Description
直径が3インチ(76.2mm)、厚みが500μmのガラスウェハを沸騰させた後、29%NH4OH:31%H2O2:純水=1:1:4の比で混合した溶液中で15分間洗浄した。沸騰した純水中で15分間ずつ2回洗浄したのち、自然乾燥させ、基板の土台とした。
大腸菌E.coli K−12 eGFP発現菌、及びDsRED発現菌の二種類の菌がそれぞれフルグロースした培養液を、1:1で混合し、2×108〜2×109/mlの混合菌液とした。混合菌液8mlが入ったシャーレに実施例1で得られた滅菌したデバイスの基板部を10秒浸漬した後、デバイスを取り出し、エアブロウで表面に残った液滴を除去した。デバイスに再度カバーし、パラフィンフィルムで密封してから、37℃、18時間で培養した。
Claims (13)
- 片側に複数の凹部が配置された基板と、
前記複数の凹部に充填されたゲル状培地と
を備える細胞培養用デバイスであって、
前記複数の凹部の内表面を除いた前記基板の前記片側の表面の少なくとも一部の、JIS 3257(1999)に従って測定した接触角が50°以上であり、
前記基板の前記凹部が配置された領域において、前記凹部の単位面積当たりの数が、1,000個/cm 2 〜10,000個/cm 2 であり、
前記基板の前記片側に、さらにカバー部材を備え、前記カバー部材は前記基板に接していない、細胞培養用デバイス。 - 前記凹部の容積が1pl〜1mlである、請求項1に記載の細胞培養用デバイス。
- 前記凹部の開口部の径が10μm〜500μmであり、前記凹部の高さが10μm〜500μmである、請求項1又は2に記載の細胞培養用デバイス。
- 前記複数の凹部同士の間に、互いに少なくとも1μmの間隔がある、請求項1〜3のいずれか一項に記載の細胞培養用デバイス。
- 前記複数の凹部が、前記基板の前記凹部が配置された領域において、同じ大きさを有し、かつ、等間隔に配置されている、請求項1〜4のいずれか一項に記載の細胞培養用デバイス。
- 前記ゲル状培地が、ゲル化材料及び培地成分を含む、請求項1〜5のいずれか一項に記載の細胞培養用デバイス。
- 前記ゲル化材料が、寒天、アガロース、アルギン酸、コラーゲン、ゼラチン、セルロース及びMatrigel(登録商標)からなる群より選択される、請求項6に記載の細胞培養用デバイス。
- 前記基板の一部は、ガラス、樹脂、金属、セラミックス及び耐水紙からなる群より選択される材料から構成されている、請求項1〜7のいずれか一項に記載の細胞培養用デバイス。
- 前記接触角が50°以上である表面が、シランカップリング剤又はフッ素樹脂によって表面処理された表面である、請求項1〜8のいずれか一項に記載の細胞培養用デバイス。
- 前記カバー部材と前記基板との間に10μm〜100mmの距離がある、請求項1〜9のいずれか一項に記載の細胞培養用デバイス。
- 前記カバー部材と前記基板とが周縁部において密封されている、請求項1〜10のいずれか一項に記載の細胞培養用デバイス。
- 前記カバー部材は、ガラス、樹脂、金属、セラミックス及び耐水紙からなる群より選択される材料から構成されている、請求項1〜11のいずれか一項に記載の細胞培養用デバイス。
- 前記複数の凹部同士の間に、表面以外の任意の位置に凹部同士を連通する連通孔をさらに備える、請求項1〜12のいずれか一項に記載の細胞培養用デバイス。
