Universityに認可されたプロトコルのもとに実行した。イソフルラン麻酔下に保った若齢(4〜6週齢)のマウスを、定位フレーム(Kopf Instruments)に配置し、ブレグマおよびラムダ頭蓋計測点を使用して高さを合わせた。皮質組織への損傷が最小となるように、開頭術を実行した。10uLのシリンジおよび35gの勾配付きの針(Word Precision Instruments)を使用して、辺縁系前頭前皮質(Infralimbic prefrontal cortex)(IL、ブレグマから前方1.8mm、側方0.35mm、腹側−2.85mm)を標的とした。ウイルスを0.1μL/分の速度で注入した。挙動研究のため、ウイルスを注入した対象に、さらに、局所送達用に付属の貫通性脳線維ありまたはなしのいずれかで、光の送達を促進するための慢性光ファイバー結合装置を埋め込み、記載のように皮質領域を標的化した(Doric Lenses,Canada)。貫通性線維を、同じ前方および側方の座標から−2.5mmの深さに定位挿入し、頭皮の接着閉鎖(Vetbond,3M)の前に接着性合着用セメント(C&B MetaBond)を使用して付着させた。手術からの回復後、動物に鎮痛剤を投与した。
個々の構築物を発現する培養海馬ニューロンからの記録および統合された蛍光読み出しを、それぞれの個々の細胞から得た。個々の細胞において、蛍光レベルは、構築物全体で、測定された光電流振幅と緊密に相関していた(図3A)。したがって、強く増加したC1V1の光電流は、主として、ニューロンにおける改善された発現からもたらされたと結論付けた。二重変異C1V1−E122T/E162Tは、試験した全ての範囲(光電流の大きさ、不活性化動態、および作用スペクトル)にわたって優れた能力を示したため、ChR2(H134R)に対する能力もまた、統合された体細胞YFP蛍光およびピーク光電流を測定することによって直接比較した。驚くべきことに、C1V1−E122T/E162T細胞は、同等の蛍光レベルで、ChR2−H134R細胞よりも強い光電流を示し(図3B)、単位コンダクタンスが増加した可能性を示唆した。
本研究により、同じ微小回路内に存在する抑制性および興奮性ニューロンを、C1V1変異形を導入することにより標的化できるかどうかを判定することにした。生体脳切片内の2つのニューロン集団の独立した活性化を調査し、この場合、CaMKIIα−C1V1−E122T/E162Tts eYFPおよびEF1a−DIO−ChR2−H134R−EYFPを、PV::CreマウスのmPFCに発現させた。
材料および方法
この一連の多重改変オプシン遺伝子を使用して、細胞のコンビナトリアル制御の可能性およびインタクトな哺乳動類系における投射を調査した。まず、同じ微小回路内に存在する興奮性および抑制性ニューロンが、C1V1変異形および従来のChRをこれらの2つの細胞集団内へ個別に導入することよって別々に標的化することができるかどうかを調べた。C1V1−E122T/E162Tを発現する培養海馬ニューロンは、2ミリ秒の緑色光パルス(560nm)に反応してスパイクするが、紫色光パルスではスパイクしないことが見出された。対照的に、ChR2−H134Rを発現する細胞は、2ミリ秒の405nmの光パルスに反応してスパイクするが、2ミリ秒の561nmの光パルスには反応しなかった(図4A)。したがって、この原理を生体脳切片内で試験し、この場合、AAV5−EFla−DIO::ChR2−H134R−EYFPとともに、AAV5−CaMKIIa::C1V1−E122T/E162T−ts−mCherryを、PV::CreマウスのmPFCに発現させた(図4B)。いずれのオプシンをも発現しない錐体ニューロンにおいて、405nmの光パルスは、PV細胞の直接的な活性化のため、強く早い抑制性シナプス後電流を引き起こした(図4C)が、一方で、561nmの光パルスは、短潜EPSC(図4D)、および局所的な抑制性ニューロンを駆動するC1V1発現錐体細胞から生じることが予測される長潜の多シナプスIPSC(図4C)の両方を引き起こした。
直接的な細胞の体細胞刺激の代わりに、軸索投射に対するコンビナトリアル制御特性を検証するために、視床網様核のニューロン(nRT)に影響を及ぼす皮質視床(CT)および視床皮質(TC)のグルタミン酸軸索の独立した活性化の効果(図5A)を、視床切片において試験した。
C57BL/6Jの野生型(出生後90〜120日)を、ペントバルビタール(100mg/kg、腹腔内)で麻酔し、頭部を切除した。視床切片の調製および全細胞パッチクランプ記録を行った。記録は、Zeiss(Oberkochen, Germany)、Axioskop顕微鏡、および赤外線カメラを有する微分コントラスト光学を使用して視覚的に識別した、nRT(視床網様)およびTC(中継視床皮質)ニューロンから取得した。EPSCおよび電流クランプ記録については、内部溶液は、(mM単位で)120 K−グルコン酸塩、11 KCl、1 MgCl2、1 CaCl2、10 Hepes、1 EGTAを含有した。pHは、KOH(290mOsm)で7.4に調節した。ECl −は、ネルンストの式に基づいて約−60mVと推定した。電位は、−15mVの液体注入電位に補正した。電圧クランプ実験については、ニューロンを、−80mVでクランプし、EPSCを、GABAA受容体アンタゴニストピクロトキシン(50μM,Tocris)の浴適用(bath application)によって薬理学的に単離した。全ての記録条件において、アクセス抵抗を監視し、細胞は、実験過程にわたってアクセス抵抗が<18MΩであり、抵抗の変化が<25%であった場合にのみ分析に含めた。
TC軸索の最小刺激は、nRTニューロンに大きく速い興奮性シナプス後電流(EPSC)を誘起し、一方で、CT軸索の最小刺激は、nRTニューロンに小さく遅いEPSCを誘起し(図5B)、いずれもこれらの経路に典型的であった。
上の2つの調製物の両方において、可視スペクトルの紫色(405nm)および緑色(560nm)レーザーを使用して、2つのオプシンの分離可能な活性化を達成した。405nmのレーザーは、安全な非紫外光を送達するが、470nmの光は、より深く組織を貫通し、散乱が少なく、より容易かつ経済的に一般的な青色光源から送達されるため、多くの用途については、青色応答性オプシンに470nmのレーザー光を使用することが好ましい場合がある。これは、470nmの光が、C1V1(図1G)、ならびにChR2を部分的に活性化させることになるため、不可能に思われる場合があるが、コンビナトリアル制御は、470nmの光でさえも達成可能であり、SSFOの時間特性およびC1V1の赤方偏移性質の両方を利用して、インタクトな哺乳動物組織内での「時間スペクトル分離」を達成することができる。この可能性を検証するために、同じ調製物内で、安定して増強する興奮性または抑制性いずれかの細胞の律動的活性への影響を直接比較することが決定された(図6A)。
ChR2−D156AおよびSSFOを、部位特異的な突然変異生成(Quikchange II XL、Stratagene)を使用してpLenti−CaMKIIα−ChR2−EYFP−WPREベクターに点突然変異を挿入することによって、生成した。ウイルス遺伝子送達、冠状脳の区分化、およびパッチクランプの記録を、上述のように行った。CaMKIIa::C1V1およびDIO−SSFOをPV::CreマウスのmPFCに発現させるため、二重ウイルス注入を行った。
SSFOは、C1V1を活性化させるものと同じ波長で効果的に非活性化することができ、強化された光感受性を有し、数分間わたるニューロンの双安定興奮を可能にする、29分の減衰定数を有する新規な多重改変チャネルロドプシンである。SSFOに関する情報は、国際特許出願公開第WO2010/056970号、ならびに米国特許出願第61/410,704号、同第61/410,711号に見出すことができ、これらの内容は、参照によりその全体が本明細書に組み込まれる。CaMKIIa::C1V1およびDIO::SSFOを、PV::CreマウスのmPFCからの急性切片に発現させるために、二重ウイルス注入を行った。これらの条件下では、興奮性錐体細胞は赤方偏移した光に、抑制性PV細胞は青色光に応答するはずである。実際に、PV細胞においてSSFOを活性化させるための1秒の470nmの光パルスに応答して、進行中のIPSCレートは、ベースラインでの8.5±1.2Hz(期間3、図6B)から青色光パルスの後の16.8±2.1Hz(期間2、n=4回の記録、p<0.005、対応t検定、図6C)に安定して増加し、SSFO発現抑制性細胞の持続的な活性化を示した。470nmの光もまた一過的にC1V1を活性化するとはいえ、この活性化は、点火後の非常に速いC1V1の非活性化のため、光パルス自体の間にのみに生じ得、点火後の持続期間は、SSFO活性のみによって特徴付けられ(図6B)、光遺伝学的制御モードの時間的分離を示す。興味深いことに、この長期間のPVニューロン活性の増加中に、IPSCパワースペクトルに著しい上昇ピークの誘発はなく、この減少した調製物におけるPVニューロン単独の直接的な活性化が、ネットワークにおけるγ同期性の誘発には不十分であることを示唆する。しかしながら、非常に対照的に、470nmの光パルス自体の間に、同じレベルのPVニューロン活性化だけでなく、C1V1発現錐体細胞の部分的な活性化もまた予測される場合、顕著なγピークが一貫して観察され(ピーク周波数39.2±3.5Hz、n=4回の記録)、高γ範囲(>60Hz)に及んだ。
本研究の過程において、C1V1変異形と称される、新しい手段の系統がもたらされた。C1V1は、注目すべきことに、単独ではニューロンに十分に発現しないが、初期のゲノム研究で特定されていた他のオプシン遺伝子(VChR1およびChR1)が結集された、赤方偏移されたオプシン遺伝子である。C1V1は、ChR2配列を全く有さないが、それにもかかわらず、ここで本明細書に報告されたその多重改変された変異形は、最も強力で、最も赤方偏移した、最も安定した既知のチャネルロドプシンを代表する。レチナール結合ポケット全体で重要なアミノ酸位置における突然変異生成は、(1)ChETA突然変異の高速相同体として生成される、高発現の赤方偏移したオプシン遺伝子であるC1V1(E162T)、(2)最も赤い作用スペクトルショルダーを示し、赤色光で活動電位を発火させるために使用可能ですらあるC1V1(E122T)、(3)最も低い脱感作、最も速い非活性化、ChR2との最小限の交差活性化のための最も少ない紫色活性化、および強い発現を有する組み合わせの変異体であるC1V1(E122T/E162T)の生成をもたらした。実際に、C1V1変異形は、電流の大きさ、非活性化動態、および作用スペクトルの検討(表1)に基づいて、異なる用途に選択することができ、例えば、二光子作業では、ChR2の2P活性化が、電流の大きさおよび速いチャネル閉鎖動態のために困難であるため、C1V1(E162T)が目的のものである可能性が高い。C1V1変異形は、細胞のE/Iバランスの上昇のレベルを増加させることが、以前は統合失調症および自閉症の両方に関連していた現象である、γ周期性の強度を増加させることにつながるという仮説の直接的な検証を可能にした。当然ながら、異なる手段もまた相乗効果があり、C1V1変異形をChR2と一緒に使用することにより、この研究で取り組む2つの異なるニューロン集団である、興奮性錐体ニューロン、および高速スパイクのパルブアルブミン発現抑制性介在ニューロンにおいてスパイクの確実で別々な駆動を可能にし、安定して上昇した細胞のE/Iバランスが、γバンド回路活性の生成に効果的であり、光遺伝学的手段における動態およびスペクトルの両方の差異を十分に利用していることを確認する。この種類のコンビナトリアル活性化は、複数の細胞型を越えて複数の神経経路型にまで及び得、例えば、システム神経科学の念願の目標である、単一の脳領域内で、異なる位置から生じる2つの軸索求心性集中経路における分離可能なスパイクの活性化にまで及び得る。
