Description
(1)皮脂腺を観察する方法であって、
(I)皮膚組織から真皮および皮下組織それぞれの全部または一部が除去された皮脂腺構造体を培地中に浮遊させた状態で培養するステップ、および
(II)ステップ(I)で得られた培養後の皮脂腺構造体を観察するステップ
を含む皮脂腺の観察方法、
(2)前記ステップ(I)を行なう前に、単離された皮膚組織から真皮および皮下組織それぞれの全部または一部を除去して前記皮脂腺構造体を得るステップをさらに含む前記(1)に記載の皮脂腺の観察方法、ならびに
(3)被験物質が有する皮脂産生調節作用を評価する方法であって、
(A)皮膚組織から真皮および皮下組織それぞれの全部または一部が除去された皮脂腺構造体を被験物質の非存在下で培地中に浮遊させた状態で培養するステップ、
(B)皮膚組織から真皮および皮下組織それぞれの全部または一部が除去された皮脂腺構造体を被験物質の存在下で培地中に浮遊させた状態で培養するステップ、および
(C)前記ステップ(A)で得られた培養後の皮脂腺構造体(A)と前記ステップ(B)で得られた培養後の皮脂腺構造体(B)とを観察し、前記皮脂腺構造体(A)における皮脂腺の動態と前記皮脂腺構造体(B)における皮脂腺の動態との違いに基づき、前記被験物質が有する皮脂産生調節作用を評価するステップ
を含み、前記皮脂腺構造体(A)における皮脂腺の動態と前記皮脂腺構造体(B)における皮脂腺の動態との違いが被験物質の有無による皮脂腺の最外層における皮脂を内包する細胞の数の違い、被験物質の有無による皮脂腺における未分化の皮脂腺基底細胞から成熟皮脂腺細胞への分化の速度の違い、被験物質の有無による皮脂腺の中心部付近に存在する崩壊した成熟皮脂腺細胞の数の違い、被験物質の有無による皮脂腺の中心部付近に存在する皮脂量の違い、被験物質の有無による分化マーカーの発現位置の違いまたは被験物質の有無による分化マーカーの発現量の違いであることを特徴とする被験物質の評価方法
に関する。
(I)皮膚組織から真皮および皮下組織それぞれの全部または一部が除去された皮脂腺構造体を培地中に浮遊させた状態で培養するステップ、および
(II)ステップ(I)で得られた培養後の皮脂腺構造体を観察するステップ
を含む皮脂腺の観察方法に関する。
(a1)単離された皮膚組織から皮下組織の全部または一部を切除するステップ、および
(a2)前記ステップ(a1)で得られた組織から膠原繊維などの繊維を除去して皮脂腺を露出させて前記皮脂腺構造体を得るステップ
を含む方法(以下、「製法A」ともいう);
(b1)単離された皮膚組織から皮下組織の全部または一部を切除するステップ、
(b2)前記ステップ(b1)で得られた組織をディスパーゼ、コラゲナーゼ、プロナーゼなどの表皮付属器官を真皮から解離させるための酵素と接触させるステップ、および
(b3)前記ステップ(b2)で得られた組織から真皮を分離するステップ
を含む方法(以下、「製法B」ともいう)などが挙げられるが、本発明は、かかる例示のみに限定されるものではない。これらの皮脂腺構造体の製法のなかでは、皮脂腺構造体を容易に製造することができ、しかも生体における皮脂腺の動態を的確に観察することができることから、製法Aおよび製法Bが好ましく、製法Aがより好ましい。
本発明は、他の側面では、被験物質が有する皮脂産生調節作用を評価する方法であって、
(A)皮膚組織から真皮および皮下組織それぞれの全部または一部が除去された皮脂腺構造体を被験物質の非存在下で培地中に浮遊させた状態で培養するステップ、
(B)皮膚組織から真皮および皮下組織それぞれの全部または一部が除去された皮脂腺構造体を被験物質の存在下で培地中に浮遊させた状態で培養するステップ、および
(C)前記ステップ(A)で得られた培養後の皮脂腺構造体(A)と前記ステップ(B)で得られた培養後の皮脂腺構造体(B)とを観察し、前記皮脂腺構造体(A)における皮脂腺の動態と前記皮脂腺構造体(B)における皮脂腺の動態との違いに基づき、前記被験物質が有する皮脂産生調節作用を評価するステップ
を含む被験物質の評価方法に関する。
(1a)前記皮脂腺構造体(A)の皮脂腺の最外層における皮脂を内包する細胞の数に比べて前記皮脂腺構造体(B)の皮脂腺の最外層における皮脂を内包する細胞の数が多いこと、
(2a)前記皮脂腺構造体(A)の皮脂腺における未分化の皮脂腺基底細胞から成熟皮脂腺細胞への分化の速度に比べて前記皮脂腺構造体(B)の皮脂腺における未分化の皮脂腺基底細胞から成熟皮脂腺細胞への分化の速度が大きいこと、および
(3a)前記皮脂腺構造体(A)の皮脂腺の中心部付近に存在する崩壊した成熟皮脂腺細胞の数に比べて前記皮脂腺構造体(B)の皮脂腺の中心部付近に存在する崩壊した成熟皮脂腺細胞の数が多いこと。
(1b)前記皮脂腺構造体(A)の皮脂腺の最外層における皮脂を内包する細胞の数に比べて前記皮脂腺構造体(B)の皮脂腺の最外層における皮脂を内包する細胞の数が少ないこと、
(2b)前記皮脂腺構造体(A)の皮脂腺における未分化の皮脂腺基底細胞から成熟皮脂腺細胞への分化の速度に比べて前記皮脂腺構造体(B)の皮脂腺における未分化の皮脂腺基底細胞から成熟皮脂腺細胞への分化の速度が小さいこと、および
(3b)前記皮脂腺構造体(A)の皮脂腺の中心部付近に存在する崩壊した成熟皮脂腺細胞の数に比べて前記皮脂腺構造体(B)の皮脂腺の中心部付近に存在する崩壊した成熟皮脂腺細胞の数が少ないこと。
<略語の説明>
BSA:ウシ血清アルブミン
DMEM:ダルベッコ改変培地
FBS:ウシ胎児血清
PBS:リン酸緩衝生理食塩水
PBSTT:ポリオキシエチレンソルビタンモノラウラート(Tween20)とポリオキシエチレン(10)オクチルフェニルエーテル(Triton X−100)とを含有するPBS溶液〔組成:0.1体積%Tween20、0.5体積%Triton X−100および残部PBS〕
PFA:パラホルムアルデヒド
DMEMとF−12培地との混合培地〔DMEM/F−12培地(体積比)が1/1〕に、FBSおよび細胞外基質〔コーニング(Corning)社製、商品名:マトリゲル〕をFBSの濃度が10体積%および細胞外基質の濃度が2体積%となるように添加して混合培地を得た。
抗K5抗体〔アブカム(abcam)(株)製、商品名:αK5 antibody〕をその濃度が0.33μg/mLとなるように2質量%BSA含有PBSTT溶液に添加して一次抗体含有溶液を得た。なお、K5は、皮脂腺の最外層に存在する未分化の皮脂腺基底細胞のマーカーである。
標識抗IgG抗体〔インビトロジェン(Invitrogen)(株)製、商品名:α IgG antibody Alexa fluor 647〕および核染色剤〔インビトロジェン(Invitrogen)(株)製、商品名:hoechst33342〕を標識抗IgG抗体の濃度が2μg/mLおよび核染色剤の濃度が10μg/mLとなるように2質量%BSA含有PBSTT溶液に添加して染色試薬Aを得た。なお、前記標識抗IgG抗体は、調製例2で用いられた抗K5抗体に対する二次抗体である。
脂肪染色試薬〔和光純薬(株)製、商品名:ナイルレッド(Nile Red)〕をその濃度が10μg/mLとなるように2質量%BSA含有PBSTT溶液に添加して染色試薬Bを得た。
カバーグラス(24mm×55mm)上にスペーサー(直径9mmおよび厚さ0.12mm)を貼り、スペーサー付カバーグラスを得た。
実体顕微鏡下に、ハサミを用い、皮膚組織から皮下組織を除去した。つぎに、ハサミおよびピンセットを用い皮下組織除去後の皮膚組織中の真皮から膠原繊維を除去することにより、皮脂腺が露出した皮脂腺構造体(以下、「皮脂腺構造体A」という)を得た。なお、皮脂腺構造体Aは、皮膚組織における皮脂腺と毛と毛包と表皮とを含有しているが、真皮および皮下組織を実質的に含有しておらず、皮脂腺が露出していた。
調製例6で得られた皮脂腺構造体Aを4質量%PFA含有PBS溶液中に4℃で30分間浸漬させることにより、皮脂腺構造体Aを固定して固定試料を得た。得られた固定試料を2質量%BSA含有PBSTT溶液中において、4℃で1時間インキュベーションすることにより、洗浄を行なった。洗浄は、3回行なった。
(1)皮脂腺構造体の外観の観察
調製例7で得られた観察用試料に含まれる皮脂腺構造体Aの外観を共焦点顕微鏡下で観察した。試験例1(1)において、調製例7で得られた観察用試料に含まれる皮脂腺構造体Aの外観を観察した結果を図2および図3に示す。図2中のスケールバーの長さは、50μmである。また、図3中のスケールバーの長さは、50μmである。図3中、1は皮脂腺、3は毛包を示す。
調製例7で得られた観察用試料に含まれる皮脂腺構造体Aの内部を共焦点顕微鏡下で観察した。試験例1(2)において、調製例7で得られた観察用試料に含まれる皮脂腺構造体の内部を観察した結果を図4、調製例7で得られた観察用試料に含まれる皮脂腺構造体の内部における皮脂の油滴を観察した結果を図5に示す。図4中のスケールバーの長さは70μm、図5中のスケールバーの長さは、50μmである。図4中、11は未分化の皮脂腺基底細胞、Nは核、Sは皮脂を示す。また、図5中、Aは皮脂腺の最外層に存在する皮脂の油滴、Bは皮脂腺の中間層に存在する皮脂の油滴、Cは皮脂腺の中心付近に存在する皮脂の油滴を示す。
(1)被験試料の調製
調製例1で得られた混合培地(以下、「被験試料A」ともいう)に、リノール酸をその濃度が1×10-4Mとなるように添加して被験試料Bを得た。また、調製例1で得られた混合培地に、リノール酸の濃度が1×10-3Mとなるように添加して被験試料Cを得た。
被験試料A1mLを24ウェルプレートの各ウェルに添加した。つぎに、調製例6で得られた皮脂腺構造体Aを、前記24ウェルプレートの各ウェル内の被験試料A中に浮かべた状態で、5体積%二酸化炭素下に37℃で24時間培養することにより、皮脂腺構造体Aと被験試料Aとを接触させた(実験番号1)。また、実験番号1において、24時間の培養を行なう代わりに48時間の培養を行なうことを除き、実験番号1と同様の操作を行ない、皮脂腺構造体Aと被験試料Aとを接触させた(実験番号2)。
調製例7において、調製例6で得られた皮脂腺構造体Aを用いる代わりに実施例1(2)で得られた培養後の皮脂腺構造体A(実験番号1〜6)を用いたことを除き、調製例7と同様の操作を行ない、実験番号1〜6の観察用試料を得た。
試験例1(1)において、調製例6で得られた観察用試料を用いる代わりに実施例1(3)で得られた実験番号1〜6の観察用試料を用いたことを除き、試験例1(1)と同様の操作を行ない、実験番号1〜6それぞれの観察用試料に含まれる皮脂腺構造体Aの内部を共焦点顕微鏡下で観察した。
試験例1(1)において、調製例6で得られた観察用試料を用いる代わりに実施例1(3)で得られた実験番号1〜6の観察用試料を用いたことを除き、試験例1(1)と同様の操作を行ない、実験番号1〜6それぞれの観察用試料に含まれる皮脂腺構造体Aの皮脂腺の最外層に含まれる細胞内の油滴の状態を観察した。つぎに、観察された皮脂腺の細胞内の油滴の状態を表1の(A)〜(G)に基づいて分類して観察した。なお、分類は、観察された細胞内の油滴の状態に基づき、一見して表1の(A)〜(G)に最も近いものを選択することによって行なった。実施例1(5)において、実験番号1〜6それぞれの観察用試料に含まれる皮脂腺構造体Aの皮脂腺の最外層に含まれる細胞内の油滴の状態を分類して観察した結果を図6に示す。図中、(A)〜(G)それぞれのスケールバーの長さは、10μmである。また、図中、(A)から(G)の順に分化が進んだ細胞を含む部分を示す。(A)〜(G)それぞれの上段と下段とは、皮脂腺構造体Aの皮脂腺中の同一の部分を示す。さらに、図中、(A)〜(G)の下段において、実線は細胞の輪郭、破線は油滴の輪郭を示す。なお、図6(A)〜(G)に示される皮脂腺の最外層に含まれる細胞内の油滴の状態は、それぞれ、表1中の(A)〜(G)の状態に対応する。
調製例1で得られた混合培地に、リノール酸および被験物質としてのリノール酸受容体アンタゴニスト〔アブカム(株)製、商品名:GSK−3787〕をリノール酸の濃度が1×10-4Mおよび被験物質の濃度が1μMとなるように添加して被験試料Dを得た。
(1)皮脂腺構造体の培養および皮脂腺構造体と被験試料との接触
被験試料A1mLを24ウェルプレートの各ウェルに添加した。つぎに、調製例6で得られた皮脂腺構造体Aを、前記24ウェルプレートの各ウェル内の被験試料A中に浮かべた状態で、5体積%二酸化炭素下に37℃で24時間培養することにより、皮脂腺構造体Aと被験試料Aとを接触させた(実験番号7)。
調製例7において、調製例6で得られた培養後の皮脂腺構造体Aを用いる代わりに実施例2(1)で得られた実験番号7〜9の皮脂腺構造体Aを用いたことを除き、調製例7と同様の操作を行ない、実験番号7〜9の観察用試料を得た。
試験例1(1)において、調製例7で得られた観察用試料を用いる代わりに実施例2(2)で得られた実験番号7〜9の観察用試料を用いたことを除き、試験例1(1)と同様の操作を行ない、実験番号7〜9それぞれの観察用試料に含まれる皮脂腺構造体Aの内部を共焦点顕微鏡下で観察した。
<評価基準>
0点:細胞内に油滴がまったく見られない〔図6(A)参照〕。
1点:細胞内にわずかに油滴が見られる〔図6(B)参照〕。
2点:細胞内に油滴のスペースができている〔図6(C)参照〕。
3点:細胞内の油滴が細胞の核を侵食しはじめている〔図6(D)参照〕。
4点:細胞内の油滴同士がつながりはじめている〔図6(E)参照〕。
5点:細胞内のすべてが油滴によって占められている〔図6(F)参照〕。
6点:細胞が消失している〔図6(G)参照〕。
実体顕微鏡下に、ハサミを用い、皮膚組織から皮下組織を除去した。つぎに、皮下組織除去後の皮膚組織を、前記60mm径ディッシュ内のディスパーゼ含有PBS溶液に浮かべた状態で、4℃で16時間インキュベーションした。インキュベーション後の皮膚組織から真皮を除去して表皮と毛と毛包と皮脂腺とを含む構造体を得た。
(1)皮脂腺構造体の培養
実施例2(1)において、調製例6で得られた皮脂腺構造体Aを用いる代わりに調製例9で得られた皮脂腺構造体Bを用いたことを除き、実施例2(1)と同様の操作を行ない、皮脂腺構造体と被験試料との接触を行なう。
調製例7において、調製例6で得られた皮脂腺構造体Aを用いる代わりに実施例3(1)で得られた培養後の皮脂腺構造体Bを用いたことを除き、調製例7と同様の操作を行ない、観察用試料を得る。
試験例1(1)において、調製例7で得られた観察用試料を用いる代わりに実施例3(2)で得られた観察用試料を用いたことを除き、試験例1(1)と同様の操作を行ない、皮脂腺構造体Bの観察を行なう。その結果、皮脂腺構造体Bが有する皮脂腺が小さい場合には、当該皮脂腺の動態を的確に観察することができることがわかる。一方、皮脂腺構造体Bが有する皮脂腺が大きい場合には、調製例6で得られた皮脂腺構造体Aを用いたときと比べ、当該皮脂腺の動態を的確に観察しにくいことがあることがわかる。
(1)被験試料の調製
調製例1で得られた混合培地(被験試料A)に、リノール酸および被験物質としてのフィチン酸をリノール酸の濃度が1×10-4Mおよび被験物質の濃度が100μMとなるように添加して被験試料Eを得た。
被験試料A1mLを24ウェルプレートの各ウェルに添加した。つぎに、調製例6で得られた皮脂腺構造体Aを、前記24ウェルプレートの各ウェル内の被験試料A中に浮かべた状態で、5体積%二酸化炭素下に37℃で24時間培養することにより、皮脂腺構造体Aと被験試料Aとを接触させた(実験番号10)。
調製例7において、調製例6で得られた培養後の皮脂腺構造体Aを用いる代わりに実施例4(2)で得られた培養後の皮脂腺構造体A(実験番号10〜12)を用いたことを除き、調製例7と同様の操作を行ない、実験番号10〜12の観察用試料を得た。
画像解析ソフトウェア〔ビットプレーン(Bitplane)社製、商品名:Imaris〕を用い、実施例4(3)で得られた実験番号10〜12それぞれの観察用試料に含まれる皮脂腺構造体Aの外観を共焦点顕微鏡下で観察することにより、皮脂腺構造体Aの3次元画像のデータを得た。つぎに、画像解析ソフトウェア〔ビットプレーン(Bitplane)社製、商品名:Imaris〕を用い、得られた3次元画像のデータから皮脂腺構造体Aの最外層の情報を抽出した。その後、抽出された情報から最外層に存在する細胞の全体積を算出するとともに、最外層に含まれる皮脂滴を計数した。最外層に存在する細胞の全体積および皮脂の油滴数を用い、式(I):

=[皮脂の油滴数]/[最外層に存在する細胞の全体積] (I)
にしたがい、皮脂生成度を求めた。

Claims (3)
- 皮脂腺を観察する方法であって、
(I)皮膚組織から真皮および皮下組織それぞれの全部または一部が除去された皮脂腺構造体を培地中に浮遊させた状態で培養するステップ、および
(II)ステップ(I)で得られた培養後の皮脂腺構造体を観察するステップ
を含む皮脂腺の観察方法。 - 前記ステップ(I)を行なう前に、単離された皮膚組織から真皮および皮下組織それぞれの全部または一部を除去して前記皮脂腺構造体を得るステップをさらに含む請求項1に記載の皮脂腺の観察方法。
- 被験物質が有する皮脂産生調節作用を評価する方法であって、
(A)皮膚組織から真皮および皮下組織それぞれの全部または一部が除去された皮脂腺構造体を被験物質の非存在下で培地中に浮遊させた状態で培養するステップ、
(B)皮膚組織から真皮および皮下組織それぞれの全部または一部が除去された皮脂腺構造体を被験物質の存在下で培地中に浮遊させた状態で培養するステップ、および
(C)前記ステップ(A)で得られた培養後の皮脂腺構造体(A)と前記ステップ(B)で得られた培養後の皮脂腺構造体(B)とを観察し、前記皮脂腺構造体(A)における皮脂腺の動態と前記皮脂腺構造体(B)における皮脂腺の動態との違いに基づき、前記被験物質が有する皮脂産生調節作用を評価するステップ
を含み、前記皮脂腺構造体(A)における皮脂腺の動態と前記皮脂腺構造体(B)における皮脂腺の動態との違いが被験物質の有無による皮脂腺の最外層における皮脂を内包する細胞の数の違い、被験物質の有無による皮脂腺における未分化の皮脂腺基底細胞から成熟皮脂腺細胞への分化の速度の違い、被験物質の有無による皮脂腺の中心部付近に存在する崩壊した成熟皮脂腺細胞の数の違い、被験物質の有無による皮脂腺の中心部付近に存在する皮脂量の違い、被験物質の有無による分化マーカーの発現位置の違いまたは被験物質の有無による分化マーカーの発現量の違いであることを特徴とする被験物質の評価方法。
