Description
本出願は、2013年9月3日に提出された米国仮出願第61/873,039号明細書の利益を主張する。尚、上記出願は、その内容を参照により本明細書に組み込むものとする。
別に定義されていない限り、本明細書で使用する技術及び科学用語は全て、本発明が属する分野の技術者によって一般に理解されている意味を有する。以下の参照文献は、当業者に、本発明で使用する多数の用語の一般的定義を提供するものである:Singleton et al.,Dictionary of Microbiology and Molecular Biology(2nd ed.1994);The Cambridge Dictionary of Science and Technology(Walker ed.,1988);The Glossary of Genetics,5th Ed.,R.Rieger et al.(eds.),Springer Verlag(1991);及びHale&Marham,The Harper Collins Dictionary of Biology(1991)。本明細書で用いる場合、以下の用語は、別に記載されていない限り、各用語に付与される下記の意味を有する。
ニューカッスル病ウイルス(NDV)は、線状、一本鎖、非分節性、マイナス鎖RNAゲノムを含むエンベロープウイルスである。NDVのマイナス鎖、一本鎖ゲノムは、RNA指令RNAポリメラーゼ、融合(F)タンパク質、ヘマグルチニン−ノイラミニダーゼ(HN)タンパク質、基質タンパク質、リンタンパク質及び核タンパク質をコード化する。ゲノムRNAは、次の順:3’−NP−P−M−F−HN−Lで遺伝子を含有する。NDV RNAゲノムの組織については、以下により詳細に説明する。ゲノムRNAは、3’末端にリーダ配列も含有する。
ウイルスは、腫瘍細胞に対して、腫瘍溶解作用を及ぼすことが知られており、治療薬としての腫瘍溶解性ウイルスの使用が報告されている。一部では、その天然の宿主に対して中度から高度の病原性を示す非ヒトウイルスを癌患者の治療に使用する取組みがなされている。本発明は、ヒト被験者において腫瘍の退縮を誘導する方法を開示し、該方法は、修飾されたFタンパク質切断部位を有するニューカッスル病ウイルス(NDV)の修飾亜病原性株を使用し、これは、家禽に対しては非病原性(長潜伏期性)であるが、腫瘍溶解性を発揮する。開示する方法は、それを必要とする個体の腫瘍の退縮を誘導する安全、有効かつ信頼できる手段を提供する。これらの方法によって、ヒトの治療法のためにウイルスの病原性株を使用する問題点が解消される。
本発明で使用するウイルスは、様々な方法によって調製することができる。例えば、NDVは、8〜10日齢の受精鶏卵(SPAFAS,Inc.,Roanoke,Ill.)を用いて調製することができる。ウイルスを単離する方法は、当分野において公知であり、例えば、Weiss,S.R.&Bratt,M.A.(1974)J.Virol,13,1220−1230を参照されたい。この方法は、以下の実施例1でさらに説明する。単離方法を用いて、約90〜95%純度のNDVを取得することができる。
療法は、癌療法が実施されるあらゆる場所:自宅、医院、診療所、病院の外来診療部門、又は病院で施すことができる。一実施形態では、本発明は、ニューカッスル病ウイルス(NDV)(例えば、r73T−R116)の使用を可能にする。
腫瘍の治療用の本発明のウイルス(例えば、NDVr73T−R116)の投与は、他の成分と併用して、腫瘍の予防、改善、又は軽減に有効な治療薬の濃度をもたらす任意の好適な手段によって行うことができる。薬剤は、任意の好適な担体物質中に任意の適切な量で含有させてよく、一般に、組成物の全重量の1〜95重量%の量で存在する。組成物は、非経口(例えば、皮下、静脈内、筋肉内、又は腹腔内)投与経路に好適な投薬形態で提供してよい。医薬組成物は、慣行的薬局業務規範(pharmaceutical practice)に従って製剤化することができる(例えば、Remington:The Science and Practice of Pharmacy(20th ed.),ed.A.R.Gennaro,Lippincott Williams&Wilkins,2000及びEncyclopedia of Pharmaceutical Technology,eds.J.Swarbrick and J.C.Boylan,1988−1999,Marcel Dekker,New Yorkを参照のこと)。
被験者が新生物を有すると診断されたら、治療方法を選択する。例えば、新生物の場合、いくつかの標準的治療レジメンが利用可能である。治療方法を選択する際、新生物のマーカプロフィールを用いる。一実施形態では、NDV(例えば、r73T−R116)による細胞殺傷に応答性の新生物細胞。
本発明の方法に有用な薬剤(例えば、NDV)は、新生物細胞の死を誘導し、及び/又は新生物細胞生存、すなわち生存性を低下させるものを含む。
本発明は、肉腫の治療又は予防のためのキットを提供する。一実施形態では、キットは、単位投与形態に有効量のNDV(例えば、r73T−R116)を含有する治療又は予防組成物を含む。別の実施形態では、キットは、単位投与形態に有効量のNDV(例えば、r73T−R116)を含有する治療又は予防組成物を含む。
高忠実度RT−PCRにより作製した6つのサブゲノムcDNA断片をpUC19ベクターに構築した。NDV73Tの完全長cDNAをp73Tと称した。73TにおけるFタンパク質切断部位(FPCS)のヌクレオチド及び推定アミノ酸配列を、NDV LaSota株(長潜伏期性、lento)の配列又はサイトメガロウイルス(CMV)の糖タンパク質B(gB)の配列(S116)に修飾した(図1;ダブルスラッシュは、Fタンパク質の切断部位を示す)。ウイルス配列を確認するために、cDNAを完全に配列決定した。Fタンパク質切断部位:Wt:ggg agg aga cag aaa cgc ttt;Lento:ggg ggg aga cag gaa cgc ctt; S116:cat aat aga acg aaa tcc ttt;S116KM:cat aat aaa atg aaa tcc ttt;R116:cat aat aga acg aaa cgc ttt。
トランスジーンをp73Tの2つの位置:P及びMの間の遺伝子間配列又はHN及びL結合部の間の遺伝子間配列に挿入した(図2A)。P−M又はHN−L結合部に単一のトランスジーンカセットを挿入するために、P−M又はHN−L結合部にトランスジーンを含有するp73TcDNAの構築を、P及びM遺伝子間(nt3148)又はHN及びL遺伝子間(nt8231)に形成したAfeI部位にトランスジーンカセットを挿入することによって実施した。挿入した遺伝子カセットは、トランスジーンの遺伝子終止
NDV73TNP、P、Lタンパク質及び抗原cDNA(p73T−lento又はp73T−S116)を、T7RNAポリメラーゼプロモータ及びターミネータの制御下でクローン化した。4つのプラスミドを、RNAポリメラーゼ発現細胞株に同時トランスフェクトした(図3A)。回収したウイルスをr73T−lento又はr73T−S116と称した。r73T−lento及びr73T−S116を、トリプシン補充有り、又はなしの培地を用いて、ベロ細胞において継代させた。r73T−lentoの増殖はトリプシン依存的であるのに対し、r73T−S116は、トリプシン補充なしの培地中で増殖することができる。P−M結合部にhGM−CSFを有する、又は有していないr73T−S116におけるFタンパク質切断配列(FPCS)を評価した。73T−S116株は、トリプシン補充なしの培地中、MOI0.01でベロ細胞及びヒト線維肉腫HT1080において10継代にわたりさらに継代させた。FPCSにおける突然変異(R113K及び/又はQ114M)が継代7で出現した。S116R突然変異は、継代9で検出された。継代10で、F、HN及びトランスジーンを配列決定したが、それ以上の突然変異は認められなかった。
P−M結合部にトランスジーン(1)を、HN−L結合部に第2トランスジーン(2)を、そして、非コード配列(3)の挿入により伸長される増大したHN−L遺伝子間領域を発現するように、ウイルスを改変することができる(図5A)。P−M結合部でのトランスジーンカセットの挿入のために図2Aと同じ設計をここで用いる。第2トランスジーンカセットは、トランスジーンのL遺伝子開始配列(GS;5’−ACGGGTAGA−3’)、オープンリーディングフレーム(ORF)、L遺伝子の3’非翻訳領域からの配列(斜体で強調)及びL遺伝子終止配列(GE;5’−TTAAGAAAAAA−3’)を含む。HN−L結合部を増大するために用いた非コード配列は、パラミクソウイルス1型(APMV−1)、RSウイルス(RSV)又はいずれの既知配列とも相同性のないランダム配列から取得した。挿入は、60〜318ntの範囲内であってよい。HN−Lでの第2トランスジーンの挿入により、ウイルスは、2つのトランスジーン、例えば、hGM−CSF及びGFPを発現することが可能になった。HN−L結合部に挿入された配列を列挙する(図5B)。
ニワトリにおけるr73T−R116ビルレンスを低減するために、様々な長さの配列の挿入によって、HN及びL遺伝子間配列を増大するように、r73T−R116を修飾した。r73T−R116誘導体を、細胞及び卵におけるプラーク形成及び複製を調べることにより、感染性について;MDT及びICPIを調べることにより、トリ病原性について、並びに腫瘍細胞殺傷について評価した(図6A)。
r73Tウイルスの増殖状態を決定するために、増殖動態試験を鶏卵(図7)及びベロ細胞(図8)において実施した。孵化鶏卵を100、1000、10,000又は100,000FFU/卵の表示ウイルスに感染させ、2、3若しくは4日にわたってインキュベートした。尿膜液を採取し、ウイルス力価をFFAによって決定した。図7に示すように、100FFU/卵は、1日目に低い力価を有したが、2日目にはピーク力価に達した。全体的に、ほとんどのウイルスが、用いた接種材料用量に関係なく、2日目に約8log/mLのピーク力価に到達した。R116i−198は、最も低い力価を有し、低い接種材料用量で2日目に最高収量を産生したが、これは、欠損粒子の蓄積があり得ることを示している。R116i−318−APMV挿入も、約7logというより低いピーク力価を有した。R116i−198−RSVは、7.5logのピーク力価に達した。逆遺伝学によって構築したS116は、卵でのウイルス産生の低減を全く示さなかった。従って、R116誘導体の中でも、RSV−198nt挿入を有すウイルスが、卵における増殖性に関して、腫瘍溶解性ウイルスの第1位の候補であった。
rT3T及びその誘導体を、非処置対照細胞に対し、正常ヒト皮膚線維芽細胞CCD1122Sk細胞と比較したヒト線維肉腫HT1080におけるその細胞殺傷について評価した(図9A及び9B)。データは、72時間にわたる1〜100,000PFUの範囲の様々な用量のr73T誘導体による感染後に取得した。HT1080癌細胞において、長潜伏期性FPCSを有するウイルスは、最小殺傷、FPCSにS116を有するウイルスは中間、また、FPCSにR116を有するウイルスは、r73Twtウイルスと同じくらい効率的な細胞殺傷を呈示した。正常CCD1122SK細胞の場合、全てのウイルスの細胞殺傷が、癌細胞の場合ほど効率的ではなかった。殺傷効率の低下は、約100倍であった。FPCS配列とは関係なく、全てのウイルスは、正常細胞において同様の殺傷効率を示した。癌及び正常細胞における細胞殺傷効率は、Prism6.0を用いた用量応答曲線から補間した50%有効濃度(EC50)として表した(図9C)。R116誘導体は、r73Twtと同様のEC50値(EC50:10PFU)を有したが、これは、FPCSの修飾が、癌細胞におけるウイルス複製及び細胞殺傷に影響を与えなかったことを示している。MOI0.01でのHT1080及びCCD1122SK細胞におけるこれらウイルスの複製を感染後3日目に決定した(図9D)。全てのウイルスは、約1.5〜2.0logの差で、正常細胞に対し、癌細胞において優先的に複製した。
in vivoで腫瘍溶解活性を評価するために、5〜6週齢のBalb/C無胸腺ヌードマウスに、5×106細胞/0.1mlの濃度でHT1080細胞を皮下注射することにより、HT1080異種移植片モデルを作製した。hGM−CSFは、マウスにおいて交差反応性がないため、この試験は、トランスジーンの評価ではなく、各種r73T構築物の腫瘍溶解能力を評価することを目標とする。R116−318i−hGM−CSFを腫瘍内(it)又は静脈内(iv)に投与して、腫瘍増殖速度を処置及び対照群の間で比較した(図10A)。
腫瘍溶解性NDVウイルスが、腫瘍組織において、選択的に複製するか否か、並びにウイルスクリアランスを決定するために、様々な臓器内でのウイルス分布を決定した。サイズ約250mm3の皮下HT1080腫瘍を保有する無胸腺ヌードマウスを、用量1×108PFUのR116i(r73T−hGM−CSF−R116i−318ntAPMV−N)で静脈内処置し、1、4、又は8日目に死なせた(1時点につきn=3)。血清、肺、脾臓、卵巣及び腫瘍を収集した。
ウイルス表面糖タンパク質は、ニワトリにおける免疫原性及びビルレンスに対する重要な抗原である。個別に又は組み合わせて、ニワトリにおいてビルレントではない他のパラミクソウイルスの対応する細胞外(エクト(ecto))ドメインによってNDVのF及びHN遺伝子を置換するために、様々な戦略を探究した。パラミクソウイルス5(PIV5)は、イヌパラミクソウイルスであり、ヒトには疾患を引き起こさない。ハトパラミクソウイルス1型(PPMV−1)変異体は、以前報告されているように、ICPIが0.025で、ニワトリにおいて非ビルレントであることが証明されており、NDVとは抗原が異なる(Dortmans et al,Veterinary Microbiology,2010,vo.143,page139−144.)。同一の短潜伏期性融合タンパク質切断部位を有するが、ビルレンスが極めて対照的な2つの遺伝子的に極めて関連するハトパラミクソウイルス1型(PPMV−1)が存在する(Veterinary Microbiology,2010,143:139−144)。F及び/又はHN糖タンパク質エクトドメインが、PPMV−1及び/又はPIV5で置換されたNDV73Tの完全長アンチゲノムcDNAを作製した(図12A)。NDV、PIV5及びPPMV−1配列は、それぞれ、青、紫又は緑で色付けしたボックスで示す)。
どの腫瘍タイプがNDV腫瘍溶解性に対して感受性であり得るかを理解するために、多種多様な腫瘍タイプ及び適応症を含む180の癌細胞株を、組換えNDV及びその変異体に対する感受性について試験した。細胞株は、American Type Tissue Collection(Manassas,VA)又はEuropean Collection of cell cultures(ECACC)から取得し、供給者により推奨される条件下で培地中に培養した。10,000個の癌細胞株を96ウェルプレートに接種し、6時間後ウイルスに感染させた。ウイルス濃度は、MOI10〜0.0001(又はウェル当たり1〜100,000pfu)の範囲であった。感染から48〜96時間後に細胞生存性を決定した。0.1のMOIのウイルスによる感染から72時間後、>30%細胞殺傷率のカットオフを用いて、感受性を決定した。図13A及び13Bは、腫瘍タイプによる感受性の概要を示す。造血系癌細胞株(白血病及びリンパ腫)は、NDV腫瘍溶解性に対して比較的非感受性であったが、試験した黒色腫、卵巣及び膵臓細胞株の大部分は、NDVに対して感受性であった。試験した全ヒト癌細胞株の約58%が、FPCSでのNDV R116iに対して感受性であったのに対し、試験した細胞株の4%が、Sベースのウイルスに対して感受性であった。これは、恐らくプロテアーゼによるFタンパク質の切断によるものと考えられる。R116iウイルスは、遍在性フリン様プロテアーゼにより切断される可能性が高いのに対し、S116Fタンパク質を切断する傾向があるプロテアーゼは、不明瞭であり、恐らくそれほど遍在的に発現されないと思われる。再誘導したS116−KM変異体は、より大きなプラークサイズをもたらし、オリジナルのS116変異体に対して腫瘍溶解能を増強した。これをさらに試験するために、NDVR116ウイルスに対して様々な感受性を有することが判明した22の癌細胞株の小さな群をGFP発現変異体に感染させて、感染から72時間後に細胞生存性を決定した。前述と同じ感受性カットオフを用いたところ、試験細胞株の41%がR116iに対して感受性で、4%がS116に対し感受性であり、再誘導S−KMは、試験細胞株の27%が感受性と、S116NDVより強力であった。
腫瘍モデル精製の後、S116−RD NDVコード化ヒト又はマウスGM−CSFを、精製B16F10同一遺伝子モデルにおける効力について試験した(図15A及び15B)。1×108pfuを3回の用量で腫瘍内に注射し、1群当たり最低8匹のマウスがいた。有意な腫瘍増殖阻害は、>80%腫瘍増殖阻害で明らかにされる(図15B)。短期腫瘍退縮が、反復腫瘍内(i.t.)投与により達成され、これは、処置群の42日に対する対照の生存期間18日にも有意な増加をもたらした。3回の用量後に処置を停止すると、腫瘍の再増殖が起こった。50日目にS116−mGM−CSF群において残留腫瘍のエビデンスがなかった唯1匹の動物を、いずれか一方の脇腹でのB16F10腫瘍細胞の移植により再攻撃した。腫瘍増殖は遅延したが、阻害されなかったことから、このモデルでは、この唯1匹の動物において十分な免疫記憶応答が達成されなかったことを示唆している。
B16黒色腫モデルにおける腫瘍溶解効果について、73T−R116i−hGM−CSF及び73T−R116i−mGM−CSFを評価した。この試験では、B16マウスにおいてウイルス耐容性を評価した。各ウイルスは、11日目及び14日目に2×107pfuで2回静脈内(i.v)若しくは腹腔内(i.p)に、又は11日目に1.1×107pfuで1回腫瘍内(i.t)に投与した。3つの異なる投与経路によりR116−hGM−SCF又はmGM−SCFで処置した群は、非処置群と比較して遅い腫瘍増殖速度を有した(図15A)。各群は3匹のマウスを含んだ。腫瘍阻害速度は、対照群から統計的に有意であった。従って、本発明のr73T誘導体は、有利には低いトリ病原性、高い腫瘍溶解活性を有し、鶏卵において高い力価まで複製した。これらの結果に基づき、本発明のウイルスは、腫瘍退縮を誘導し、癌患者治療成果を高める上で有用である(図16)。

(2)OX40L、CD40L、ICOSL、Flt3:、B.1(CD80)、CD137L、CXCL10(IP−10)、CCL5、CXCL9などの細胞表面リガンド及びケモカイン。(3)Myc阻害物質:Omomyc。(4)例えば、放射線ウイルス療法(radiovirotherapy)のためのヨウ化ナトリウムシンポータ(Sodium iodide symporter)(NIS)媒介放射線ウイルス療法などのin vivoイメージングを目的とするトランスジーン。(5)腫瘍殺傷を増強するための腫瘍細胞生存のさらなる調節物質(例えば、限定はしないが、細胞周期進行の阻害物質、抗アポトーシスタンパク質の阻害、アポトーシス促進性タンパク質の増強、悪性形質転換の重要な発癌ドライバの阻害など)。これらは、腫瘍細胞における選択的NDV複製後のタンパク質のトランスジーン送達、選択的若しくは広範な活性siRNAの生成、miRNAの送達又は選択されたmiRNAの阻害を含み得る。(6)E6、E7などの腫瘍抗原、癌・精巣抗原、癌胎児抗原、人工若しくは過剰発現タンパク質、例えば、単独又は他のトランスジーンと組み合わせた新規の腫瘍抗原。(7)負の調節を阻止するか、又はT細胞機能を増強するアゴニスト作用シグナルを提供するために、免疫調節タンパク質をターゲティングする抗体又は組換え融合タンパク質。このような抗体の例として、限定はしないが、以下:PD−L1、CTLA4、CD−137(4−1BB)、OX40、GITR、TIM−3、CD73、PD−1、HVEM、LIGHTが挙げられ得る。(8)タンパク質をrecNDV上に輸送する細胞においてrecNDVを改変又は発現することよって、補体によるクリアランスを低減するか、又はNDVに対する適応免疫応答を低減することによる、recNDVの薬力学/薬動態活性の増大。
NDV腫瘍溶解性ウイルスは、必要に応じて、治療抗体又はアゴニスト作用融合タンパク質(例えば、抗PD−L1、抗CTLA4、抗OX40、抗GITR、抗TIM−3、抗PD−1及び抗ICOS)と同時に、又は順次投与することができる。本明細書に記載する新規のNDV構築物と組み合わせて、腫瘍モデルにおけるNDVの活性を増強する分子の最も有効な用量及びスケジュールを確立する前臨床データを取得する。トランスジーンは、例えば、新規のNDV変異体により誘導される腫瘍細胞死を増強するために、複数のモードの活性を送達するように、単独で又は組み合わせて、発現用の組換えNDVに挿入することができる。免疫調節アプローチと組み合わせた腫瘍細胞抗原の放出の増大は、これらの遊離した腫瘍抗原に対する適応免疫応答を増大する能力を有する。
Fタンパク質切断部位の差が、感染細胞におけるFタンパク質切断及び融合活性へのその影響に作用するかどうかを理解するために、Fタンパク質プラスミドを293細胞にトランスフェクトして、Fタンパク質切断を調べた。さらに、F及びHNタンパク質のいずれも融合形成に必要であるため、F及びHNプラスミドを同時トランスフェクトして、トランジェントアッセイで融合活性を調べた(図3B及び3C)。wtFタンパク質は、宿主プロテアーゼによって、Fタンパク質切断部位にR、S又はS−KM突然変異を有するFタンパク質よりも効率的に切断され、S切断部位を有するFにはFタンパク質切断産物(F1)がほとんど検出されず、また、長潜伏期性Fプラスミドトランスフェクト細胞には切断産物が全く検出されなかった。R及びS構築物のFタンパク質は、切断部位にN結合グリコシル化部位(NXT)を有することから、KM突然変異がグリコシル化部位を無効にしたために、SDS−PAGEでの移動運動性がlento及びS−KMより遅くなる。これらのデータは、Fタンパク質切断部位でのR及びS突然変異が、Fタンパク質切断に作用して、S−KMが、Sより効率的になることを立証した。F及びHN遺伝子は各々、EGFP遺伝子をコード化するpVitro2−neo−MCSベクターにクローン化されたことから、融合した緑色細胞により、合胞体形成を視覚化することができる。wtF及びHNプラスミドトランスフェクト293細胞に、大きな合胞体形成が観察された。Fタンパク質切断部位にR、S又はS−KM残基を有するF突然変異体は全て、合胞体形成を大幅に減少させた。
HN−L結合部間に198ntが挿入されたR116iウイルスは、図6B及び6Dに示すように、高いmoi条件下で、ニワトリDF−1細胞において、より低速の増殖動態を示した。198nt挿入を有するR116iの増殖の差は、wt及びRウイルス(感染の早期時点(10〜20時間)で遺伝子間挿入を含まない)と比較して、小さくなった。ベロ細胞では、これらのウイルス同士の増殖の差は、あまりはっきりとしなかった(図6C及び6E)。R116iウイルスにおける遺伝子間挿入が、ウイルスRNA転写及び複製に影響を及ぼしたかどうかを理解するために、感染DF−1細胞におけるウイルスRNA及びタンパク質合成を、それぞれ、ノーザン及びウエスタンブロッティング分析により調べた(図6F及び6G)。198nt挿入を有するR116iのRNA及びタンパク質合成は、感染DF−1細胞において大幅に低減した。対照的に、感染ベロ細胞においては、NPmRNAのような上流遺伝子転写物は、R116i−198又は198RSV感染細胞において大幅に増加したが、これら2つのウイルスのゲノムRNAは減少した。これら2つのウイルスのLタンパク質のレベルは低下したが、図6H及びIに示すように、NP、F及びHNなどの他のウイルスタンパク質産物は増加した。図6F〜Iにおいて得られたデータは、高いmoi条件(moi=5)で実施した。さらに、RNA及びタンパク質合成は、低いmoi条件(moi=0.001)で実施した。ここでも、R116i−198又は198RSVは、DF−1及びベロ細胞において差次的なRNA及びタンパク質合成プロフィールを有した。R116i−198又は198RSVのRNA及びタンパク質合成のいずれも、ニワトリDF−1細胞において低減した(図6J)。対照的に、上流のRNA及びタンパク質は、ベロ細胞において増加したが、LmRNA及びLタンパク質のレベルは、198nt配列の遺伝子間挿入のために低下した(図6K)。ウイルスRNA及びタンパク質合成をさらに、ヒト細胞、ヒト線維肉腫HT1080及びヒーラ細胞、及びDF−1細胞の場合と比較した(図6L)。これらのデータによって、R116i−198又は198RSVが差次的発現を有しており、上流のRNA及びタンパク質合成は増大したのに対し、Lタンパク質発現は下方制御されたことが確認された。これらのデータは、R116i−198又は198RSVウイルスが、何故ベロ、ヒトHT1080及びヒーラ細胞などの哺乳動物細胞株において良好に複製したのに、孵化鶏卵及びニワトリDF−1細胞では良好に増殖しなかったのかを説明するものであった。これはまた、その低い脳内病原性インデックス(ICPI)によって示されるように、これらのR116iウイルスのニワトリ病原性低減の根拠も提供した。
図11Cでは、HT1080異種移植片マウス腫瘍モデルにおけるR116i−198RSV及びS116−KMの腫瘍溶解活性に対する、mGM−CSFの寄与をhGM−CSFトランスジーン発現と比較した。hGM−CSFトランスジーンは、mGM−CSFと交差反応しないため、対照として使用した。腫瘍内投与されたR116i−198におけるmGM−CSFトランスジーンは、hGM−CSFと比較して、腫瘍増殖阻害に低い効力を示した。mGM−CSF及びhGM−CSFの同様の腫瘍増殖阻害が、S116ウイルスについて観察された。ウイルス処置腫瘍増殖阻害におけるウイルス力価をプラークアッセイにより決定した(図11D)。腫瘍注射後4日目に、ペアとしてmGM−CSF又はhGM−CSFを有するR116i−198RSV及びS116−KMについて同様の力価が検出された。しかし、注射後7日目に、mGM−CSFを有するR116i−198RSVは検出されなかったが、hGM−CSFを有するR116i−198RSVは、ウイルス力価:約4.5log/g組織を有した。hGM−CSF又はmGM−CSFを有するS116のウイルス力価に差はなかった。これらのデータは、R116iによるmGM−CSFの発現が、ウイルスクリアランスを促進したことを示している。
補体(C’)系は、宿主において微生物侵入に対する主要な防御系である。ヒトの補体系には約30の異なる糖タンパク質があり、そのうちの20が、血漿において作用し、10が細胞膜に対する調節物質又は受容体である。膜結合C’調節物質(RCA)は、4つの十分に特性決定された分子:hCD46、hCD55、hCD59及びhCD35を含む。これらの主要な機能は、外来因子を排除するC’の役割に作用することなく、オートロガス補体攻撃からヒト細胞を防御することである。これらのRCAタンパク質は、宿主種特異的である。過去にウイルス療法のために用いられたNDVは、一般に、孵化鶏卵において生産された。癌患者に対し静脈内注射により投与されたNDV腫瘍溶解性ウイルスは、急速に排泄されるため、有効なウイルス用量が低下すると予想される。ヒト細胞から産生されるエンベロープウイルスは、感染細胞からのその放出中に、RCAタンパク質を取り込むことから、C’媒介のウイルス溶解又は不活性化を低減するようにヒト細胞培養物中にNDVを生産することが望ましい。
以下の細胞株及び対応する培地を用いた:アフリカミドリザル腎臓ベロ細胞株(ATCC)及びヒト線維肉腫(HT1080、ATCC)、10%ウシ胎仔血清(FBS)を含むイーグル最小必須培地(EMEM、Hyclone);ベロクローン51D11株(MedImmune)、1%グルタミンを含む無血清培地(SFMMegaVir、Hyclone);正常ヒト皮膚線維芽細胞(CCD1122Sk、ATCC)、10%FBSを含むATCC製剤化イスコフ改変ダルベッコ培地(IMEM)。組換えニューカッスル病ウイルス(NDV)を10〜11日齢特定病原体除去(SPF)孵化鶏卵の尿膜腔、ベロ細胞、又はベロクローン51D11細胞において増殖させた。
NDV株73TのウイルスRNAは、Mark Peeles博士(Nationwide Children’s Hospital)から取得した。ウイルスRNAのRT−PCR用のDNAオリゴヌクレオチドを設計する目的で、共通配列を取得するために、NDV配列(GenBank)をアラインメントした。NDVゲノム全体にわたる6つのサブゲノムcDNAオーバーラップ断片を高忠実度RT−PCRにより生成した(図1)。NDV73T株の完全長アンチゲノムcDNAの連続的アセンブリのために、EcoRI及びHindIII部位の間に導入された制限部位を含有する88ntオリゴヌクレオチドリンカーを含むように、pUC19ベクターを修飾した。さらに、73T株cDNAプラスミド(p73T)は、5’末端に27ヌクレオチド(nt)T7RNAポリメラーゼプロモータを、3’末端にHDVアンチゲノムリボザイム配列を有する189ntと、T7RNAポリメラーゼ転写終結シグナルを含む。非ビルレントNDVを作製するために、融合タンパク質のプロテアーゼ切断部位をコード化する配列を、部位指定突然変異誘発により、非ビルレントNDV LaSota株(長潜伏期性、lento)又はサイトメガロウイルス(S116)の糖タンパク質B(gB)の配列に修飾した。NPの構築のために、P及びL発現プラスミド、タンパク質オープンリーディングフレーム(ORF)をRT−PCRにより増幅してから、T7RNAポリメラーゼの制御下でプラスミドpCITE2aにクローン化した。
P−M結合部でのトランスジーンの挿入のために、SacII−PmlI断片を含有するサブクローンプラスミドのnt3148に、AfeI制限部位を導入した(図2A)。ヒト若しくはマウス顆粒球−マクロファージコロニー刺激因子(GM−CSF)又はインターロイキン(IL−2)をコード化するcDNAをコドン最適化した後、DNA2.0により合成した。遺伝子カセットは、Nの遺伝子終止(GE)、Pの遺伝子開始(GS)及びトランスジーンのオープンリーディングフレーム(ORF)を含有し、これをAfeI部位に挿入した。得られたプラスミドからのSacII−PmlI断片をプラスミドr73Tにシャッフルして、p73T−P1と名付けた。
NDVのF及びHNをハトパラミクソウイルス1(PPMV−1)のそれらで置換することにより、キメラNDVゲノムDNAを作製した。NDV73TFの細胞質尾部及び膜貫通部分のC末端コード配列(アミノ酸残基503〜553)をPPMV−1のエクトドメインFタンパク質コード配列(残基1〜502)と連結し、NDV HNのN末端コード配列(アミノ酸配列残基1〜45)を、GeneArt kit(Invitrogen)を用いたオーバーラップPCR反応により、HN(残基46〜577)と融合させた。増幅した断片を消化し、PmlI−AgeI消化NDV cDNAにクローン化した。同様のクローン化戦略により、パラインフルエンザウイルス5(PIV−5)F又はHNをNDV73TアンチゲノムcDNAに導入した。PIV5F(残基1〜486)エクトドメインをNDV73TFの膜貫通部分及び細胞質尾部(残基503〜553)と融合させた。NDV HN(残基1〜45)をPIV5 HNエクトドメイン(残基36〜565)と連結させた。cDNA断片をPmlI−AgeI消化NDVアンチゲノムcDNAにクローン化した。
BHK−T7細胞などのT7RNAポリメラーゼを発現する哺乳動物細胞株を、NDV NP、P及びLタンパク質を発現する3つのプラスミド(それぞれ、6ウェルディッシュのウェル当たり0.4μg、0.4μg、および0.2μg)並びにNDVアンチゲノムcDNAをコード化するプラスミド(1.6μg)で、Lipofectamine 2000を用いてトランスフェクトした。トランスフェクションから3日後に、細胞培養物上清を10〜11日齢のSPF孵化鶏卵の尿膜腔に注入するか、又はベロ細胞中で継代させて、レスキューされたウイルスを増幅した。ウイルスの回収は、1%ニワトリ赤血球(RBC)を用いた血球凝集反応アッセイによって確認した。ウイルスのレスキューは、以前記載されているように、T7RNAポリメラーゼを発現するプラスミドと一緒に、NP、P、L、アンチゲノムcDNAプラスミドをベロ細胞にエレクトロポレーションすることによっても実施することができる(Kaur et al.,Optimization of plasmid−only rescue of highly attenuated and temperature−sensitive respiratory syncytial virus(RSV)vaccine candidates for human trials.2008 J.Virol.Methods 153:196−202)。回収したウイルスをRT−PCR増幅cDNAの配列決定により確認した。
Fタンパク質切断部位(FPCS)が安定しているかどうか、並びに組織培養物中での継代後にいずれかの安定化突然変異を選択することができるかどうかを調べるために、MOI:0.01で、ベロ及びヒト線維肉腫HT1080細胞においてr73T−S116を連続的に10回継代させた。2〜3継代毎に、ウイルスRNAを培地から単離し、cDNAをRT−PCRにより増幅して、F及び/又はHN遺伝子を配列決定した。
6ウェルプレート上のベロ細胞を、階段希釈したウイルスに感染させた後、ウイルス力価定量用のトリプシン(TrpyLE(商標),Invitrogen)の存在下でプラーク形態学のために、1%メチルセルロースオーバーレイの下、37℃で3日又は6日にわたりインキュベートした。細胞単層をメタノールで固定し、全不活性化NDVウイルスに対してニワトリ抗NDVポリクローナル抗体で染色した後、セイヨウワサビペルオキシダーゼ(HRP)結合抗ニワトリ抗体(Dako)に暴露した。
10日齢SPF孵化鶏卵における平均致死時間(MDT)により、r73Tウイルスの病原性を決定した。1日齢SPFニワトリにおけるICPI試験は、USDA’s National Veterinary Service Laboratory(NVSL,Ames,Iowa)で実施した。MDT試験のために、10-6〜10-9の10倍希釈シリーズの0.1mlを、希釈物当たり8〜10個の9〜10日齢鶏卵の尿膜腔に接種した後、37℃でインキュベートした。7日間にわたって鶏卵を毎日2回検査し、胚致死時間を記録した。MDTは、ウイルスの最小致死用量が、全ての接種胚を死滅させるのにかかる平均時間(hr)として計算した。MDTアッセイは、ウイルスの病原性の妥当な予測を提供する。MDT<60時間のウイルスは、通常短潜伏期性(ビルレント)株であり;MDT=60〜90時間のウイルスは、亜病原性(中間)株であり;>90時間は、長潜伏期性(非ビルレント)株である。ICPI試験のために、各ウイルスについて、新鮮な感染尿膜液の1:10希釈物を0.05mlずつ、10羽の1日齢のSPFニワトリの群に脳内経路により接種した。8日間にわたり、8時間ごとに1回臨床的症状及び死亡についてニワトリを観察した。観察のたびに、ニワトリを次のようにスコア付けした:正常であれば、0、病的状態であれば、1、致死は2。ICPIは、8日間にわたる観察毎の1羽当たりのスコアの平均値である。ICPI値は、0.0〜2.0の範囲である。低ビルレント(LoND):ICPI<0.7;ビルレント(vND):ICPI≧0.7。
細胞を5×103細胞/ウェルで96ウェルプレートに塗布し、様々なMOIでr73Tに一晩感染させた。CellTiter Glo kit(Promega)により、製造者のマニュアルに従って細胞生存性を決定した。各試験サンプルのATPレベルを、100%生存の非処置サンプル対照と比較することにより、生存細胞の相対パーセントを決定した。表に示すデータは、死滅細胞の相対パーセントである。
無胸腺NCR均質ヌードマウス(Taconic)に、5×106HT1080細胞(100μLのPBS中)を一方の脇腹に皮下(s.c.)移植した。腫瘍が65〜300mm3の体積に達したら、ウイルス処置を開始した。100μL中の組換え73Tを様々な用量で、腫瘍内(i.t.)注射により局所的に、又は尾静脈への腫瘍内(i.t.)注射により全身に、のいずれかでそれぞれ投与した。対照マウスには、100μLのPBSのみを注射した。デジタルノギスを用いて、腫瘍増殖を測定し、腫瘍体積を0.5×(高さ)×幅×長さ(mm3)として計算した。体重が元の体重の20%減少するか、又は腫瘍体積が2000mm3を超えたら、マウスを死なせた。
HT1080ヒト線維肉腫異種移植片皮下腫瘍を担持する9匹のヌードマウスに、108pfuのr73T−R116i−hGM−CSFをi.vで注射した。注射から1、4及び8日後に3匹のマウスを死なせた。PBSを注入した1匹のマウスは8日目に死なせた。腫瘍、肺、脾臓、卵巣及び血清サンプルを収集した。組織ホモジネートの感染性ウイルス力価をプラークアッセイにより定量した。
gentle MACS Dissociator(Miltenyi Biotec)を用いて、製造者の指示に従い、PBSにおけるNDV感染及びPBS注入マウスからの腫瘍を均質化した。マウスから収集した均質化組織又は血清の上清を、Duoset ELISA kit(R&D)により、GM−CSFのレベルについて試験した。
統計的分析は全て、GraphPad Prism 6.0ソフトウェアを用いて実施した。独立t検定を用いて、群同士の腫瘍退縮の差を評価した。GraphPad Prismソフトウェアは、正常及び腫瘍細胞におけるin vitro細胞殺傷についてrNDV73TのIC50を計算するのにも用いた。
以上の説明から、様々な用途及び条件に合わせて、本明細書に記載した本発明に変更及び修正を加え得ることは明らかであろう。こうした実施形態も、以下に示す特許請求の範囲に含まれる。

- 前記ウイルスが、60〜318ヌクレオチドの長さの非コード配列を含む増大したHN−L遺伝子間領域を有する、請求項1又は2に記載の弱毒化ニューカッスル病ウイルス。
- 前記非コード配列が、パラミクソウイルス1型(APMV−1)、RSウイルス(RSV)又はランダム配列から得られる、請求項3に記載の弱毒化ニューカッスル病ウイルス。
- 前記HN−L遺伝子間非コード配列が、60、102、144、198又は318ヌクレオチドの長さである、請求項3に記載の弱毒化ニューカッスル病ウイルス。
- 前記ウイルスが、P−M結合部及び/又はHN−L結合部に挿入された1つ又は複数の異種ポリヌクレオチド配列を有する、請求項1〜5のいずれか一項に記載の弱毒化ニューカッスル病ウイルス。
- 前記異種ポリヌクレオチド配列が、前記ウイルスの腫瘍溶解性を増強するポリペプチドをコード化するトランスジーンである、請求項6に記載の弱毒化ニューカッスル病ウイルス。
- 前記トランスジーンが、サイトカイン、細胞表面リガンド、及び/又はケモカインをコード化する、請求項7に記載の弱毒化ニューカッスル病ウイルス。
- 前記サイトカインが、GM−CSF、IL−2、IL−21、IL−15、IL−12、及びIL−12p70からなる群から選択される、請求項8に記載の弱毒化ニューカッスル病ウイルス。
- 前記サイトカインが、ヒトGM−CSFである、請求項9に記載の弱毒化ニューカッスル病ウイルス。
- 前記ウイルスが、73T−R116i−hGM−CSFである、請求項1又は2に記載の弱毒化ニューカッスル病ウイルス。
