Claims (15)
- 修飾を含む組換えポリペプチドの存在を検出する方法であって、
前記修飾を含む組換えポリペプチドを含む試料を、前記修飾に選択的に結合する捕捉剤と、前記修飾に対する捕捉剤の選択的結合を可能にする条件下でインキュベートし、それによって、ポリペプチド-剤複合体を形成する工程と、
前記試料から前記ポリペプチド-剤複合体を精製する工程と、
前記修飾を含む組換えポリペプチドの存在を示す前記組換えポリペプチドの活性を検出することによって前記組換えポリペプチドの存在についてアッセイする工程と
を含み、前記修飾はPEG化、HES化、シリル化、又はポリシアル化の少なくとも1つにより前記組換えポリペプチドと会合するものであり、前記捕捉剤は修飾認識抗体であり、前記組換えポリペプチドは第VIII因子又は第IX因子である、上記方法。 - 前記試料が、前記修飾を有しないポリペプチド及び/又は修飾の程度の異なるパターンを有するポリペプチドを含む、請求項1に記載の方法。

- PEG化組換え第VIII因子の存在を検出する方法であって、前記PEG化組換え第VIII因子に対する抗PEG抗体の選択的結合を可能にする条件下で、前記PEG化組換え第VIII因子を含む試料を、抗PEG抗体とインキュベートし、それによって、第VIII因子-抗体複合体を形成する工程と、前記試料から前記第VIII因子-抗体複合体を精製する工程と、前記組換え第VIII因子の存在及び/又は第VIII因子活性についてアッセイする工程とを含み、前記第VIII因子及び/又は前記第VIII因子活性の検出が前記PEG化組換え第VIII因子の存在を示し、前記PEG化組換え第VIII因子が第VII因子及び/又は第VIIIa因子である、上記方法。
- 前記アッセイする工程が、定性アッセイ又は定量アッセイを使用して実施され;又は前記アッセイする工程が、インビトロアッセイ、細胞ベースのアッセイ、又はインビボアッセイを使用して実施される、請求項1又は10に記載の方法。
- 前記アッセイする工程が、非特異的ポリペプチドアッセイ又は特異的ポリペプチドアッセイを使用して実施され、前記特異的ポリペプチドアッセイが発色アッセイである、請求項1又は10に記載の方法。
- 前記捕捉剤の選択的結合が、中性からアルカリ性のpHで生じる、請求項1又は10に記載の方法。
- 前記組換えポリペプチドが、第VIII因子(FVIII)である、請求項1又は10に記載の方法。
- 請求項1〜14のいずれか1項に記載の方法を実施するのに有用な1種以上の成分を含むキットであって;前記1種以上の成分が、1種以上の捕捉剤、1種以上の固相支持体、並びに/又は組換えポリペプチドの存在及び/若しくは活性を検出するのに必要な1種以上の試薬を含む、前記キット。
