Description
50w/v%に希釈した蜂蜜を菌培養培地に添加し、形成する阻止円の大きさで抗菌活性を比較するディスク拡散法を用いて、各種蜂蜜の黄色ブドウ球菌(Staphylococcus aureus subsp. aureus(NBRC13276))に対する抗菌活性をフェノール水溶液濃度に換算して算出する方法にて確認した。具体的には、固化した黄色ブドウ球菌含有ミュラーヒントンII寒天培地の中心部に8mm径の孔を開け、その孔に濃度50%の検体(蜂蜜)を播種し、37℃にて18時間培養した。培養後、阻止円の直径を測定した。検体としてフェノールを用いた場合の、フェノール濃度と阻止円直径の相関より、検体(蜂蜜)のフェノール濃度換算値(質量%)を算出した。蜂蜜としては、Quercus hartwissianaを蜜源とする蜂蜜(アレキサンダー株式会社より入手)(実施例1)、蜂蜜1kg中のメチルグリオキサール濃度がそれぞれ302.3mg/kg(商品名マヌカハニーUMF10+、コンビタジャパン株式会社)、458.5mg/kg(商品名マヌカ蜜、双日食料株式会社)、790.9mg/kg(商品名ストロングマヌカハニー、株式会社TCN)であるマヌカ蜂蜜(比較例1〜3)を用いた。各蜂蜜中のメチルグリオキサール濃度は、0.05%(w/v)o−フェニレンジアミン水溶液を溶媒としてHPLCを用いて測定した。結果を表1に示す。フェノール濃度換算値は、数値が大きいほど抗菌活性が高いことを意味する。
最小発育阻止濃度(MIC)法にて、大腸菌(Escherichia coli(ATCC 10536))、薬剤耐性(ペニシリン耐性)黄色ブドウ球菌(Staphylococcus aureus(ATCC 6538P))、緑膿菌(Pseudomonas aeruginosa(ATCC 43504))に対する各種蜂蜜の抗菌活性を確認した。具体的には、検体(蜂蜜)の濃度が53〜900mg/mLになるようにCAMHB培地にて希釈し、各種菌液を播種した。36℃にて20時間培養後、各種菌の発育状態を確認し、最少発育阻止濃度を評価した。結果を表1に示す。最小発育阻止濃度は、数値が低いほど抗菌活性が高いことを意味する。
Claims (7)
- クエルクス・ハルトウィッシアナ(Quercus hartwissiana)を蜜源とする蜂蜜を有効成分として含み、薬剤耐性黄色ブドウ球菌に対する抗菌活性を有し、かつ緑膿菌及び大腸菌からなる群より選ばれる少なくとも1種に対する抗菌活性を有する、抗菌剤。
- 黄色ブドウ球菌、薬剤耐性黄色ブドウ球菌、緑膿菌及び大腸菌に対する抗菌活性を有する、請求項1に記載の抗菌剤。
- 前記クエルクス・ハルトウィッシアナを蜜源とする蜂蜜が、クエルクス・ハルトウィッシアナの花蜜、樹液又はこれらの組合せを蜜源とする蜂蜜である、請求項1又は2に記載の抗菌剤。
- 薬剤耐性黄色ブドウ球菌に対する最小発育阻止濃度が120mg/ml以下である、請求項1〜3のいずれか一項に記載の抗菌剤。
- 緑膿菌に対する最小発育阻止濃度が180mg/ml以下である、請求項1〜4のいずれか一項に記載の抗菌剤。
- 大腸菌に対する最小発育阻止濃度が180mg/ml以下である、請求項1〜5のいずれか一項に記載の抗菌剤。
- 黄色ブドウ球菌に対する抗菌活性を有し、当該抗菌活性が、フェノール濃度換算値として4.0質量%以上である、請求項1〜6のいずれか一項に記載の抗菌剤。
