Description
タダラフィルが十分溶解せず、分散または懸濁によって製造される場合、水に対する強い反発性のためフィルム製造液の層分離、活性成分の非均質化などの問題が生じる恐れがある。このような層分離及び非均質化はフィルム製造液の製造、塗布のための製造液の移送、塗布後の乾燥などの工程で生じ得る。
タダラフィル遊離塩基は強い疎水性を呈し、水溶液状態でフィルムを製造することがかなり困難である。そこで、本発明はタダラフィル遊離塩基の強い疎水性を克服する代わりに、かえってタダラフィル遊離塩基の強い疎水性を用いて、タダラフィル遊離塩基を高分子溶液に実質的に溶解させず分散(懸濁)させることで、所望の含量のタダラフィル遊離塩基を含みながらも、服用に適した厚さと大きさを有し、さらに所望の物性を有するフィルムが得られるという知見に基づく。
本発明において、分散安定化剤としてビニルピロリドン系高分子及び/またはポリエチレングリコール系高分子を使用することで、その他の添加剤(additive)、例えばタダラフィルの遊離塩基を高分子溶液内に単に懸濁させることに留まらず、高分子鎖の間にタダラフィル遊離塩基をさらに安定的に分散させるか、タダラフィル遊離塩基粒子間の凝集を減少させるか、または層分離を抑制するために添加する添加剤の使用量を減らすことができる。例えば、フィルムを製造するために必要な分散安定化剤、可塑剤(plasticizer)、または界面活性剤(surfactant)の総使用量を減らすことができる。したがって、乾燥したフィルムの総重量を基準にした分散安定化剤、可塑剤、及び界面活性剤の総含量は、1〜90重量%、より望ましくは1〜70重量%、さらに望ましくは1〜50重量%、最も望ましくは10〜15重量%であり得る。また、前記フィルム製造時に用いられる界面活性剤、可塑剤及び分散安定化剤の総重量を基準にした分散安定化剤としてのビニルピロリドン系高分子及び/またはポリエチレングリコール系高分子の含量は、5〜90重量%、最も望ましくは10〜20重量%であり得る。5重量%未満であるか又は90重量%を超える場合、タダラフィル遊離塩基を安定的に分散させ難い。本発明によるフィルム製造液に含まれる可塑剤としては、例えばグリセリン、ソルビトール、プロピレングリコール、またはこれらの混合物が使用できるが、これらに制限されない。また、本発明によるフィルム製造液に含まれる界面活性剤及び/または分散安定化剤としては、例えば、ポリソルベート、ポリオキシエチレンアルキルエテル、ポリオキシエチレンヒマシ油、ポリオキシエチレンステアレート、ドクセートナトリウム、ラウリル硫酸ナトリウム、ソルビタンエステル、またはこれらの混合物が使用できるが、これらに制限されない。
タダラフィル遊離塩基含有フィルム製剤は、以下のようにして製造した。精製水に可塑剤、添加剤、甘味剤、界面活性剤及び分散安定化剤を添加した後、撹拌して溶解または分散させ、そこにタダラフィル遊離塩基を添加した。その後、ホモジナイザー(Ultra turrax T−25、IKA)を用いて均質化した。そこに高分子を添加し、同じホモジナイザーを用いて再度均質化した。次いで、45℃、真空条件でフィルム製造液内のガスを除去し、室温に冷却した後、ポリエチレン(PE)フィルム上に適当な厚さにコーティングした。その後、80℃で乾燥してタダラフィル遊離塩基を含むフィルム製剤を製造した。
上述した「タダラフィル遊離塩基含有フィルム製剤の製造」と同じ方法でフィルムを製造した。ただし、フィルムに含まれる成分及びその含量は下記表1のように異ならせてフィルムを製造し、フィルム内のタダラフィル遊離塩基の分散安定度を測定した結果をRSD%(相対標準偏差)で示した。
Claims (5)
- フィルム剤形であって、
タダラフィル遊離塩基と、
ヒドロキシプロピルセルロースからなるフィルム形成用高分子と、
ポリエチレングリコール、ポリビニルピロリドン、ビニルアルコール−エチレングリコール共重合体、若しくはビニルピロリドン−ビニルアセテート共重合体、またはこれらの混合物からなる分散安定化剤と、を含み、
前記タダラフィル遊離塩基、前記ヒドロキシプロピルセルロース及び前記分散安定化剤はそれぞれ、乾燥したフィルムの総重量を基準として、10重量%以上、30重量%以下、20重量%以上、80重量%以下及び0.3重量%以上、10重量%以下含まれる、フィルム剤形。 - 前記分散安定化剤は、分子量が200g/mol以上、10,000g/mol以下のポリエチレングリコールである、請求項1に記載のフィルム剤形。
- 有効成分であるタダラフィル遊離塩基分散体と、
ヒドロキシプロピルセルロースからなるフィルム形成用高分子と、
ポリエチレングリコール、ポリビニルピロリドン、ビニルアルコール−エチレングリコール共重合体、若しくはビニルピロリドン−ビニルアセテート共重合体、またはこれらの混合物からなる分散安定化剤とを含み、
前記タダラフィル遊離塩基、前記ヒドロキシプロピルセルロース及び前記分散安定化剤はそれぞれ、乾燥したフィルムの総重量を基準として、10重量%以上、30重量%以下、20重量%以上、80重量%以下及び0.3重量%以上、10重量%以下含まれる、タダラフィル遊離塩基含有フィルムの製造液。 - フィルム製造液中、溶媒の90%以上が水である、請求項3に記載のタダラフィル遊離塩基含有フィルムの製造液。
- タダラフィル遊離塩基が分散し、フィルム形成用高分子と分散安定化剤とが溶解したフィルム形成溶液を調整する工程と、
前記フィルム形成溶液を乾燥させる工程とを備え、
前記分散安定化剤は、ポリエチレングリコール、ポリビニルピロリドン、ビニルアルコール−エチレングリコール共重合体、若しくはビニルピロリドン−ビニルアセテート共重合体、またはこれらの混合物であり、
前記フィルム形成用高分子は、ヒドロキシプロピルセルロースであり、
前記タダラフィル遊離塩基、前記ヒドロキシプロピルセルロース及び前記分散安定化剤はそれぞれ、乾燥したフィルムの総重量を基準として、10重量%以上、30重量%以下、20重量%以上、80重量%以下及び0.3重量%以上、10重量%以下含まれる、タダラフィル遊離塩基含有フィルムの製造方法。
