Description
− 対象物またはサンプルを発光共役オリゴチオフェンに接触させるステップと、
− 発光共役オリゴチオフェンの少なくとも1つの検出シグナルを検出するステップと、
− 前記検出された検出シグナルに基づいて、前記対象物上または前記サンプル中の1種または複数種の炭水化物の存在、正体、および/または量を決定するステップと、
を含む、1種以上の炭水化物の検出、同定、および/または定量のための方法が提供される。
共役オリゴチオフェンは、硫黄ヘテロ環チオフェンのオリゴ体化から生じる。電子は、その共役骨格に沿って非局在化され、このオリゴ体に伝導性および/または光学的性質を付与する。共役オリゴチオフェンは、ドーピングにより電子を共役π軌道に追加したりまたはそれから除去したりしたときに伝導性になりうる。標的へのLCOプローブの結合は、静電相互作用により駆動される。また、このオリゴ体と標的分子との相互作用は、その骨格構造の捩れを引き起こして、電子歪みおよびその光学的性質の劇的変化をもたらしうる。したがって、オリゴチオフェンは、対応する特異なオリゴ体骨格関連シグナルにより個別に同定可能な広範にわたる結合標的を有する。
本発明は、以下のステップ、すなわち、
− 対象物またはサンプルを発光共役オリゴチオフェンに接触させるステップと、
− 発光共役オリゴチオフェンの少なくとも1つの検出シグナルを検出するステップと、
− 前記検出された検出シグナルに基づいて、前記対象物上または前記サンプル中の1種または複数種の炭水化物の存在、正体、および/または量を決定するステップと、
を含む、1種以上の炭水化物の検出、同定、および/または定量のための方法を提供する。
炭水化物は、多くの領域内で有用であり、たとえば、薬剤、栄養補助剤、調味剤、および甘味剤、さらには材料として、種々の用途で利用される。したがって、LCOは、そのような用途の炭水化物化合物の製造および品質評価のためのインジケーターとして有用である。炭水化物の検出、同定、および定量におけるLCOの使用は、主に、医薬品産業内および食品産業内で、さらには研究で、行われるであろう。
研究目標:
共焦点分析を用いてプロトタイプLCO h−FTAAがバイオフィルム中の炭水化物成分すなわちセルロースを検出可能であることを実証すること。
I.伝統的コンゴーレッドプレートアッセイを用いた既知のバイオフィルムモルフォロジーの確認
カーリーおよび/またはセルロースの産生に関連付けられるバイオフィルムモルフォロジーを検証するために、コンゴーレッド(40μg/mL)およびクーマシーブリリアントブルーG−250(20μg/mL)が追加されたLB寒天プレート(塩を用いない)上で、S.エンテリティディス(S.enteritidis)wt株3934ならびにアイソジェニック突然変異株ΔbscA(カーリー+、セルロース−)、ΔcsgA(カーリー−、セルロース+)、およびΔcsgD(カーリー−、セルロース−)を培養した。プレートを28℃で48時間培養した。
ガラスカバースリップを6ウェルプレートのウェルに導入し(図2Aに示される構成による)、バイオフィルム形成のための表面を提供した。これは、実験の終了時、顕微鏡分析のために容易に取出し可能であった。バイオフィルム実験の準備をするために、S.エンテリティディス(S.enteritidis)wt株3934ならびにアイソジェニック突然変異株ΔbscA、ΔcsgA、およびΔcsgDの個別の培養物をフラスコ内のLB培地中で一晩増殖させた。各培養物を新しいLB中に100倍希釈し、OD600=0.6になるまで振盪インキュベーター(230rpm)中37℃で培養した。塩を用いないLB中に培養物を105CFU/mlの培養密度に希釈し、8mlのアリコートでカバースリップ含有6ウェルプレート中に分配した。プレートを28℃で48時間培養した後、ガラスカバースリップを取り出し、PBSで2回洗浄し、その後、4mlの4%ホルムアルデヒド中で1時間固定した。固定サンプルを2回洗浄し、次いで、それぞれh−FTAA(2μg/mL)、h−HTAA(2μg/mL)、およびPBSの溶液中に暗所で30分間浸漬した。PBSは、陰性対照として機能し、自己蛍光レベルの評価に使用された。次いで、処理されたスライドをPBSで2回洗浄し、蛍光に基づく共焦点レーザー走査型顕微鏡法分析のためにVectashield(登録商標)を用いて封入した。特定的には、液体空気界面に形成されたバイオフィルムのエッジを適切な蛍光フィルターを用いて可視化した。図2C〜Dでは、各スライドは、励起されたときにバイオフィルム結合LCOにより生成された蛍光を示す画像により表される(左側)。右側は、LCO蛍光との位相差による細菌存在の光学検査のオーバーレイである。位相差は、表面上へのバイオフィルム付着の目視確認のための伝統的方法である。
株はすべて、コンゴーレッドアッセイで予想された形態型を示した(図2B)。各株の形態型の特性は、カーリーおよびセルロースの産生に関してそれらのバイオフィルム成形能に関連付けられ、この関係は、これまでに他のものにより報告されてきた。
LCOプローブは、バイオフィルムに結合し、蛍光分析下でその可視化を可能にした。それに加えて、それらは、異なる細菌表現型に由来するバイオフィルムモルフォロジーの識別を可能にした。LCOプローブh−FTAAは、セルロースを発現するがカーリーを発現しない株ΔcsgAの蛍光検出シグナルを生成することが示されたことから、h−FTAAは、カーリー以外の他の成分おそらくセルロースを検出可能であることが示唆される。
各バイオフィルムでLCOが有する特異なスペクトルシグネチャーを示すことにより、異なるカーリー含有量およびセルロース含有量を含むバイオフィルムモルフォロジーを識別するLCOの能力を示すこと。
I.96ウェルプレート中でのバイオフィルム増殖
臨床由来S.エンテリティディス(S.enteritidis)wt株3934ならびにアイソジェニック突然変異株(ΔbscA、ΔcsgA、およびΔcsgD)の新しい一晩培養物を新しいLB中に接種し、OD600=0.6になるまで37℃で培養した。それぞれの各培養物をLB(塩を用いない)で105CFU/mlの細胞密度に希釈した後、3つの個別のフラスコ内に分注した。h−FTAA(2μg/mL)およびh−HTAA(2μg/mL)をそれぞれフラスコの2つに添加し、一方、対照として使用されたPBSを第3のフラスコに添加した。次いで、50μlの各培養物を96ウェルプレートの個別のウェル内にトリプリケート方式で接種し、28℃で48時間インキュベートした。
インキュベーターからバイオフィルム培養物を取り出した後、処理ステップを実施せずにLCOシグナルの検出を行った。Synergy Mxモノクロメーター式マルチモードマイクロプレートリーダーを用いてプレートを読み取った。300〜500nmでサンプルを励起して545nmで発光を検出することにより、LCOの励起スペクトルを収集した。サンプルを405nmで励起したとき、500〜700nmで発光シグナルを読み取ることにより、カーリー結合LCOに対する発光スペクトルを収集した。サンプルを500nmで励起したとき、520〜700nmで発光シグナルを読み取ることにより、セルロース結合LCOに対する発光スペクトルを収集した。
h−HTAAのスペクトルプロファイルは、異なるアイソジェニック突然変異株により形成されたバイオフィルムモルフォロジーを識別しなかった(図3A)。h−HTAAの励起スペクトルパターンは、すべてのアイソジェニック株にわたり同一であった。一方、h−FTAAは、各株で励起ピークおよびショルダーの識別可能なスペクトルパターンを生成した(図3B)。h−FTAAをカーリーの存在下で380nmで励起したとき(wtおよびΔbscA)、特異な発光スパイク(励起ショルダー)が検出される。h−FTAAをカーリーの不在下で380nmで励起したとき(ΔcsgAΔcsgD)、その代わりに約355nmの発光スパイク(励起ショルダー)が検出される。最後に、h−FTAAをセルロースの存在下で480nmで励起したとき(wtおよびΔcsgA)、特異なピーク発光が検出される。
LCO h−FTAAは、各バイオフィルムモルフォロジーに特異なスペクトルシグネチャーを生成する。h−FTAAは、ECM成分のカーリーおよびセルロースとの相互作用を介して生成される特異なスペクトルプロファイルに基づいてバイオフィルムを識別する。本方法は、生の培養物からのバイオフィルムの物理的分離を必要としない。LCO h−HTAAは、洗浄ステップを用いることなく異なる細菌表現型により形成された異なるバイオフィルム量を識別することができなかった。h−HTAAは、クリスタルバイオレットと類似の洗浄ステップを用いてバイオフィルム検出のための一般的なプローブとしてより良好に適用されうる。しかしながら、h−HTAAは、実験全体を通じて増殖培地中に存在しうる非殺細菌プローブであるという利点を有する。
研究目標:
セルロースを標的としてスペクトルシグネチャーを与えるLCO h−FTAAの能力を検証すること、およびセルロースの定量におけるその有用性を検証すること。
6.25mg/mlから0.0488までの純粋不溶性微結晶セルロース懸濁液の二倍段階希釈液を調製し、これにh−FTAAを3μMの濃度になるように添加した。100μlアリコートを96ウェルプレート内に分配した。Synergy Mxモノクロメーター式マルチモードマイクロプレートリーダーを用いてプレートを読み取った。300〜500nmでサンプルを励起して545nmで発光を検出することにより、LCOの励起スペクトルを収集した。ここに示されるセルロース濃度は、6.25mg/ml、3.125mg/ml、1.5625mg/ml、および0.78125mg/mlである。
純粋セルロースの存在下で、約480nmでh−FTAAを励起すると、545nmで検出される発光ピークを与える(図4)。このシグナル強度は、セルロース濃度と比例して増加したことから、h−FTAAが実際にセルロースに結合することが検証される。
七量体LCO h−FTAAは、炭水化物セルロースの検出および定量に使用可能である。
研究目標:
バイオフィルム形成のリアルタイム追跡でh−FTAAの使用を実証して、培養物増殖との関連で時間に対するバイオフィルム相関RFU(相対蛍光単位)の変化を示すこと。
OD600での吸光度により測定される細菌培養物の濁度を用いて、培養物密度を規定する。GFPの発現は、細菌増殖および培養密度の直接表現を提供する。蛍光検出による培養密度のより直接的表現のためにgfp遺伝子を有するプラスミドP2777を用いて、S.エンテリティディス(S.enteritidis)3934wtならびにアイソジェニック突然変異株ΔbscA、ΔcsgA、およびΔcsgDをトランスフォームした。各株の新しい一晩培養物を新しいLB中に接種して、OD600=0.6になるように37℃で培養した。培養物をLB(塩を用いない)で105CFU/mlの細胞密度に希釈した後、2つの個別のフラスコ内に分注した。h−FTAA(2μg/mL)を1つのフラスコに添加し、一方、対照として使用されたPBSを第2のフラスコに添加した。50μlの各培養物混合物を4つの96ウェルプレート上にトリプリケート方式で接種して28℃で培養した。フルオロフォアの漂白を回避すべく4時間間隔で各プレートのスキャンを行うようにして、GFP発現さらにはカーリー結合h−FTAAシグナル(励起405nm、発光556nm)およびセルロース結合h−FTAAシグナル(励起500nm、発光600nm)に関して、48時間にわたり1時間ごとにタンデムにプレートを読み取った。4つのプレートからのGFPおよびh−FTAAのRFUデータを単一のプロットで組み合わせて、時間に対してシグナルの1時間ごとの変化を可視化した。
バイオフィルム培養でOD600の吸光度に対してプロットされたGFPシグナルの増加傾向の比較は、密接な相関を示した(図5A)。GFPシグナルに対する吸光度のプロットが0.9423のR2値を与えたことから、2つのシグナルの密接な相関が示唆される(図5B)。このことから、GFP発現は、細菌増殖をモニターする手段として使用可能であることが示唆された。
LCOプローブh−FTAAは、バイオフィルム形成のリアルタイム分析を可能にする。セルロースおよびカーリーに特異的なシグナルは、培養進行中、1時間おきに検出可能であった。経時的なシグナルの増加は、形成されるバイオフィルム量の増加を反映する。培養増殖(GFPシグナルにより表される)とh−FTAAからのバイオフィルムシグナルとの比較から、バイオフィルム形成は、培養物が定常期に達したときに行われることが示唆され、これまでの研究と一致する。カーリーおよびセルロースの検出のために選択されるチャンネルは、GFP蛍光からの有意なノイズを受けない。カーリー/セルロース結合h−FTAAの広い発光スペクトルは、GFPチャネル中にブリードする。しかしながら、これは、分析に影響を及ぼさない。
最大発光の励起波長(λmax)の変化およびシグナル強度と炭水化物濃度との相関を研究することにより、β−1,3−グルカン、セルロース、キチン、ナトリウムアルギネート、α−D−グルコース、セルロビオース、アミロース、グリコーゲン、ヘパリン、およびコンドロイチン硫酸Aを識別する際の五量体LCOの使用を実証すること。
炭水化物懸濁液の段階希釈液をdH2O中に調製した。β−1,3−グルカン、セルロース、およびキチンに対しては、10〜0.039mg/mlの範囲内の濃度を使用し、一方、ナトリウムアルギネート、グルコース、アミロース、グリコーゲン、セルロビオース、ヘパリン、コンドロイチン硫酸Aに対しては、5〜0.019mg/mlを使用した。3μMの各LCOプローブを各濃度の炭水化物の1mlアリコートに添加し、その後、100μlを96ウェルプレート中にトリプリケート方式で分配した。300〜500nmでサンプルを励起して545nmで発光を検出することにより、励起スペクトルを収集した。陰性対照に対してλmaxおよび/またはRFU強度の再現性のある変化が存在する場合、炭水化物の検出が結論付けられる。それぞれの図に明記されるように、3つの濃度さらには陰性対照からのデータが各炭水化物に対して示される。

- 請求項2または3に記載の方法において、前記発光共役オリゴチオフェンが1つ以上の官能性側鎖を含むことを特徴とする方法。
- 請求項4に記載の方法において、前記官能性側鎖が、アミノ酸、アミノ酸誘導体、神経伝達物質、単糖、多糖、核酸、およびそれらの誘導体さらには組合せから選択されることを特徴とする方法。
- 請求項1乃至5の何れか1項に記載の方法において、前記七量体の発光共役オリゴチオフェンが、h−FTAAまたはh−HTAAであり、かつ前記五量体の発光共役オリゴチオフェンが、pHTA−His、pHTA−Lys、pHTEA、pHTIm、pHTA−Tyr、pHTA−Arg、pHTA−Asp、およびpHTA−Gluの何れかであることを特徴とする方法。
- 請求項1乃至6の何れか1項に記載の方法において、前記検出シグナルが蛍光シグナルや比色シグナルなどの光シグナルまたは伝導率などの電気シグナルであることを特徴とする方法。
- 請求項1乃至7の何れか1項に記載の方法において、前記発光共役オリゴチオフェンが少なくとも2種の異なる炭水化物を識別可能であることを特徴とする方法。
- 請求項1乃至8の何れか1項に記載の方法において、前記炭水化物の少なくとも1種が不溶性炭水化物であることを特徴とする方法。
- 請求項9に記載の方法において、前記不溶性炭水化物が、セルロース、キチン、β−グルカン、アルギネート、アミロース、およびグリコーゲン、またはそれらの組合せの何れかであることを特徴とする方法。
- 請求項1乃至9の何れか1項に記載の方法において、前記炭水化物の少なくとも1種が可溶性炭水化物であることを特徴とする方法。
- 請求項11に記載の方法において、前記可溶性炭水化物が、グルコース、セルロビオース、ヘパリン、コンドロイチン硫酸A、またはそれらの組合せの何れかであることを特徴とする方法。
- 請求項1乃至12の何れか1項に記載の方法において、前記炭水化物の少なくとも1種が、構造炭水化物、貯蔵炭水化物、グリコアミノグリカン、炭水化物変換の中間生成物、および/または代謝基質であることを特徴とする方法。
- 請求項1乃至13の何れか1項に記載の方法において、少なくともステップa)および/またはステップb)が、in vivo、in vitro、またはin situで行われることを特徴とする方法。
- 1種以上の炭水化物の検出、同定、および/または定量のためであることを特徴とする発光共役オリゴチオフェンの使用。
