Description
本出願は、参照によりその全容が本明細書に組み込まれている、2012年10月30日に出願されたUS61/720,218号、2013年8月27日に出願されたUS61/870,418号、2013年8月27日に出願されたUS61/870,435号に対する優先権を主張するものである。
抗体および試薬
トリスジブチルベンジリデンアセトンパラジウム(トリスDBA)はDr.Arbiser(U.Alabama)の親切な提供物であった。DDD85646は[J.A.Frearson et al(2010)Nature.464.728〜723頁]中に記載されたように合成し、および/またはDDD86481はDr.David Gray and Paul Wyatt、Dundee Universityから得た。
エドマン分解配列決定(Alphalyse)により同定したNMT1とNMT2のカスパーゼ切断部位を部位特異的突然変異誘発により突然変異させた。したがって本発明者らは、事前にクローニングしたV5-NMT1およびV5-NMT2ゲートウェイベクターを使用して、同定したカスパーゼ切断部位を突然変異させた。したがって、V5-NMT1のAsp-72残基ならびにV5-NMT2のAsp-25、Asp-67および両Asp-25,67(二重突然変異)残基を、製造者の説明書に従いQuickchange(登録商標)部位特異的突然変異誘発キット(Agilent Technologies社)を使用して突然変異させた。
以下のカスパーゼ切断型、切断型NMT、His-73NMT1、His-26NMT2およびHis-68NMT2を分子クローニングにより作製した。切断型DNA配列を、N末端ヘキサヒスチジン(His)-タグ配列を有するpET-19b(Novagen(登録商標))ベクターにクローニングした。
アポトーシスの誘導前に1時間、EMD Chemicals社から購入した10μMの確立されているカスパーゼ阻害剤を用いて細胞を事前処置した。使用したカスパーゼ阻害剤は一般的カスパーゼ阻害剤(z-VAD-FMK)、カスパーゼ-3(z-DEVD-FMK)、カスパーゼ-8(z-IETD-FMK)、カスパーゼ-9(z-LEHD-FMK)および陰性対照(z-FA-FMK)であった。対照細胞はDMSO(1:1000)で処置した。
IM9、KMH2、BL2およびRamos細胞を平板培養し(6ウェルディッシュ中にウェル当たり3×106個の細胞)、5時間10μMのMG-132で処置した。等量のDMSOを対照サンプルに加えた。次いで細胞を溶解しSDS-PAGE/ウエスタンブロット分析を施した。
IM9、BL2およびRamos細胞を6ウェルディッシュ中にウェル当たり3×106個の細胞で平板培養し、クラスIおよびクラスIIヒストンデアセチラーゼ(HDAC)の可逆的阻害剤であるスベロイルアニリドヒドロキサム酸(SAHA)1μMを用いて24時間処置した。等量のDMSOを対照として細胞に加えた。細胞を溶解しSDS-PAGE/ウエスタンブロット分析を施した。
150または300ng/mLのマウス抗Fas抗体を使用し外部経路を介してアポトーシスを刺激し、2.5μMのスタウロスポリン(STS)を使用し内部経路を介してアポトーシスを刺激した。示した場合、シクロヘキシミド(5μg/mL)を使用しタンパク質翻訳を阻害し、細胞死をさらに助長した。
HMAを鹸化し、細胞に加える前にウシ血清アルブミン(BSA)と結合させ、以前に記載されたように(Yap et al.,2010)その細胞への取り込みを容易にした。簡単に言うと、15分間65℃で20%モル過剰な水酸化カリウム(KOH)と共にHMAをインキュベートした。その後、37℃で20%脂肪酸無BSAを含有する無血清RPMI培地にKOH鹸化HMAを溶解し、次に37℃でさらに15分間インキュベートすることにより20×溶液を作製した。ミリスチン酸ナトリウムを対照として使用したので、それも細胞に加える前に、37℃で20%脂肪酸無BSAを含有する無血清RPMI培地でインキュベートした。その後、ジャーカットT細胞(1×107個の細胞)を、(FBS、サプリメントまたは抗生物質を含まない)RPMI培地中で37℃において1時間、脂肪酸無BSAと結合した1mMのHMAまたは1mMのミリスチン酸ナトリウムのいずれかとインキュベートした(細胞培養中、最終濃度1%)。インキュベーション後、シクロヘキシミド(5μg/mL)または媒体(DMSO)のみで抗Fas(150ng/mL)またはSTS(2.5μM)を使用して、アポトーシスを誘導した。サンプルは8時間の間2時間毎に回収し、冷却PBSで洗浄し、0.3mLの1%ドデシル硫酸ナトリウム(SDS)放射性免疫沈降アッセイ(RIPA)バッファーで溶解し、Branson Sonifierにおいて5.5〜6.5のアウトプットで15秒間2ラウンドの超音波処理を施し、各サイクル間は氷上に2分間置いた。
2×106個の細胞(CEM、L0、BL2およびRamos)を6ウェルプレート中で増殖させ、多量(0、1、2および5μg/ml)のトリス(ジベンジリデンアセトン)ジパラジウム(トリスDBA)とインキュベートした(Bhandarkar et al.,2008)。トリスDBAで処置した細胞の生存率はトリパンブルー色素排除アッセイで測定した(Hudson,1976)。
1×105個の細胞[KMH2(ホジキンリンパ腫)、IM9(「正常」EBV不死化B細胞)、BL2、Ramos](100μL/ウェル)を96ウェルプレートに平板培養し、24、48および72時間、多量のピラゾールスルホンアミド系NMT阻害剤DDD85646[分子量(MW)495.43](Frearson et al.,2010)および低効能ピラゾールスルホンアミド系アナログDDD73228(MW=362.5)で処置した。各ウェル中のDMSOの最終体積が1%になるように、薬剤をDMSOに溶解し、各試験濃度に関して100×ストック溶液を作成した。これらの阻害剤で処置した細胞の生存率はトリパンブルー色素排除アッセイで測定した(Hudson,1976)。
1×105個の細胞[KMH2(ホジキンリンパ腫)、IM9(「正常」EBV不死化B細胞)、BL2、Ramos](100μL/ウェル)を96ウェルプレートに平板培養し、多量のDDD86481[MW610.5]で処置した。各ウェル中のDMSOの最終体積が1%になるように、薬剤をDMSOに溶解し、各試験濃度に関して100×ストック溶液を作成した。MTS[(3-(4,5-ジメチルチアゾール-2-イル)-5-(3-カルボキシメトキシフェニル)-2-(4-スルホフェニル)-2H-テトラゾリウム)]アッセイを使用して薬剤の細胞毒性を測定した(Cory et al.,1991)。
典型的に、細胞を冷却PBSで洗浄し、採取し、1×完全プロテアーゼ阻害剤を補充した0.1%のSDS-RIPAバッファーまたは0.1%のSDS-RIPA-HEPESバッファーに溶解した(このとき細胞はアルキン-脂肪酸で標識した)。細胞懸濁液は4℃で15分固定した。次いで細胞溶解物を4℃において10分間16,000gで遠心分離し、分裂後期核の上清を回収した。タンパク質濃度はPierceTMビシンコニン酸(BCA)タンパク質アッセイキット(Thermo Fisher Scientific社、Waltham、MA)を使用して測定した。
ヒトびまん性大細胞型B細胞リンパ腫(DLBCL)および濾胞性リンパ腫(FL)組織は、Dr. Raymond Lai(Cross Cancer Institute社、Alberta、カナダ)からの親切な進物であった。凍結腫瘍組織は小片(約1〜3mm)に切断し、1×完全プロテアーゼ阻害剤を含む1%SDS-RIPAと混合した。サンプルは小型ダウンス型ホモジェナイザーを使用して均質化し、次いで腫瘍組織が溶解バッファーに溶解するまで、6.0のアウトプット(Branson Sonifier450)で、(氷上において)1分間隔で2分間の間繰り返し超音波処理した。次いでサンプルは4℃において10分間16,000gで遠心分離し、ウエスタンブロッティング分析用に分裂後期核の上清を回収した。
この方法は提供された切断型カスパーゼ-3ブロットのみに使用し、全ての他のウエスタンブロッティングは標準的な手順を使用して行った。電気泳動後、ゲルはニトロセルロース膜に移し、0.1%Tween20を含むPBS(PBS-T)中5%NFMで1時間ブロッキングした。一次抗体(ウサギ抗切断型カスパーゼ-3、Cell Signaling社)もPBS-T中5%NEMに1:2000で希釈し、膜に加え24時間インキュベートした。次に、膜には6洗浄サイクルを施し、それぞれ5分間続け、PBSで2回洗浄、PBS-Tで2回洗浄、および最後にPBSで2回洗浄した。二次抗体(Alex Fluor680ヤギ抗ウサギ、Life Technologies社)はPBS-Tで(1:5000)希釈した後に加え、ブロットはアルミホイルで覆い、二次抗体と1時間インキュベートし、前と同じPBSとPBS-Tを用いた6×洗浄サイクルを施した。(ウサギ用の)チャンネル700において84μmのスキャン解像度(強度:5〜7)で、LI-COR Inc社からのOdyssey(登録商標)蛍光イメージングシステムでブロットをスキャンした。
HeLa細胞を150mmプレート中で密集状態まで増殖させ、5時間の間抗Fas(300ng/mL)(外部)またはSTS(内部)でアポトーシスの過程を経るように誘導した。次いで細胞を、(以下のセクションで詳細に記載するように)(Yap et al.,2010)アルキン-C14で代謝的に標識しハイポトニック溶解を施した。
ω-ミリスチン酸アルキニルを使用した細胞の代謝標識
細胞を採取する前に、100〜25μMのω-ミリスチン酸アルキニルで30分間細胞を標識し、0.1%SDS-RIPA-HEPESバッファーに溶解した。生成した細胞溶解物由来のタンパク質(50〜30μg)を、クリックケミストリーを使用して100μMのアジド-ビオチンと反応させ、以前に記載されたように(Yap et al.,2010)処理した。簡単に言うと、標識前に1時間、5%デキストランコーティングチャコール処置FBS(DCC-FBS)を補充した培地(RPMIまたはDMEM)でインキュベートすることにより、細胞を脂肪酸枯渇状態にした。次に、ω-ミリスチン酸アルキニルをDMSOに溶解し25または100mMストック溶液を作製した。典型的にω-ミリスチン酸アルキニルの細胞内取り込みは、鹸化およびBSAとの結合で典型的に改善された。したがって標識前に、ω-ミリスチン酸アルキニルを、15分間65℃で20%モル過剰な水酸化カリウムで鹸化した。次に、20%脂肪酸無BSAを含有する(予め温めた)無血清培養培地を鹸化ω-ミリスチン酸アルキニルに37℃で加え、37℃でさらに15分間インキュベートした。
ω-ミリスチン酸アルキニルで細胞を標識した後、生成した溶解物にアジド-ビオチンを用いたクリックケミストリーを施し、以前に記載されたような(Yap et al.,2010)ECLによるミリストイル化タンパク質の検出を可能にした。典型的に、細胞溶解物(タンパク質30〜50μg)が1%SDSを含有するように調節し、100μMのトリス-(ベンジルトリアゾリルメチル)アミン(TBTA)、1mMのCuSO4、1mMのトリス-カルボキシエチルホスフィン(TCEP)、および100μMのアジド-ビオチンと共に暗所で37℃において30分間インキュベートし、クリック反応を進行させた。次に、10体積の氷冷アセトンを加えてクリック反応を停止させ、タンパク質は一晩-20℃で沈殿させた。その後、アセトン沈殿タンパク質は16,000gで10分間遠心分離し、20mMのDTTを含む1×SDS-PAGEサンプルバッファーに再懸濁した。次いでサンプルは95℃で5分間加熱しSDS-PAGEに施し、ウエスタンブロット分析用にPVDF膜に移した。
KOHまたは中性トリス-HClでPVDF膜を処置した実験用に、タンパク質サンプルを二連SDS-PAGEゲルに充填した。電気泳動後、軽く振とうしながら45分間室温で、KOHで処置したPVDF膜を0.1N KOH、メタノール中100ml[1N KOH、H2O:メタノール中1:9(v/v)]でインキュベートし、一方他の膜は0.1Nトリス-HCl、pH7.0メタノール中[1Nトリス-HCl、pH7.0:メタノール1:9(v/v)]でインキュベートした。その後、処置した膜はPBSで完全に洗浄し、ストレプトアビジン-HRPまたはNeutravidin-HRPでプロービングしECLで検出した。
Sharma研究室(King and Sharma,1991;Raju and Sharma,1999)により開発されたプロトコールを採用することによってNMT活性を測定した。[3H]-ミリストイル-CoAのストック溶液を、以前に記載されたように(Towler and Glaser,1986)新たに調製し合成した。[3H]-ミリストイル-CoAの200μLストック溶液を作製するため、97μLのミリストイル-CoA生成バッファーを、20μLの50mM ATP、10μLの20mM LiCoA、60μLのシュードモナスアシル-CoAシンテターゼおよび13μLの[9,10-3H]-ミリスチン酸と組み合わせた。溶液は軽く混合し、37℃で30分間インキュベートした。
完全長および切断型HisタグNMTのNMT活性を、前に記載したのと同じプロトコールを使用して測定した。しかしながら、このアッセイでは、わずか1μgの精製タンパク質を使用した(体積はNMTアッセイバッファーにおいて最大10μLにした)。
in vitroでのNMT切断アッセイを、カスパーゼ切断アッセイ用バッファー中、100μLの反応で1時間37℃において、10 gの精製His-NMT1およびHis-NMT2と1μgの組換えヒト活性カスパーゼ-3または-8をインキュベートすることにより実施した。10μMの一般的カスパーゼ阻害剤z-VAD-FMKを加えて反応を停止させ、その後5×サンプルローディングバッファーを加えた。反応サンプルは10%SDS-PAGEゲル上に分離させPVDF膜に移した。バンドはクーマシーブルー染色により可視化し、切断断片は膜から切除し、エドマン分解のためAlphalyse社、Palo Alto, CAに送った。
Lemo21(DE3)pLysSコンピテントセル(New England Biolabs社)を、製造者のプロトコールに従いHis-NMT1およびHis-NMT2ベクターで形質転換した。NMT1およびNMT2タンパク質を以下のように調製した。3mLの開始培養物をLBブロス(1%のトリプトン、0.5%の酵母エキス、0.5%のNaCl、100μg/mLのアンピシリンおよび34μg/mLのクロラムフェニコール)中で、225rpmで振とうしながら37℃において4時間増殖させた。全培養物を使用して、37℃において16時間増殖させた50mL培養物を接種した。次に細菌細胞を、室温における10分間6000×gでの遠心分離によってペレット状にした。細菌ペレットは10mLのLBに再懸濁し、5mLの再懸濁液を使用して、0.5〜0.6のOD600に達するまで激しく振とうしながら(225rpm)37℃でインキュベートした500mLのLBを接種した。
製造者のプロトコールに従いTRIzol(登録商標)試薬を使用してIM9、KMH2、RamosおよびBL2細胞からRNAを単離した。次に、Applied Biosciences社からのランダムプライマースキームを備える高性能cDNA逆転写キットを使用して、製造者の説明書に従い単離したRNAからcDNAを合成した。定量リアルタイムPCR(qRT PCR)反応を、TaqMan(登録商標)Universal Master Mix II、およびNMT1、NMT2、およびLife Technologies社(Carlsbad,CA)から購入した18のTaqman(登録商標)プローブを使用して設定し、三反復の各反応を供給者のガイドラインに従い設定した。qRT PCRはMastercycler(登録商標)ep realplex thermocycler(Eppendorf社)を使用して実施し、結果はRealplexソフトウエア(Eppendorf社)を使用して分析した。
トリスDBA、DDD73228、およびDDD85646で処置した細胞の生存率を、製造者の説明書に従い、TC10TMトリパンブルー色素(Biorad社)と細胞をインキュベートすることによって測定した(Hudson,1976)。次いで細胞生存率を、TC10TM自動セルカウンター(Biorad社)を使用して定量化した。
DDD86481で処置した細胞の生存率を、製造者の説明書に従い、Promega社からのCellTiter96AQueous Non-Radioactive細胞増殖アッセイ(MTS)(Cory et al.,1991)を使用して測定した。
B細胞リンパ腫腫瘍をホルマリンで固定し、パラフィン包埋し、所望の厚さ(約5ミクロンまたはμm)までミクロトームで切断し、Superfrost(登録商標)プラス正帯電スライド(Fisher Scientific社)上に固定した。染色前に、腫瘍組織を含有するスライドを、キシレン中にそれぞれ10分間3回の浸漬、エタノール中での一連の洗浄[100%エタノール中に20回浸漬(4回反復)、80%エタノール中に20回浸漬、および50%エタノール中に20回浸漬]および冷たい流水で10分間の最終洗浄によって脱パラフィン処理した。
図1は、正常細胞および様々なB細胞リンパ腫およびT細胞白血病における、NMT1およびNMT2発現の分析を示す。この図は、正常B細胞(EBV形質転換ヒトBリンパ球、L0)およびヒト白血病T細胞系(ジャーカット、MOLT-4、CEM)と比較した、NMT1のみを発現するBリンパ腫細胞系(BL-2、Ramos)における、NMT2の発現のほぼ完全な不在を示す。
図2は、NMT阻害剤、トリス-ジベンジリデンアセトン-ジパラジウム(トリス-DBA)に対する、様々なB細胞リンパ腫およびT細胞白血病の感受性を示す。様々なB細胞およびT細胞は、高濃度のトリスDBAと共に24時間インキュベートした。トリパンブルー色素排除法を使用して細胞生存率を測定し、対照に関しては100%に調整した。トリパンブルー色素排除によって測定した細胞生存曲線は、B細胞リンパ腫がNMT阻害剤、トリス-ジベンジリデンアセトン-ジパラジウム(トリス-DBA)に対してより感受性があることを示す。
図3は、トリス-ジベンジリデンアセトン-ジパラジウム(トリス-DBA)による、N-ミリストイルトランスフェラーゼ(NMT)の阻害を示す。
図4は、NMT1(パネルA)およびNMT2(パネルB)に対する抗体でリンパ腫細胞系をプロ−ビングした、イムノブロットの結果を示す。図4の凡例は、以下に相当する:IM9:Bリンパ芽球;BL2:バーキットリンパ腫;CEM:T細胞白血病;Karpas299:T細胞リンパ腫;Sup-M2:ALCL;UCONN:ALCL(ALCL:未分化大細胞リンパ腫);DAUDI:バーキットリンパ腫;Ramos:バーキットリンパ腫BJAB:バーキットリンパ腫;HD-MYZ:ホジキンリンパ腫;KM-H2:ホジキンリンパ腫;L428:ホジキンリンパ腫;ジャーカット:T細胞白血病。
図5は、異なる濃度で48時間後に不死化正常Bリンパ球性細胞系(IM9)と比較した、バーキットリンパ腫細胞系Ramosに対するNMT阻害剤の有効性を示す。
この実施例では、pcDNA3.1-V5-NMT2による(本明細書中で示すようにNMT1を発現する)RamosBリンパ腫細胞のトランスフェクションによって、空プラスミドベクターでトランスフェクトした対照細胞に対して、トリスDBA(5ug/ml)に対する生存が2.5倍増大した。図6では、Ramos細胞系に関して勧められるプロトコール(1,350ボルト、30ms)に従いNeonトランスフェクションシステム(Invitrogen)を使用して、20×106個のRamosBリンパ腫細胞を32μgのDNA(pcDNA3.1-V5空またはpcDNA3.1-V5-NMT2)でトランスフェクトした。トランスフェクト細胞は5分間1200rpmで遠心分離して、死細胞および細胞残骸を除去した。上清中の細胞は完全RPMIにおいて6時間回収した。PBSで洗浄した後、細胞を再縣濁し、トリスDBA(5ug/ml)を含有するRPMI中で24時間増殖させ、次いでトリパンブルー色素排除法を使用して計数した(パネルA)。細胞を溶かし、NMT2に対する抗体および充填対照用にGAPDHを用いて、ウエスタンブロッティング(ECL)を実施してNMT2の発現を確認した(パネルB)。
この実施例では、内部対照として18Sプローブを使用し、TaqmanのNMT1およびNMT2プローブを用いてqRT-PCRを実施した。サイクル時間数の差(Δct)は、本発明者らが各細胞型に関する18S内部対照の発現の指数関数的増加を見たサイクル時間(ct)を、シグナルの指数関数的増加を再度見る地点でのNMTサイクル時間から差し引くことによって計算した。以下のTable 1(表1)中に示すように、Bリンパ腫細胞系においてNMT2発現とNMT1発現の比は(最大60倍)減少する。理論によって束縛されることは望まないが、ウエスタンブロッティングにより評価して、これらの結果は、NMT2をコードするmRNAの低減は、NMT2タンパク質レベルの低減の原因であり得ることを示唆することができる。
この実施例中、図7中では、様々なリンパ球性細胞系および固形リンパ腫腫瘍に存在したNMT2タンパク質のレベルの差を示す。(A)NMT1およびNMT2のレベルを、様々なタイプのヒト固形リンパ腫(びまん性大細胞型B細胞リンパ腫(DLBCL)および濾胞性リンパ腫(FL)腫瘍溶解物におけるウエスタンブロッティングにより評価する。示した両パネルに関して同じ抗体濃度、ウサギ抗NMT1(1:2500)、マウスモノクローナル抗NMT2(1:2500)を使用して、ウエスタンブロッティングによりNMTを検出した。各タイプの多数の腫瘍サンプルが、平均NMT2発現レベル(0.392+/-0.30)の範囲内にあることが示される。
この実施例中、図8のパネルAおよびパネルB中では、様々な固形リンパ腫腫瘍に存在したNMT2タンパク質のレベルの差を免疫組織化学法により決定した。
この実施例中、図9中では、72時間DDD85646で処置した様々なBリンパ球性細胞系の残存率を示す(図9A)。以下の等式を使用するGraFit解析に関するLeatherbarrow,R.J.(2009)GraFit Version6,Erithacus Software Ltd.,Horley,U.K中に記載された4パラメータ等式に従い、図9A中の曲線をプロットした。
この実施例中、図10中では、24時間のNMT阻害剤トリスDBAに対する、様々な不死化正常L0Bリンパ球(Dr.Riabowol,U.of Calgaryから入手)、悪性Bリンパ腫細胞(RamosとBL2)、およびT細胞白血病(CEM)の感受性を示す。(A)24時間トリスDBAで処置した様々な細胞系の残存率(n=9)(B)トリスDBAと24時間インキュベートしたNMT1またはNMT2を一過的に発現するCOS7細胞における、N-ミリストイルトランスフェラーゼ活性に対するトリスDBAの影響。in vitroにおいて、トリスDBAに関するIC50は、NMT1に関して約2μMおよびNMT2に関して5μMである。バーキットリンパ腫細胞系RamosおよびBL-2は、NMT阻害剤トリスDBAに対する感受性がより高い。
この実施例中、図11中では、967の癌細胞系に関する専門書(CCLE)のデータベースにおけるNMT発現のレベルを決定した。パネルA)CCLEのデータベースをNMT1およびNMT2発現に関して調べ、各NMTレベルをプロットした。NMT1(約6〜9)と比較してNMT2は広範囲の発現を示す(約3〜11)。パネルB)数種の癌亜型由来の癌細胞系のボックスおよびウィスカープロットをプロットし、967の癌細胞系全てのプロットと比較する。スチューデントのT検定を、全腫瘍と各群の比較により実施した。**は0.001より小さいP値を示す。パネルC)CCLEのデータベースを最も低いNMT2レベルで発現した50の細胞系に関して調べ、それを表形式で示した。様々な造血およびリンパ組織由来の腫瘍細胞系は最小NMT2発現細胞系の76%(50中38)を表し、様々な色で強調する。
この実施例中、図12中では、NMTはアポトーシス中に切断されるが依然活性状態である。パネルAおよびパネルB。抗Fasまたはスタウロスポリン媒介アポトーシスの過程を経るよう誘導したジャーカット細胞の溶解物を、ウエスタンブロッティングにより抗NMT1および抗NMT2を用いてプロービングし、両酵素の時間依存的切断を示す。ウエスタンブロッティングは、PAK2およびGAPDHに対する抗体を使用して同じサンプルで実施した。パネルCおよびパネルD。抗Fasまたはスタウロスポリン媒介アポトーシスの過程を経るよう誘導したジャーカット細胞におけるω-アルキニル-ミリステートの組み込みは、細胞が死滅し始めるとミリストイル化プロファイルの変化を示し、NMTは切断されるがそれらは依然活性状態であることを示唆する。ジャーカット細胞は、抗FAS(100ng/ml)とシクロヘキシミド(5μg/ml)(C)またはスタウロスポリン(2.5μM)とシクロヘキシミド(5μg/ml)(D)を用いたアポトーシスの誘導後に、25μMのアルキニル-ミリステートで代謝的に標識した。タンパク質サンプルはクリックケミストリーを使用してアジド-ビオチンと反応させ、NeutrAvidin(商標)-HRPを用いたウエスタンブロッティングにより可視化した。ウエスタンブロッティングによる標識組み込みの評価の前に、膜を0.1MのKOHでインキュベートした(BおよびD)。パネル(E)示した様々な時点で、ct-Bid N末端デカペプチドおよび[3H]-ミリストイル-CoAを使用してNMT活性をアッセイした。両NMT酵素が切断され両方共に依然触媒活性状態であるが、NMT1のみが有意な活性の消失を示す。(F)in vitroでのカスパーゼ-8および-3によるNMT1切断、ならびにカスパーゼ-3によるNMT2切断の再構築(クーマシー染色ゲル)。精製His-NMT1およびHis-NMT2(それぞれ5ug)を、活性カスパーゼ3および8(それぞれ500ng)と37℃で1時間インキュベートした。一般的カスパーゼ阻害剤(z-VAD-fmk)をインキュベーションの最後に加えて、反応がさらに進行するのを止めた。β-メルカプトエタノールを含む5×サンプルローディングバッファーと共にサンプルをすぐに煮沸し、10%アクリルアミドゲル上に載せPVDF膜上に移した。次いで膜をクーマシーブルーで染色し、タンパク質を可視化した。エドマン分解のため送付された切断断片に箱印をつけ、D72後のNMT1ならびにD25およびD67後のNMT2におけるカスパーゼ切断部位の同定を可能にした。

この実施例中、図14中では、精製組換え完全長His6-NMT1と組換え「カスパーゼ切断型」ct-His6-NMT1の間の酵素活性の比較を示す。完全長His6-NMT1(アミノ酸73〜496)。King et al.1999,Anal Biochemから取り入れたペプチドミリストイル化アッセイを使用してNMT活性をアッセイした。実験は二連。
この実施例中、図15中では、様々なNMT阻害剤および異なる細胞系のEC50とIC50の比較を示す。グラフは、生化学的酵素アッセイにおける8個のNMT阻害剤の生化学的効能(IC50)と、7個の細胞系を使用したアラマーブルー細胞生存率アッセイにおけるそれらの効能(EC50)の間の相関関係を示す。回帰分析を各細胞系で実施し0.9の平均R2値(範囲0.82〜0.96)を得た。様々なIC50とEC50の間のこの高レベルの相関関係は、細胞生存率アッセイにおける分子の活性は、NMTでのそれらの阻害活性によるものであることの強力な証拠である。2対の分子は生化学的アッセイでは類似した効能があることを記す。
この実施例中、図16中では、BL細胞でのDDD86481によるアポトーシスの誘導を示す。
この実施例中、図17中では、ヒト腫瘍におけるNMT2の消失に関する閾値の推定値を与える。Table 7(表7)中では、利用可能な場合CCLEデータセットの表示細胞系に関するlog2(マイクロアレイ蛍光強度NMT2)としてNMT2 mRNAの発現レベルを示す。
この実施例中、図18中では、ストレプトアビジン-セファロースビーズを使用し白血病ジャーカットT細胞において、翻訳後ω-アルキニル-ミリストイル化タンパク質をプルダウンするためのデスチオビオチン-PEG-アジドプローブの使用を示す。
この実施例中、図19中では、ストレプトアビジン-磁気ビーズを使用し白血病ジャーカットT細胞において、翻訳後ω-アルキニル-ミリストイル化タンパク質をプルダウンするためのデスチオビオチン-PEG-アジドプローブの拡大使用を示す。ジャーカットT細胞を、3時間2.5μMのスタウロスポリンでアポトーシスの過程を経るよう誘導し、次いで37℃で1時間いずれもBSAと結合した100μMのミリステート(C14)(パネルA)またはアルキニル-ミリステート(Alk C14)(パネルB)で標識した。細胞を回収し、溶解しクリックケミストリーを使用してアジド-デスチオビオチンと反応させた。デスチオビオチン-アルキニル-ミリストイル化タンパク質をストレプトアビジン磁気ビーズと結合させ、洗浄し50mMビオチンおよび2%SDSで15分間37℃で溶出した。溶出液1(パネルB)は多量のデスチオビオチン-ミリストイル化タンパク質を含有する一方で、ミリステートで標識した細胞由来の溶出液1ではほとんど見られない(パネルA)。露出時間:5秒間。
この実施例中、図20中では、アルキニル-ミリステートで標識した「正常」不死化B細胞(IM9)およびBL細胞(BL2とRamos)のミリストイル化プロファイルを示す。採取前に1時間、100μMのアルキニル-ミリステートで細胞を代謝的に標識した。クリックケミストリーを使用してアジド-ビオチンとタンパク質サンプルを反応させ、各細胞溶解物由来のタンパク質25μgをSDS-PAGEにより分離し、NeutrAvidin(商標)-HRPを使用したウエスタンブロッティングにより可視化した。矢印は、「正常」不死化Bリンパ球IM9には存在したがBL細胞(BL2とRamos)では見られなかった、ビオチニル化-アルキニル-ミリストイル化タンパク質を示す。
この実施例中、図21中では、DDD86481とドキソルビシンの組合せからの時間および用量依存的細胞毒性グラフを示す。Bリンパ球性細胞系IM-9(A)およびBL-2(B)を、24、48または72時間(上部〜底部)、0(青色)、17(赤色)および86nM(緑色)のヒドロキシドキソルビシンで高濃度のDDD86481(0、0.001、0.01、0.1、1.0、10および100μM)と共に処置した。MTSアッセイは、個別では非常に毒性が低い濃度のドキソルビシンとDDD86481により誘導された細胞殺傷を実証し、この影響は相乗的相互作用と一致する。これは、24時間と48時間の時点におけるDDD86481の単独使用に関するEC50と比較して、両化合物を一緒に使用したときのEC50の6倍の低下によって特徴付けられる。
この実施例中、以下のTable 8(表8)中では、北アメリカでの多種のヒトの癌におけるNMT2消失の大まかな広がりを評価する。本発明者らは、NMT2がない細胞(Toledo細胞、実施例18参照)に相当するlog2(マイクロアレイ蛍光強度)に関する5.64の最小カットオフ値を確立し、967細胞系の114(約12%)がこの閾値より下の範囲にあることを発見した。北アメリカでの各腫瘍タイプの発生率、NMT阻害療法からの利点を示す患者の数(閾値未満の発生率X%)、および選択した癌のメディカルニーズに関する根拠も示す。
この実施例中、図22中では、DMSO、スタウロスポリン、αFAS、または担体のみとインキュベートした細胞でイムノブロッティングを実施した。生成したイムノブロットは、抗NMT1抗体、抗NMT2抗体、抗PARP-1抗体、および抗GAPDH抗体でプロービングした。これらのデータは、NMTがアポトーシスの誘導により切断されることを示す。
この実施例中、図23中では、カスパーゼ切断型NMT2が完全長NMT2より3〜4倍活性があることを示す。精製完全長およびカスパーゼ切断型ヘキサヒスチジン(His)-NMTのNMT活性は、ペプチドミリストイル化アッセイを使用してアッセイした。N-ミリストイルトランスフェラーゼ活性は生成した放射標識ミリストイルペプチドの量から計算し、(King et al.1991,Anal Biochem.から取り入れた)ホスホセルロースペーパーで検出した。それぞれのNMTアッセイは三連で実施した。使用した対照はHis-NMT1精製からの溶出液5である。カスパーゼ切断型NMT2は完全長NMT2より3〜4倍活性がある。
この実施例中、図24中では、BL細胞におけるNMT2レベルの低下はヒストンデアセチラーゼの作用と関係があることを示す。
この実施例中、図25中では、様々なBL細胞系においてNMT2タンパク質レベルが低下することを示す。様々なリンパ球性細胞系由来の溶解物を地方の研究者から集め、それらのNMT含有量を分析した。結果は、「正常」不死化リンパ芽球B細胞系、IM9、L0およびVDSと比較したとき、試験した全てのバーキットリンパ腫(BL2、Daudi、RamosおよびBJAB)細胞系においてNMT2タンパク質レベルが低下したことを実証する。
この実施例中、図26中では、プロテオソームによる分解がBL細胞におけるNMT2枯渇の原因ではないことを示す。これらの実験では、「正常」不死化Bリンパ球(IM9)および悪性Bリンパ腫細胞[ホジキンリンパ腫(KMH2)およびBL(Ramos、BL2)]を、5時間10μMのプロテオソーム阻害剤(MG-132)で処置した。ウエスタンブロッティングを細胞溶解物で実施して、NMT1、NMT2、Mcl-1、およびGAPDHのレベルをモニタリングした。
この実施例中、図27中では、NMT2ではなくNMT1がカスパーゼ-8により切断されることが示される。ジャーカットT細胞(A)およびカスパーゼ-8ドミナントネガティブ突然変異体発現ジャーカットT細胞(B)をDMSOまたは抗Fas(150ng/mL)で処置し、サンプルは0、4、および8時間の時点で回収した。細胞を溶解し、NMT1、PARP-1および切断型カスパーゼ-8の存在を、ECLを使用したウエスタンブロッティングにより評価した。*は非特異的バンドを示す。
この実施例中、図28中では、NMT1とNMT2の両方がカスパーゼ-3により切断された。MCF7(A)およびカスパーゼ3発現MCF7(MCF7/カスパーゼ3)(B)を、DMSOまたはSTS(2.5μM)およびシクロヘキシミド(5μg/mL)で処置し、サンプルは0、4、および8時間の時点で回収した。細胞を溶解し、NMT2、PARP-1および切断型カスパーゼ3の存在を、ECLを使用したウエスタンブロッティングにより評価した。
この実施例中、図29中では、エドマン分解により同定したNMT1とNMT2のカスパーゼ切断部位を太字で示し、正に帯電したリシン(K)ボックスをNMT1およびNMT2アミノ酸配列(アミノ酸1〜80)で強調する。
この実施例中、図30中では、部位特異的突然変異誘発によるNMT切断部位の確認を示す。野生型および突然変異V5-NMT構築物を一過的に発現したHeLa細胞をSTS(2.5μM)とインキュベートした。V5-NMT1またはV5-NMT2構築物でトランスフェクトした細胞は、それぞれ4時間と5時間の時点で溶解した。ウエスタンブロッティングは、V5およびα-チューブリン(充填対照)抗体を使用してこれらのサンプルで実施した。
この実施例中、図31中では、細胞がアポトーシスの過程を経るときのミリストイル化プロファイルに対する変化を示す。抗Fas(150ng/mL)およびシクロヘキシミド(5μg/mL)を用いたアポトーシスの誘導後、25μMのアルキニル-ミリステートでジャーカット細胞を代謝的に標識した。クリックケミストリーを使用してアジド-ビオチンとタンパク質サンプルを反応させ、NeutrAvidin(商標)-HRPを用いたウエスタンブロッティングにより可視化した。ウエスタンブロッティングにより標識の組み込みを評価する前に、膜を0.1Mのトリス-HClまたは0.1MのKOHでインキュベートした。
この実施例中、図32中では、細胞がアポトーシスの過程を経るときのNMTレベルに対する変化を示す。抗Fas(150ng/mL)およびシクロヘキシミド(5μg/mL)を用いたアポトーシスの誘導後、25μMのアルキニル-ミリステートでジャーカット細胞を代謝的に標識した。ウエスタンブロッティングは、NMT1、NMT2およびGAPDHに対する抗体を使用して図31中と同じサンプルで実施した。(*)は非特異的バンドを示す。
この実施例中、図33中では、スタウロスポリンおよびシクロヘキシミドを用いてアポトーシスの過程を経るための、V5-NMT1およびV5-NMT2を一過的に発現するCOS7細胞の誘導を示す。NMT活性はペプチドミリストイル化アッセイを使用してアッセイし、ウエスタンブロッティングは、v5およびα-チューブリンに対する抗体(充填対照)を使用してこのサンプルで実施した。
この実施例中、図36中では、組換えヘキサヒスチジン(His)-タグ完全長およびカスパーゼ切断型hNMT1の精製を示す。精製はNi-NTAクロマトグラフィーを使用して実施した(材料および方法参照)。精製タンパク質は、クーマシーブルーゲル染色液でゲルを染色することにより可視化した。(FT:フロースルー、W:洗浄物およびE:溶出分画)。
この実施例中、図39中では、アポトーシス中のHeLa細胞における内在性NMTの細胞成分分画化を示す。HeLa細胞はDMSO、STS(2.5μM)または抗Fas(300ng/mL)およびシクロヘキシミド(5μg/mL)で処置し、サンプルは5時間の時点で回収し、材料および方法中に記載したように細胞成分分画化を施した。同体積のP100分画とS100分画を、NMT1およびNMT2抗体を使用したウエスタンブロッティングに施した。
この実施例中、図42は、アポトーシスの誘導に対する2-ヒドロキシミリスチン酸(HMA)の影響を示す。ジャーカットT細胞はHMA(1mM)有りまたは無しで処置し、アポトーシスは抗Fas(150ng/ml)およびシクロヘキシミド(5μg/ml)で誘導した。対照細胞はDMSOで処置した。サンプルは0、2、4、6および8時間の時点で回収した。細胞は溶解し、サンプルはSDS-PAGEにより分離して、PARP-1、PAK2、NMT1およびNMT2に対する抗体でイムノブロッティングした(コンポジットゲル)。

- NMT2合成、レベル、活性、もしくは機能の阻害、低下または排除、ならびにNMT2のタンパク質の合成、レベル、活性、もしくは機能の誘導または刺激の阻害によって定義される、NMT2に欠陥がある癌を有する対象を治療するための組成物であって、

癌を有する対象または癌を有する疑いがある対象由来のサンプルを処理する工程、
処理サンプルとサンプル内のミリストイル化タンパク質またはアジド-ビオチン標識ミリストイル化タンパク質に結合する抗体を接触させてサンプル中に存在する検出可能な標識とミリストイル化タンパク質の間で複合体を形成する工程を含み、前記複合体を示す少なくとも1つのミリストイル化プロファイルをもたらす結合アッセイを実施する工程を含み、
対照と比較して、前記ミリストイル化プロファイルが、前記癌が、NMT2合成、レベル、活性、もしくは機能の阻害、低下または排除、ならびにNMT2のタンパク質の合成、レベル、活性、もしくは機能の誘導または刺激の阻害によって定義される、NMT2に欠陥があることを示すとき、前記対象への前記組成物の投与が示され、
