治療法で、前記薬剤学的組成物は、癌の疑いのある個体に薬剤学的に有効な量で投与されてもよい。前記個体は、犬、牛、馬、ウサギ、マウス、ラット、鶏及びヒトのような哺乳類であってもよいが、これに限定されるのではない。薬剤学的組成物は、非経口的に、皮下に、腹腔内に、肺内にまたは、鼻内に、必要ならば、局所治療のために病変内投与を含む適切な方法によって投与されてもよい。本発明の薬剤学的組成物の好ましい服用量は、個体の健康状態及び体重、疾患の重症度、薬剤の種類、投与経路及び投与時間を含んだ多様な要因により変わり、本分野の当業者によって容易に決定される。

HUVEC(Human umbilical vein endothelial cells、Lonza, Baltimore, MD, USA)をEGM−2(endothelial growth medium-2)に置いた。MAEC(Mouse aortic endothelial cells)及びCOS−7細胞は、10%(v/v)FBS(fetal bovine serum)及び1%(v/v)ペニシリン/ストレプトマイシンを含んだDMEM(Dulbecco's modified Eagle medium)中で培養した。細胞を37℃及び5%CO2で湿度及びCO2が調節されたインキュベーター((Sanyo, Panasonic Healthcare Company, Secaucus, NJ, USA)に維持した。HEK293F細胞は、湿度調節されたMultitronインキュベーション振とう機(Infors HT, Bottmingen (Switzerland))中に1%(v/v)ペニシリン/ストレプトマイシンを添加したFreestyleTM293発現培地(Invitrogen)で37℃及び8%CO2で維持した。Lipofectamine2000(Invitrogen, Carlsbad, CA, USA)を用いて製造者の指示に従って、GFP、clec14a−GFP(野生型clec14a)またはclec14aΔCTLD−GFP(clec14a CTLD欠失変異体)をコードするベクターでHUVECs及びCOS−7細胞をトランスフェクションした。
プライマー5’−TCGCGCGGCCGCTGCTCGGCCTCGGGGGCCTGC−3’(配列番号1)及び5’−TCGCCTCGAGCTTGCACAGGTAGCCGTTGG−3’(配列番号2)を用いてhCTLDの31−172番アミノ酸残基をコードするDNAを増幅した。mCTLDの同じ残基をコードするDNAは、プライマー5’−TCGCGGCCCAGGCGGCCTGTTCGGCCTCGGGGGCTTG−3’(配列番号3)及び5’−TCGCGGCCGGCCTGGCCCTTGCATAGGTAGCCATCGG−3’(配列番号4)を用いて増幅した。SfiI(NEB, Ipswich, MA, USA)を処理してPCR断片を作製して、ヒンジ及びヒトIgG1のCH2−CH3ドメインをコードする変形哺乳動物発現ベクターpCEP4(Invitrogen)の3'部位のクローニングサイトにクローニングした(gift of Dr. Chung, Seoul National University, Seoul, South Korea)。ライゲーションされた産物を反応性Escherichia coli DH5α細胞に形質転換し、プラスミドDNAを作製した。1.5mgポリエチレンイミン(polyethylenimine: Polysciences, Inc., Warrington, PA, USA)を用いて、各DNA0.75mgでHEK293F細胞(6×108cells)をトランスフェクションした。トランスフェクションされた細胞を1%(v/v)ペニシリン/ストレプトマイシンが添加されたFreestyleTM293発現培地に維持した。7日間培養した後、培養培地を回収して、タンパク質Aセパロード(RepliGen, Waltham, MA, USA)の親和度クロマトグラフィーを介して融合タンパク質を精製した。
NanoDrop分光光度計(Wilmington, DE, USA)を用いてタンパク質濃度を定量した。サンプルをPBSで透析してSDS−PAGE及びクマシーブリリアントブルー染色(Coomassie brilliant blue staining)を介して分析した。最終収集された分画の分液(aliquot)を−80℃に保管した。
ヒト合成scFvライブラリー(Barbas CF. Phage display: a laboratory manual. Cold Spring Harbor, N.Y.: Cold Spring Harbor Laboratory Press; 2001)を再増幅した。Fcバインダーを事前除去するために、タンパク質Aセパロード上にヒトイムノグロブリン−G(Green Cross Pharma Derivatives Corp, Yong-in, Korea)を固定させて、37℃で抗体−タンパク質A複合体を2時間インキュベーションした。簡単な遠心分離後、上澄液を収集して、ペレットは除去した。このような過程を2回繰り返した。最終上澄液をファージELISAで分析して除去の程度を評価した。
4μg組換えヒト(hCTLD−Fc)またはマウス(mCTLD−Fc)融合タンパク質でコートされたイムノチューブ(ImmunoTM tube maxisorp, Nunc, Rochester, NY, USA)または、マグネチックビーズ(Dynabeads M-270 epoxy, Invitrogen)で3回のバイオパンニングを行って、Barbas CF. Phage display: a laboratory manual. Cold Spring Harbor, N.Y.: Cold Spring Harbor Laboratory Press; 2001; Vestweber D. Lymphocyte trafficking through blood and lymphatic vessels: more than just selectins, chemokines and integrins. European Journal of Immunology. 2003; 33(5): 1361-4に記述された通り、種交差反応性を持つクローンを選別した。アウトプットプレートで育ったコロニーから無作為で96個のファージクローンを選別して、ファージ酵素免疫アッセイを介してヒトとマウスCTLDに対する反応性をテストした。最終scFvクローンのDNAをシーケンスして、異なる相補性決定領域配列を持つと確認された四つのscFvクローンに分類した。
プライマー5’−CGGGAATTCGCCGCCACCATGGAATGGAGCTGGGTCTTTCTCTTCTTCCTGCTGTCAGTAACTACAGGTGTCCTCTCCGAGGTGCAGCTGTTGGAGTCTG−3’(配列番号5)及び5’−GGCGGGCCCTTGGTGGAGGCTGAGCTCACGGTGACCAGTGCCCTTGGCCCC−3’(配列番号6)を用いて選別されたscFvクローン(クローン1−4)の重鎖可変領域(VH)遺伝子を増幅した。クローンの軽鎖可変領域(VL)は、プライマー5’−CCCAAGCTTGCCGCCACCATGGAGACACATTCTCAGGTCTTTGTATACATGTTGCTGTGGTTGTCTGGTGTTGAAGGACCAGTCTGTGCTGACTCAGCC−3’(配列番号7)及び5’−GGCCGTACGTAGGACCGTCAGCTTGGTGCCTCCGCCTAAGACATAACCACC−3’(配列番号8)を用いて増幅した。5'及び3'両末端にEcoRI及びApaI制限酵素サイトを追加して、VHプライマーを設計した。両末端にHindIII及びBsiWIサイトを追加して、VLプライマーを設計した。PCR断片を適切な制限酵素(NEB, Ipswitch, MA, USA)で処理して、ヒンジ及びヒトlgG1のCH2−CH3ドメインをコードするpCDNA3.1(bicistronic mammalian expression vector, Invitrogen)(gift of Dr. Hong, Kangwon National University, Chuncheon, Kangwon, South Korea)の3'VHクローニングサイトにクローニングした(Sambrook J, Russell DW. Molecular cloning: a laboratory manual. 3rd ed. Cold Spring Harbor, N.Y.: Cold Spring Harbor Laboratory Press; 2001)。Clec14a−CTLD IgGを産生して、Kim HY, Tsai S, Lo SC, Wear DJ, Izadjoo MJ. Production and characterization of chimeric monoclonal antibodies against Burkholderia pseudomallei and B. mallei using the DHFR expression system. PLoS ONE [Electronic Resource]. 6(5): e19867に記述された通り精製した。
製造者の指示に従って、CytoSelectTM24−well創傷治癒アッセイキット(wound healing assay kit: Cell Biolabs Inc, San Diego, CA, USA)を用いた。簡単に、創傷治癒挿入口の一番上に開放された末端を介してピペットチップを注意深く挿入して、GFPでトランスフェクションされたCOS−7細胞、clec14a−GFPまたはclec14aΔCTLD−GFPを各ウェルに入れた。細胞が育って合流すれば、挿入口をウェルから除去して、細胞をPBS(phosphate-buffered saline)で2回洗浄した。損傷20時間が過ぎた後、イメージをキャプチャーした。
内皮細胞移動性を分析するために、単一層が形成されるまでHUVECを挿入口で培養して、20μg/mLのclec14a−CTLD IgGまたはセツキシマブの存在あるいは不在下で9時間の間37℃でインキュベーションした。細胞を2回PBSで洗浄して、クリスタルバイオレット(Sigma, St. Louis, MO, USA)で染色した。定量分析のために、光学顕微鏡(Nikon TS 100, Melville, NY, USA)下でプレート当たり5個のフィールドを撮影して、移動距離をマニュアルに従って測定した。
Lee S, et al., Hydrogen peroxide increases human leukocyte adhesion to porcine aortic endothelial cells via NFkappaB-dependent up-regulation of VCAM-1. Int Immunol. 2007 Dec; 19(12): 1349-5926に記述された通り、免疫細胞化学を行った。簡単に、GFP、clec14a−GFP、またはclec14aΔCTLD−GFPでトランスフェクションされて、1日間コラーゲンでコートされたカバースリップで成長したCOS−7細胞及びHUVECを30分間37℃で4%(w/v)パラホルムアルデヒドで固定した。細胞をPBSで2回洗浄して、5%(w/v)BSA(bovine serum albumin)及び0.1%TX−100を含んだPBSで1時間37℃でインキュベーションして遮断した。0.1unit/wellローダミンパロイジン及び2μg/mLヘキストと共に1時間細胞をインキュベーションして、蛍光顕微鏡(Leica DM2500, Wetzlar, Germany)下で観察した。
PBSに含まれた25nMの組換えhCTLD−Fc、mCTLD−FcまたはIgG1のFc断片をマイクロタイタープレートの96ウェルに入れた。37℃で一夜インキュベーションした後、0.05%(v/v)Tween20(PBST)を含んだPBSで3回洗浄して、PBSTに3%(w/v)BSAを含んで1時間37℃でインキュベーションした。以後、プレートを3%(w/v)BSAを含んだPBSTに67nM clec14a−CTLD IgGを含み、3時間37℃でインキュベーションした。プレートをPBSTで2回洗浄して、3%(w/v)BSAを含むPBST中でHRP(horseradish peroxide)が接合された抗−ヒトラムダ軽鎖抗体(Bethyl Laboratories, Montgomery, TX, USA)(1:1000に希釈)と1時間37℃でインキュベーションした。
6−wellマイクロタイタープレートで育ったHUVEC(3×105)を20ng/mL hTNFα(Millipore, Billerica, MA, USA)、または、20μg/mL clec14a−CTLD IgGまたはセツキシマブの存在または不在下で24時間インキュベーションした。細胞を収穫して、フローサイトメトリーバッファー中で1時間37℃で20μg/mL抗−VCAM−1またはICAM−1ポリクローナル抗体と共に染色した。細胞をフローサイトメトリーバッファーで3回洗浄して、1000Xgで10分間遠心分離して、フローサイトメトリーバッファー中でAlexa Fluor 488でラベリングされた抗−ウサギ抗体(1:1000;Jackson ImmunoResearch)と1時間37℃でインキュベーションした。

Biochemical & Biophysical Research Communications. 404(1): 103-8に記述された通り、チューブ形成アッセイを行った。簡単に、250μL Matrigel(BD Biosciences, Bedford, MA)を24−wellプレートのウェルに入れて、37℃で20分間重合されるようにした。EGM−2で培養されたHUVECを収穫して、EGM−2に再懸濁した後、Matrigel(1×105cells/well)に接種した。培養物をclec14a−CTLD IgGまたはセツキシマブの存在または不在下で37℃でインキュベーションして、21時間後撮影した。チューブはマニュアルに従ってカウントした。
HUVEC(104)を96−wellマイクロタイタープレートに接種して、20μg/mL clec14a−CTLD IgG、セツキシマブまたは5−FU(Sigma)と2日間37℃でインキュベーションした。細胞生存能は、セルカウンティングキット−8(Cell Counting Kit-8:Dojindo, Kumamoto, Japan)を用いて製造者の指示に従って測定した。VICTORTMX4分光光度計(Perkin Elmer)を用いて450nmで吸光度を測定した。
細胞−細胞接触法をRho SS, et al., Clec14a is specifically expressed in endothelial cells and mediates cell to cell adhesion. Biochemical & Biophysical Research Communications. 404(1): 103-8に記述された通り行った。簡単に、懸濁液のうち1.5×107HEK293F細胞をGFP、Clec14a−GFP及びClec14aΔCTLD−GFPを発現するプラスミドでトランスフェクションして、FreestyleTM293発現培養液で一夜の間培養した後、6ウェルプレートに接種した(5×105cells/well)。20μg/mL clec14a−CTLD IgGまたはセツキシマブの存在または不在下で8時間細胞を維持した。細胞凝集体(mass>4cells)を10個以上のフィールドでカウントして、平均±標準偏差(SD)を計算した。
Van Meter KE, et al., A monoclonal antibody that inhibits translation in Sf21 cell lysates is specific for glyceraldehyde-3-phosphate dehydrogenase. Archives of Insect Biochemistry & Physiology. 2008; 69(3): 107-17に記述された通り免疫ブロット分析を行った。簡単に、12%ポリアクリルアミドゲルを介して電気泳動してGFP−、clec14a−GFP−、またはclec14aΔCTLD−GFPを分離した。ウェットトランスファーシステム(wet transfer system: GE Healthcare Life Sciences, Pittsburgh, PA, USA)を用いてタンパク質をニトロセルロース膜に移動した。5%(w/v)脱脂乳を含み、10mM Tris/HCl、pH7.5、150mM NaCl、及び0.05(v/v)%Tween20(TTBS)で1時間室温でインキュベーションした後、同じ溶液で抗−GFP単クローン抗体(1:5000;Santa Cruz Biotechnology, Santa Cruz, CA, USA)または、抗−β−アクチン単クローン抗体(1:5000;Applied Biological Materials, Richmond, BC, Canada)と共に4℃で一夜でインキュベーションした。TTBSで膜を洗浄して、5%(w/v)脱脂乳を含むTTBSでHRP−接合されたアフィニピュアヤギ抗−マウスIgG(HRP-conjugated Affinipure goat anti-mouse IgG)(1:5000;Jackson ImmunoResearch, West Grove, PA, USA)と共に1時間室温でインキュベーションした。TTBSで数回洗浄した後、製造者の指示に従って、SuperSignal West Pico Chemiluminescent Substrate(Pierce, Rockford, IL, USA)を用いてタンパク質バンドを視覚化した。
96ウェルプレートに置かれたHUVECs(104)を20μg/mL clec14a−CTLD IgGまたはFcと共に0、10、30、60、120、または、180分間37℃でインキュベーションした。細胞を氷冷PBSで2回洗浄して、1時間4℃で3%(w/v)BSA含むPBSで遮断した後、遮断溶液中でヒツジ抗−clec14aポリクローナル抗体(1:1000)と2時間4℃でインキュベーションした。氷冷PBSで細胞を3回洗浄して、HRPと接合された抗−ヒツジIgG(1:5000;Santa Cruz Biotechnology)と1時間4℃でインキュベーションした。PBSで数回洗浄後、100μL TMB基質溶液を各ウェルに追加した。マイクロタイタープレートリーダーを用いて、450nmで光学密度を測定した。
PBSにある10μg/mL hCTLD−Fc、FcまたはhCTLD−FcのN−末端またはC−末端断片の各々を96ウェルプレートに入れた。プレートを37℃で一夜でインキュベーションして、0.05%(v/v)Tween20(PBST)を含むPBSで3回洗浄した後、3%(w/v)BSAを含んだPBSTと1時間37℃でインキュベーションした。3%(w/v)BSAを含むPBST中10μg/mL clec14a−CTLD IgGと共に2時間37℃でプレートをインキュベーションした。PBSTで2回洗浄した後、3%(w/v)BSAを含むPBST中HRPで接合された抗−ヒトラムダ軽鎖抗体(1:1000;Bethyl Laboratories, TX, USA)を1時間37℃でインキュベーションした。
250μLのMatrigel(BD Biosciences)を24ウェルプレートに入れて、20分間37℃で重合されるようにした。EGM−2で培養されたHUVECを収穫して、EGM−2に再懸濁した後、Matrigelに接種した(1×105cells/well)。10μg/mL及び25μg/mL hCTLD FcまたはFcの存在または不在下で培養物を37℃インキュベーションして、8時間後撮影した。
clec14a媒介細胞移動でのCTLDの役割を説明するために、COS−7細胞をGFP、clec14a−GFP(wild-type clec14a fused to GFP)またはclec14aΔCTLD−GFP(clec14a CTLD deletion mutant fused to GFP)でトランスフェクションして、0時間及び20時間となった時創傷治癒アッセイでの移動を分析した。野生型clec14aを発現する細胞の移動程度は、GFP単独発現細胞に比べて約1.6倍高い反面、clec14aΔCTLD欠失変異体の発現は、移動にほとんど影響を与えなかった(図1A及び1B)。
細胞移動は、アクチン細胞骨再配列によって精巧に調節されるので、免疫細胞化学を用いてGFP、clec14a−GFPまたはclec14aΔCTLD−GFPを発現するCOS−7細胞及びHUVECでのアクチンネットワーク組織を調べた。二つの細胞類型で共に、clec14a−GFP発現は、顕著にフィロポジウム形成を増加させた反面、GFPまたはclec14aΔCTLD−GFPは最小の影響を示した(図2A及び2B)。
まとめると、このような結果は、アクチン細胞骨格再配列を介してCTLDが内皮細胞移動に重要な役割を果たすことができることを示す。
ヒト、マウスまたはチンパンジーclec14a(表1)のCTLDは、Fc融合タンパク質であるため、ヒト合成scFvライブラリーからFcバインダーを除去するために、2工程の連続的事前除去過程を行った;ファージ酵素結合免疫吸着法によって約90%が除去されたことを確認した。

scFvクローンをIgGに変換して、HEK293F(human embryonic kidney 293F)細胞で発現した後、精製した。四つのIgGクローンは、SDS−PAGE及びクマシー染色によって90%超の純度であることを示した。ELISAは、精製されたCTLD特異的IgG(clec14a−CTLD IgGs)がヒト及びマウスCTLD−Fc二つに共に特異的に結合するが、Fc単独はそうでないことを示した。クローン1及び2は、クローン3及び4よりはるかに大きい親和度を示した(図4)。
clec14a−CTLD IgGが内皮細胞移動に阻害作用をするか否か調べるために、HUVECと共にclec14a−CTLD IgGの存在または不在下で創傷治癒アッセイを行った。セツキシマブ、抗−EGFR抗体を対照IgGとして用いた。選別されたclec14a−CTLD IgG中、クローン1及び2は、各々約44%及び54%でHUVEC細胞移動を顕著に抑制した反面、クローン3及び4とセツキシマブ単独は、ほとんど効果がなかった(図5A及び5B)。
clec14a−CTLD IgGがチューブ形成に及ぼす影響を確認するために、clec14a−CTLD IgG及びセツキシマブの存在または不在下でチューブ形成をアッセイした。クローン1及び2とセツキシマブは、前述の通りHUVECチューブ形成を顕著に遮断する反面、残りのクローンは大きな影響がなかった(図5C及び5D)。
このような結果は、clec14a−CTLD IgGが腫瘍血管新生を顕著に抑制することを強く示す。
clec14a−CTLD IgGが、内皮細胞生存能に及ぼす影響を調べるために、clec14a−CTLD IgG、セツキシマブまたは5−FU(5-fluorouracil)、アポトーシス誘導剤存在または不在下で2日間HUVECを培養して、細胞カウンティングキットを用いて細胞生存の可能性を確認した。抗体は、HUVEC生存能に影響をほとんど及ぼさない反面、5−FUは特異的に生存能を減少させた(図6A)。
内皮細胞活性にclec14a−CTLD IgGが及ぼす影響を確認するために、clec14a−CTLD IgGs、セツキシマブまたはhTNFα(human tumor necrosis factor alpha)の存在または不在下でHUVECを培養した。活性は、フローサイトメトリーによって測定されたVCAM−1(vascular cell adhesion molecule-1)及びICAM−1(intercellular cell adhesion molecule-1)の発現に基づいて決定された。細胞をhTNFαで処理して陽性対照群でVCAM−1及びICAM−1の上向き調節を誘導した。抗体及びセツキシマブは、HUVEC活性に影響が殆どなかった(図6B)。
まとめると、このような結果は、clec14a−CTLD IgGが内皮細胞生存能または活性に影響をほとんど及ぼさないことを示す。
内皮細胞−細胞接触でclec14a−CTLD IgGの役割を確認するために、GFPまたはclec14a−GFPでトランスフェクションされたHEK293F細胞によって形成されて、clec14a−CTLD IgGまたはセツキシマブの存在または不在下で培養されて、clec14a媒介細胞−細胞接触のマーカーである細胞凝集体の数を測定した。細胞凝集体の数は、GFP単独発現細胞よりclec14a−GFP発現細胞で約4倍以上多かった(図7A及び7B)。さらに、クローン1及び2は、clec14a−GFPでトランスフェクションされた細胞の凝集を大きく抑制した反面、他のクローン及びセツキシマブはほとんど影響がなかった。
まとめると、このような結果は、clec14a−CTLD IgGは、腫瘍血管新生の間clec14a媒介内皮細胞−細胞接触で阻害作用を行うことができることを示す。
免疫分析を用いて、トランスフェクションされたCOS−7細胞の溶解物中でGFP、clec14a−GFP及びclec14aΔCTLD−GFPの発現を確認した(図8A)。以後、溶解物をhCTLD−FcまたはFcと共にインキュベーションして、CTLDの間の相互作用を観察した。hCTLD−Fcが、COS−7細胞溶解物中でいくつかの他のタンパク質と相互作用できても、clec14aΔCTLD−GFPでないclec14a−GFPに強く結合しており、これは内皮細胞で特異的CTLD−CTLD相互作用を示す(図8B)。
clec14a−CTLD IgGが、CTLD−CTLD相互作用を遮断するのか否かを決めるために、hCTLD−Fcは増加する濃度のclec14a−CTLD IgG(クローン1)とインキュベーションして、タンパク質複合体は、GFP−またはclec14a−GFP−トランスフェクションされた細胞の溶解物とインキュベーションした。競争的ELISAを介して、clec14a−CTLD IgGが、濃度依存的方式でCTLD−CTLD相互作用を特異的に阻害することを示し(図8C)、これはclec14a−CTLD IgGが内皮細胞でclec14a CTLD−CTLDを特異的に遮断できることを示す。
新生血管形成表現型(pro-angiogenenic phenotype)を抑制するのに、clec14a−CTLD IgGの可能な役割を調べるために、フローサイトメトリー法を用いてclec14a−CTLD IgG(クローン1)で処理前後にHUVEC膜でclec14a発現レベルを比較した。IgG処理時生細胞でclec14aは顕著に減少したが、パラホルムアルデヒドに固定された細胞はそうではなかった(図9A)。
clec14a−CTLD IgGによってHUVEC膜でのclec14a下向き調節を確認するために、細胞をIgG(クローン1)またはFcで処理して、clec14a膜を細胞ELISAで測定した。Clec14a−CTLD IgGは、HUVEC膜でclec14aを時間依存的方式で下向き調節した(図9B)。
このような結果は、clec14aに対するclec14a−CTLD IgGの交差反応が内皮細胞膜でのclec14aを特異的に下向き調節することを強く示す。また、このような結果は、clec14a−CTLD IgGが癌細胞のようなclec14a発現細胞に内在化できることを示す。
エピトープの細部マッピングのために、ELISAでClec14awtCTLD及びhCTLD−Fc断片に対するclec14a−CTLD IgGの特異度(図10A)を試験した。
その結果、図10Bに示した通り、クローン1及び2二つ共にclec14a−CTLD IgGは、wthCTLDだけでなく、hCTLDのN−末端及びC−末端に結合した。
このような結果は、CTLDのN−末端またはC−末端に、例えばクローン1及び2における全てのclec14a−CTLD IgGのように、hCTLDで1番アミノ酸〜42番アミノ酸のアミノ酸断片を含む融合タンパク質、hCTLDで1番アミノ酸〜62番アミノ酸のアミノ酸断片を含む融合タンパク質、hCTLDで82番アミノ酸〜142番アミノ酸のアミノ酸断片を含む融合タンパク質、hCTLDで62番アミノ酸〜142番アミノ酸のアミノ酸断片を含む融合タンパク質、hCTLDで122番アミノ酸〜142番アミノ酸のアミノ酸断片を含む融合タンパク質に特異的に結合して、エピトープとして用いられる可能性があることを強く示す。
血管新生にhCTLD−Fcの抑制効果を確認するために、実施例16によりhCTLD−FcまたはFcの存在下でチューブ形成をアッセイした。
その結果、図11に示した通り、hCTLD−Fcが濃度依存的方式でチューブ形成を抑制した。
このような結果は確かにhCTLD−Fcが腫瘍血管新生を特異的に抑制することを示している。
具体的な構成を参照して本発明を詳細に記載したが、このような記載は、好ましい具現例に関しており、発明の範囲を制限しないことは、当業者にとっては自明である。よって、本発明の実質的範囲は、出願された請求項及びその等価物によって定義される。

- 前記血管新生関連疾患は、癌、転移、糖尿網膜病症、未熟児網膜病症、角膜移植拒否、黄斑変成、血管新生緑内障、全身紅色症、増殖性網膜症、乾癬、血友病性股関節炎、動脈硬化性プラークの毛細血管形成、ケロイド、創傷肉芽、血管癒着、リューマチ関節炎、退行性関節炎、自己免疫疾患、クローン病、再狭窄、粥状動脈硬化症、腸狭窄、猫引っかき病、潰瘍、肝硬変症、腎臓炎、糖尿病性腎臓疾患、真性糖尿病、炎症疾患及び神経変成疾患で構成された群から選択されることを特徴とする請求項16に記載の薬剤学的組成物。
- 前記癌は、食道癌、胃癌、大腸癌、直膓癌、口腔癌、咽頭癌、喉頭癌、肺癌、結腸癌、乳癌、子宮頸部癌、子宮内膜癌、卵巣癌、前立腺癌、睾丸癌、膀胱癌、腎臓癌、肝臓癌、すい臓癌、骨癌、結合組織癌、皮膚癌、脳腫瘍、甲状腺癌、白血病、ホジキンリンパ腫瘍、リンパ腫及び多発性骨髄血液癌で構成された群から選択されることを特徴とする請求項17に記載の薬剤学的組成物。
- 請求項1〜7のいずれか一項に記載の抗体を含む血管新生抑制用組成物。
- clec14aのCTLDの単離されたN−末端またはC−末端を含むポリペプチドを、血管新生を抑制する抗体の生産を誘導できるエピトープとして用いてバイオパンニングする工程を含む、請求項1〜7のいずれか一項に記載のclec14a(C−タイプレクチンドメインファミリー14、メンバーA)のCTLDに特異的に結合する抗体を製造する方法。
