化合物の生成
6−MSAS遺伝子は、真菌、細菌もしくは植物等の任意の天然源から得ることができるか、または発現された6−MSAS PKSの最適な生合成特徴をもたらす合成、ハイブリッド、または改変された誘導体であり得る。一実施形態では、使用される6−MSAS遺伝子は、狭葉サンタ草(Eriodyction pavonina)等の植物に由来し得る(Khaden S.and Marles,RJ.Molecules15;7985−8005(2010)。酵素活性の改変および最適化は、当該技術分野において周知である。

Claims (17)
- フェノール化合物を生成するための改変された組換え宿主細胞であって、前記細胞から単離される生成物が、フェノール化合物であるように、
i)発現されることができる6−メチルサリチル酸シンターゼ(6−MSAS)又はオルセリン酸シンターゼ(OSAS)をコードする少なくとも1つのヌクレオチド配列を含む第1の発現系と、
ii)前記6−MSAS又はOSASをパンテテイニル化するホロアシルキャリアタンパク質(ACP)シンターゼをコードする少なくとも1つのヌクレオチド配列を含む第2の発現系と、
iii)前記フェノール化合物を形成するための脱カルボキシル化を触媒する官能性デカルボキシラーゼをコードする少なくとも1つのヌクレオチド配列を含む第3の発現系と、
を含み、
6−MSAS又はOSASの第1の発現系およびACPシンターゼの第2の発現系は、単一のベクターまたは別個のベクター上に存在し、官能性デカルボキシラーゼの第3の発現系は、6−MSAS又はOSAS、またはホロACPシンターゼ、または双方と同一のベクター上に存在するか、または別個のベクター上に存在するか、または;
第1、第2、および第3の発現系は、宿主細胞染色体内に組み込まれるか、または酵母人工染色体(YAC)から発現される、
改変された組換え宿主細胞。 - 前記フェノール化合物が、メタ−クレゾールまたはオルシノールである、請求項1に記載の改変された細胞。
- 前記芳香族6−MSASが、真菌または原核生物由来である、請求項1に記載の改変された細胞。
- 前記デカルボキシラーゼをコードする前記ヌクレオチド配列が、プリムネシウム・パツルム(P.Patulum)由来の6−MSAデカルボキシラーゼ遺伝子、アスペルギルス・クラバタス由来のPatG遺伝子、グリオクラジウム・ロゼウム由来のOSAデカルボキシラーゼ遺伝子、アスペルギルス種由来の2,3−ジヒドロキシ安息香酸デカルボキシラーゼ遺伝子、およびスフィンゴモナス・パウシモビリスSYK−6由来の5−カルボキシバニリン酸デカルボキシラーゼ遺伝子からなる群から選択される、請求項1〜3のいずれか1項に記載の改変された細胞。
- 前記6−MSAS又はOSASおよび前記ホロACPシンターゼが、2シストロン性メッセンジャーRNAから発現される、請求項1に記載の改変された細胞。
- 前記宿主生物が、サッカロマイセス・セレヴィシエ、ピキア・パストリスおよびアスペルギルス種からなる群から選択される、請求項1に記載の改変された細胞。
- フェノール化合物を生成する方法であって、
a)改変された組換え宿主細胞であって、
i)発現されることができる6−メチルサリチル酸シンターゼ(6−MSAS)又はオルセリン酸シンターゼ(OSAS)をコードする少なくとも1つのヌクレオチド配列を含む第1の発現系と、
ii)前記6−MSAS又はOSASをパンテテイニル化するホロアシルキャリアタンパク質(ACP)シンターゼをコードする少なくとも1つのヌクレオチド配列を含む第2の発現系と、
iii)官能性デカルボキシラーゼをコードする少なくとも1つのヌクレオチド配列を含む第3の発現系と、を含む、改変された組換え宿主細胞を提供することと、
b)前記組換え宿主細胞において第1のフェノール化合物を生成することと、
c)前記第1のフェノール化合物から第2のフェノール化合物を合成することであって、組換えデカルボキシラーゼが、前記第1のフェノール化合物の脱カルボキシル化を触媒して、前記第2のフェノール化合物を形成することと、
を含み、
6−MSAS又はOSASの第1の発現系およびACPシンターゼの第2の発現系は、単一のベクターまたは別個のベクター上に存在し、官能性デカルボキシラーゼの第3の発現系は、6−MSAS又はOSAS、またはホロACPシンターゼ、または双方と同一のベクター上に存在するか、または別個のベクター上に存在するか、または;
第1、第2、および第3の発現系は、宿主細胞染色体内に組み込まれるか、または酵母人工染色体(YAC)から発現される、
方法。 - 前記第1のフェノール化合物が、6−MSAまたはオルセリン酸(OSA)であり、第2のフェノール化合物が、メタクレゾールまたはオルシノールである、請求項7に記載の方法。
- フェノール化合物を生成する方法であって、
a)改変された組換え宿主細胞であって、
i)発現されることができる6−メチルサリチル酸シンターゼ(6−MSAS)又はオルセリン酸シンターゼ(OSAS)をコードする少なくとも1つのヌクレオチド配列を含む第1の発現系と、
ii)前記6−MSAS又はOSASをパンテテイニル化するホロアシルキャリアタンパク質(ACP)シンターゼをコードする少なくとも1つのヌクレオチド配列を含む第2の発現系と、を含む、改変された組換え宿主細胞を提供することと、
b)前記組換え宿主細胞において生成された第1のフェノール化合物を単離することと、
c)金属触媒で加熱し、続いて、蒸留することによって、ステップb)の前記第1のフェノール化合物を脱カルボキシル化して、第2のフェノール化合物を形成することと、
を含む、方法。 - 前記第1のフェノール化合物が、6−MSAであり、前記第2のフェノール化合物が、メタクレゾールである、請求項9に記載の方法。
- 前記金属触媒が、亜鉛触媒である、請求項9または10に記載の方法。
- フェノール化合物がアルキル化フェノール化合物である請求項1に記載の改変された細胞であって、前記細胞から単離される生成物が、アルキル化フェノール化合物であるように、改変された細胞が
(iv)O−メチルトランスフェラーゼ(OMT)をコードする少なくとも1つのヌクレオチド配列を含む第4の発現系を更に含む、改変された細胞。 - 前記アルキル化フェノール化合物が、3−メチルアニソールまたは3,5−ジメトキシトルエンである、請求項12に記載の改変された細胞。
- OMTが、OOMT1、OOMT2、OOMT3、OOMT4、COMT1からなる群から選択されるロサ・キネンシス(R.Chinensis)遺伝子によってコードされる、請求項12に記載の改変された細胞。
- アルキル化フェノール化合物を生成するための、請求項7に記載の方法であって、
d)前記組換え宿主細胞において生成された第2のフェノール化合物を単離することと、
e)アルキル化剤での処理によって、ステップd)の前記第2のフェノール化合物をアルキル化して、前記アルキル化フェノール化合物を形成することと、を更に含む、方法。 - 前記第2のフェノール化合物が、メタクレゾールまたはオルシノールである、請求項15に記載の方法。
- デカルボキシラーゼをコードするヌクレオチド配列は、プリムネシウム・パツルム(P.patulum)由来の6−MSAデカルボキシラーゼ遺伝子、アスペルギルス・クラバタス(Aspergillus clavatus)由来のPatG遺伝子、グリオクラジウム・ロゼウム(Gliocladium roseum)由来のOSAデカルボキシラーゼ遺伝子、アスペルギルス種由来の2,3−ジヒドロキシ安息香酸デカルボキシラーゼ遺伝子、およびスフィンゴモナス・パウシモビリス(Sphingomonas paucimobilis)SYK−6由来の5−カルボキシバニリン酸デカルボキシラーゼ遺伝子からなる群から選択される、請求項7、8、15または16の方法。
