Description
(a)試料を、膜貫通孔および2つ以上のプローブのパネルと接触させるステップであって、

本発明は、試料中の2つ以上の分析物の群の1つまたは複数の分析物メンバーの存在または非存在を決定する方法を提供する。従って、本発明は、分析物の群の各分析物メンバーについての多重アッセイに関する。群中の2つ以上の分析物は、予め決定されるよって、。本発明は、具体的で予め決定された分析物の群の各分析物メンバーについての多重アッセイに関する。
試料は、任意の適切な試料であってよい。本発明は、2つ以上の分析物を含有することがわかっているかまたは含有することが疑われる試料に対して典型的に行われる。本発明は、その正体がわかっていない2つ以上の分析物を含有する試料に対して行うことができる。代わりに、本発明は、試料に対して行って、試料中のその存在がわかっているかまたは期待される2つ以上の分析物の正体を確認することができる。
本発明の方法は、試料中の2つ以上の分析物の群の1つまたは複数の分析物メンバーの存在または非存在を決定するためである。2つ以上の分析物の群は、2、5、10、15、20、30、40、50、100以上の分析物などの任意の数の分析物を含んでよい。群は、好ましくは、約5から約80までの分析物、約10から約60までの分析物または約20から約50までの分析物などの約4から約100までの分析物を有する。
本発明の方法は、試料を膜貫通タンパク質孔と接触させるステップを含む。膜貫通孔は、印加された電位により駆動される水和イオンが膜の一方の側から膜の他方の側へと流れることを可能にする構造である。
本発明の方法は、試料を、2つ以上のプローブのパネルと接触させるステップを含む。言い換えると、本発明の方法は、試料を、2つ以上の型のプローブのパネルと接触させるステップを含む。パネルは、2、5、10、15、20、30、40、50、100以上のプローブなどの任意の数の2つ以上のプローブを含んでよい。パネルは、好ましくは、約5から約80までのプローブ、約10から約60までのプローブまたは約20から約50までのプローブなどの約4から約100までのプローブを有する。
パネル中の各プローブ(すなわちそれぞれの型のプローブ)は、分析物メンバーの1つまたは複数を認識し、1つまたは複数の分析物メンバーと結合するアプタマーを含む。プローブによる分析物メンバーの認識については、上記している。
パネル中の各プローブ(すなわちそれぞれの型のプローブ)は、孔に侵入できる尾部を含む。尾部は、典型的に、孔のバレルまたはチャネルに侵入して、それを通過できる直鎖状分子である。尾部を形成するために適切な分子については、以下で論じる。尾部が孔に侵入すると、尾部は、バレルまたはチャネル中に存在する尾部の部分に特異的な様式で孔を流れる電流に影響する。
各プローブ(すなわちそれぞれの型のプローブ)は、孔を流れる電流に示差的な様式で影響する。言い換えると、プローブは、孔を流れる電流に、異なるプローブが孔を流れる電流に影響する方式とは区別または識別できる様式で影響する。このことにより、本発明に従って各プローブ(すなわちそれぞれの型のプローブ)の正体を測定できる。プローブと分析物メンバーとの結合を、次いで、上記のようにして測定できる。各プローブの正体および各プローブと分析物メンバーとの結合を測定できるので、各分析物メンバーの存在または非存在を決定できる。
電気的測定は、Stoddart, D. S.ら、(2009)、Proceedings of the National Academy of Sciences of the United States of America 106、7702〜7707頁、Lieberman KRら、J Am Chem Soc. 2010;132(50):17961〜72頁および国際出願WO−2000/28312に記載されるような標準的な1チャネル記録装置を用いて行ってよい。代わりに、電気的測定は、多重チャネル系、例えば国際出願WO−2009/077734および国際出願WO−2011/067559に記載されるようなものを用いて行ってよい。
本発明の方法は、好ましくは、1つまたは複数の分析物メンバーと結合するプローブについて、各プローブが結合しているときおよび未結合のときに孔を流れる異なる電流を比較するステップをさらに含む。このことは、全般的に、検量線を参照することにより、平衡定数を用いることによりまたは対照データを参照することにより、試料中に存在する分析物メンバーの濃度を決定するための助けとなる。濃度を算出する方法は、当該技術において公知である。
(i)上記の本発明の方法を行うステップと、
(ii)試料中に存在することが示された1つまたは複数の分析物メンバーについて、ステップ(b)における孔を流れる電流を、各分析物メンバーについての対照または参照データと比較し、そのことにより試料中の1つまたは複数の分析物メンバーの濃度を決定するステップと
を含む方法も提供する。ステップ(i)は、好ましくは、1つまたは複数の分析物メンバーと結合するプローブについて、各プローブが結合しているときおよび未結合のときに孔を流れる異なる電流を比較することを含む。対照または参照データは、既知の濃度の分析物メンバーを用いてアッセイを校正する対照実験を行うことにより作製できる。このことについては、実施例2に記載する。
本発明は、試料中の2つ以上の分析物の群の1つまたは複数の分析物メンバーの存在、濃度または非存在を決定するためのプローブのパネルも提供する。パネルは、2つ以上のプローブを含み、
各プローブは、分析物メンバーの1つまたは複数を認識し、(i)1つまたは複数の分析物メンバーと結合するアプタマーと、(ii)膜貫通孔に侵入でき、プローブ中のアプタマーが分析物メンバーの1つと結合しているか否かに応じて孔を流れる電流に異なる影響を与える尾部とを有し、
各プローブは、孔を流れる電流に示差的な様式で影響し、
2つ以上の分析物の群中の各分析物メンバーは、パネル中の少なくとも1つのプローブにより認識される。本発明の方法に関して上記の任意の実施形態は、本発明のパネルに対して等しく当てはまる。パネル中の各プローブは、好ましくはプローブが膜にカップリングされることを可能にする化学基をさらに含む。化学基は、好ましくは、コレステロールである。
本発明は、試料中の2つ以上の分析物の群の1つまたは複数の分析物メンバーの存在、濃度または非存在を決定するための装置も提供する。装置は、複数の孔と、本発明のプローブのパネルとを含む。装置は、好ましくは、本発明の方法を行うための指示書をさらに含む。装置は、アレイまたはチップなどのポリヌクレオチド分析のための任意の従来の装置であってよい。本発明の方法に関して上記の任意の実施形態は、本発明の装置に対して等しく当てはまる。
膜と複数の孔とを支持でき、孔を用いて分析物メンバーの特徴決定を行うために作動可能なセンサデバイスと
特徴決定を行うための材料を保持するための少なくとも1つの貯蔵器と、
少なくとも1つの貯蔵器からセンサデバイスに材料を制御可能に供給するように構成された流体システムと、
それぞれの試料を受容するための複数の容器と
を含み、流体システムは、容器からセンサデバイスに選択的に試料を供給するように構成されている。装置は、国際出願第PCT/GB08/004127号(WO2009/077734として公開)、第PCT/GB10/000789号(WO2010/122293として公開)、国際出願第PCT/GB10/002206号(未公開)または国際出願第PCT/US99/25679号(WO00/28312として公開)に記載されるもののいずれであってもよい。
電気的測定は、銀めっきした(WO2009/077734)標準的な128ウェルシリコンチップ(フォーマット直径75μm、深さ20μmおよびピッチ250μm)を用いて得た。チップは、最初に、20mLのエタノール、次いで20mLのdH2O、次いで20mLのエタノールを用いて洗浄した後にCF4プラズマ処理した。用いたチップを、次いで、浸漬被覆により前処理し、真空シールして4℃にて貯蔵した。使用前に、チップを少なくとも20分間室温まで温めた。
上記の方法を用いて、トロンビンの存在および非存在下で特徴的なブロックレベルを検出可能であった。プローブ5A15x10A_TBA(配列番号7)の添加により、レベル1にて1つのブロックレベルのみが観察された(図2aを参照されたい)。これに対して、プローブ5A15x10A_TBAをトロンビンとプレインキュベートした場合に、さらなるレベル(レベル2;図2b)も検出された。トロンビンとプローブ5A15x10A_TBAとの結合により、平均電流より高い流れが測定され、このことは、脱塩基領域(x)がβ−バレル内にあることを示した。2つのレベル間の差は、30.5pAであった(図2b)。同じ実験を、配列番号8〜18を用いて行った(8=11x19A_TBA、9=13x17A_TBA、10=15x15A_TBA、11=17x13A_TBA、12=19x11A_TBA、13=21x9A_TBA、14=23x7A_TBA、15=25x5A_TBA、16=27x3A_TBA、17=29x1A_TBAおよび18=30x0A_TBA)。
電気的測定は、銀めっきした(WO2009/077734)標準的な128ウェルシリコンチップ(フォーマット直径75μm、深さ20μmおよびピッチ250um)を用いて得た。トロンビンの濃度の決定を可能にする実験を行うために、同じ実験構成手順を、実施例1に記載するようにして行い(ナノポアαHL−(E111N/K147N)7(配列番号2)および緩衝剤1M KCl、10mM TRIS pH7.5を用いる)、同じ初期制御プログラム(0mVにて10秒、170mVにて50秒、20回まで反復)を5分間運転した。
上記の方法を用いて、プローブ5A15x10A_TBA(配列番号7)と結合するトロンビンに相当する事象の頻度を測定することが可能であった(図2を参照されたい)。このデータを次いで用いて、トロンビンの濃度対遮断事象の総数(結合と未結合とを組み合わせたもの)と比較したトロンビン結合事象の比率の校正プロットを作成した。図3を参照されたい。これらの校正プロットを次いで用いて、トロンビンの未知試料の濃度を決定できる。
電気的測定は、銀めっきした(WO2009/077734)標準的な128ウェルシリコンチップ(フォーマット直径75μm、深さ20μmおよびピッチ250um)を用いて得た。より低い濃度のコレステロールタグ付加プローブ(コレステロールTEGを3’端に有する配列番号19)を用いるトロンビンの検出を可能にする実験を行うために、同じ実験構成手順を、実施例1に記載するようにして行い(ナノポアαHL−(E111N/K147N)7(NN)(配列番号2)および緩衝剤1M KCl、10mM Tris pH7.5を用いる)、初期制御プログラム(0mVにて10秒、170mVにて50秒、20回まで反復)を5分間運転した。
上記の方法を用いて、著しくより低い濃度のプローブ15x15A_TBA_3’CholTEG(コレステロールTEGを3’端に有する配列番号19)を用いて、トロンビンの存在および非存在下で特徴的なブロックレベルを検出可能であった。上記の例では、プローブ15x15A_TBA_3’CholTEGを用いてレベル1を、そしてプローブ15x15A_TBA_3’CholTEGをトロンビンとプレインキュベートした場合にレベル1および2を観察できた(図4)。ステップレベルの変化が、100pM濃度だけのコレステロールタグ付加プローブ15x15A_TBA_3’CholTEGを用いて観察された。この濃度は、実施例2で試験したものよりもかなり低い。
電気的測定は、銀めっきした(WO2009/077734)標準的な128ウェルシリコンチップ(フォーマット直径75μm、深さ20μmおよびピッチ250um)を用いて得た。多様な異なるプローブを用いてトロンビンの検出を可能にする実験を行うために、同じ実験構成手順を、実施例1に記載するようにして行い(ナノポアαHL−(E111N/K147N)7(NN)(配列番号2)および緩衝剤1M KCl、10mM Tris pH7.5を用いる)、同じ初期制御プログラム(0mVにて10秒、170mVにて50秒、20回まで反復)を5分間運転した。
上記の方法を用いて、トロンビンの存在下および非存在下の両方で用いたプローブのそれぞれについての示差的ステップレベルを検出することが可能であった(図5)。この実験は、観察されるステップレベルが、プローブ尾部配列に特異的であり、よって、複数のプローブ尾部を用いて1つより多いタンパク質を検出できたことを示す。
電気的測定は、1,2−ジフィタノイル−グリセロ−3−ホスホコリン脂質(Avanti Polar Lipids)2重層中に挿入された単一αHL−(E111N/K147N)7(NN)(配列番号2)ナノポアから得た。2重層を、20μm厚のPTFEフィルム(カスタムDelrinチャンバ中)中の直径およそ100μmの開口を横切ってMontal−Mueller技術により形成して、2つの1mL緩衝溶液(1M KCl、10mM Tris pH7.5)を分けた。単一チャネル電流を、1440Aデジタイザを備えるAxopatch 200B増幅器(Molecular Devices)で測定した。Ag/AgCl電極を緩衝溶液に接続して、シス区画(ナノポア、プローブおよびPDGFの両方が添加された)をAxopatchヘッドステージの接地と接続し、トランス区画をヘッドステージの活性電極と接続した。
上記の方法を用いて、PDGFの存在下および非存在下で特徴的なステップレベルを検出可能であった。プローブ17x8A_PDGF(配列番号20)を採用して、レベル1にてブロックレベルが観察された(図6aを参照されたい)。これに対して、17x8A_PDGFとプレインキュベートしたPDGFを実験溶液に加えた場合に(レベル2)も検出された(図6bを参照されたい)。図7aおよび7bは、別のプローブ25x_PDGF(配列番号21)を用いた場合に、PDGFの存在下および非存在下で生じるステップレベルを示す。
電気的測定は、銀めっきした(WO2009/077734)標準的な128ウェルシリコンチップ(フォーマット直径75μm、深さ20μmおよびピッチ250um)を用いて得た。PDGFの濃度の決定を可能にする実験を行うために、実験構成手順を、実施例1に記載するようにして行い(ナノポアαHL−(E111N/K147N)7(NN)(配列番号2)および緩衝剤1M KCl、10mM Tris pH7.5を用いる)、同じ初期制御プログラム(0mVにて10秒、170mVにて50秒、20回まで反復)を5分間運転した。
上記の方法を用いて、プローブ25x_PDGF(配列番号21、図7aおよびbを参照されたい)と結合するPDGFに相当する事象の頻度を測定することが可能であった。このデータを次いで用いて、PDGFの濃度対遮断事象の総数(PDGF結合と未結合とを組み合わせたもの)と比較した場合のPDGF結合事象のみの比率の校正プロットを作成した。図8を参照されたい。この校正プロットを次いで用いて、PDGFの未知試料の濃度を決定できる。
電気的実験は、脂質2重層に挿入された単一孔(ナノポアαHL−(E111N/K147N)7(NN)(配列番号2)および緩衝剤1M KCl、10mM Tris pH7.5を用いる)を達成するために実施例5に記載するようにして設定し、次いで、+180mVの印加電位にて対照を5分間運転した。PDGFの代わりにATPを検出するために異なるアプタマーを用いた以外は、実施例5と同じ手順ステップを行った。アプタマーのみの運転について、本実施例で用いたアプタマーは、1M KCl、10mM Tris pH7.5中の25x_ATP(配列番号22、0.5μM)であり、アプタマープラスATPの運転について、以下の濃度の試薬を用いた:25x_ATP(配列番号22、0.5μM)およびATP(10mM)(Cat No:A6559、Sigma−Aldrich、Dorset、UK)。
上記の方法を用いて、ATPの存在下および非存在下で特徴的なブロックレベルを検出可能であった。プローブ25x_ATP(配列番号22)を採用して、レベル1にてブロックが観察された(図9aを参照されたい)。これに対して、プローブ25x_ATPをATPとプレインキュベートした場合に、さらなるレベル(レベル2;図9b)も検出された。ATPの場合、プローブ転位により生じる事象は非常に短く、結合したATPに相当する事象は著しく長い(図9b)。図10aおよび10bは、プローブ17x8A_ATP(配列番号23)を用いた場合に、ATPの存在下および非存在下で生じるステップレベルを示す。
電気的実験は、脂質2重層に挿入された単一孔(ナノポアαHL−(E111N/K147N)7(NN)(配列番号2)および緩衝剤1M KCl、10mM Tris pH7.5を用いる)を達成するために実施例5に記載するようにして設定し、次いで、+180mVの印加電位にて対照を5分間運転した。スクリプトを次いで停止し、1M KCl、10mM TRIS pH7.5緩衝剤中のプローブ17x8A_ATP(配列番号23、0.5μM)を電気生理学的チャンバのシス区画に加え、実験を20分間運転した。長い遮断事象(>15s)が検出された場合、電位を−180mVに即座に切り替えて、遮断している分析物を手動で排出した。スクリプトを次いで停止し、1M KCl、10mM TRIS pH7.5緩衝剤中で最小限で5分間一緒にプレインキュベートしたプローブ17x8A_ATP(0.5μM)およびATP(1μM)(Cat No:A6559、Sigma−Aldrich、Dorset、UK)をシス区画に次いで加えて、実験をさらに10分間運転した。このプロセスを、3.3μM、10μMおよび100μMの濃度のATPについて反復した。実験の最後に、制御プログラムを停止した。
上記の方法を用いて、プローブ17x8A_ATP(配列番号23、図10aおよびbを参照されたい)と結合するATPに相当する事象の頻度を測定することが可能であった。このデータを次いで用いて、ATPの濃度対遮断事象の総数(ATP結合と未結合とを組み合わせたもの)と比較した場合のATP結合事象の比率の校正プロットを作成した。図11を参照されたい。この校正プロットを次いで用いて、ATPの未知試料の濃度を決定できる。マイクロモル範囲においてKd値を有するのでより高い濃度のATPを加えることが必要であった以外は、同様の事象比率の増加が、トロンビンと同様にATPについて観察された(図11)。
電気的実験は、脂質2重層に挿入された単一孔(ナノポアαHL−(E111N/K147N)7(NN)(配列番号2)および緩衝剤1M KCl、25mM Tris pH7.5を用いる)を達成するために実施例5に記載するようにして設定し、次いで、制御プログラム(0mVにて10秒、+180mVにて50秒、20回まで反復)を5分間運転した。スクリプトを次いで停止し、1M KCl、25mM TRIS pH7.5緩衝剤中のプローブ15x15A_TBA(配列番号10、166nM)、17x8A_PDGF(配列番号20、166nM)および17x8A_ATP(配列番号23、166nM)を電気生理学的チャンバのシス区画に加え、制御プログラムを10分間運転した。ATP(10mM)(Cat No:A6559、Sigma−Aldrich、Dorset、UK)を次いでシス区画に加え、制御プログラムをさらに10分間運転した。このプロセスを、1μM、PDGF(Biorbyt、Cat No:orb80544、Cambridge、UK)および0.5μMトロンビン(Cat No.T6884、Sigma−Aldrich、Dorset、UK)について反復した。実験の最後に、制御プログラムを停止した。
上記の方法を用いて、3つの分析物全てについて溶液中に存在する場合に特徴的なステップレベル変化を検出可能であった。個別に試験した場合に分析物のそれぞれについて観察された3つ全てのステップシグナルを、上記の実験においてみることができた(図12を参照されたい)。ATPおよびPDGFはともに、トロンビンよりも短い結合事象を生じるが、このことは、これらの分析物を用いて4重鎖構造が欠如していることによる可能性がある。よって、本方法を用いて、未知の混合物中の3つの分析物のいずれも検出可能である。
電気的実験は、脂質2重層に挿入された単一孔(ナノポアαHL−(E111N/K147N)7(NN)(配列番号2)および緩衝剤1M KCl、10mM Tris pH7.5を用いる)を達成するために実施例5に記載するようにして設定し、次いで、制御プログラム(0mVにて10秒、+180mVにて50秒、20回まで反復)を5分間運転した。この後に、1M KCl、10mM Tris pH7.5中で少なくとも5分間一緒にプレインキュベートした5A15x10A_TBA(配列番号7、1.25μM)およびトロンビン(10nM)(Cat No.T6884、Sigma−Aldrich、Dorset、UK)をチャンバに流し、制御プログラムをさらに15分間運転した。チップの上の全容積の溶液を、次いで、1M KCl、10mM Tris pH7.5中で最小限で5分間プレインキュベートした5A15x10A_TBA(1.25μM)およびトロンビン(33nM)の溶液で置き換え、制御プログラムをさらに15分間運転した。このプロセスを、100nM、330nMおよび1000nMの濃度のトロンビンについて反復した。実験の最後に、制御プログラムを停止した。

電気的測定は、各ウェル構造中に白金電極を有する標準的な128ウェルシリコンチップ(フォーマット直径75μm、深さ20μmおよびピッチ250um)を用いて得た。プローブを用いるストレプトアビジンの検出を可能にする実験を行うために、同じ実験構成手順を、実施例1に記載するようにして行い(ナノポアαHL−(E111N/K147N)7(NN)(配列番号2)および緩衝剤625mM NaCl、100mM HEPES、75mMフェロシアン化カリウム、25mMフェリシアン化カリウム、pH8.0を用いる)、同じ初期制御プログラム(0mVにて10秒、170mVにて50秒、20回まで反復)を5分間運転した。
上記の方法を用いて、ストレプトアビジンの存在下(図14b)および非存在下(図14a)で特徴的なブロックレベルを検出可能であった。strepプローブ1(配列番号24)の添加により、レベル1のブロックレベルだけが観察された(図14aを参照されたい)。これに対して、strepプローブ1をストレプトアビジンとプレインキュベートした場合に、さらなるレベル(レベル2;図14b)も検出された。図15は、ストレプトアビジンの存在下(2)および非存在下(1)でアプタマー配列strepプローブ1(配列番号24)について観察される異なるブロックレベルの拡大図を示す(y軸=電流(pA)およびx軸=(a)および(b)についての時間(s))。これは、表示の目的のために、Besselフィルタにかけた。
電気的測定は、白金めっきした(WO2009/077734)標準的な128ウェルシリコンチップ(フォーマット直径75μm、深さ20μmおよびピッチ250um)を用いて得る。K2 EDTA(Harlan Scientific、コード−S.B−0009)中に20%ウサギ全血を含有する試料中でタンパク質トロンビン、PDGFおよびATPの存在の検出を可能にする実験を行うために、同じ実験構成手順を、実施例1に記載するようにして行い(ナノポアαHL−(E111N/K147N)7(NN)および緩衝剤625mM KCl、100mM HEPES、75mMフェロシアン化カリウム、25mMフェリシアン化カリウムpH8.0を用いる)、同じ初期制御プログラム(0mVにて10秒、170mVにて50秒、20回まで反復)を3分間運転する。
上記の方法を用いて、K2 EDTA(Harlan Scientific、コード−S.B−0009)80%緩衝液中の20%ウサギ全血中に存在する場合に、3つ全ての分析物についての特徴的なステップレベル変化を検出することが可能である。

パネル中のプローブは、孔にカップリングされておらず、
各プローブが、分析物メンバーの1つまたは複数を認識し、(i)分析物メンバーの1つまたは複数と結合するアプタマーと、(ii)孔に侵入でき、プローブ中のアプタマーが分析物メンバーの1つと結合しているか否かに応じて孔を流れる電流に異なる影響を与える尾部とを含み、

(b)群が、約4から約100までの分析物メンバーを有する;および/または
(c)分析物メンバーが、金属イオン、無機塩、重合体、アミノ酸、ペプチド、ポリペプチド、タンパク質、ヌクレオチド、オリゴヌクレオチド、ポリヌクレオチド、色素、漂白剤、医薬品、診断剤、レクリエーショナルドラッグ、爆発物および環境汚染物質から独立して選択される;および/または

(e)アプタマーが、ペプチドアプタマーまたはオリゴヌクレオチドアプタマーである、請求項1から9のいずれか一項に記載の方法。 - 孔が、前庭またはバレルを含み、場合によって、
(a)バレルが、2本鎖ポリヌクレオチドが孔を通過できないように十分に狭い;および/または
(b)前庭およびバレルが、それぞれ少なくとも2ヌクレオチドを含有するように十分に長い;および/または

- 試料中の2以上の分析物の群の1つまたは複数の分析物メンバーの濃度を決定する方法であって、
(i)請求項1から12のいずれか一項に記載の方法を行うステップと、
(ii)試料中に存在することが示された1または複数の分析物メンバーについて、ステップ(b)における孔を流れる電流を、各分析物メンバーについての対照または参照データと比較し、そのことにより試料中の1つまたは複数の分析物メンバーの濃度を決定するステップと
を含む方法。 - 試料中の2つ以上の分析物の群の1つまたは複数の分析物メンバーの存在、濃度または非存在を決定するためのプローブのパネルであって、パネルが、2つ以上のプローブを含み、
各プローブが、分析物メンバーの1つまたは複数を認識し、(i)分析物メンバーの1つまたは複数と結合するアプタマーと、(ii)膜貫通孔に侵入でき、プローブ中のアプタマーが分析物メンバーの1つと結合しているか否かに応じて孔を流れる電流に異なる影響を与える尾部とを含み、
パネル中のプローブは、孔にカップリングされておらず、
各プローブが、孔を流れる電流に示差的な様式で影響し、
アプタマーが、3次元構造に折り畳まれているときには孔を通って移動するには大きすぎ、折り畳みがほどかれると孔を通って移動することが可能であり、
尾部が、孔に侵入し、孔を流れる電流に影響を与えることができる直鎖状の重合体を含み、
2つ以上の分析物の群中の各分析物メンバーが、パネル中の少なくとも1つのプローブにより認識される、プローブのパネル。 - パネルが、請求項2から9のいずれか一項で定義するとおりである、および/または、各プローブが、プローブと膜とのカップリングを可能にする化学基をさらに含む、請求項14に記載のプローブのパネル。

複数の孔を支持でき、孔を用いて分析物メンバーの特徴決定を行うために作動可能なセンサデバイスと
特徴決定を行うための材料を保持するための少なくとも1の貯蔵器と、
少なくとも1つの貯蔵器からセンサデバイスに材料を制御可能に供給するように構成された流体システムと、
それぞれの試料を受容するための複数の容器と
