溶媒は、さらなる精製を行うことなく規定の品質で使用した。アセトニトリル(勾配グレード、Sigma−Aldrich);ジクロロメタン(AnalaR Normapur、VWR);酢酸エチル(研究所試薬グレード、Fisher Scientific);N,N−ジメチルホルムアミド(ペプチド合成グレード、Biosolve);1−メチル−2−ピロリドン(バイオテクノロジーグレード、Sigma−Aldrich);1,4−ジオキサン(Emplura、Merck);メタノール(p.a.、Merck)。水:Milli−Q Plus、Milliporeで脱塩されたもの。
化学物質は文献手順に従って、もしくは文献手順と同様にして合成し、またはSigma−Aldrich−Fluka(Deisenhofen、Germany)、Bachem(Bubendorf、Switzerland)、VWR(Darmstadt、Germany)、Polypeptide(Strasbourg、France)、Novabiochem(Merck Group、Darmstadt、Germany)、Acros Organics(distribution company Fisher Scientific GmbH、Schwerte、Germany)、Iris Biotech(Marktredwitz、Germany)、Amatek Chemical(Jiangsu、China)、Roth(Karlsruhe、Deutschland)、Molecular Devices(Chicago、USA)、Biochrom(Berlin、Germany)、Peptech(Cambridge、MA、USA)、Synthetech(Albany、OR、USA)、Pharmacore(High Point、NC、USA)およびAnaspec(San Jose、CA、USA)あるいは他の会社から購入し、それ以上精製を行うことなく指定の品質で使用した。
HT29(Cat.No.91072201)はECACCから購入し、Capan−1はATCC(Cat No.HTB−79)細胞から入手した。ヒト、マウスおよびラットNTR1を発現するHEK293細胞は、Trenzyme(Konstanz、Germany)によって産生された。細胞は、pExoIN2プラスミドベクター(図1を参照)によってコードされており、ユビキチンのN末端に結合されている赤血球凝集素エピトープ(HA)−タグ付きピューロマイシンN−アセチルトランスフェラーゼからなる発現系を使用して、安定的にトランスフェクトし、次いで、これをNTR1のN末端に結合させる。この系は、トランスフェクトされたタンパク質の効果的な発現を確実にする。安定した細胞株およびpExoINベクターの産生は、Matentzoglu et al.,BioTechniques,2009,46,21−28に記載されている。
分取HPLC分離は、個別の実施例に記載されているカラムおよび勾配を用いて実施した。勾配については、次の溶媒を使用した:
A:H2O中0.05%TFA
B:ACN中0.05%TFA。
直線バイナリ勾配をすべての分離において使用した。例えば:勾配が「30分間で20から60%のB」と記載されている場合、これは、30分以内に20%のB(および80%のA)から60%のB(および40%のA)までの直線勾配を意味する。流速はカラムサイズに依存する:それぞれ、カラム25mm直径に関しては、流速は30ml/minであり、カラム50mm直径に関しては60ml/minである。
本発明の化合物は、下記の方法を、有機合成化学の技術分野において公知の合成法または当業者には明らかであるようなその変更法と一緒に使用して合成することができる。好ましい方法としては、限定するものではないが、下記の方法が挙げられる。下記に引用したそれぞれの参考文献は、参照により本明細書に援用される。
トリチルクロライドポリスチレン樹脂(初期充填1.8mmol/g、1.11g、2mmol、1.0当量)を30分間DCM中で膨化させた。次いで、この樹脂にDCM(6.5ml)中のN,N−ビス[3−(メチルアミノ)−プロピル]メチルアミン(1.6ml、8mmol、4当量)を加え、混合物を一晩振盪した。その後、樹脂をDMF、DCMおよびジエチルエーテル(5/3/1)で連続的に洗浄し、真空で乾燥させた。
N,N−ビス[3−(メチルアミノ)−プロピル]メチルアミンを充填したトリチル樹脂(1g、1.8mmol、1.0当量)を30分間DMF中で膨化させた。樹脂をDMF/DIPEA(9/1)(残留物N,N−ビス[3−(メチルアミノ)−プロピル]メチルアミン塩酸塩を除去するため)およびDMF(3/3)で洗浄した。1−(4−カルボキシ−2−イソプロピル−フェニル)−5−(2,6−ジメトキシ−フェニル)−1H−ピラゾール−3−カルボン酸メチルエステル(X)(1.15g、2.7mmol、1.5当量)[US5723483で開示されているように調製したもの]、HATU(1.03g、2.7mmol、1.5当量)およびDIPEA(937μl、5.4mmol、3当量)をDMF(18ml)中に溶解し、1分間完全に混合した。活性化した構成成分を添加した後、樹脂を一晩振盪した。樹脂を洗浄し(DMF5回、DCM3回およびジエチルエーテル)、真空で乾燥させた。反応の完了は以下のように確認した:樹脂試料は、30分間、安息香酸、HATUおよびDIPEA(1/1/2)のDMF溶液で処理した。DMFおよびDCMで洗浄した後、TFAを樹脂に添加した。LC−MSにおいて安息香酸N,N−ビス[3−(メチルアミノ)−プロピル]メチルアミドが存在しないことは、樹脂上に遊離アミノ画分が存在しないことを示し、したがって、1−(4−カルボキシ−2−イソプロピル−フェニル)−5−(2,6−ジメトキシ−フェニル)−1H−ピラゾール−3−カルボン酸メチルエステルのカップリングが完了している証拠を提供した。
前述の樹脂(1.64g、1.75mmol、1.0当量)を、ジオキサン(35ml)およびLiOH水和物(689mg、16mmol、10当量)の水(12ml)溶液で一晩処理した。この手順を一回繰り返し、樹脂を水、DMFおよびDCM(3/3/3)で連続して洗浄し、真空乾燥させた。
前述の樹脂(0.7g、0.75mmol、1.0当量)を30分間DMF中で膨化させた。その後、HOAt(153mg、1.13mmol、1.5当量)、DIC(232μl、1.5mmol、2.0当量)および2−アミノ−アダマンタン−2−カルボン酸tert−ブチルエステル(942mg、3.75mmol、5.0当量)をDMFおよびDCM(2:1)の混合物(6ml)中に溶解し、続いて樹脂に添加した。2.5時間後、さらなるDIC(232μl、1.5mmol、2.0当量)を添加した。樹脂を60分間振盪させ、その後反応は完了した。次いで、樹脂をDMFおよびDCM(3/3)で洗浄し、真空乾燥させた。
残留物をHPLC精製(30分で15から45%のB、Agilent PLRP−S 25x150mm)に供し、表題化合物(210mg、26.3μmol、35.0%)を得た。HPLC:Rt=5.5分。MS:m/z=799.4([M+H]+、計算値799.5)。C46H66N6O6(MW=799.05)。
残留物をHPLC(30分で20から50%のB、Agilent PLRP−S 25x150mm)により精製し、表題化合物(155.5mg、0.21mmol、28%)を得た。HPLC:Rt=4.7分。MS:m/z=743.4([M+H]+、計算値742.4)。C42H57N6O6(MW=741.94)。
DOTA(tBu)3−OH(500mg、0.873mmol、1.0当量)を乾燥DMF(5ml)に溶解した。N,N’−ジメチル−N−(3−メチルアミノ−プロピル)−プロパン−1,3−ジアミン(3.5ml、17.5mmol、20当量)およびDIPEA(0.389ml、2.27mmol、2.6当量)を添加した後、混合物を0℃まで冷却した。PyBOP(590mg、1.13mmol、1.3当量)を乾燥DMF(5ml)に溶解させた。このPyBOP溶液の0.5mlを、すべての溶液を添加するまで、反応混合物に5〜10分毎に添加した。1時間後、DMFを真空下で除去した。残りの残留物をEtOAc(100ml)に溶解させ、水(5×5ml)で抽出した。有機層をNa2SO4で脱水し、蒸発させ、1.01gの粗製物質を得た。
式(XI)のメチルエステル(294mg、0.259mmol)を1,4−ジオキサン(1.35ml)に溶解した。LiOH(1.04ml、1.04mmol、4当量)の1Mの水溶液を滴下して加えた。5時間撹拌した後、HOAc(0.373ml)でpHを5〜6に調整した。ACN(18ml)および水(225ml)を添加した後、混濁溶液を固相抽出カラムに供し(60mlポリスチレンシリンジ中、3.0gのVarian Bondesil−ENV)、メタノール(3×20ml)および水(3×20ml)で予備洗浄した。第1のフラクションとしてカラムを10%ACN水溶液60mlで溶出し、次のそれぞれのフラクションは、0.1%TFAを含有する50%ACN水溶液60mlで溶出した。3〜8のフラクションを凍結乾燥した後、表題化合物(248mg、86%)を得た。HPLC:Rt=4.9min。MS:m/z=1120.7([M+H]+、計算値1120.7)。C59H93N9O12(MW=1120.42)。
式(XII)のカルボン酸(248mg、0.222mmol)を乾燥NMP(3ml)に溶解させた。HATU(84.3mg、0.222mmol、1.0当量)を固体として添加した。この混合物に、DIPEA(76μl、0.443mmol、2.0当量)を添加した。5分間撹拌した後、この溶液は、5分以内に、2−アミノ−アダマンタン−2−カルボン酸(43.3mg、0.222mmol、1.0当量)の乾燥NMP(6.5ml)懸濁液に移した。室温で1時間置いた後、フラスコを浴温65℃で油浴により加熱した。6時間後、DIPEA(38μl、0.222mol、1.0当量)を添加し、加熱をさらに18時間継続した。冷却した後、1:1のACN/水を添加し、溶液を凍結乾燥した。100μlのDMSO/200μlのHOAcおよび1mlのACNを残りの固形物に加え、懸濁液を濾過した。濾液をprep.HPLC(30分で20から60%のB、Agilent PLRP−S 25×150mm)によって分離し、表題化合物(74mg、0.057mmol、収率26%)を得た。HPLC:Rt=5.1min。MS:m/z=1297.7([M+H]+、計算値1197.8)。C70H108N10O13(MW=1297.67)。
TFA(9ml)を式(XIII)のトリス−tBu−エステル(74mg、0.057mmol)およびトリイソブチルシラン(600μl)の乾燥DCM(2.4ml)溶液に添加した。室温で5時間後、混合物を減圧下で蒸発させ、prep.HPLC(30分で15から50%のB、Agilent PLRP−S 25x150mm)によって精製した。これによって、TFA塩として表題化合物(43mg、0.038mmol、66%収率)を得た。HPLC:Rt=5.3min。MS:m/z=1129.7([M+H]+、計算値1129.6)。C58H84N10O13(MW=1129.35)。
A.DOTA−遷移金属複合体を合成する一般的手順
対応する金属塩(3.0当量から5.0当量)の1mM溶液を、5倍容量の酢酸緩衝液(pH5.0、0.4M)で希釈した。この溶液をDOTA含有化合物(3から10mg、1.0当量)に添加した。反応物は油浴(浴温90℃)に置いた。20分後、反応混合物をRTに冷却し、固相抽出カラム(15mlポリスチレンシリンジ中250mgのVarian Bondesil−ENV)に供し、メタノール(1×5ml)および水(2×5ml)で予備洗浄した。このカラムを水(2×5ml)で溶出し、第1フラクションとしての50%ACN水溶液5mlおよび次のそれぞれのフラクションを0.1%TFA含有水中の50%ACN5mlで溶出した。純生成物を含有するフラクションをプールし、凍結乾燥した。
複合体形成は、一般的手順(実施例6A)に従って実施したが、次の試薬:式(IIIa)の化合物(5.0mg)、InCl3×4H2O(3.9mg)を使用し、表題化合物(4.26mg、3.4μmol、78%)を得た。HPLC:Rt=4.4min。MS:m/z=1241.6([M+H]+、計算値1241.5)。C58H81InN10O13(MW=1241.14)。
複合体形成は、一般的手順(実施例6A)に従って実施したが、次の試薬:式(IIIa)の化合物(3.0mg)およびGa(NO3)3水和物(3.9mg)を使用し、表題化合物(2.61mg、2.2μmol、82%)を得た。HPLC:Rt=4.4min。MS:m/z=1195.6([M+H]+、計算値1195.5)。C58H81GaN10O13(MW=1196.05)。
複合体形成は、一般的手順(実施例6A)に従って実施したが、次の試薬:式(IIIa)の化合物(3.0mg)およびY(NO3)3×6H2O(3.1mg)を使用し、表題化合物(2.54mg、2.1μmol、79%)を得た。HPLC:Rt=4.5min。MS:m/z=1215.6([M+H]+、計算値1215.5)。C58H81N10O13Y(MW=1216.24)。
複合体形成は、一般的手順(実施例6A)に従って実施したが、次の試薬:式(IIIa)の化合物(3.0mg)およびLuCl3(2.2mg)を使用し、表題化合物(2.88mg、2.2μmol、83%)を得た。HPLC:Rt=4.4min。MS:m/z=1301.5([M+H]+、計算値1301.5)。C58H81LuN10O13(MW=1301.30)。
クロロトリチル樹脂(167mg、0.3mmol、1.0当量)を1時間DCM中で膨化させた後、Fmoc−Ttds−OH(326mg、0.6mmol、2.0当量)およびDIPEA(155μl、0.9mmol、3.0当量)のDCM(4ml)溶液を添加した。2.5時間後、溶液を濾過し、DCM、MeOH、DCMおよびDMF(1/1/1/3)で連続的に樹脂を洗浄した。樹脂をDMF中の20%ピペリジンで2回処理し(2分および20分)、その後、DMFで5回洗浄した。次に、Tri−tert−ブチル1,4,7,10−テトラアザシクロ−ドデカン−1,4,7,10−テトラアセテート(DOTA(tBu)3−OH)(322mg、0.56mmol、1.9当量)、HATU(214mg、0.56mmol、1.9当量)およびDIPEA(195μl、1.13mmol、3.8当量)の混合物を5分間振盪し、続いて、樹脂に添加した。2時間撹拌した後、樹脂をDMFおよびDCM(5/2)で洗浄し、続いて、真空で乾燥した。樹脂を4回、5分間、TFA、TIPSおよびDCM(5/5/90)の混合物で処理した。DOTA保護基の早期喪失を防止するため、得られた溶液を水性緩衝液(10ml、pH=8、100mM NH4(CO3)2)に直ちに注いだ。混合物のpH値は、4NのNaOH溶液を添加することによってpH=7以上に保持した。標的化合物を含有するDCM緩衝混合液を合わせ、相を分離させ、水相をDCMで2回抽出し、有機相を蒸発乾固した。残留物をACN/水(1/1)に再融解し、凍結乾燥した。
残留物をHPLC(30分で15から45%のB、Agilent PLRP−S 25×150mm)により精製し、表題化合物(118.6mg、0.136mmol、45%)を得た。HPLC:Rt=4.3min。MS:m/z=875.5([M+H]+、計算値875.6)。C42H78N6O13(MW=875.10)。
2−({5−(2,6−ジメトキシ−フェニル)−1−[2−イソプロピル−4−(メチル−{3−[メチル−(3−メチルアミノ−プロピル)−アミノ]−プロピル}−カルバモイル)−フェニル]−1H−ピラゾール−3−カルボニル}−アミノ)−アダマンタン−2−カルボン酸tert−ブチルエステル(XIX)(24.9mg、31.1μmol、1当量)をDMF(0.5ml)中に溶解した。DIPEA(32.4μl、187μmol、6当量)を溶液に添加してpH値をpH=7に調整した。N−{3−[2−(2−{3−[2−(4,7,10−トリス−tert−ブトキシカルボニルメチル−1,4,7,10−テトラアザ−シクロドデカ−1−イル)−アセチルアミノ]−プロポキシ}−エトキシ)−エトキシ]プロピル}−スクシンアミド酸(DOTA(tBu)3−Ttds−OH)(XX)(30.0mg、34.3μmol、1.1q当量)を溶液に加え、続いてHOAt(16.9mg、124.4μmol、4当量)およびDIC(14.5μl、93.3μmol、3当量)を加えた。24時間混合物を撹拌した後、溶媒をエバポレーションで除去した。残りの残留物に、水(1ml)およびEtOAc(2ml)を添加した。有機相を分離し、乾燥し、蒸発させた。残留物を8時間、TFA、フェノール、水およびTIPS(18/1/1/2)(330μl)で処理した。すべての揮発性物質を真空下で除去した。
複合体形成は、次の試薬:式(IIIb)の化合物(4.0mg)およびLuCl3(2.35mg)を使用し、一般的手順(実施例6A)に従って実施し、表題化合物(2.61mg、1.6μmol、57%)を得た。HPLC:Rt=4.7min。MS:m/z=1603.8([M+H]+、計算値1603.7)。C72H107LuN12O18(MW=1603.66)。

2−({5−(2,6−ジメトキシ−フェニル)−1−[2−イソプロピル−4−(メチル−{3−[メチル−(3−メチルアミノ−プロピル)−アミノ]−プロピル}−カルバモイル)−フェニル]−1H−ピラゾール−3−カルボニル}−アミノ)−アダマンタン−2−カルボン酸tert−ブチルエステル(XIX)(12.7mg、15.9μmol、1当量)をDMF(0.3ml)中に溶解した。DIPEA(16.6μl、95.4μmol、6当量)を溶液に添加してpH値をpH=7に調整した。6−[2−(4,7,10−トリス−tert−ブトキシカルボニルメチル−1,4,7,10−テトラアザ−シクロドデカ−1−イル)−アセチルアミノ]−ヘキサン酸(DOTA(tBu)3−Ahx−OH)(XXI)(16.4mg、23.85μmol、1.5q当量)を溶液に加え、続いてHOAt(8.7mg、63.6μmol、4当量)およびDIC(7.4μl、47.7μmol、3当量)を加えた。72時間混合物を撹拌した後、溶媒をエバポレーションで除去した。残りの残留物に、水(1ml)およびEtOAc(2ml)を添加した。有機相を分離し、乾燥し、蒸発させた。残留物を8時間、TFA、フェノール、水およびTIPS(18/1/1/2)(330μl)で処理した。すべての揮発性物質を真空下で除去した。
複合体形成は、次の試薬:式(IIId)の化合物(10.0mg)およびGa(NO3)3水和物(7.47mg)を使用し、一般的手順(実施例6A)に従って実施し、表題化合物(7.46mg、6.4μmol、70%)を得た。HPLC:Rt=4.8min。MS:m/z=1166.6([M+H]+、計算値1166.5)。C57H78GaN9O13(MW=1167.0)。
クロロトリチル樹脂(556mg、1.0mmol、1.0当量)を1時間、DCM中で膨化させた後、Fmoc−Ttds−OH(1085mg、2.0mmol、2.0当量)およびDIPEA(516μl、3.0mmol、3.0当量)のDCM(10ml)溶液を添加した。2.5時間後、溶液を濾過し、DCM、MeOH、DCMおよびDMF(1/1/1/3)で樹脂を連続的に洗浄した。樹脂を、20%ピペリジンを含むDMFで2回処理し(2分および20分)、DMFおよびDCM(5/2)で洗浄し、真空で乾燥させ、760mgのH−Ttds−トリチル樹脂を得た(質量増加に基づいて充填:約0.8mmol/g)。H−Ttds−トリチル樹脂(375mg、0.3mmol、1.0当量)は、30分間DMF中で膨化させた。次に、2−(4,7−ビス−tert−ブトキシカルボニルメチル−[1,4,7]トリアゾナン−1−イル)−ペンタン二酸1−tert−ブチルエステル(NODAGA(tBu)3−OH)(245mg、0.45mmol、1.5当量)、HATU(171mg、0.45mmol、1.5当量)およびDIPEA(150μl、0.9mmol、3.0当量)の混合物を5分間振盪し、続いて、樹脂に添加した。24時間撹拌した後、樹脂をDMFおよびDCM(5/2)で洗浄し、続いて、真空で乾燥させた。まず、樹脂をTFA、TIPSおよびDCM(2/5/93)の混合物で1回処理し、続いて、4回、TFA、TIPSおよびDCM(5/5/90)の混合物で5分間処理した。NODAGA保護基の早期喪失を防止するため、得られた溶液を水性緩衝液(10ml、pH=8、100mM NH4(CO3)2)へ直ちに注いだ。混合物のpH値は、4NのNaOH溶液を添加することによってpH=7以上に維持した。標的化合物を含有するDCM緩衝混合液(第1処理と第2処理で得た溶液)を合わせ、相を分離させ、水相をDCMで2回抽出し、有機相を蒸発乾固した。残留物をACN/水(1/1)に再融解させ、凍結乾燥した。
2−(4,7−ビス−tert−ブトキシカルボニルメチル−[1,4,7]トリアゾナン−1−イル)−4−[3−(2−{2−[3−(3−カルボキシ−プロピオニルアミノ)−プロポキシ]−エトキシ}−エトキシ)−プロピルカルバモイル]−酪酸tertブチルエステル(NODAGA(tBu)3−Ttds−OH)(XXII)(65mg、76μmol)をDMF(0.5ml)に溶解させた。この溶液(39mgのNODAGA(tBu)3−Ttds−OH(XXII)を含有、46μmol、1.3当量)の0.3mlを使用し、2−({5−(2,6−ジメトキシ−フェニル)−1−[2−イソプロピル−4−(メチル−{3−[メチル−(3−メチル−アミノ−プロピル)−アミノ]−プロピル}−カルバモイル)−フェニル]−1H−ピラゾール−3−カルボニル}−アミノ)−アダマンタン−2−カルボン酸tert−ブチルエステル(XIX)(32.0mg、35μmol、1当量)を溶解させた。DIPEA(42μl、250μmol、7当量)を溶液に添加し、pH値をpH=7に調整した。その後、HOAt(22mg、162μmol、4.5当量)およびDIC(19μl、122μmol、3.5当量)を混合物に添加し、これを続いて6時間撹拌した。その後、最初に調製した溶液(6.5mgのNODAGA(tBu)3−Ttds−OH(XXII)を含有する50μl、7.7μmol、0.2当量)およびDIC(10μl、64μmol、1.8当量)のさらなる量を添加し、この混合物を一晩撹拌した。すべての揮発性物質を真空で除去し、残留物をDCMおよびクエン酸水溶液(10%)で溶解した。有機層を分離し、乾燥させ、蒸発乾固した。残留物をTFA、TIPSおよび水(95/2.5/2.5)で処理した。
2−({5−(2,6−ジメトキシ−フェニル)−1−[2−イソプロピル−4−(メチル−{3−[メチル−(3−{1−メチル−3−[4−(3−DFO−チオウレイド)−フェニル]−チオウレイド}−プロピル)−アミノ]−プロピル}−カルバモイル)−フェニル]−1H−ピラゾール−3−カルボニル}−アミノ)−アダマンタン−2−カルボン酸(IIIf)(9.85mg、6.58μmol、1.0当量)およびジルコニウム(IV)アセチルアセトナート(12.95mg、26.3μmol、4.0当量)をMeOHに溶解した。1時間撹拌した後、溶媒を蒸発させた。
残留物をHPLC精製(30分で25から50%のB、Agilent PLRP−S 25×150mm)に供し、表題化合物(2.5mg、1.6μmol、24.2%)を得た。HPLC:Rt=5.1min。MS:m/z=1581.6([M]+、計算値1581.7)。C75H107N14O14S2Zr+(MW=1584.09)。
トリチルクロライド樹脂(初期充填1.8mmol/g、334mg g、0.6mmol、1.0当量)は、30分間DCM中で膨化させた。次いで、DCM(4ml)中のN,N−ジメチルジプロピレントリアミン(0.54ml、3mmol、5当量)およびDIPEA(0.2ml、1.2mmol、2.0当量)を樹脂に加え、混合物を一晩振盪した。その後、樹脂をDMF、DCM、MeOHおよびジエチルエーテル(5/3/1)で洗浄し、真空乾燥した。
N,N−ジメチルジプロピレントリアミンを充填したトリチル樹脂(0.6mmol、1.0当量)を30分間、DMF中で膨化させた。1−(4−カルボキシ−2−イソプロピル−フェニル)−5−(2,6−ジメトキシ−フェニル)−1H−ピラゾール−3−カルボン酸メチルエステル(382mg、0.9mmol、1.5当量)、HATU(342mg、0.9mmol、1.5当量)およびDIPEA(312μl、2.7mmol、3当量)をDMF(6ml)に溶解させ、1分間完全に混合した。活性化した構成成分を添加した後、樹脂を3時間振盪した。樹脂を洗浄し(DMF/DCM/ジエチルエーテル5/3/1)、真空乾燥した。
前述の樹脂(0.6mmol、1.0当量)を30分間ジオキサン中で膨化させ、その後、50℃で、ジオキサン(30ml)およびLiOH水和物(504mg、12mmol、20当量)を含む水(4ml)で処理した。この手順をRTで一晩継続し、樹脂を水、DCMおよびEt2O(3/3/3)で連続して洗浄し、真空乾燥させた。
前述の樹脂(0.6mmol、1.0当量を1時間、DMF中で膨化させた。次いで、HOAt(327mg、2.4mmol、4.0当量)、DIC(279μl、1.8mmol、3.0当量)および2−アミノ−アダマンタン−2−カルボン酸tert−ブチルエステル(453mg、1.8mmol、3.0当量)をDMFおよびDCM(2:1)の混合物(6ml)に溶解させ、樹脂に加えた。樹脂を60時間振盪させ、その後、反応は完了した。樹脂をDMFおよびDCM(3/3)で洗浄し、真空乾燥した。

DOTA(tBu)3−OH(200mg、0.349mmol、1.0当量)およびPyBOP(236mg、0.454mmol、1.3当量)を乾燥DMF(5ml)に溶解させた。1分後、N1−(3−ジメチルアミノ−プロピル)−プロパン−1,3−ジアミン(0.315ml、1.75mmol、5当量)およびDIPEA(0.155ml、0.98mmol、2.6当量)を含む乾燥DMF(2ml)を添加した。90分後、DMFを真空下で除去した。残りの残留物をEtOAc(30ml)に溶解させ、水で2回抽出した。有機層をNa2SO4で脱水し、蒸発させ、0.41gの粗製物質を得た。
式(XIV)のメチルエステル(370mg、0.330mmol)を1,4−ジオキサン(1.72ml)に溶解させた。LiOH(1.32ml、1.32mmol、4当量)の1M水溶液を滴下して加えた。5時間撹拌した後、HOAc(0.475ml)でpHを4に調整した。ACN(18ml)および水(100ml)を添加した後、混濁溶液を凍結乾燥した。この物質をACN(24ml)および水(300ml)に溶解させ、固相抽出カラム(60mlポリスチレンシリンジ中4.0gのVarian Bondesil−ENV)にアプライし、メタノール(3×25ml)および水(3×25ml)で予備洗浄した。第1のフラクションとしてカラムを10%ACNの水溶液80mlで溶出し、次のそれぞれのフラクションは、0.1%TFAを含有する50%ACN水溶液80mlで溶出した。フラクション4から6を凍結乾燥した後、表題化合物(313mg、86%)を得た。HPLC:Rt=4.4min。MS:m/z=1106.5([M+H]+、計算値1106.7)C58H91N9O12、(MW=1106.40)。
式(XV)のカルボン酸(287mg、0.260mmol)を乾燥NMP(3.7ml)に溶解させた。HATU(98.7mg、0.260mmol、1.0当量)を固体として加え、この混合物に、DIPEA(89μl、0.52mmol、2.0当量)を加えた。5分撹拌した後、この溶液を5分以内に、2−アミノ−アダマンタン−2−カルボン酸(50.7mg、0.260mmol、1.0当量)およびDIPEA(44μl、0.26mmol、1.0当量)を含む乾燥NMP(7.6ml)の懸濁液に移した。室温で1時間後、フラスコを浴温65℃の油浴で加熱した。6時間後、さらなる2−アミノ−アダマンタン−2−カルボン酸(50.7mg、0.260mmol、1.0当量)およびDIPEA(44μl、0.26mmol、1.0当量)を加え、さらに18時間加熱を継続した。冷却後、1:1のACN/水を添加し、溶液を凍結乾燥した。残留固形物をprep.HPLC(30分で20から60%のB、Agilent PLRP−S 25×150mm)により分離し、表題化合物(40mg、0.031mmol、12%収率)を得た。HPLC:Rt=5.0min。MS:m/z=1283.7([M+H]+、計算値1283.8)C69H106N10O13、(MW=1283.64)。
TFA(4.8ml)を、式(XVI)のトリス−tBu−エステル(40mg、36μmol)およびトリイソブチルシラン(320μl)を含む乾燥DCM(1.3ml)の溶液に添加した。室温で3.5時間後、混合物を減圧下で蒸発させ、prep.HPLC(30分で15から55%のB、Agilent PLRP−S 25×150mm)により精製した。これにより、TFA塩として表題化合物(24mg、19μmol、52%)を得た。HPLC:Rt=4.0min。MS:m/z=1115.6([M+H]+、計算値1115.6)C57H82N10O13、(MW=1115.32)。
2−{[1−{4−[(3−アミノ−プロピル)−(3−ジメチルアミノ−プロピル)−カルバモイル]−2−イソプロピル−フェニル}−5−(2,6−ジメトキシ−フェニル)−1H−ピラゾール−3−カルボニル]−アミノ}−アダマンタン−2−カルボン酸tert−ブチルエステル(XXIV)(24.4mg、31.1μmol、1.0当量)をDMF(0.5ml)に溶解させ、DIPEA(33μl、187μmol、6当量)を溶液に添加してpH値をpH=7に調整した。トリ−tert−ブチル−1,4,7,10−テトラアザシクロドデカン−1,4,7,10−テトラアセテート(DOTA(tBu)3−OH、16.9mg、34.3μmol、1.1当量)を添加した。次いで、HOAt(16.9mg、125μmol、4.0当量)およびDIC(14.5μl、95μmol、3.0当量)を混合物に加え、続いてこれを24時間撹拌した。すべての揮発性物質を真空で除去し、残留物をEtOAcおよび水に溶解させた。有機層を乾燥させ、蒸発させた。残留物を12時間、TFA、フェノール、水およびTIPS(18/1/1/2)(0.3ml)と一緒に撹拌した。

DOTA(tBu)3−OH(100mg、0.175mmol、1.0当量)を乾燥DMF(0.5ml)に溶解させ、HATU(66.4mg、0.175mmol、1.0当量)を乾燥DMF(0.5ml)に溶解させ、コリジン(46.1μl、0.350mmol、2.0当量)を添加した。5分後、この混合物は、エチレンジアミン(0.873mmol、5.0当量)の0℃乾燥DMF(1.5ml)冷却溶液にゆっくりと添加した。19時間撹拌した後、DMFを蒸発させ、残留油状物をEtOAc(5ml)に溶解させ、水(0.5ml)、sat.aq.NaHCO3(0.5ml)およびsat.aq.NaCl(0.5ml)で抽出した。有機層をNa2SO4で脱水し、蒸発させ、溶離剤としてDCM、DCM/メタノール20/1およびDCM/メタノール10/1を使用するフラッシュクロマトグラフィーによって残留物を精製した。これにより、45mgのモノアシル化エチレンジアミンを得た。この物質(18mg、29μmol)の乾燥DMF(0.2ml)溶液を、SR−142948(20mg、29μmol)[HATU(11.1mg、29μmol)およびDIPEA(10μl、58μmol、2当量)を含む乾燥DMF(0.4ml)]の10分間予備活性化した溶液に添加した。15時間後、モノアシル化エチレンジアミン(9mg、15μmol、0.5当量)を含む乾燥DMF(0.1ml)を添加した。5時間後、反応混合物を30分間60℃に加熱した。次いで、溶媒を蒸発させ、この物質をprep.HPLC(30分で15から55%のB、Agilent PLRP−S 25×150mm)によって精製した。これにより、式(XVIII)の表題化合物(15mg、12μmol、40%)を得た。HPLC:Rt=3.9min。MS:m/z=1282.7([M+H]+、計算値1282.8)C69H107N11O12、(MW=1282.65)。
TFA(1.5ml)を、式(XVIII)のトリス−tBu−エステル(15mg、11μmol)およびトリイソブチルシラン(100μl)の乾燥DCM(0.4ml)溶液に添加した。室温で4時間後、混合物を減圧下で蒸発させ、prep.HPLC(30分で15から45%のB、Agilent PLRP−S 25×150mm)によって精製した。これにより、TFA塩として表題化合物(6.3mg、5.7μmol、48%)を得た。HPLC:Rt=3.4min。MS:m/z=1114.6([M+H]+、計算値1114.6)C57H83N11O12、(MW=1114.33)。
Ca2+イオンは、通常、細胞のサイトゾルにナノモルのレベルで維持されており、多数のシグナル伝達経路において二次伝達物質として作用する。ニューロテンシン受容体をはじめとする多くのGPCRは結合してカルシウムイオンシグナル伝達を誘導し、多くの主要な細胞アッセイは、GPCR活性化の機能的読み出しとして細胞内カルシウムイオン濃度の測定を使用する。標準アッセイプロトコルにおけるカルシウムイオン濃度の変化は、細胞内のCa2+イオン濃度の変化が生じた時に発光する蛍光色素を用いて容易に検出することができる。これらの応答の一時的な性質を前提として、それらは、多くの場合、「注入し読み取る」能力を有する機械で読み取られる。本実施例は、本発明の化合物がNTR1発現細胞に対してアゴニスト活性を有していないことを示す。さらに、本実施例は、本発明の化合物がNTR1に結合し、さらに存在しているNTR1アゴニストの活性を阻害することを示す。
NTR1に対する放射性標識を含む化合物の結合親和性を決定するため、放射性リガンド結合アッセイを実施した。放射性リガンドは、身体の細胞受容体系の診断用または研究向けの試験用に使用される放射性生化学物質である。インビボ系において、これは、多くの場合、放射性リガンドの結合部位への試験分子の結合の定量するために使用される。分子の親和性が高くなるほど、より多くの放射性リガンドが結合部位から置き換えられる。結合した放射性リガンドの量はシンチレーション計測により測定し、それによって定量することができる。このアッセイは、一般に、分子の受容体への結合定数を計算するために使用される。この実施例は、本発明の化合物が高親和性でNTR1に結合することを示している。
血漿安定性アッセイは、血漿での本発明の化合物の分解を測定するために実施した。これは、プロドラッグを除き、血漿で急速に分解する化合物が一般にインビボにおいて効果が低く見える場合の化合物の重要な特徴である。

薬剤の効果は、血漿内のタンパク質にそれが結合する程度によって影響を受ける可能性がある。血液中の薬剤は、結合型および非結合型の2つの形態で存在する。血漿タンパク質に対する特異的な薬剤の親和性に応じて、一部の薬剤は血漿タンパク質に結合され、残りは未結合となり得る。注目すべきは、薬理効果を示す非結合フラクションである。また、代謝され、かつ/または排出され得るのもこのフラクションである。タンパク質結合は、薬剤の体内における生物学的半減期に影響を及ぼし得る。結合型の部分は、薬剤が非結合型の形態としてゆっくり放出されるリザーバーまたはデポーとして作用し得る。
特異性スクリーニングは、本発明の化合物の明確でない結合について試験するために実施した。NTR1についての特異性は、GPCR、イオンチャネルおよび輸送タンパク質に対する55のアッセイを含む標準の一連のアッセイ(「ExpresSProfile」)を使用して試験した。このアッセイは、Cerep(Celle l’Evescault、France;カタログ参照番号P1)によって実施した。
オートラジオグラフィーによって、組織切片上のその受容体への物質の結合を決定することができる。したがって、この方法を使用して本発明の一部の化合物の結合を決定し、組織当たりの受容体結合部位の数を定量した。驚いたことに、受容体に対するアゴニストおよびアンタゴニストの同様の親和性を比較した場合、アンタゴニストはアゴニストよりも多くの受容体結合部位を認識する。これは、診断上または治療上有効な薬剤としての本発明の化合物の特定の適合性を強調するものである。
診断活性薬または治療活性薬として機能するためには、化合物を放射性同位体で標識化する必要がある。標識化の手順は、本発明の放射性標識化合物の高放射化学的収率および高純度を確保するために適切であることが必要である。この実施例は、本発明の化合物が放射性標識に適切であり、高放射化学的収率および高純度で標識化することができることを示している。
放射性標識化合物は、SPECTおよびPETなどの画像診断法によって検出することができる。さらに、こうした技術によって取得されるデータは、本発明の放射性標識化合物を注射した動物から調製された、個々の器官に含まれている放射活性を直接測定することによって確認することができる。したがって、放射性標識化合物の生体内分布を決定し分析することができる。この実施例は、本発明の化合物が腫瘍の画像診断および治療的処置に適切な生体内分布を示すことを示している。

Claims (34)
- 式(I)の化合物:
R1は、水素、メチルおよびシクロプロピルメチルからなる群から選択され;
AA−COOHは、2−アミノ−2−アダマンタンカルボン酸、シクロヘキシルグリシンおよび9−アミノ−ビシクロ[3.3.1]ノナン−9−カルボン酸からなる群から選択されるアミノ酸であり;
R2は、(C1〜C6)アルキル、(C3〜C8)シクロアルキル、(C3〜C8)シクロアルキルメチル、ハロゲン、ニトロおよびトリフルオロメチルからなる群から選択され;
ALKは、(C2〜C5)アルキリデンであり;
R3、R4およびR5は、それぞれ独立して、水素および(C1〜C4)アルキルからなる群から選択され、但し、R3、R4およびR5の1つは次式(II)

ALK’は、(C2〜C5)アルキリデンであり;
R6は、水素および(C1〜C4)アルキルからなる群から選択され;
R7は、エフェクター部分であり、
前記エフェクター部分がアクセプター、−[アクセプター−エフェクター]、−[リンカー−アクセプター]、および−[リンカー−アクセプター−エフェクター]からなる群から選択され、
−[アクセプター−エフェクター]は、前記エフェクターが前記アクセプターと複合体形成されているか、または共有結合されている部分であり、
−[リンカー−アクセプター]は、前記リンカーが前記アクセプターにコンジュゲートされている部分であり、
−[リンカー−アクセプター−エフェクター]は、前記リンカーが前記アクセプターにコンジュゲートされており、前記エフェクターが前記アクセプターと複合体形成されているか、または共有結合されている部分であり、
前記エフェクターが診断上有効な薬剤、治療上有効な薬剤およびそれらの組み合わせからなる群から選択され、
前記アクセプターがキレート剤または芳香族化合物であり、
前記リンカーは、前記アクセプターを式(II)のN原子に連結する部分であり、前記リンカーと式(II)の基のN原子の間の共有結合の種類がアミド、尿素、チオ尿素およびアルキルアミンからなる群から選択され;リンカーとアクセプターの間の共有結合の種類がアミド、アルキルアミン、尿素、エーテル、チオエーテル、チオ尿素およびカルバメートからなる群から選択されるものである]
またはそれらの薬理学的に許容される塩、溶媒和物もしくは水和物。 - R1がメチルである、請求項1に記載の化合物。
- AA−COOHが2−アミノ−2−アダマンタンカルボン酸およびシクロヘキシルグリシンからなる群から選択されるアミノ酸である、請求項1または2に記載の化合物。
- R2がイソプロピルである、請求項1、2および3のいずれか1項に記載の化合物。
- R3、R4およびR5が、それぞれ独立して、水素およびメチルからなる群から選択され、但し、R3、R4およびR5の1つは次式(II)

- 前記エフェクターが治療上有効な核種である、請求項1、2、3、4、5および6のいずれか1項に記載の化合物。
- 前記エフェクターが治療上有効な放射性核種である、請求項9に記載の化合物。
- 前記放射性核種が、18F、113mIn、99mTc、67Ga、52Fe、68Ga、72As、111In、97Ru、62Cu、51Cr、52mMn、157Gd、64Cu、89Zr、177Lu、47Sc、67Cu、86Y、124I、131I、186Re、188Re、203Pb、211At、212Pb、225Ac、213Bi、227Thからなる群より選択される、請求項8または10のいずれか1項に記載の化合物。
- 前記キレート剤が、DOTA、NOTA、DTPA、TETA、EDTA、NODAGA、NODASA、TRITA、CDTA、BAT、DFOおよびHYNICからなる群より選択される、請求項1〜11のいずれか1項に記載の化合物。
- アクセプターが電子豊富な芳香族化合物である、請求項1、2、3、4、5、6、8、9、10、11、および12のいずれか1項に記載の化合物。
- 請求項1に記載の化合物であって、前記化合物が、式(IIIa)
- 請求項1に記載の化合物であって、前記化合物が、式(IIIb)
- 請求項1に記載の化合物であって、前記化合物が、式(IIIc)
- 請求項1に記載の化合物であって、前記化合物が、式(IVa)

- 請求項1から24のいずれか1項に記載の化合物を使用する診断方法を実施することを含む、疾患の処置に応答する可能性が高い対象、および疾患の処置に応答しない可能性が高い対象への対象群の層化方法において使用するための、請求項25または26のいずれか1項に記載の組成物。
- 前記疾患がニューロテンシン受容体関連疾患である、請求項27から30のいずれか1項に記載の組成物。
- 前記疾患がニューロテンシン受容体1関連疾患である、請求項31に記載の組成物。
