1,5-ビス{[2-(ジメチルアミノ)エチル]アミノ}アントラセン-9,10-ジオンを、WO99/65992の実施例1に従って合成した。クロロホルム(2mL)を、1,5-ビス{[2-(ジメチルアミノ)エチル]アミノ}アントラセン-9,10-ジオン(58mg、0.152mmol)に添加した。暗紫色溶液に、MeCN(1ml)、続いてヨウ化メチル(95μl、1.524mmol)を添加した。10分間の撹拌後、沈殿物が形成する。4時間の撹拌後、揮発性物質を蒸発させた。残渣をクロロホルム(5ml)から摩砕し、濾過によって回収し、ジクロロメタン(20ml)およびジエチルエーテル(10ml)で洗浄した。暗いピンク色の固体が単離された(0.09g、0.135mmol、89%収率)。1H NMR(400MHz, d6-DMSO) δ: 9.69(2H, t)、7.72(2H, dd)、7.52(2H, dd)、7.30(2H, dd)、3.91(4H, q)、3.62(4H, t)、3.18(18H, s)
1,5-ビス{[2-(ジメチルアミノ)エチル]アミノ}-5,8-ジヒドロキシアントラセン-9,10-ジオンを、WO99/65992の実施例1に従って合成した。クロロホルム(2mL)を、1,5-ビス{[2-(ジメチルアミノ)エチル]アミノ}-5,8-ジヒドロキシアントラセン-9,10-ジオン(44mg、0.107mmol)に添加した。暗紫色溶液に、MeCN(2ml)、続いてヨウ化メチル(66.6μl、1.067mmol)を添加した。1分間の撹拌後、沈殿物が形成する。4時間の撹拌後、揮発性物質を蒸発させた。残渣をクロロホルム(5ml)から摩砕し、濾過によって回収し、ジクロロメタン(20ml)およびジエチルエーテル(10ml)で洗浄した。暗青色固体が単離された。収率=173-012(0.05g, 0.068mmol, 63.9%収率) 1H NMR(400MHz, d6-DMSO) δ: 13.97(2H, s)、9.77(2H, br t)、7.49(2H, d)、7.40(2H, d)、3.96(4H, br q)、4.09(4H, t)、3.19(18H, s)
化合物(IA)を、実施例2に記載されている原理を使用して合成し、緩衝液中の5mM化合物(IA)希釈のストック溶液として+4℃で保存した。吸光度スペクトルを、分光計およびPBS中に溶解させた作用物質の20μM溶液を使用して得、および1cm光路石英シリカキュベットにおいて測定した。1cm光路長セミマイクロ石英キュベットにおける20μM化合物(IA)の溶液についての蛍光スペクトルを、633、589、534、488nmでの励起によって決定した。蛍光測定は、Perkin Elmer LS50分光蛍光計においてスリット幅を10nmに設定して行った。分光蛍光計は、赤色感受性の光電子倍増管(タイプR928;浜松ホトニクス株式会社(Hamamatsu Photonics KK)、日本)を備えていた。データを蓄積し、スプレッドシートにエクスポートして、緩衝液対照について補正し、発光最大を決定した。結果を、図8に示す。化合物(IA)の488nm(フローサイトメトリーにおいて)から647nmまでの波長(Exλmax 646nm)による励起は、最適に及ばなくてもよい。典型的には、細胞イメージングでは、励起は633nmまたは647nmのいずれかの波長で実施される。発光スペクトルは、励起波長と無関係であり、すなわち、すべての発光スペクトルは、励起波長に関係なく同一である。
この実施例は、細胞毒性薬物(VP-16)に曝露させたヒトB細胞リンパ腫(DoHH2)細胞中の原形質膜のホスファチジルセリン分子の内部(細胞質の)小葉からの移動と関連する、より後の段階の細胞死に関する。アポトーシスの用量依存的な誘導の検出を、フローサイトメトリーを使用して実施した。
VP-16(VP-16-213;VEPESID;エトポシド)は、34mMストック溶液(Bristol Meyers Pharmaceuticals、Syracuse、NY)として提供され、4℃で保存した。フルオレセインコンジュゲートアネキシンV(アネキシンV-FITC)は、Pharmingen(Becton Dickinson UK、Oxford、UK)から購入した。ヨウ化プロピジウム(PI)は、H2O中の1mg/ml溶液(Molecular Probes Europe、Leiden、オランダ)として得た。化合物(IA)は、水中に5mM溶液として配合し、4℃で保存した。
本発明者らは、核のDNAを効率的に標的化する化合物(IA)の特性を求めた。
この実施例は、フローサイトメトリーによって分析されるヒトJurkat細胞集団におけるスタウロスポリンによる細胞死誘導の分析を示す。本発明者らは、スタウロスポリンにより誘導された細胞死の結果として膜透過性が増強された細胞に起因する生存不能な部分を決定する細胞生存可能性マーカーとしての、化合物(IA)の適用を実証しようとした。
細胞培養および処理:ヒト濾胞性Bリンパ腫細胞系をこの研究において使用した。SU-DHL-4を、10%FCSおよび100 U ml-1ペニシリン、100μg ml-1ストレプトマイシン、および2mMグルタミンを添加したRPMI1640中で常法に従って維持した。細胞を、37℃で5%CO2/95%空気の加湿した雰囲気中で培養した5×104細胞ml-1の開始濃度で、1週間に2回継代した。細胞をフラスコ当たり5mlで、0.5×105細胞/mlに設定した。各培養物を、3つの化合物(IA)用量(0, 3および10μM)のうちの1つに、標準的な培養条件下で96時間、継続的に曝露させた。時間(t)0、24、48、72および96時間で、細胞密度を、各フラスコの0.4ml試料のCoulter計数によって決定した。データは、所与の濃度の化合物(IA)について時間(時間)に対する相対的細胞数(Nt/N0)(695LP)の増加として表示する。結果を、図12に示す。
インタクト細胞と損傷細胞とを区別する手段を提供するための、アッセイへの同時組み込みのための利点を有する、細胞の完全性の喪失の報告における、対象とする特性を有する別の蛍光試薬を使用した、典型的なマルチパラメータ分析への好ましい細胞不透過性色素(式(IA))の化合物の組み込み。
細胞培養および薬物処理:ヒト濾胞性Bリンパ腫細胞系をこの研究において使用した。DoHH2を、5%FCSおよび100U ml-1ペニシリン、100μg ml-1ストレプトマイシン、および2mMグルタミンを添加したRPMI1640中で常法に従って維持した。細胞を、37℃で5%CO2/95%空気の加湿した雰囲気中で培養した5×104細胞ml-1の開始濃度で、1週間に2回継代した。細胞を5×105細胞/mlに設定し、VP-16用量(0.25μM)に曝露させてアポトーシスを誘導した。20μM化合物(IIA)および4μM化合物(IA)を、1mlの細胞に添加し、標準的な培養条件下で10分間インキュベートした。試料をフローサイトメトリーによって分析した。
Claims (11)
- 式(I)の化合物:
X1、X2およびX3は、水素、ヒドロキシル、NR1-A-NR2R3R4+(Zm-)1/m、ハロゲノアミノ、C1〜4アルコキシまたはC2〜8アルカノイルオキシから独立して選択され;
(Zm-)1/mは、電荷mのアニオンである]
または基NR1が四級化されている誘導体と、
少なくとも第2の蛍光色素または発光化合物との混合物を含む組成物であって、
前記第2の蛍光色素または発光化合物が、式(II)の化合物:
X1、X2およびX3は、水素、ヒドロキシル、NR1-A-NR2R3、ハロゲノアミノ、C1〜4アルコキシまたはC2〜8アルカノイルオキシから独立して選択される]
またはそのN-オキシド誘導体
である、組成物。 - 式(I)の化合物において、X1、X2およびX3のうちの少なくとも1つが、NR1-A-NR2R3R4+(Zm-)1/mである、請求項1に記載の組成物。
- 式(I)の化合物において、X1およびX3ではなくX2が、NR1-A-NR2R3R4+(Zm-)1/mである、請求項2に記載の組成物。
- X1およびX3が両方ともヒドロキシルである、請求項1から3のいずれか一項に記載の組成物。
- X1およびX3が両方とも水素である、請求項1から4のいずれか一項に記載の組成物。
- R1が水素である、請求項1から5のいずれか一項に記載の組成物。
- R2、R3およびR4がC1〜4アルキルである、請求項1から6のいずれか一項に記載の組成物。
- R2、R3およびR4がメチルである、請求項7に記載の組成物。
- Aが(CH2)2である、請求項1から8のいずれか一項に記載の組成物。
- 式(II)の化合物が、1,5アミノ置換アントラキノンである、請求項1から9のいずれか一項に記載の組成物。
