Description
そのため、微生物感染症を予防・治療するために、本分野では、操作が簡単で、結果が正確な、新しい診断・治療技術を開発することによって、このような疾患の実験室診断および臨床治療を改善し、患者の病状の対応の遅れを避け、患者の身体および経済的な負担を軽減することが切望されている。

ここで用いられるように、用語「本発明のmilRNA」、「サルモネラ菌特異的milRNA」とは、sal−1、sal−2、sal−3、sal−4、sal−5のいずれかまたはこれらの組み合わせを指し、入れ替えて使用することができる。
ここで用いられるように、用語「阻害剤」、「拮抗剤」および「遮断剤」は、意味が同様で、入れ替えて使用することができる。
ここで用いられるように、用語「sal−1遮断剤」とは、sal−1の機能を抑制または遮断する物質を指し、たとえばアンチセンス配列や核酸スポンジなどが挙げられる。これらの阻害剤は、sal−1と標的遺伝子iNOSのmRNAの結合を抑制し、そしてsal−1の標的遺伝子の発現に対する下方調節を抑制することができる。
ここで用いられるように、用語「微生物」は、ウイルス、細菌、古細菌、衣原体、原生生物などの様々な微小生物を含む。本発明において、「体内微生物」とは、宿主(ヒトまたはほかの動物)の体内に存在する、感染微生物、寄生微生物、および共生微生物を含む様々な微生物を指す。一種類の典型的な体内微生物は、疾患を引き起こす病原体である。
本発明において、病原体と宿主細胞の関係によって、病原体は、細胞外寄生菌と細胞内寄生菌の2種類に分かれる。細胞外寄生菌とは、宿主細胞外の細胞の隙間、血液、リンパ液、組織液などの体液の中で生長・繁殖する細菌を指す。細胞内寄生菌は、また2種類に分かれる。通性細胞内寄生菌とは、宿主の体内では主に細胞内に寄生して生長・繁殖し、体外では活細胞のない培地でも生長することができる。偏性細胞内寄生菌は、体内でも体外でも、活細胞内でしか生長・繁殖しない。
ここで用いられるように、用語「sRNA」、すなわちsmall non-coding RNAとは、微小の非コードRNA配列を指す。本発明において、sRNAは、microRNAおよびmilRNAを含む。
iNOSとは、誘導型一酸化窒素合成酵素を指す。
非コードRNA(non-coding RNA、ncRNA)とは、タンパク質に翻訳されないRNAを指し、rRNA、tRNA、snRNA、snoRNA、microRNAなどの様々なRNAを含み、これらのRNAはゲノムから転写され、タンパク質に翻訳されないが、タンパク質の翻訳過程に関与し、RNAレベルでそれぞれの生物学的機能を発揮する。たとえば、snRNA、snoRNAは、RNAスプライシングとRNA修飾に関与する。
snoRNAは、最初核小体で発見された小さなRNAで、核小体低分子RNAと呼ばれ、最初発見された生物的機能はrRNAの修飾である。大多数の核小体低分子RNAは、2種類に分かれる。一種類はC/D box snoRNAで、この種類のsnoRNAはRNAの塩基に対してメチル化修飾を行うものである。もう一種類はH/ACA boxで、この種類のsnoRNAはRNAの塩基に対してメチルウラシル化修飾を行うものである。その特徴は、一つの二本鎖双ステムが形成し、途中に一つのループ領域があり、途中のループ領域に一つのbox Hがある。また、尾部のテールに一つのbox ACAがある。box Hおよびbox ACAの一次配列の特徴の線引きが比較的に緩い。
eRNAは、イントロンまたは非コードDNAから転写されたRNA分子で、精確に遺伝子の転写および翻訳の効率を調節する。
シグナル認識粒子RNAとは、細胞質においてシグナルペプチドを含むmRNAを認識し、分泌を決めるRNA機能分子である。
pRNAとはファージのRNAである。たとえば、研究によると、fi29ファージにおいて、6個の同様の小さいRNA分子がATPを利用してDNAのパッケージングに関与する。
tmRNAとは、tRNAおよびmRNAの両方の性質を持つ複合体のRNAである。tmRNAは、幅広く細菌に存在し、翻訳またはコードの解読が間違ったリボソームを認識し、機能を休止したリボソームも認識し、これらの問題のあるリボソームの崩壊を仲介する。
また、mRNAにおける非コード領域は、イントロン領域および5’−UTR、3’−UTRなどのリボソーム認識エレメントを含み、非コードRNAと見なすこともできる。
本発明は、サルモネラ菌由来の新しい種類のmilRNAを提供する。この種類のmilRNAは、miRNAと非常に似ていて、RNA分子で、milRNA前駆体を形成できる転写物から加工して得られる。長さは、通常、18〜28個のヌクレオチド(nt)を有する(特に約20〜26 nt)。miRNAと同様に、milRNAは、通常、ノーザンブロッティングによって検出することができる。
milRNAは、前駆体RNA(Precursor milRNA、Pre-milRNA)から加工して得られ、前記の前駆体RNAはフォルディングして安定したステム・ループ(ヘアピン)構造になり、前記のステム・ループ構造の長さは、通常、50〜100 bpの間である。前記の前駆体milRNAは、フォルディングして安定したステム・ループ構造になり、ステム・ループ構造のステム部の両側は基本的に相補的な二本の配列を含む。前記の前駆体milRNAは、天然のものでも人工合成されたものでもよい。
本発明によって提供されるmilRNA配列に基づき、そのアンチセンスオリゴヌクレオチドを設計し、前記のアンチセンスオリゴヌクレオチドは体内で相応のmilRNAの発現を下方調節する。ここで用いられるように、「アンチセンスオリゴヌクレオチド(antisense-oligonucleotides、AS−OnまたはASO)」は、「アンチセンスヌクレオチド」とも呼ばれ、長さが約18〜28 nt(特に約20〜26 nt)のDNA分子またはRNA分子またはその類似体を指す。
本発明に係るアンチセンスオリゴヌクレオチドを動物(たとえばサルモネラ菌の感染者)の体内に導入すると、関連milRNAの発現を顕著に下方調節する。
本発明によって提供されるmilRNA配列に基づき、導入された後加工されて相応のmRNAの発現に影響するmilRNAになるようなポリヌクレオチド構築物を設計することができ、すなわち、前記ポリヌクレオチド構築物は体内で相応のmilRNAの量を上方調節することができる。そのため、本発明は、単離されたポリヌクレオチド(構築物)であって、ヒト細胞によって前駆体milRNAに転写され、前記の前駆体milRNAがヒト細胞内によって切断され、かつ前記のmilRNAとして発現される、ポリヌクレオチドを提供する。
本発明の一つの好適な様態として、前記ポリヌクレオチド構築物は、式Iで表される構造を有し、

Seqフォワードは、細胞において前記のmilRNAとして発現されるヌクレオチド配列で、Seqリバースは、Seqフォワードに基本的に相補的なヌクレオチド配列である。あるいは、Seqリバースは、細胞において前記のmilRNAとして発現されるヌクレオチド配列で、Seqフォワードは、Seqフォワードに基本的に相補的なヌクレオチド配列である。
式Iで表される構造は、細胞に導入された後、式IIで表される二次構造になる。

前記の発現ベクターは、好ましくは1つまたは2つ以上の選択性マーカー遺伝子を含み、形質転換された宿主細胞を選択するための形質、たとえばカナマイシン、ゲンタマイシン、ハイグロマイシン、アンピシリン耐性を提供する。
本発明がわかりやすくなるように、本発明者は以下の機序を提供する。しかし、もちろん、これらの機序は本発明の何らかの限定にはならない。
本発明の研究によって、微生物が人体に感染した後、自身のncRNAをすこし分泌して循環系に入ることが示され、これらのncRNAは微生物自身由来のものであるため、微生物感染症の診断のマーカーとすることができる。
典型的に、血清に存在する、微生物由来のncRNAは微生物感染症の診断のマーカーとすることができる。これらの血液または血清における病原体の特有のncRNAは、微生物感染症の診断の効率を大幅に向上させ、疾患自身の治療に大いに役立ち、重要な臨床上の意義があると同時に、微生物由来のncRNAの更なる研究はこのような疾患の発症または感染の機序に新しい手がかりを提供する。
また、本発明は、汎用の病原体由来のncRNAを同定する方法であって、
血清または血液に存在する病原体のncRNAの同定を例として、
a.各種の微生物(細菌、古細菌、ウイルス、マイコプラズマ、クラミジア、原生生物などを含む)の感染者の血液または血清の標本を収集する工程と、
b.ハイスループットシーケンシング技術と生物情報学的分析・比較を合わせた手段によって、患者の血液または血清における微生物由来の1組のsRNA(miRNAを含む)の初期選別を行う工程と、
c.さらに高感度で、正確なリアルタイム蛍光定量PCRによって初期選別されたsRNAの検証を行い、各種の微生物の感染者の血清におけるそれぞれの特異的な微生物自身由来の血清sRNAを見つけ、かつ疾患そのものに対する診断上の価値を評価する工程と、
を含む方法を提供する。
本発明において、同定された病原体由来のncRNAに対し、さらに前記ncRNAの病原体感染における作用を評価することができる。
a.各種の微生物(細菌、古細菌、ウイルス、マイコプラズマ、クラミジア、原生生物などを含む)で細胞に感染させ、感染後の細胞の培養液を収集する工程と、
b.ハイスループットシーケンシング技術(Solexaシーケンシング技術)と生物情報学的分析・比較を合わせた手段によって、細胞培養液における微生物由来の1組のsRNA(miRNAを含む)の初期選別を行う工程と、
c.さらに高感度で、正確なリアルタイム蛍光定量PCRによって初期選別されたsRNAの検証を行う工程と、
を含む。
本発明によって提供される上記方法では、微生物感染症の患者の体液、血液(または血清)における微生物自身由来の一つまたは複数のncRNA(miRNAを含む)の微生物感染症の診断に対する特異性、正確性を同定して評価し、かつ既存の臨床診断方法と比較することができる。
本発明の研究は、初めて、体内の感染微生物、寄生微生物、共生微生物の宿主を調節する新たな手段を開示する。分泌経路を通し、異なる体内の病原体(たとえば体内の感染微生物、寄生微生物、共生微生物)は自身が生成した非コードRNA(non-coding RNA、ncRNA)を宿主細胞に提示し、宿主のタンパク質を「借り」、宿主の免疫系に影響することによって、体内の病原体の宿主の体内における生活に貢献する。そのため、病原体の特異的なncRNAを干渉または抑制するのは、微生物感染の抑制および微生物性疾患の治療の新たなアプローチである。
本発明の発見に基づき、病原体の特異的なncRNAを検出(または診断)のマーカーとして使用し、かつ治療の標的とすることができる。
サルモネラ菌を例として、サルモネラ菌は細菌自身によってコードされる非コードRNA(ncRNA)を宿主細胞内に提示し、細胞のmicroRNA切断システムによって、microRNAと類似のmilRNAを生成し、milRNAで免疫系(たとえばiNOS)を調節することで、細菌に利用され、宿主による除去から自身を守る。milRNAの阻害剤でmilRNAを吸着すると、細菌の細胞内の生存能力を有効に抑制し、細菌の生存能力を低下させるため、細菌感染の治療に使用することができる。
また、本発明は、サルモネラ菌またはサルモネラ菌感染を検出キットであって、本発明の検出試薬、または検出チップを含むキットを提供する。前記のキットは、本発明のサルモネラ菌感染の特異的なmiRNAの発現プロファイルの検出、あるいはサルモネラ菌またはサルモネラ菌感染の検出に使用することができ、好ましくは、前記のキットは、さらに、RNAサンプルを標識するための標識物、および前記標識物に相応する基質を含む。
また、前記のキットは、さらに、RNAの抽出、PCR、ハイブリダイゼーション、呈色などに必要な様々な試薬を含んでもよく、抽出液、増幅液、ハイブリダイゼーション液、酵素、コントロール液、呈色液、洗浄液、抗体などを含むが、これらに限定されない。
また、前記のキットは、さらに、使用説明書および/またはチップ画像分析ソフトを含んでもよい。
microRNAの発現プロファイルチップは、通常、百、千またはそれ以上のプローブを含み、複数種類のmicroRNAに関し、二本鎖の相同対合の原理で標本に含まれる各種のmicroRNAの含有量を検出する。そのため、一回で標本におけるmicroRNAの転写レベルを測定することができる。
本発明に係るmilRNA配列では、相応のmilRNAチップを製造し、さらにその発現プロファイルおよびmilRNAの調節方式を研究することができる。
一方、本発明は、さらに、milRNAの発現プロファイルを分析するためのチップであって、サルモネラ菌またはサルモネラ菌感染の検出に使用できるチップを提供する。
具体的に、本発明に係るmilRNAに基づき、適切なプローブを設計し、固相担体に固定化し、「オリゴヌクレオチドアレイ」を形成することができる。前記の「オリゴヌクレオチドアレイ」とは、アドレス指定可能な位置(すなわち区別可能なアクセスできるアドレスを特徴とする位置)を有するアレイで、各アドレス指定可能な位置はいずれもそれに連結する特徴的なオリゴヌクレオチドを1つ含む。必要により、オリゴヌクレオチドアレイをいくつかのサブアレイに分けてもよい。
前記のmilRNAチップの製造は、本分野で既知のバイオチップの通常の製造方法を使用してもよい。たとえば、固相担体を修飾されたガラスシートまたはシリコンシートとする場合、プローブの5’末端にアミノ基で修飾されたポリdT鎖が含まれ、オリゴヌクレオチドプローブを溶液とした後、スポッターでそれを修飾されたガラスシートまたはシリコンシートにスポットし、所定の配列またはアレイに並べた後、一晩置いて固定化させることで、本発明のmilRNAチップが得られる。
その後、標識シグナルのmilRNAチップにおける位置、強度などの情報から測定したい情報を獲得する。増幅産物を蛍光基で標識する場合、直接蛍光検出装置(たとえば共焦点レーザー走査装置Scanarray 3000など)で測定したい情報を獲得してもよい。
また、本発明は、薬学的に許容される担体あるいは有効量の本発明のmilRNA(たとえばsal−1)の阻害剤、遮断剤または拮抗剤を含む薬物組成物を提供する。
ここで用いられるように、用語「有効量」または「有効投与量」とは、ヒト及び/又は動物に機能や活性があり、且つヒト及び/又は動物にとって受けられる量である。
ここで用いられるように、「薬学的に許容される」成分は、哺乳動物に適用する場合、過度の不良な副反応(例えば毒性、刺激とアレルギー反応)がない、すなわち、合理的なベネフィット/リスク比を持つ物質である。用語「薬学的に許容される担体」とは、治療剤の給与のための担体で、各種の賦形剤と希釈剤を含む。
本発明に係る薬学的に許容される担体は、水、塩水、リポソーム、脂質、タンパク質、タンパク質−抗体複合体、ペプチド系物質、セルロース、ナノゲル、またはこれらの組み合わせを含むが、これらに限定されない。担体の選択は投与様態に応じ、これらはすべて当業者に熟知のことである。
(a)病原体(たとえばサルモネラ菌)感染を検出する簡単で、迅速でかつ正確な新たな方法を提供する。
(b)微生物感染の抑制および微生物性疾患の治療の新たなアプローチを提供する。
サルモネラ菌の菌種を白金耳でLB平板に画線分離し、中等の大きさの集落を取ってLB液体培地に接種し、37℃で一晩培養した後、遠心でLB培地を除去し、RPMI−1640+10%FBS培地で菌体を再懸濁させた。
細菌感染の前日に、パンクレアチンで消化されたヒト腸管上皮細胞HT−29細胞をシャーレに展開させ、5% CO2インキュベーターで37℃で培養した。
その後、Trizol LSで溶解液における全RNAを抽出し、Solexaディープシーケンシング後、得られたデータを分析し、サルモネラ菌の特異的な小さいRNA断片を見つけた。
solexaシーケンシングで得られた断片を分析し、コピー数の多いmilRNA断片配列を選別し、これらの配列に対し、定量PCR検出プローブおよびノーザンブロッティングの検出プローブを設計して合成した。
Trizol LS試薬でそれぞれLBで培養したサルモネラ菌の全RNA、およびサルモネラ菌を細胞に感染させた後のサイトソルの全RNAを抽出し、全RNAを1×RNA loading buffer(Takara)で再懸濁させた後、15%TBE-Urea polyacrylamidegelelectrophoresis (PAGE)で電気泳動を行い、電気泳動後RNAをナイロン膜に転写し、紫外線(UV)で架橋させた後ジゴキシゲニン(DIG)で標識されたプローブでハイブリダイゼーションを行い、milRNAの発現を分析した。
Trizol LS試薬でそれぞれLBで培養したサルモネラ菌の全RNA、およびサルモネラ菌を細胞に感染させた後のサイトソルの全RNAを抽出し後、DEPCで処理された超純水でRNAを溶解させ、まずプローブの下流プライマーでcDNAに逆転写し、さらに合成したプローブ(ABI)で定量PCR増幅を行い、増幅後各サンプルにおけるmilRNAの含有量を分析した。
実験結果は図1、図2および図3に示した。
ノーザンブロッティングおよびqRT−PCの検出では、図1および2の結果から、サルモネラ菌を細胞に感染させた後だけ、19〜30 ntの小さいRNA(本発明においてmilRNAと呼ぶ)が現れたことがわかり、本発明では、milRNA(sal−1)を選んで全面的に深く研究した。

milRNAに対する阻害剤(inhibitor)の宿主への細菌感染を抑制する能力
2.1 体外実験
本実験において、同定されたサルモネラ菌の特異的なsal-1に対し、それと標的遺伝子inducible nitric oxide synthase(iNOS)の関連性を研究した。
PCRでmilRNAと結合する標的サイト領域を増幅し、Spe I+Hind IIIで二重酵素切断を行った後、pMIR−REPORTルシフェラーゼベクター(上海英為信科技有限公司から購入)に挿入し、組み換えプラスミド(iNOS)を構築した。一方、前記構築した組み換えプラスミドのmilRNA結合サイトを突然変異させ、組み換え突然変異プラスミド(iNOS mut)を構築した。
milRNAコントロールプラスミドまたはpre−sal−1とiNOSまたはiNOS mutをHEK−293T細胞にコトランスフェクトし、レポーター遺伝子の検出を行い、同時にβ−galプラスミドを参照とした。
具体的な結果は図4に示す。図4のルシフェラーゼの結果から、sal−1はiNOSにおける2つの標的サイトのいずれも調節することによって、iNOSを下方調節することができることがわかる。
野生型の全長のiNOS遺伝子を真核発現プラスミドにクローンし、組み換えiNOS発現プラスミド(iNOS WT)を構築してiNOS発現を行ったと同時に、milRNA結合サイトを突然変異させ(同義突然変異で、アミノ酸配列を変えずに塩基配列だけを変える)、iNOS発現プラスミド(iNOS MUT)を構築して突然変異iNOSの発現を行った。pre−sal−1をそれぞれiNOS WTおよびiNOS MUTとマウスRAW264.7細胞(当該細胞は刺激を受けないとiNOSを発現しない)にコトランスフェクトして発現させ、milRNAのiNOSに対する調節能力を検出した。
具体的な結果は図5に示す。図5の結果から、RAW264.7細胞に全長iNOSの発現プラスミドを導入する場合、sal−1が同様にiNOSの発現を調節する(下方調節)ことがわかる。
HT−29細胞をサルモネラ菌感染の前日に24ウェルプレートに置き、次の日にanti−sal−1をlipofectin 2000で転写し、anti−sal−1をトランスフェクトしてから24h後、MOI(5〜10:1)でサルモネラ菌を感染させ、コントロール群ではランダムの配列を使用し、感染後24hで細胞内の細菌の生存能力を検出した。

pre-sal-1配列を酵素切断を経て通常のlentivirusベクターに挿入することで、milRNAを過剰発現するレンチウイルスをパッケージングした。
milRNAに相補的に結合する配列を遺伝子工学的手段によってmilRNAと結合するスポンジ体(3回繰り返す相補断片を含有する)を構築した後、milRNAのスポンジ体もlentivirusベクターに挿入してmilRNAを吸着するレンチウイルスをパッケージングした。
さらに、マウスの体内でもiNOSが発現されるので、milRNAのiNOSに対する調節機能を検証するために、本発明者はマウスのiNOSにおけるmilRNAの結合サイトがすべて突然変異した発現プラスミドを構築した後、このようなプラスミドを酵素切断を経てentivirusベクターに挿入することで、マウスiNOSを過剰発現するレンチウイルスをパッケージングした。
図7Aの結果から、マウスにサルモネラ菌SE2472を感染させた後、sal−1の発現量が上昇し、lenti−sal−1+SE2472群ではsal−1の発現量がさらに上昇し、lenti−sal−1 sponge+SE2472群のsal−1の発現レベルはsal−1 sponge(sal−1に対する阻害剤)に吸着されて低下したことがわかる。マウスiNOSのsal−1結合サイトを同義突然変異させてレンチウイルスにパッケージングしてマウスに感染させた後、sal−1が若干低下し(突然変異したiNOSNo発現が細菌のマウス体内における生存能力に影響し、数が若干低下したため)、lenti−sal−1+ iNOS MUT SE2472群ではsal−1の発現量がある程度回復し、lenti− sal−1 sponge+ iNOS MUT SE2472群ではsal−1の発現量がさらに低下した。
図7Bは、この8群のマウスのSE2472に感染した場合の臨床症状の変化を示し、これらの変化は前記sal−1の変化による細菌感染症状の変化と一致する。
図7Eは、この8群におけるマウスの大腸組織(腸腔内のものがすでに除去された)における細菌含有量を検出した。sal−1の発現に対する上方調節は細菌の腸組織における感染能力を促進し、sal−1がspongeに吸着して除去された後、細菌感染能力が低下した。iNOS同義突然変異群では、sal−1は調節作用を発揮せず、これらの結果はいずれもsal−1がiNOSを調節し、かつiNOSを介して細菌の宿主体内における侵入と生存能力に影響することを示した。
サルモネラ菌milRNAの応用
本発明に係るmilRNAの使用は、(a)サルモネラ菌感染を検出する試薬、検出チップまたはキットの製造、あるいは(b)iNOSの発現または活性を調節する調節剤の製造に用いられる。
ここで、サルモネラ菌感染を検出する核酸チップは、固相担体と、前記固相担体に規則正しく固定化された、特異的に本発明に係るmilRNAを捕獲するオリゴヌクレオチドプローブとを含む。
さらに、本発明に係る核酸チップの使用は、サルモネラ菌感染を検出するキットの製造に用いられる。サルモネラ菌感染を検出するキットは、本発明に係る核酸チップまたは本発明に係るmilRNAを含む。
まず、シーケンシングの方法またはPCR法によって正常の血清/血漿にコピー数が1個以上のmilRNAを確定した。そして、定量PCR技術とバイオチップ技術で疾患と正常の生理状態における発現量および差異の大きな血清/血漿milRNAを選別し、サルモネラ菌感染の有無の予測および病変の程度の診断の指標とした。最後に、相応の血清/血漿milRNAの数を選別したが、これはチップのプローブライブラリーに基づいて最適な簡略化である。このキットは、血清/血漿milRNAプライマー、Taq酵素、dNTPなどの試薬を含む。このキットの価値は、ほかの組織のサンプルが不要で、必要なのは血清/血漿だけで、最も簡略化したプローブライブラリーでmilRNAの変化傾向を検出し、さらに当該変化傾向からサルモネラ菌感染の発生の可能性を予測し、またはサルモネラ菌感染の病理的段階を診断することにある。そのため、このキットを実践に投入すると、サルモネラ菌感染の早期発見の可能性が増加し、診断と治療の指導に役立つ。
Claims (11)
- 単離されたマイクロRNA様RNA(milRNA)であって、
(i) 配列番号1で示される配列のmilRNA、
(ii) (i)における前記milRNAのヌクレオチド配列に相補的なmilRNA、
から選ばれるmilRNA。 - milRNAは、サルモネラ菌由来のものである請求項1に記載のmilRNA。
- 単離された前駆体milRNAまたは人工的に構築された前駆体milRNAであって、動物細胞内において切断され、請求項1または2に記載のmilRNAとして発現される、前駆体milRNA。
- 単離されたポリヌクレオチドであって、動物細胞によって前駆体milRNAに転写され、前記の前駆体milRNAがヒト細胞内において切断され、請求項1または2に記載のmilRNAとして発現される、ポリヌクレオチド。
- 請求項1または2に記載のmilRNA、または請求項4に記載のポリヌクレオチドを含むベクター。
- (a)サルモネラ菌感染を検出する試薬、検出チップまたはキットの製造、あるいは(b)iNOSの発現または活性を調節する調節剤の製造に用いられる請求項1または2に記載のmilRNAの使用。
- 固相担体と、
前記固相担体に規則正しく固定化された、特異的に請求項1または2に記載のmilRNAを捕獲するオリゴヌクレオチドプローブと、
を含む核酸チップ。 - 請求項7に記載の核酸チップまたは請求項1または2に記載のmilRNAを含むキット。
- 請求項1または2に記載のmilRNAを特異的に抑制または遮断する阻害剤。
- 薬学的に許容される担体と、請求項1または2に記載のmilRNAを特異的に抑制または遮断する阻害剤とを含む薬物組成物。
- サルモネラ菌感染の治療に用いられる候補薬物を選別する方法であって、
(a)テスト群とコントロール群を提供し、前記テスト群では、候補物質をテスト群の細胞または動物に施用し、かつ使用後の前記テスト群におけるsal−1の発現レベルを測定し、一方、前記のコントロール群では、テスト群と同様の条件を使用するが、候補物質をコントロール群の細胞または動物に施用しない工程と、
(b)テスト群のsal−1とコントロール群のsal−1の発現レベルを比較する工程と、
を含み、
ここで、sal−1の配列は配列番号1で示され、テスト群のsal−1の発現レベルがコントロール群のsal−1の発現レベルよりも顕著に低い場合、当該候補物質はサルモネラ菌感染の治療に用いられる候補薬物であることを示す方法。
