試験細胞である角質形成細胞（細胞株名：ＨａＣａＴ、入手先：Ｄｒ．Ｎ．Ｅ． Ｆｕｓｅｎｉｇ， Ｄｅｕｔｓｃｈｅｓ Ｋｒｅｂｓｆｏｒｓｃｈｕｎｇｓｚｅｎｔｒｕｍ， Ｈｅｉｄｅｌｂｅｒｇ， Ｇｅｒｍａｎｙ）を、１０％のウシ胎児血清（Ｆｅｔａｌ ｂｏｖｉｎｅ ｓｅｒｅｕｍ， ＦＢＳ、入手先：Ｇｉｂｃｏ， ＵＳＡ）と１％ペニシリン−ストレプトマイシン（Ｐｅｎｉｃｉｌｌｉｎ−Ｓｔｒｅｐｔｏｍｙｃｉｎ、入手先：Ｇｉｂｃｏ， ＵＳＡ）を含有したＤＭＥＭ（Ｄｕｌｂｅｃｃｏ’ｓ Ｍｏｄｉｆｉｅｄ Ｅａｇｌｅ Ｍｅｄｉｕｍ、入手先：Ｌｏｎｚａ，ＵＳＡ）培地を利用して９６ウェルプレート（９６−ｗｅｌｌ ｐｌａｔｅ）に約４×１０４細胞数／ウェルの密度で分注した後、３７℃、５％ＣＯ２の条件下で２４時間培養した。培養液を除去した後、リン酸塩緩衝液（Ｐｈｏｓｐｈａｔｅ ｂｕｆｆｅｒｅｄ ｓａｌｉｎｅ， ＰＢＳ）２００μｌで１回洗浄し、ウシ胎児血清が含有されていないＤＭＥＭ培地２００μｌを入れて６時間培養した。その後、１％ウシ胎児血清が含有されたＤＭＥＭに、前記実施例において製造した烏梅抽出物を下記表に言及した濃度で３０分間処理した後、１０μＭ濃度のレチノイン酸（ｒｅｔｉｎｏｉｃ ａｃｉｄ、入手先：Ｓｉｇｍａ−Ａｌｄｒｉｃｈ， ＵＳＡ）又は１０ｐｐｍ濃度のラウリル硫酸ナトリウム（Ｓｏｄｉｕｍ Ｌａｕｒｙｌ Ｓｕｌｆａｔｅ、ＳＬＳ）を処理し、２４時間培養した。培養液５０μｌを取ってから、ＥＬＩＳＡ法を利用して、ＩＬ−６、ＩＬ−８エライザキット（ＥＬＩＳＡ ｋｉｔ）（入手先：ＢＤ ｐｈａｒｍｉｇｅｎ， ＵＳＡ）を利用してこれら炎症媒介因子の遊離抑制効果を評価した。抑制能は、標準物質であるｒｈＩＬ−６、ｒｈＩＬ−８（入手先：ＢＤ ｐｈａｒｍｉｇｅｎ， ＵＳＡ）の吸光度を基に標準曲線を描いて吸光度と標準物質との間の反応式を求めた後、処理物質の吸光度を代入してこれら炎症媒介因子の分泌量を得、これを図１及び図２に示した。また、以下の公式により炎症媒介因子抑制率（％）、すなわち刺激緩和率（％）を評価し、これを表１に示した。 [Experimental Example 1] Evaluation of Inhibition Effect on Inflammation-Mediated Factor Secretion by Stimulation Keratinocytes (cell line name: HaCaT, source: Dr. NE Fusenig, Deutsches Krebsforschungszentrum, Heidelberg, Germany), which are test cells, are 10 % DMEM (Dulbecco's Modi Fed, Gbco, USA) and 1% penicillin-streptomycin (Gibco, USA) containing 1% fetal bovine serum (Fetal bovine serum, FBS, available from Gibco, USA) (First: Lonza, USA) medium using 96-well plate at a density of about 4 × 10 4 cells / well.
