Description
本出願は、2012年5月3日に出願された米国仮出願第61/642,441の優先権を主張するものであり、該仮出願は、引用により本明細書に組み込まれる。
本開示は一般的に、口腔、歯、及び歯肉の疾患、並びに他の有害な口腔の疾病に関する障害の処置と関係する、薬物療法の分野に関係する。より具体的に、本発明は、歯の疾患に関連するものを含む、様々な有害な口腔の疾病の予防及び処置のための製剤に属する。本発明は、歯科、老人医学、免疫障害、腫瘍学、及び美容医学(cosmeceuticals)を含む様々な分野で適用可能な、口腔の健康状態及び口腔の組織の表面的な外観の改善における製剤の使用に属する。
キレート剤:キレート化は、金属が環の一部である複合体中の金属との化合である。有機リガンドは、キレート化剤又はキレート剤と呼ばれ、キレートは金属複合体である。金属原子への閉環が多くなるほど、化合物はより安定する。キレートの安定性もキレート環における原子の数に関係する。H2O又はNH3のような1つの同位原子を有する単座リガンドは、他の化学プロセスによって容易に粉砕され、一方でしかし、多座のキレート化剤は、金属イオンへの複数の結合に寄与し、より安定した複合体を提供する。クロロフィル(緑色植物の色素)は、4つの複合キレート剤(ピロール環)と結合された中央のマグネシウム原子から成るキレートである。ヘムは、ポルフィリンの中心において鉄(II)イオンを含む鉄キレートである。キレート剤は、水溶液系において金属イオンを制御するために広範囲の金属イオン封鎖剤を提供する。多価性の金属イオンを備える安定した水溶性複合体の形成によって、キレート剤は、金属イオンの標準の反応性の遮断によって、望まれない相互作用を予防する。EDTA(エチレンジアミンテトラアセテート)は、チッ素原子及び短鎖カルボン酸基を有する一般のキレート剤の優れた例である。
典型的なアリール基は、1つの芳香環又は2つの融合した或いは連結した芳香環(例えば、フェニル、ナフチル、ビフェニル、ジフェニルエーテル、ジフェニルアミン、ベンゾフェノンなど)を含む。「アリール」は非置換及び置換のアリールを含み、ここで、置換基は、任意に置換した「アルキル」基に関して上述される通りのものであり得る。用語「アラルキル」はアリール置換基を備えたアルキル基を指し、ここで、「アリール」及び「アルキル」は上に定義された通りである。好ましいアラルキル基は6〜14の炭素原子を含み、特に好ましいアラルキル基は6〜8の炭素原子を含む。アラルキル基の例は、限定されないが、ベンジル、2−フェニル−エチル、3−フェニル−プロピル、4−フェニル−ブチル、5−フェニル−ペンチル、4−フェニルシクロヘキシル、4−ベンジルシクロヘキシル、4−フェニルシクロヘキシルメチル、4−ベンジルシクロヘキシルメチルなどを含む。用語「アシル」は、式−(CO)−アルキル、−(CO)−アリール、又は−(CO)−アラルキルを有する置換基を指し、ここで「アルキル」、「アリール」、及び「アラルキル」は上に定義された通りである。用語「ヘテロアルキル」及び「ヘテロアラルキル」はそれぞれ、ヘテロ原子含有のアルキル基及びアラルキル基(即ち、1以上の炭素原子が炭素以外の原子(例えば、窒素、酸素、硫黄、リン、又はシリコン、典型的には、窒素、酸素、又は硫黄)と置換されるアルキル基及びアラルキル基)を指すために使用される。
歯垢バイオフィルムの形成は、薄膜の初期のコロニー形成から始まり、成熟したバイオフィルムの複合体形成で終わる、一連の工程を含む。歯垢バイオフィルムは、割目、表面平滑、及び歯肉縁罅隙を含む様々な歯表面上に存在するが、それらは、割目と隙間のようなより不活発な部位における、それらの成熟した状態で最も見られる可能性があり、なぜなら、これらの場所が歯ブラシのような除去の力からの保護を提供するためである。加えて、プラークバイオフィルムの成長プロセスを介し、微生物の組成物は、主としてグラム陽性であり且つ連鎖球菌が豊富なものから、そのより成熟した状態におけるグラム陰性嫌気性生物で満たされる構造へと変化する。
様々な手段が、本発明の組成物を処方するために使用され得る。製剤と投与についての技術は、「“Remington: The Science and Practice of Pharmacy,” Twentieth Edition, Lippincott Williams & Wilkins, Philadelphia, PA (1995)」に見出され得る。ヒト又は動物への投与のために、調製は、FDAによって要求されるものに匹敵する、無菌性、発熱性、一般的安全性、及び純度の基準を満たさねばならない。医薬製剤の投与は、本明細書に記載されるように、様々な方法で行なわれ得る。
製剤を含むローション剤を、Sigmaから購入したEDTA(四ナトリウム塩)及びMSMを使用して調製した。全ての実施例は、2.6%のEDTA及び5.4%のMSMを含む製剤Aを使用した。
Claims (20)
- 有害な口腔疾病を処置するための製剤であって、該製剤は、
少なくとも0.1重量%の濃度の負に荷電した生体適合性のキレート剤;
荷電マスキングする浸透エンハンサーとしての効果的な濃度のメチルスルホニルメタン(MSM);及び、
水膨潤可能なポリマーを含有する薬学的に許容可能な担体を含み、
該製剤は、ヒドロゲルを含み、該製剤を、口腔内の金属イオンレベルを低下させるのに十分な量および時間で、口腔に接触させることにより、有害な口腔疾病を処置するものであることを特徴とする、製剤。 - 製剤が投与後にインサイツでヒドロゲルを形成するように、ヒドロゲルは熱可逆性ヒドロゲル形成ポリマーである、ことを特徴とする請求項1に記載の製剤。
- 請求項1に記載の製剤のリポソーム分散液を含む、経口送達システム。
- 生体適合性のキレート剤は、エチレンジアミン四酢酸(EDTA)、シクロヘキサンジアミン四酢酸(CDTA)、ヒドロキシエチルエチレンジアミン三酢酸(HEDTA)、ジエチレントリアミンペンタ酢酸(DTPA)、ジメルカプトプロパンスルホン酸(DMPS)、ジメルカプトコハク酸(DMSA)、アミノトリメチレンホスホン酸(ATPA)、クエン酸、クルクミン、キレート抗生物質、2以上のキレート窒素原子を含有するキレート剤、リン酸塩、デフェロキサミン、及びそれらの許容可能な塩、及び前述の何れかの組み合わせから選択される、ことを特徴とする請求項1に記載の製剤。
- 許容可能なEDTA塩は、二アンモニウムEDTA、二ナトリウムEDTA、二カリウムEDTA、三アンモニウムEDTA、三ナトリウムEDTA、四ナトリウムEDTA、三カリウムEDTA、カルシウム二ナトリウムEDTA、及びそれらの組み合わせから選択される、ことを特徴とする請求項4に記載の製剤。
- キレート剤は、クロロキン及びテトラサイクリンから選択されるキレート抗生物質である、ことを特徴とする請求項4に記載の製剤。
- キレート剤は、ピロリン酸塩、トリポリリン酸塩、ヘキサメタリン酸塩、及びそれらの組み合わせから選択される、ことを特徴とする請求項4に記載の製剤。
- 6.8乃至7.8の範囲のpHを有する、請求項1に記載の製剤。
- 生体適合性のキレート剤は、製剤の0.1乃至15重量%を示す、ことを特徴とする請求項4に記載の製剤。
- メチルスルホニルメタンは、製剤の0.1重量%乃至40重量%を示す、ことを特徴とする請求項8に記載の製剤。
- 口腔への製剤の送達のための無菌の挿入物であって、該挿入物は:
請求項1に記載の製剤を収容し、且つ口腔の任意の部分への移植に適切な制御放出インプラント
を含む、無菌の挿入物。 - インプラントは、拡散及び/又はマトリックス劣化を介して口腔組織に製剤を徐々に放出するポリマーマトリックスで構成される、ことを特徴とする請求項11に記載の無菌の挿入物。
- ポリマーマトリックスは完全に可溶性及び/又は完全に生物分解性である、ことを特徴とする請求項12に記載の無菌の挿入物。
- インプラントは積層構造で構成され、該構造内で、製剤を収容する内部コアは、製剤が徐々に分散する浸透可能なポリマーの外層の間に含まれる、ことを特徴とする請求項11に記載の無菌の挿入物。
- インプラントは、製剤を収容し、且つ、製剤が徐々に拡散するポリマー膜に含まれるリザーバーを含む、ことを特徴とする請求項11に記載の無菌の挿入物。
- インプラントは、口腔組織への移植後、システム内の浸透圧の増加の結果として製剤が徐々に放出される、浸透圧システムを含む、ことを特徴とする請求項11に記載の無菌の挿入物。
- 有害な口腔疾病は、加齢、歯肉炎、歯周病、ミネラル堆積、汚い歯、歯石、又は酒石の形成、細菌のバイオフィルム又はプラークの形成、歯腔、齲歯、及びAIDSに関連する疾病から選択される、口腔の疾病、疾患、又は障害である、ことを特徴とする請求項1に記載の製剤。
