1つの態様において、本発明は、IgG CH2ドメインおよびIgG CH3ドメインを含む単量体Fc含有ポリペプチドであって、CH3ドメインが、CH3−CH3二量体化界面において1つまたは複数の操作されたN連結型グリコシル化部位を含み、操作されたN連結型グリコシル化部位が、XがProを除く任意のアミノ酸であるAsn−X−SerまたはAsn−X−Thrのコンセンサス配列を提供するための少なくとも1つのアミノ酸修飾を含む、単量体Fc含有ポリペプチドを提供する。
本発明はまた、本明細書において記載される単量体Fc含有ポリペプチドをコードする核酸分子および核酸配列を包含する。一部の実施形態において、異なる核酸分子が、本明細書において記載される単量体Fc含有ポリペプチドの1つもしくは複数または一部をコードする。他の実施形態において、同一の核酸分子が、本明細書において記載される単量体Fc含有ポリペプチドをコードする。
1つの態様において、本発明は、単量体Fc含有ポリペプチド、または組換えによって連結した少なくとも2つの本明細書において記載される単量体Fc含有ポリペプチドを含むポリペプチドを生産する方法を提供する。

1つの態様において、本発明は、薬学的に許容できる担体内に、単量体Fc含有ポリペプチド、または組換えによって連結した少なくとも2つの単量体Fc含有ポリペプチドを含むポリペプチドを含む、医薬組成物を提供する。特定の実施形態において、本発明のポリペプチドは、中性形態(両性イオン形態を含む)で、または正もしくは負に荷電した種として、存在し得る。一部の実施形態において、本明細書において記載されるポリペプチドは、対イオンと複合して「薬学的に許容できる塩」を形成し得、この塩は、1つまたは複数のポリペプチドおよび1つまたは複数の対イオンを含む複合体を指し、対イオンは、薬学的に許容できる無機および有機の酸および塩基に由来する。
CH3−CH3界面内にN−グリコシル化部位を組み込むことによる、本来は二量体であるFcの単量体化
糖鎖工学戦略を用いて、抗体のFcドメインの安定な単量体形態を操作した。さらに具体的には、嵩高い親水性の糖質部分を、Fcの二量体形態を単量体形態に分離していたCH3−CH3界面内に導入した。驚くべきことに、この糖鎖工学はまた、CH3ドメインの暴露された界面を安定化させた。4つの基準を採用して、(Asn−X−Ser/Thrの基準のN連結型グリコシル化シグナル配列を生じさせるために)どこにN−グリコシル化突然変異部位を組み込むかを決定した。第一に、Igドメイン核内に埋まっている残基、または溶媒に暴露されている残基が選択されることを回避するために、CH3−CH3の界面上に位置する残基を同定した(図1)。ヒトガンマ1Fc(PDB ID:1HZH)の結晶構造を用いて、プログラムMOE(Chemical Computing Group)で、両Fc二量体(非変性形態)における、およびFc二量体の一方の鎖(すなわち、仮想のFc単量体)における各残基のアクセス可能な表面のパーセント(%ASA)を計算した。より高い%ASA(二量体)を有する残基は、溶媒に暴露されている残基である。高い%ASA(単量体)値を有する残基ほど、溶媒に暴露されているかまたはCH3−CH3界面内に埋まっている可能性が高いと仮定された。したがって、界面の関与の程度を、ΔASA=%ASA(単量体)−%ASA(二量体)としてFc二量体の%ASAから単量体の%ASAを差し引くことによって決定し、ΔASAが閾値である10%を超えた22の界面残基を選択した。表1を参照されたい。第二に、プロリン残基、グリシン残基、およびシステイン残基は通常、タンパク質の構造的フレームワークの維持において重要な役割を有するため、これらの残基の突然変異生成を回避した。第三に、グリコシル化の占有の可能性を考慮した。残基Asn−X−Ser/Thr−Yを、X(AsnとSer/Thrとの間に位置するアミノ酸)もY(Ser/Thrの次に位置するアミノ酸)もプロリン残基ではない領域内に組み込み、それは、これらの位置にあるプロリンはグリコシル化効率を強力に阻害するためである。Asn−X−Ser/Thrの基準のN連結型グリコシル化シグナル配列の第3の位置(すなわち、Ser/Thr)にある残基が突然変異していなくてはならない場合、スレオニンがセリンよりも選択され、それは、第3の位置にあるスレオニンが第3の位置にあるセリンよりもアスパラギン残基でのより高いグリカン占有をもたらすことが示されているためである。最後に、残基をFcドメインの一方の鎖の三次元構造上でマッピングし、手作業で調べ、操作された糖質がCH3−CH3界面を分離するためにほとんど影響を有し得なかった位置(Leu256およびAsp276)を排除した。したがって、全部で9つの位置を、N−グリコシル化のために合理的に選択した。これらの選択された残基は、ヒトIgGの全アイソタイプ(1、2、3、および4)ならびにマウスIgG(2、2a、2b、および3)アイソタイプの間でよく保存されている(図2)。個々のN−グリコシル化突然変異体は、ヒンジ領域(Gly226からLys497)を有さないヒトIgG1およびIgG4のFcドメインを鋳型として用いることによって構築した。これらのペプチドをコードする核酸を、HEK293細胞において一過性発現させた。N−グリコシル化の発現および効率を評価するために、培地上清を、還元条件下でのSDS−PAGE、およびウェスタンブロット分析にかけた。全ての突然変異体の発現レベルは、366位の突然変異体の発現が弱かったことを除いて、野生型Fcドメインに類似していた。N−グリコシル化変異体はおよそ25kDaの分子量に対応する移動度で移動したが、非グリコシル化変異体はおよそ22kDa移動したようであった。5つの部位(364、366、368、405、および407)は効率的なN−グリコシル化を示したが、4つの部位(347、390、401、409)はN−グリカンのおよそ50%以下の組み込みを示した。IgG1突然変異体およびIgG4突然変異体の発現およびグリコシル化のプロファイルは類似であったため、IgG1突然変異体をさらなる研究のために用いた。

完全なN−グリコシル化を有する4つの突然変異体(364位、368位、405位、および407位)を、さらなる精製および特徴付けのために選択した。CH23−N364(Fc−S364N)、CH23−N368(Fc−L368N/K370T)、CH23−N405(Fc−F405N/Y407T)、およびCH23−N407(Fc−Y407N/K409T)として示されるこれらのN−グリコシル化突然変異体を、本明細書の他の箇所に記載されるように精製し、SDS−PAGEによって判断される95%超の純度を得た。精製タンパク質の収量は、培地1リットル当たり20〜30mgの範囲であった。各位置でのグリコシル化変異体および非グリコシル化変異体の相対的収量を評価するために、PNGase Fの処理を行わない、および行う、精製タンパク質のキャピラリーゲル電気泳動(CGE)アッセイを行った。本明細書において開示されているデータは、これらの4つの突然変異体が最大10%の非グリコシル化変異体を含有することを示唆しており、これは、野生型Fcに類似している(表2)。分析用サイズ排除カラム(SEC)を用いて、N−グリコシル化変異体の見かけの分子量を推定した。4つの突然変異体の全ては、野生型Fc(約48kDa)よりも低い見かけの分子量(25〜30kDa)を示し、このことは、組み込まれたN−グリコシル化がFc二量体のCH3−CH3界面を破壊するのに成功したことを示唆する。N−グリコシル化突然変異体はSECによって単量体であると思われたが、SEC−MALS(サイズ排除クロマトグラフィー多角度光散乱検出器)を用いて、これらの突然変異体のオリゴマー種の分布のさらに厳密な分析を行った。屈折率のシグナル全体にわたる光散乱により決定された分子量は、CH23−N405が完全に単量体であることを示したが、CH23−N364、CH23−N405、およびCH23−N407は単量体形態と二量体形態との混合物であることが分かった。これらの4つのN−グリコシル化突然変異体の熱安定性を、示差走査熱量測定(DSC)によってさらに特徴付けした。野生型Fcのサーモグラムは、72℃および83℃の融解温度での2つの転移を示した。これらの値は、CH2ドメインおよびCH3ドメインの融解に割り当てられている70.8℃および83.3℃の値と同程度であった。逆に、個々のN−グリコシル化突然変異体は、より低い融解温度での単一の転移を示した(表2)。
複数のN−グリコシル化部位の組み合わせ
各変異体の非グリコシル化部分をさらに最小にするために、CH3ドメイン内の2つのN−グリコシル化部位を、364位、368位、405位、および407位での個々の突然変異を介して導入し、その結果、グリコシル化の90%から95%の占有を得た。図3は、CH3ドメインの界面におけるSer364、Leu368、Phe405、およびTyr407の特別なアラインメントを強調する。これらの所見に基づいて、368、405、および407のうちの2つの部位にある2つの糖質部分が構造をさらに不安定化させることが仮定され、それは、これらの3つの残基が互いにごく接近して位置しているためである。したがって、これらのN連結型部位の3つの組み合わせ、すなわち、N364およびN368、N364およびN405、ならびにN364およびN407を、2つのN−グリコシル化部位の組み込みのために選択した。これらの3つの二重N−グリコシル化突然変異体、すなわち、CH23−N364/N368、CH23−N364/N405、およびCH23−N364/N407を、HEK293細胞において発現させ、ウェスタンブロットによって発現およびグリコシル化について試験した。単一N−グリコシル化突然変異体と比較したサイズの増大が、CH23−N364/N368およびCH23−N364/N407で観察され、一方、CH23−N364/N405は検出可能なレベルで分泌されなかった。CH23−N364/N405は細胞によって発現されるが排出はされないことが分かったため、この二重突然変異はタンパク質の分泌を無効にし得るか、またはタンパク質構造の不安定性を生じさせ得ると仮定される。PNGase Fの処理の結果、Fcドメインの還元形態に対応する分子サイズが生じたため、CH23−N364/N368およびCH23−N364/N407のサイズの増大は、各分子上の複数のN連結型グリカンの存在に寄与した。CH23−N364/N368タンパク質およびCH23−N364/N407タンパク質を精製し、オリゴマー状態、非グリコシル化分子の収量、および熱安定性を、本明細書の他の箇所に記載されるように調べた。CH23−N364/N368とCH23−N364/N407との両方の非グリコシル化分子を、検出不可能なレベルまで減少させた(表2)。生産されたCH23−N364/N407は、CH23−N364およびCH23−N407の個々の突然変異体が検出可能な量の二量体を形成したにもかかわらず、完全に単量体であった。注目すべきことに、CH23−N364/N368は、凝集傾向ならびにSEC−MALS分析およびDSC分析に基づく熱安定性の減少を示した。それにもかかわらず、364位および407位での二重突然変異は、安定性、グリコシル化効率、および単量体傾向に関して、単量体Fcの特性を向上させた(表2)。グリコシル化はまた、(タンパク質構成要素にのみ感受性の)280nMでのUV検出器、および(タンパク質構成要素と糖質構成要素との両方に感受性の)RI検出器を用いる分析的SEC−MALSによって確認された。
CH23−N364/N407の結晶構造
CH23−N364/N407の結晶を成長させ、これは、1.9Åに対して回折した。構造を、Fc二量体の一方のポリペプチド鎖の座標(3DTS)をサーチモデルとして用いて、分子置換によって解明した。データの収集項目、ならびにデータセットおよびモデルについての精密化統計を、表3に示す。CH23−N364/N407の実験マップは、Gly224からSer447までの骨格全体についての明らかな密度をもたらし、側鎖の95%超は、電子密度にフィットした。S364N、Y407N、およびK409Tの置換は、CH3ドメイン上ではっきりと見ることができた。Asn297に接続している糖質鎖のトポロジーを、その電子密度から決定した。8つの糖残基(GlcNAc1〜GlcNAc5、Man7、GlcNAc8、およびFuc)を同定した。逆に、各糖残基のみが、操作されたAsn364残基およびAsn407残基上で同定された。その電子密度に従ってAsn364側鎖に付着したGlcNAc1を、位置決定した。しかし、密度マップはAsn407側鎖にごく接近して、水分子よりも大きな重原子が存在することを示唆していたが、Asn407に付着したGlcNAc1は、電子密度が低いために位置付けることができなかった。それでも、CH23−N364/N407結晶の非対称の単位内容物は、2つの同一のグリコシル化ポリペプチド鎖の二量体として存在する正規のFcドメインの1つのみの単量体単位を示した(図4)。これらの結晶学的データは、CH3−CH3界面上の操作されたグリコシル化がFcドメインの単量体形態を安定化させ得ることを実証する。
Fab−CH23変異体の生産およびインビトロでの特徴付け
CH23の血清寿命におけるFcRnの関与を解明するために、抗KLH抗体に由来するFab断片をCH23に融合した(本明細書において「Fab−CH23」と呼ばれ、これはまた、Fab−CH23−N364/N407(S364N−L355−T366およびY407N−S408−K409T(例えば、配列番号71)とも呼ばれる)。同一のFab断片をまた、FcRnノックアウト変異体(Fab−CH23[H310A/H433A](例えば、配列番号75))およびFcRn増強変異体(Fab−CH23[M428L/N434S](例えば、配列番号73))にも融合した。2つの操作されたFcポリペプチドを含むタンデムCH23構築物(Fab−CH23−CH23(例えば、配列番号85))、および正規のIgG1フォーマット対照(すなわち、抗KLH抗体)もまた構築して、アビディティーがCH23の薬物動態学的特性を向上させ得るという仮説を試験した(図5)。全ての構築物をCHO細胞内にトランスフェクトし、タンパク質をプロテインGカラムクロマトグラフィーによって精製した。対照として、HEK293細胞一過性発現系において生産されるFab−CH23(「Fab−CH23−HEK」と呼ばれる)を生産した。1対1の結合フォーマット(BIAcoreチップの表面に結合した「Fc変異体」およびチップ表面に浮遊する可溶性FcRn)と、アビディティーフォーマット(BIAcoreチップ表面に結合したマウスFcRnタンパク質およびチップ表面に浮遊する各Fc変異体)との両方のこれらの構築物のFcRnの結合を調べた。平衡結合データを表4にまとめる。Fab−CH23またはFab−CH23−HEKの1対1の結合アフィニティー(表4)は、CH23−N364/N407の結合アフィニティー(表1)に類似しており、このことは、Fab融合がFcRnへのFcの結合に影響しなかったことを示唆する。予想した通り、Fab−CH23[M428L/N434S]でFcRnへのより高い結合アフィニティーが観察されたが、一方、FcRnの結合は、Fab−CH23[H310A/H433A]では観察されなかった。1対1の結合アッセイにおいて、チップ表面上に局在化したFab−CH23−CH23は、FcRnへのFab−CH23の結合アフィニティーと類似の結合アフィニティーを示した。驚くべきことに、しかし、アビディティーアッセイ(チップ表面上に局在化したFcRn)において、Fab−CH23−CH23は、Fab−CH23よりもおよそ40倍高い、FcRnへのアフィニティー結合を示した。IgG FcおよびタンデムCH23−CH23のアーキテクチャは異なるが、Fab−CH23−CH23は、単一Fcドメインを含むIgGのアフィニティーに類似の、バイオセンサー表面上に局在化するFcRnに高いアフィニティーで結合することが分かった。Fab−CH23−CH23が中性pHでFcRnから解離したことを評価するために、解離相の間にpH7.4およびpH6.0で結合したFab−CH23−CH23のパーセントを測定した。Fab−CH23−23とIgGとの両方の結合%がほとんど同一であることが観察された。これらの結果は、Fab−CH23−CH23が、酸性pHでFcRnに堅く結合するだけではなく、野生型IgG Fcに類似して、中性pHで効率的にFcRnから解離することを実証する。
マウスにおけるFab−CH23変異体の薬物動態学
Balb/c雄マウスにおいて、二価抗体、または抗体と同一のFabを含む単量体Fc含有ポリペプチドの、5mg/kgの単回IV用量後の、平均血漿濃度のプロファイルを決定し、データを図6に示し、PKパラメータを表5にまとめる。対象間のばらつきはFab−CH23で比較的高く、血漿濃度の有意な低下が投薬の96時間後に観察され、このことは、免疫原性(例えば、構築物に対するマウス抗ヒト抗体、AHAによるクリアランス)の可能性を示唆する。Fab−CH23についてのクリアランス(CL)は低く、0.3mL/時間/kgでの典型的なIgGのCLと同程度であり、T1/2は173時間(約7.2日)であった。18および14mL/時間/kgでのFab−CH23−HEKおよびFab−CH23[H310A/H433A]についてのCL率は、それぞれ、0.3mL/時間での野生型IgGについてのCL率よりもはるかに高かった。結果として、T1/2は、12時間および11時間でのこれらの2つの変異体について、それぞれ、野生型IgG(173時間)と比較して、はるかに短かった。Fab−CH23−HEKを、腹腔内経路を介して10mgのマンナン(マンノース受容体の天然阻害剤)と同時投与すると、CLはマンナンを伴わないFab−CH23と比較して2倍増大し、このことは、マンノース受容体介在性のクリアランスがFab−CH23−HEKの主要なクリアランスメカニズムであることを示す。Fab−CH23およびFab−CH23[M428L/N434S]は、CLが約9mL/時間/kgでありT1/2がそれぞれ32時間および42時間であるFab−CH23−HEKおよびFab−CH23[H310A/H433A]よりも向上したPKを有していた。タンデムFab−CH23−CH23は、全ての単量体Fab融合体の間で、最も遅いCL(3mL/時間/kg)および最も長いT1/2(97時間)を有していた。野生型IgGに類似して、Fab−CH23−CH23の血漿濃度もまた、投薬の96時間後に劇的に低下し、このことは、潜在的な免疫原性もまたFab−CH23−CH23のクリアランスに関与し得ることを示す。
実験手順
プラスミドの構築およびタンパク質の発現
野生型Fc断片のための発現プラスミドを、Gly236で始まるヒトガンマ1定常領域の前にあるN末端ヘキサヒスチジンタグとして構築した。全てのプラスミド構築および突然変異生成は、In−FusionドライダウンPCRクローニングキット(Clontech、マウンテンビュー、カリフォルニア州)で行った。突然変異構築物は、所望のアミノ酸置換を生じさせるプライマーを用いてPCRによって生成した。得られたPCR産物を、In−Fusionクローニングエンハンサーで処理し、その後、XbaIおよびEcoRI(New England Biolab、イプスウィッチ、マサチューセッツ州)で処理した発現ベクター内に挿入した。Fab−monoFc変異体の発現ベクターを、抗KLH抗体のFab断片およびmonoFcをコードする構築物のPCR増幅によって構築した。N−グリコシル化Fc変異体のタンパク質生産のために、HEK293F細胞を、293fectin試薬を用いることによって発現プラスミドで一過的にトランスフェクトし、製造者のプロトコルに従って(Invitrogen)、FreeStyle293培地において成長させた。馴化培地を、トランスフェクションの6日後に、2000×gで10分間の遠心分離によって回収した。IgG変異体およびFab−monoFc変異体のために、CHO細胞を、リポフェクタミン2000(Invitrogen、グランドアイランド、ニューヨーク州)によって発現プラスミドをトランスフェクトした。安定なクローンを、50ug/mLのG418および50nMのメトトレキサートで2から3週間、選択した。馴化培地を遠心分離によって回収し、上清を、その後の精製のために、0.2umのフィルターによって濾過した。発現を、還元条件下でSDS−PAGEによって確認し、その後、抗His G HRPコンジュゲート(Invitrogen、グランドアイランド、ニューヨーク州)または抗ヒトFc抗体(Sigma−Aldrich、セントルイス、ミズーリ州)でブロットした。
野生型Fcドメインおよび操作されたN−グリコシル化Fc変異体の精製のために、馴化培地を、リン酸緩衝溶液(PBS)(1mMのKH2PO4、10mMのNa2HPO4、137mMのNaCl、および2.7mMのKCl、pH7.2)で事前に平衡化したHiTrapキレート化カラム(GE healthcare、ピスカタウェイ、ニュージャージー州)にロードした。非特異的結合タンパク質を、緩衝液A(1mMのKH2PO4、10mMのNa2HPO4、137mMのNaCl、および2.7mMのKCl、10mMのイミダゾール、pH7.6)で洗浄除去し、タンパク質を、緩衝液Aから緩衝液B(1mMのKH2PO4、10mMのNa2HPO4、137mMのNaCl、および2.7mMのKCl、250mMのイミダゾール、pH7.6)への線形勾配で溶出した。プールした画分を、PBS緩衝液で事前に平衡化したHiTrapプロテインAカラム(GE healthcare、ピスカタウェイ、ニュージャージー州)にロードし、プロテインA溶出緩衝液(50mMのクエン酸、pH3.3)で溶出した。得られたタンパク質溶液を、1Mのtris−HCl溶液(pH8.0)によって中和し、緩衝液をPBSに交換し、濃縮し、−80℃で保存した。純度を、SDS−PAGE(4〜20%線形勾配ゲル、Invitrogen、グランドアイランド、ニューヨーク州)によって確認した。Fab−monoFc変異体のために、馴化培地を、PBS緩衝液で事前に平衡化したHiTrapプロテインGカラム(GE healthcare、ピスカタウェイ、ニュージャージー州)にロードし、プロテインG溶出緩衝液(100mMのクエン酸、pH2.5)で溶出した。プールした画分を、1Mのtris−HCl溶液(pH8.0)で中和し、10mLまで濃縮し、PBS緩衝液で事前に平衡化したSuperdex200カラム(Hiload 26/60 prep grade、GE healthcare、ピスカタウェイ、ニュージャージー州)にロードした。純度を、SDS−PAGEによって確認し、N−グリコシル化タンパク質の単量体状態を、10×300mmのカラムを用いて、Superdex 200 10/300 GL(GE Healthcare、ピスカタウェイ、ニュージャージー州)によって評価した。精製タンパク質溶液を、0.22umのフィルターによって滅菌し、−80℃で保存した。タンパク質の定量を、280nmでの吸光度を測定すること、およびPaceら、Protein Sci.4、2411〜2423(1995)に従ったモル吸光係数を用いて濃度を計算することによって達成した。
サイズ排除クロマトグラフィー多角度光散乱検出器(SEC−MALS):野生型Fcドメインおよび操作されたN−グリコシル化Fc変異体の平均モル質量およびオリゴマー化状態を、SEC−MALSを用いて決定した。タンパク質試料を、PBS緩衝液中で4.5〜7.0mg/mlの範囲の濃度で調製した。各試料(200μg)を、Agilent 1100 HPLC系(フォスターシティー、カリフォルニア州)に接続した分析用Superdex 200 10/300 GLカラム(GE Healthcare、ピスカタウェイ、ニュージャージー州)に注入した。サイズ決定カラム上で分解されたタンパク質ピークを、HPLC系に一列に接続されたWyatt’s MiniDawn 3角度光散乱検出器およびOptilab−REX屈折計(サンタバーバラ、カリフォルニア州)を用いて分析した。クロマトグラフィー分析および光散乱分析は、25℃で行った。MiniDawn光散乱系を、製造者の指示に従ってトルエンで校正し、ウシ血清アルブミン(Thermo Scientific、ロックフォード、イリノイ州)を用いて正規化した。データの収集および分析は、タンパク質についてのΔn/Δc値が0.185ml/gであるWyatt’s Astraソフトウェアを用いて行った。グリカン質量の寄与を、糖部分についての近似Δn/Δc値0.14ml/gを用いて、Astraソフトウェアにおいてタンパク質コンジュゲーション鋳型を適用することによって決定した。
FcRn結合アッセイを、表面プラズモン共鳴(SPR)バイオセンサー、Biacore 3000(Biacore、ウプサラ、スウェーデン)を用いて行った。センサーチップCM5、界面活性剤P20、N−エチル−N−(3−ジメチルアミノプロピル)カルボジイミド(EDC)、N−ヒドロキシスクシンイミド(NHS)、および1Mのエタノールアミン(pH8.5)を、GE Healthcare(ピスカタウェイ、ニュージャージー州)から購入した。SPR実験を、25℃で、0.005%のP20を有するPBS緩衝液(pH6.0)において行った。マウスFcRnタンパク質を、ARVYS Proteins,Inc.(スタンフォード、コネティカット州)から購入した。全ての実験は、3回反復した。
タンパク質を、monoFcの結晶化試験のために(Fc−CH23−N364/N407)、tris緩衝液(25mMのtris−HCl、150mMのNaCl、pH7.5)中に30mg/mlまで濃縮した。結晶化は、貯蔵器溶液に対して平衡化された0.2μLのタンパク質溶液および0.2μLの貯蔵器溶液を含有する滴剤を用いて、ハンギングドロップ蒸気拡散方法を18℃で用いて行った。大きな三方晶系結晶が、2.2Mの硫酸アンモニウムおよび200mMのフッ化ナトリウムを沈殿剤として用いて得られた。結晶を、母液中の20%グリセロールの存在下で凍結防止処理し、液体窒素中で即座にフラッシュ冷却した。X線回折データを、SER−CATビームライン22−ID、Advanced Photon Source(APS)、アルゴンヌ、イリノイ州で、最大1.9Åの分解能まで、単結晶から回収した。データにインデックスを付け、積分し、HKL2000でスケーリングした(統計を表2に示す)。結晶は、格子定数がa=b=64.22Åでありc=146.94Åである空間群P312に属していた。構造を、突然変異したADCC増強ヒトFcドメイン(PDB ID:2QL1)(Oganesyanら、Mol.Immunol.45、1872〜1882(2008))の結晶構造をサーチモデルとして用いて、PHASERでの分子置換によって解明した。monoFc単量体を位置決定した後、最初のモデルをBUSTERでの最小化にかけ、COOT(分子グラフィックスプログラム)を用いてさらに再構成した。再校正を伴う数回の精密化改変によって、0.25のRcrystおよび0.259のRfreeに対応する最終精密化モデルが得られた(精密化統計を表6に示す)。
動物研究 − 雄Balb/cマウス(約8週齢の雄)を、Charles River(ウィルミントン、マサチューセッツ州)から購入した。全ての研究は、National Institutes of Healthの動物資源のケアおよび使用についてのガイドに従って行った。1群当たり6頭のマウスに、単回用量のFab−monoFc変異体を静脈内経路を介して投与した。5mg/kgの投与用量は、最近の予定体重に基づくものであった。試験試料をPBS中に調製し、投薬容積は4mL/kgであった。投薬の0分、10分、6時間、24時間、2日、3日、4日、7日、14日、および21日後、10μLの血液試料を毛細管を介して尾静脈から回収し、90μlのRexxip A緩衝液(Gyros AB、ウプサラ、スウェーデン)中に即座に希釈した。試料を3000×gで10分間、4℃で遠心分離し、上清を別の試験管に移し、今後の分析のために−80℃で凍結した。
