Description

プロリンは、cisまたはtransコンフォメーションのいずれかをとる能力に関してユニークなアミノ酸残基である。その異性化のエネルギー障壁(εu=14〜24kcal mol−1)が比較的大きいため、非触媒異性化は、低速プロセスであるが、PPIアーゼにより加速されうる。PPIアーゼは、タンパク質のフォールディングを促進し、例としては、シクロフィリン(Cyps)、FK506結合タンパク質(FKBP)、およびパルブリン様PPIアーゼ(たとえば、Ess1およびPin1)が挙げられる。
本発明には、コンフォメーション特異的抗体またはそのフラグメントの発生および使用のための方法および組成物が記載されている。コンフォメーション特異的抗体は、たとえば、ポリペプチドのcisまたはtransコンフォメーションに特異的に結合しうる。特定の実施形態では、本発明に係るコンフォメーション特異的抗体は、ポリペプチドのリン酸化または非リン酸化Xaa−Proモチーフのcisまたはtransコンフォメーションに結合しうる(たとえば、cis pT231−tauまたはtrans pT231−tau)。Xaa−Proモチーフは、ポリペプチドのリン酸化Ser/Thr−Proモチーフでありうる(たとえば、Pin1基質、たとえば、pT231−tau)。コンフォメーション特異的抗体のその抗原(たとえば、Pin1基質、たとえば、pT231−tau)への結合は、障害または障害の進行の治療、診断、またはモニタリングに有用でありうる。

本発明に係る抗原は、当技術分野で公知の任意の方法により、たとえば、モノクローナル抗体、ポリクローナル抗体、キメラ抗体、ヒト化抗体、または組換えコンフォメーション特異的抗体を発生するために、使用されうる。これらの方法としては、KohlerおよびMilstein(Nature 256:495−497,1975 and Eur.J.Immunol.6:511−519,1976)ならびにCampbell(“Monoclonal Antibody Technology,The Production and Characterization of Rodent and Human Hybridomas,”in Burdon et al.,Eds.,Laboratory Techniques in Biochemistry and Molecular Biology,Volume 13,Elsevier Science Publishers,Amsterdam,1985)により記載された免疫学的方法、さらにはHuseら(Science 246:1275−1281,1989)により記載された組換えDNA法が挙げられる。
本発明は、ヒト化抗体を包含する。非ヒト抗体をヒト化する種々の方法が、当技術分野で公知である。たとえば、ヒト化抗体は、非ヒト源から導入された1個以上のアミノ酸残基を有しうる。これらの非ヒトアミノ酸残基は、多くの場合、「輸入」残基として参照され、典型的には、「輸入」可変ドメインから得られる。ヒト化は、本質的には、Winterら(Jones et al.,Nature 321:522−5,1986、Riechmann et al.,Nature 332:323−7,1988、Verhoeyen et al.,Science 239:1534−6,1988)の方法に従って、ヒト抗体の対応する配列を超可変領域配列で置換することにより、行うことが可能である。したがって、そのような「ヒト化」抗体は、無傷ヒト可変ドメインよりも実質的に小さい部分が非ヒト種の対応する配列により置換されたキメラ抗体である(米国特許第4,816,567号明細書)。実際には、ヒト化抗体は、典型的には、少なくともいくつかの超可変領域残基さらには他の可変領域残基が、齧歯動物抗体中の類似の部位の残基により置換されるヒト抗体である。
本発明に係るヒト抗体は、ヒト由来ファージディスプレイライブラリーから選択されるFvクローン可変ドメイン配列と、既知のヒト定常ドメイン配列と、を組み合わせることにより構築可能である(Hoogenboom et al.,J.Mol.Biol.227:381−8,1992、Marks et al.,J.Mol.Biol.222:581−97,1991)。他の選択肢として、本発明に係るヒトモノクローナル抗体は、ハイブリドーマ法により作製可能である。ヒトモノクローナル抗体を産生するためのヒト骨髄腫細胞系およびマウス−ヒトヘテロ骨髄腫細胞系は、たとえば、Kozbor,J.Immunol.133:3001−5,1984、Brodeur et al.,Monoclonal Antibody Production Techniques and Applications,pp.51−63(Marcel Dekker,Inc.,New York,1987)、およびBoerner et al.,J.Immunol.147:86−95,1991により記載されている。
本発明はまた、無傷抗体の一部を含む、好ましくはその抗原結合領域を含む抗体フラグメントを特徴とする。抗体フラグメントの例としては、Fab、Fab’、F(ab’)2、およびFvフラグメント、ダイアボディー、線状抗体、一本鎖抗体分子、ならびに抗体フラグメントから形成される多重特異的抗体が挙げられる。
本発明に係るコンフォメーション特異的抗体は、タウオパチー、TBI、または脳卒中の治療、阻害、または予防に使用されうる。コンフォメーション特異的抗体はまた、タウオパチー、TBI、または脳卒中の症状を改善するために使用されうる。
本発明に係る抗体は、たとえばp−Tauのcisまたはtransコンフォメーションいずれかが病理学的に高レベルであることにより特徴付けられる障害を治療するのに有用である。そのような治療は、たとえばp−Tauのcisまたはtransコンフォメーションいずれかが病理学的に高レベルであるとして同定される患者(たとえば、本明細書に記載の方法により診断された患者)で、またはそのような病理学的レベルに関連することが知られている疾患と診断された患者で、使用可能である。そのような障害としては、神経障害、たとえば、アルツハイマー病(AD)、軽度認識障害(MCI)、パーキンソン病(PD)、多発性硬化症(MS)、筋ジストロフィー、皮質基底核変性症、拳闘家認知症、ダウン症候群、前頭側頭型認知症、筋緊張性ジストロフィー、ニーマン・ピック病、ピック病、プリオン病、進行性核上性麻痺、亜急性硬化性全脳炎、痙攣性疾患(たとえば、癲癇)、血管性認知症、加齢性認知症、頭部外傷、脳卒中、神経繊維腫症、レビー小体病、筋萎縮性側索硬化症(ALS)、末梢神経症、および黄斑変性が挙げられる。本発明に係る抗体により治療可能な他の障害としては、外傷性脳傷害(TBI)、慢性外傷性脳症(CTE)、進行性核上性麻痺、前頭側頭葉変性、リティコ・ボディグ病、縺れ優位認知症、髄膜血管腫症、亜急性硬化性全脳炎が挙げられる。
本発明は、本明細書に記載の障害(たとえば、タウオパチー、TBI、または脳卒中)を治療、診断、およびその進行をモニターする方法および組成物を特徴とする。方法および組成物は、たとえば、リン酸化Ser/Thr−Proモチーフを、cisまたはtransコンフォメーション(たとえば、cis pT231−tauおよび/またはtrans pT231−tau)で、含有するPin1基質(またはその任意のフラグメントもしくは誘導体、たとえば、pT231−tau)の検出および測定を含みうる。本方法は、正常参照と比較してcisまたはtransコンフォメーションのpT231−tauの絶対レベルの測定を含みうる。5ng/ml未満、4ng/ml、3ng/ml、2ng/ml)、または1ng/ml血清未満のcisまたはtransコンフォメーションのpT231−tauの血清中レベルは、障害(たとえば、タウオパチー)と診断された患者において転帰良好と予測されると考えられる。5ng/ml超、10ng/ml、20ng/ml、30ng/ml、40ng/ml、または50ng/mlのcisまたはtransコンフォメーションの基質の血清中レベルは、障害とすでに診断された被験体において転帰不良と診断されると考えられる。
本発明はまた、診断検査キットを提供する。たとえば、診断検査キットは、ポリペプチド(たとえば、cis pT231−tauまたはtrans pT231−tau、それらのフラグメントに特異的に結合するコンフォメーション特異的抗体)と、ポリペプチド(たとえば、抗体)とp−Tauとの結合を評価するおよび/または検出するためのコンポーネントと、を含みうる。他の選択肢として、キットは、被験体サンプルの血清、血液、またはCSFに存在するcis pT231−tauまたはtrans pT231−tauを検出するために、cis pT231−tauポリペプチドもしくはtrans pT231−tauポリペプチド、またはcis pT231−tauフラグメントもしくはtrans pT231−tauフラグメントを含みうる。他の例では、本発明に係る診断用キットは、正常対照に存在するレベルなどの参照と比較して、cis pT231−tauまたはtrans pT231−tauポリペプチドのレベルの変化を同定するために使用されうる。そのようなキットは、陽性参照または正常対照参照の指標となる参照サンプルまたは標準曲線を含みうる。
本明細書に記載の診断方法はまた、治療時に障害(たとえば、MCI、TBI、タウオパチー、たとえば、AD、CTE)の進行をモニターするために、または治療用化合物の投与量を決定するために、使用可能である。一実施形態では、たとえばcisまたはtransコンフォメーションのpSer/Thr−Proモチーフを有するポリペプチド(例えばpT231−tau)のレベルは、障害を診断し、障害の治療もしくは管理をモニターする方法として、繰返し測定される。被験体における障害の進行をモニターするために、いくつかの時間点で被験体サンプルを取得し、次いで、比較しうる。たとえば、診断方法を用いて、治療前、治療時、治療後、被験体をモニターすることが可能である。この例では、本発明に係るコンフォメーション特異的抗体を用いて、被験体においてcisコンフォメーションのpT231−tauのレベルを厳密にモニターし、cisコンフォメーションのpT231−tauのレベルが治療時に減少し始めたら、障害を治療するための治療レジメンを、臨床医により決定されるように変更可能である(たとえば、治療剤の投与量を変化させたり、または異なる治療剤を投与したりしうる)。本発明に係るモニター方法はまた、たとえば、被験体において特定の薬剤または療法の有効性を評価したり、投与量を決定したり、または感染の進行、状態、もしくは病期を評価したりするのに使用されうる。
cisおよびtrans pT231−tauマウスmAbを発生するために、Nakamura et al.,Cell 149:232−244,2012に記載されるように、74%のcis pT231−Pip tauペプチドを用いてマウスを免疫した。この手順により480のハイブリドーマクローンを産生し、次いで、特徴付けた。Nakamura et al.,Cell 149:232−244,2012に記載されるcis(pT231−Dmp)もしくはtrans(pT231−Ala)にロックされた野生型cis+trans(pT231−Pro)または非リン酸化(T231−Pro)であるpT231−tauペプチドを用いてELISAによりスクリーニングされた最初の96のハイブリドーマ培養上清のうち、野生型およびcis pT231−tauペプチドを認識したが、transも非リン酸化も認識しなかった2つのcisハイブリドーマクローン#113および#74を同定し(図3A、3B)、野生型およびtrans pT231−tauペプチドを認識したが、cisも非リン酸化も認識しなかった2つのtransハイブリドーマクローン#25および#69を同定した(図3C)。mAbの重鎖および軽鎖のCDRを含む核酸配列およびタンパク質配列を以下に示す。さらに、不活性Cpd1Aを対照として、Pin1阻害剤Cpd1およびそれほど活性でない1Eで処理された細胞内で、cis mAb#113およびtrans mAb#25により検出したとき、cisは安定であったが、transは減少した(図3D)。図3Eは、アイソタイプ決定ELISAキット(Eagle)を用いて決定したとき、mAbのIgGのサブクラスを示している。mAbの重鎖および軽鎖のDNA配列は、Bradbury et al.,Neurobiol Aging 16:465−475,1995に記載の5’RACE RT−PCR技術を用いて決定された。BLAST検索から、これらの4つのmAbクローンが完全に新規であることが示された。予測タンパク質配列は、フレームワーク領域では高度に保持されているが、相補性決定領域(CDR)1〜3では明確に異なる(図8および9)。とくに、2つのcis mAbクローンは、2つのtrans mAb中には存在しないいくつかの保存残基を含有した。また、その逆も同様であった(図8および9)。
cis pT231−tauがADおよびCTEの脳においていつどこで現れるかを調べるために、cisおよびtrans mAb、続いて、アイソタイプ二次抗体を用いて、脳セクションの二重免疫染色を行った。trans mAbは、正常脳でさえも少数のニューロンの細胞体でのみ染色し、cis mAbは、正常脳を染色しなかったが、MCIニューロンの直線状神経突起でロバストにシグナルが検出とれ、さらにADニューロンの歪んだ神経突起に蓄積し局在した(図4A〜4C)。注目すべきことに、このパターンは、Nakamura et al.(Cell 149:232−244,2012)に記載のポリクローナルcisおよびtrans抗体により検出されたものに類似しており、cisおよびtrans mAbの特異性が確認されるだけでなく、ヒトMCIおよびADにおいて初期病原性コンフォメーションを標的とするのはtransではなくcisであることが示唆される。Dr.McKee(Liliang et al.,J Surg Res 160:302−307,2010、Chen et al.,Cancer Res 73:3951−3962,2013)により提供された8名の運動家および8名の退役軍人に関連するCTEの染色脳セクションから、transではなくロバストなcis p−tauが、II期に神経突起で容易に検出され、III期にさらに蓄積したことが示された(図4D〜4I)。これらのcis陽性神経突起は、軸索マーカー神経細線維または樹状突起マーカーMAP2と共染色することにより、軸索であることが確認された。注目すべきことに、cisおよびtrans p−tauの両方を有しているニューロンは非常に稀であったので、それらはin vivoで容易に相互変換されることが裏付けられる。したがって、cisは、ADおよびCTEにおいてごく初期に現れる。
cisおよびtrans mAbがニューロンに進入して予想されたニューロン区画に達することが可能であるかを調べるために、ヒトSY5Yニューロンをtau、p25/Cdk5(tauキナーゼ)、またはベクター対照でトランスフェクトし、続いて、培養培地にmAbを48時間添加し、その後、二次抗体または免疫ブロッティングのみを用いて免疫染色した。cisおよびtrans mAbは両方とも、ニューロンだけでなく、さまざまなニューロン区画で容易に検出された。cis mAbは、神経突起で主に検出されたが、trans mAbは、MCIおよびADの脳で予想されるとおり(図4)、細胞体で主に検出された(図5Aおよび5B)。さらに興味深いことに、cis mAbは、p25/Cdk5過剰発現後にのみ、内因性および外因性のtauの全レベルを有意に低減し、tau T231A突然変異体には明らかな影響を及ぼさなかった(図5C)。また、ヒト野生型Tau−Tgマウス海馬スライスの培養培地にcis mAbを添加したところ、tauレベルが劇的に低減した(図5D)。それに加えて、SY5Yニューロンを外傷性脳傷害(TBI)に類似したストレス条件である血清枯渇に付した時、SY5Yニューロンにおいて、cis pT231−tauが、血清枯渇により時間依存的に顕著に増加したが、そのような増加は、transではなくcis mAbにより効率的に抑止されたことが判明した(図5E)。したがって、cis mAbは、in vitroおよびex vivoでtauレベルを低減した。
cis mAbが、微小管破壊および神経毒性を誘導するp−tauの能力に影響を及ぼしうるか調べるために、SY5Y細胞をp25/Cdk5、tau、およびGFPで共トランスフェクトし、cisまたはtrans mAbを48〜72時間添加し、続いて、チューブリンおよびDNAの免疫染色を行った(図6A)。それに加えて、SY5YニューロンをCdk5−p25およびGFP−tauまたはそのT231A突然変異体で共トランスフェクトし、次いで、cisまたはtrans mAbを添加し、続いて、細胞形態および神経毒性を調べるために生細胞共焦点イメージングを行った(図6Bおよび6C)。両方のアッセイから、transではなくcis mAbが、ニューロンにおいてp−tauにより誘導される微小管破壊(図6A)および神経毒性(図6Bおよび6C)を強力に抑制することが明確に示された。ほとんどのGFP−tau陽性細胞は、対照細胞およびtrans mAb処理細胞において死滅し、微小管(MT)ネットワークは、核の周りに圧潰された(図6A)。ほとんどのcis mAb処理GFP−tau陽性細胞は、神経突起のMTネットワークでさえも、良好に生存した図6B、図6Cの矢印)。培養培地に添加した時、transではなくcis mAbはまた、血清枯渇により誘導されるSY5TYニューロンの微小管破壊および神経毒性を効率的に抑制した(図6Dおよび6E)。
CSF pT231−tauは、初期ADバイオマーカーである。CSF pT231−tauのコンフォメーションをアッセイするために、CSFにおいて、検出抗体としてcisまたはtrans Abを用いてINNOTEST hTau ELISAキット(Innogenetics)によりcisおよびtrans pT231−tauを測定した。8つすべての対照CSFにおいて、検出可能なcis pT231−tauは存在しなかったが、8つののうちの3つで少量のtrans pT231−tauが検出された(図7A)。これは、transが神経原線維の縺れ(NFT)に関連していないので、予想されうることである。注目すべき点として、進行性AD患者では、transおよびとくにcis pT231−tauは、脳組織(図4)で見られるものに一致して、顕著に増加した(図7A)。さらに、cisまたはtransのレベルには広範な個体間変動が存在したが、cis:transの比は、AD患者間で非常に類似していた(図7B)。
Pin1触媒コンフォメーション変化を可視化するために、第一cisおよびtrans pT231−tau抗体を形成する革新的ペプチド化学を開発した(図11)。注目すべきことに、transではなくcis pT231−tauは、MCIニューロンに初期に現れ、さらに、ADの進行に伴って変性ニューロンのみの軸索中に蓄積し、認知障害と良好に相関する。さらに、transではなくcis pT231−tauは、その正常微小管集合能力を喪失させ、毒性機能を取得し、脱リン酸化および分解に耐性であり、かつ凝集を起こしやすい(図11)。したがって、cis pT231−tauは、MCIおよびADにおいて初期病原性イベントである。発明者は、この度、ニューロンさらにはマウスTBI脳においてそれぞれのp−tau異性体を排除するのにきわめて有効な中和cisおよびtrans pT231−tauモノクローナル抗体(mAb)を開発した。
発明者は、生(緑色)/死(赤色)細胞アッセイキット(Abcam)およびアネキシン5FACS(図12A〜12E)より示されるように、低酸素や栄養枯渇などの種々のニューロンストレスが時間依存的にcis pT231−tauをロバストに誘導し、次いで、MTネットワークの破壊および最終的にアポトーシスによる細胞死を誘導することを見いだした。また、我々の生細胞ビデオ撮影から、時間依存的MT破壊およびミトコンドリアの軸索輸送の異常(データは示されていない)が確認された。注目すべき点として、cis mAb治療は、cis p−tau誘導をほぼ完全に排除し、さらに軸索MT破壊、ミトコンドリア輸送障害、およびアポトーシスを効率的にレスキューし、一方、trans mAbは、trans p−tauを除去し、クロス枯渇を伴うことなく表現型を促進した(図12)。アポトーシスを誘導するcis p−tauの能力は、カスパーゼおよびアポトーシスがヒトADニューロンに存在するというこれまでの証拠と一致する(Gervais et al.,Cell 97:395−406,1999)。同様に、cis p−tauを排除するcis mAbの能力は、cis p−tauがtransよりも安定であり(Nakamura et al.,Cell 149:232−244,2012)、タンパク質分解のために抗体複合体がTRIM21により認識される(Mallery et al.,PNAS 107:19985−1998590,2010;McEwan et al.,Bioessays 33:803−809,2011)という我々の知見と一致する。
AD患者のCSFには豊富なpT231−tauが存在し、培養ニューロンは非従来的機序で培地中にtauを分泌するので、発明者は、ニューロンがストレスを受けてcisおよびtrans p−tauを分泌するかを調べた。実際に、ストレスを受けたニューロンは、40hrでcis pT231−tauを分泌したが、trans pT231−tauを分泌しなかった。72hrで細胞死となり、cisおよびtrans p−tauの両方さらにはアクチンが放出され時(図13A)。より重要なこととして、健常ニューロンに3日間添加した時、cis含有培地は、アポトーシスによりニューロンを死滅させた。trans mAbではなくcis mAbによる培地の前処理、続いて、プロテインGによるmAbの枯渇を行ったところ、ニューロン死は完全にレスキューされた(図13B)。
また、ヒトAD脳が毒性cis pT231−tauを含有していたか調べるために、発明者は、ヒトAD脳溶解物を培養ニューロンに添加し、レシピエントニューロンにおいてcis pT231−tauを検出した。ただし、対照脳溶解物を使用した時、cis−tauを添加しなかった。より重要なこととして、正常ではなくADの脳溶解物は、レシピエントニューロンにおいてアポトーシスを誘導する。これは、transではなくcis mAbでAD脳溶解物を前処理することにより完全にレスキューされた(図14)。ヒトCTE脳溶解物を用いた場合にも、類似の結果が得られた。
cis p−tauとTBI重症と度の関係を調べるために、発明者は、スポーツ関連TBIを模倣して、マウスにおいてさまざまな高さで落錘デバイスを用いてさまざまな重症度の閉鎖性頭部脳傷害を誘導し、TBIの48hr後、脳内のcis p−tauを試験した。TBIの48hr後、TBI重症度に伴って、cisおよび全tauは、ロバストに増加した(図15A)。これは、cis p−tauが分解に対して耐性があるという我々の知見と一致する。ロバストなcis p−tauはまた、軍事関連TBIを模倣して爆風誘導TBIの48hr後に見いだされた。時間経過研究から、重篤なTBI後、驚くべきことに、12hrs後、transではなくcis p−tauは誘導され、時間と共に増加し、少なくとも2週間維持された(図15Bおよび15C)。cis陽性神経突起は、この場合も、樹状突起ではなく軸索であった。注目すべきことに、発明者は、AT8、AT180、TG3、AT100、MC1、Alz50、またはPHF1を含めて、使用したmAbにより、縺れ関連エピトープをなんら見いだすことができなかった(図15D)。仮に見いだされたとしても、WTマウスでは、TBIのかなり後である。
cis mAbがTBIマウス脳においてcis pT231−tauおよびその毒性を導入し排除するかを調べるために、発明者は、最初に、ビオチン化cis mAbをB6マウスの腹腔内または静脈内に投与し、3日後cis mAbを検出した。注入されたmAbは、脳内で容易に検出された(図9A)。次いで、重篤なTBI後、4日ごとに250μgのcis mAbを用いて3回にわたりマウスを腹腔内治療しに、14日後、脳を分析した。注目すべき点として、cis mAbは、マウス脳においてTBI誘導cis pT231−tau誘導を効率的に防止し、全tauを低減した(図9B)。さらに、cis mAb治療は、PARP切断アッセイにより評価したとき、軸索MT破壊およびミトコンドリア破壊(図10A)さらにはアポトーシスでさえも効率的に回復したが(図9C)、対照IgGではそうはならなかった。
以上の説明から、種々の使用法および条件に合わせて本明細書に記載の発明に変更および修正を加えうることは、明らかであろう。そのような実施形態もまた、以下の特許請求の範囲内にある。

(22)タウオパチーを有するまたはその素因を有するとして被験体を診断する方法であって、a.前記被験体から得られたサンプルでcis pT231−tauのレベルまたはcis:trans pT231−tauの比を決定することと、b.前記サンプルのcis pT231−tauの前記レベルまたはcis:trans pT231−tauの比を正常参照サンプルと比較することであって、前記正常参照サンプルと比較して、cis pT231−tauのレベルの上昇またはcis:trans pT231−tauの比の増加が、前記タウオパチーを有するまたはその素因を有するとして前記被験体を診断することにつながる、比較することと、c.前記タウオパチーを治療するのに十分な量で、(1)〜(8)のいずれか一に記載の結合部分を、前記被験体に投与することと、を含む、方法。
(23)前記サンプルが、尿、血液、血清、血漿、唾液、羊水、および脳脊髄液(CSF)からなる群から選択される、(20)または(22)に記載の方法。

Claims (31)
- それぞれ配列番号1〜3の重鎖相補性決定領域(CDR)1、CDR2及びCDR3配列を含む重鎖可変領域とそれぞれ配列番号4〜6の軽鎖CDR1、CDR2及びCDR3配列を含む軽鎖可変領域とを含む、単離されたコンフォメーション特異的結合部分であって、リン酸化トレオニン231-タウタンパク質(pT231-tau)のcisコンフォメーションに特異的に結合する、前記結合部分。
- 前記重鎖可変領域が配列番号22の配列を含み、且つ前記軽鎖可変領域が配列番号23の配列を含む、請求項1記載の単離された結合部分。
- それぞれ配列番号7〜9の重鎖相補性決定領域(CDR)1、CDR2及びCDR3配列を含む重鎖可変領域とそれぞれ配列番号10〜12の軽鎖CDR1、CDR2及びCDR3配列を含む軽鎖可変領域とを含む、単離されたコンフォメーション特異的結合部分であって、リン酸化トレオニン231-タウタンパク質(pT231-tau)のcisコンフォメーションに特異的に結合する、前記結合部分。
- 前記重鎖可変領域が配列番号24の配列を含み、且つ前記軽鎖可変領域が配列番号25の配列を含む、請求項3記載の単離された結合部分。
- 前記結合部分が抗体又はその抗原結合フラグメントである、請求項1〜4のいずれか1項記載の単離された結合部分。
- 前記抗体又はその抗原結合フラグメントが一本鎖抗体である、請求項5記載の単離された結合部分。
- 前記抗体又はその抗原結合フラグメントがキメラ抗体、ヒト化抗体又はそれらの抗原結合フラグメントである、請求項5又は6記載の単離された結合部分。
- 請求項1〜7のいずれか1項記載の結合部分と薬学的に許容可能な担体とを含む医薬組成物。
- 前記結合部分がモノクローナル抗体である、請求項8記載の医薬組成物。
- 必要とされる被験体においてタウオパチー、外傷性脳傷害(TBI)又は脳卒中を治療するための請求項8又は9記載の医薬組成物。
- 前記タウオパチーが、進行性核上性麻痺、慢性外傷性脳症(CTE)、前頭側頭型認知症、前頭側頭葉変性、リティコ・ボディグ病、縺れ優位認知症、髄膜血管腫症、亜急性硬化性全脳炎、ピック病、皮質基底核変性症及びアルツハイマー病から成る群より選択される、請求項10記載の医薬組成物。
- 前記被験体が前記タウオパチーの素因を有するか又は初期段階にある、請求項10又は11記載の医薬組成物。
- 前記タウオパチーの素因又は初期段階が、前記被験体から得られたサンプルでcis pT231-tauのレベルの上昇又はcis:trans pT231-tauの比の増加により決定される、請求項12記載の医薬組成物。
- CSF t-tau、pT181-tau、Aβ42又はApoE4のレベルを決定することをさらに含む、請求項13記載の医薬組成物。
- 前記サンプルが、尿、血液、血清、血漿、唾液、羊水及び脳脊髄液(CSF)から成る群より選択される、請求項13記載の医薬組成物。
- 前記被験体が、繰り返された脳外傷の病歴により素因が与えられる、請求項13記載の医薬組成物。
- 請求項1〜7のいずれか1項記載の結合部分を用いて治療された被験体において治療反応をモニターする方法であって、前記被験体から得られたサンプルでcis pT231-tauのレベル又はcis:trans pT231-tauの比を決定することを含み、cis pT231-tauのレベル又はcis:trans pT231-tauの比の減少が、前記結合部分に有効な治療反応をもたらす、前記方法。
- CSF t-tau、pT181-tau、Aβ42又はApoE4のレベルを決定することをさらに含む、請求項17記載の方法。
- CSF t-tau、pT181-tau、Aβ42又はApoE4の前記レベルが減少する、請求項18記載の方法。
- 前記サンプルが、尿、血液、血清、血漿、唾液、羊水及び脳脊髄液(CSF)から成る群より選択される、請求項17記載の方法。

b.前記サンプルのcis pT231-tauの前記レベル又はcis:trans pT231-tauの比を正常参照サンプルと比較することであって、前記正常参照サンプルと比較して、cis pT231-tauのレベルの上昇又はcis:trans pT231-tauの比の増加が、前記タウオパチーを有する又はその素因を有するとしての前記被験体の診断において補助する、比較することと、
を含む、前記使用。 - 前記キットが、前記タウオパチーを有する又はその素因を有するとして同定された被験体に請求項1〜7のいずれか1項記載の結合部分を投与するための説明書をさらに含む、請求項21記載の使用。
- 前記サンプルが、尿、血液、血清、血漿、唾液、羊水及び脳脊髄液(CSF)から成る群より選択される、請求項21記載の使用。
- それぞれ配列番号13〜15の重鎖相補性決定領域(CDR)1、CDR2及びCDR3配列を含む重鎖可変領域とそれぞれ配列番号16〜18の軽鎖CDR1、CDR2及びCDR3配列を含む軽鎖可変領域とを含む、単離されたコンフォメーション特異的結合部分であって、pT231-tauのtransコンフォメーションに特異的に結合する、前記結合部分。
- 前記重鎖可変領域が配列番号26の配列を含み、且つ前記軽鎖可変領域が配列番号27の配列を含む、請求項24記載の単離された結合部分。
- 前記結合部分が抗体又はその抗原結合フラグメントである、請求項24又は25記載の単離された結合部分。
- 前記抗体又はその抗原結合フラグメントが、キメラ抗体又はヒト化抗体である、請求項26記載の単離された結合部分。
- 前記抗体又はその抗原結合フラグメントが一本鎖抗体又は抗体フラグメントである、請求項26又は27記載の単離された結合部分。
- 請求項24〜28のいずれか1項記載の結合部分と薬学的に許容可能な担体とを含む医薬組成物。
- 前記結合部分がモノクローナル抗体である、請求項29記載の医薬組成物。
- a.pT231-tauのcisコンフォメーションに特異的に結合する、請求項1〜7のいずれか1項記載の結合部分と、
b.pT231-tauのtransコンフォメーションに特異的に結合する、請求項24〜28のいずれか1項記載の結合部分と、
c.タウオパチーを有する又はその素因を有するとして被験体を診断するためのa.の結合部分及びb.の結合部分を使用するための説明書と、
を含む、タウオパチーを有する又はその素因を有するとして被験体を診断するためのキット。
