Description
(1)霊芝、ハナビラタケ菌、食用蟻及びマカの4成分を添加した液体種菌培養基又は固体培養基に、蛹虫草の種菌を接種して培養することを特徴とする、蛹虫草の生産方法。
(2)前記液体種菌培養基又は固体培養基が、培養基全体に対して、霊芝を0.1〜5質量%、ハナビラタケ菌を1〜30質量%、食用蟻を0.1〜5質量%、及びマカを1〜25質量%含有する、(1)に記載の生産方法。
(3)前記蛹虫草の種菌を接種した後の固体培養基に100〜200lxの光を所定期間照射する、(1)又は(2)に記載の生産方法。
(4)前記光照射期間中、前記固体培養基の温度を16〜22℃の範囲内に、相対湿度を80〜90%の範囲内に保持する、(3)に記載の生産方法。
(5)前記所定期間の経過後、成熟した子実体を収穫し、陰干し又は40℃以下で、含水量が12質量%以下になるまで乾燥させる、(4)に記載の生産方法。
(6)虫草素の含有量が3.0〜12.2mg/g、虫草多糖の含有量が117〜183mg/g、有機セレンの含有量が0.038〜0.078mg/kg、トリテルペン化合物の含有量が0.072〜1.76質量%、macamidesの含有量が0.26〜0.43質量%、及びmacaenesの含有量が0.31〜0.51質量%である、蛹虫草。
・液体種菌培養基(製品名:蛹虫草液体培養基、製造業者:大連百祥聚生物科技有限公司)
・固体培養基(製品名:蛹虫草固体培養基、製造業者:大連百祥聚生物科技有限公司)
・霊芝の粉体(製品名:赤霊芝粉、製造業者:大連百祥聚生物科技有限公司)
・霊芝の胞子の粉体(製品名:赤霊芝胞子粉、製造業者:大連百祥聚生物科技有限公司)
・ハナビラタケ菌(製品名:ハナビラタケ3号、製造業者:大連百祥聚生物科技有限公司)
・食用蟻の粉体(製品名:擬黒多刺蟻、製造業者:吉林省洋康薬業有限公司)
・食用蟻エキスの液体(製品名:蟻濃縮液、製造業者:大連百祥聚生物科技有限公司)
・マカ(製品名:マカ、製造業者:ウエルネス四万十研究所)
・蛹虫草の種菌(製品名:蛹虫草CO5、製造業者:大連百祥聚生物科技有限公司)
・酵素(製品名:フミンゲン、製造業者:ウエルネス四万十研究所)
各種薬効成分の測定方法を下記に示す。
・虫草素:紫外可視近赤外分光光度法及び高速液体クロマトグラフィー
・虫草多糖:フェノール硫酸法
・有機セレン:原子蛍光分光光度法
・トリテルペン化合物:薄層クロマトグラフィー
・マカ(macamidesとmacaenes):高速液体クロマトグラフィー及び質量分析法
液体種菌培養基(1リットル)に、霊芝の粉体(0.5質量%)、ハナビラタケ菌(15質量%)、食用蟻の粉体(0.6質量%)及びマカ(10質量%)を添加した。その液体種菌培養基をオートクレーブ(120℃)で25分間滅菌処理した後、室温(25℃以下)にまで自然冷却した。次いで、液体種菌培養基に蛹虫草の種菌(5グラム)を接種した。種菌接種後の液体種菌培養基を振動発生器にセットし、室温で7日間(168時間)、振動培養した。
液体種菌培養基(1リットル)に、霊芝の粉体(1.0質量%)、ハナビラタケ菌(10質量%)、食用蟻の粉体(2質量%)及びマカ(15質量%)を添加したことを除き、実施例1と同一条件で複合型蛹虫草を培養した。得られた複合型蛹虫草の成分を測定したところ、虫草素が4.2mg/g、虫草多糖が117mg/g、有機セレンが0.057mg/kg、トリテルペン化合物が0.79質量%、macamidesが0.32質量%、そしてmacaenesが0.35質量%であった。
固体培養基(50グラム)に、霊芝の粉体(1質量%)、ハナビラタケ菌(10質量%)、食用蟻エキスの液体(1.5質量%)及びマカ(15質量%)を添加した。その固体培養基をオートクレーブ(120℃)で1時間滅菌処理した後、室温(25℃以下)にまで自然冷却した。次いで、固体培養基に蛹虫草の種菌(5グラム)と、酵素(0.02ミリリットル)とを接種した。接種後の固体培養基を20℃±5℃の恒温室内で10日間培養した。次いで、菌糸体が菌床に蔓延したことを確認し、100〜200lxの光を45日間照射した。光照射期間中、培養温度を16〜22℃の範囲内に、また相対湿度を80〜90%の範囲内に保持し、かつ、風通しをよくするため培養容器に小さな孔を数カ所開けた。45日経過後、子実体が成熟した。成熟した子実体を収穫し、40℃以下で乾燥させて含水量を12質量%以下にした。得られた複合型蛹虫草の成分を測定したところ、虫草素が9.3mg/g、虫草多糖が150mg/g、有機セレンが0.063mg/kg、トリテルペン化合物が0.83質量%、macamidesが0.36質量%、そしてmacaenesが0.37質量%であった。
固体培養基(50グラム)に、霊芝の胞子の粉体(0.8質量%)、ハナビラタケ菌(10質量%)、食用蟻エキスの液体(2質量%)及びマカ(15質量%)を添加したことを除き、実施例3と同一条件で複合型蛹虫草を培養した。得られた複合型蛹虫草の成分を測定したところ、虫草素が12.2mg/g、虫草多糖が183mg/g、有機セレンが0.078mg/kg、トリテルペン化合物が1.76質量%、macamidesが0.43質量%、そしてmacaenesが0.51質量%であった。
Claims (6)
- 霊芝、ハナビラタケ菌、食用蟻及びマカの4成分を添加した液体種菌培養基又は固体培養基に、蛹虫草の種菌を接種して培養することを特徴とする、蛹虫草の生産方法。
- 前記液体種菌培養基又は固体培養基が、培養基全体に対して、霊芝を0.1〜5質量%、ハナビラタケ菌を1〜30質量%、食用蟻を0.1〜5質量%、及びマカを1〜25質量%含有する、請求項1に記載の生産方法。
- 前記蛹虫草の種菌を接種した後の固体培養基に100〜200lxの光を所定期間照射する、請求項1又は2に記載の生産方法。
- 前記光照射期間中、前記固体培養基の温度を16〜22℃の範囲内に、相対湿度を80〜90%の範囲内に保持する、請求項3に記載の生産方法。
- 前記所定期間の経過後、成熟した子実体を収穫し、陰干し又は40℃以下で、含水量が12質量%以下になるまで乾燥させる、請求項4に記載の生産方法。
- 虫草素の含有量が3.0〜12.2mg/g、虫草多糖の含有量が117〜183mg/g、有機セレンの含有量が0.038〜0.078mg/kg、トリテルペン化合物の含有量が0.072〜1.76質量%、macamidesの含有量が0.26〜0.43質量%、及びmacaenesの含有量が0.31〜0.51質量%である、蛹虫草。
