Description
a)間葉系幹細胞の培養のために通常用いられる基礎培地
b)ヒト白血球/血小板コート溶解物(バフィーコート;LBC)
c)インスリン
d)亜セレン酸ナトリウム
e)エタノールアミン
f)塩基性線維芽細胞成長因子(bFGF)。
(実施例1)
基礎培地としてDMEM/F12+ピルビン酸塩、10%(v/v)ヒト白血球/血小板コート溶解物(バフィーコート)、10mg/Lのインスリン、0.0067mg/Lの亜セレン酸ナトリウム、2mg/Lのエタノールアミンおよび20ng/mLのbFGFを含有する、本発明による培養培地を調製した。この培地をLV2と称した。培地LV2は単独で用いたか、間葉系幹細胞(MSCQ)に適する10%(v/v)のウシ胎児血清(FBS)、またはヒト血漿に由来する15%(v/v)の細胞培養添加物(CCS)を追加した。Lonza(Basel、Switzerland)またはPromoCell(Heidelberg、Germany)から市販されているhMSCを培養した。製造業者によって推奨される培養培地を、陽性対照として用いた。1つの市販基礎培地だけを、陰性対照として用いた。結果をTable 1(表1)に示す。
本発明による培養培地の全ての構成成分を組み合わせることの効果を判定するために、LV2培養培地を実施例1に記載のように調製し、これに加えて、本発明による培養培地の少なくとも1つの構成成分を欠く様々な培地も調製した。調製した培地、および得られた結果をTable 2(表2)に示す。
Claims (13)
- ヒト間葉系細胞のための培養培地であって、
a)DMEM/F12+ピルビン酸塩である間葉系幹細胞基礎培地、
b)バフィーコート、
c)インスリン、
d)亜セレン酸ナトリウム、
e)エタノールアミン
f)塩基性線維芽細胞成長因子(bFGF)および
g)ウシ胎児血清(FBS)またはヒト血漿に由来する細胞培養添加物(CCS)
を含むことを特徴とする、培養培地。 - 前記バフィーコートが培養培地中に5から20%(v/v)の量で存在することを特徴とする、請求項1に記載のヒト間葉系細胞のための培養培地。
- 前記バフィーコートが培養培地中に7から15%(v/v)の量で存在することを特徴とする、請求項2に記載のヒト間葉系細胞のための培養培地。
- 前記インスリンが2から20mg/Lの量で存在することを特徴とする、請求項1から3のいずれか一項に記載のヒト間葉系細胞のための培養培地。
- 前記インスリンが5から15mg/Lの量で存在することを特徴とする、請求項4に記載のヒト間葉系細胞のための培養培地。
- 前記亜セレン酸ナトリウムが0.005から1.730mg/Lの量で存在することを特徴とする、請求項1から5のいずれか一項に記載のヒト間葉系細胞のための培養培地。
- 前記エタノールアミンが1から8mg/Lの量で存在することを特徴とする、請求項1から6のいずれか一項に記載のヒト間葉系細胞のための培養培地。
- 前記エタノールアミンが2から5mg/Lの量で存在することを特徴とする、請求項7に記載のヒト間葉系細胞のための培養培地。
- 前記bFGFが5から25ng/mLの量で存在することを特徴とする、請求項1から8のいずれか一項に記載のヒト間葉系細胞のための培養培地。
- 抗生物質をさらに含むことを特徴とする、請求項1から9のいずれか一項に記載のヒト間葉系細胞のための培養培地。
- 前記抗生物質がゲンタマイシン/アンホテリシンの混合物であることを特徴とする、請求項10に記載のヒト間葉系細胞のための培養培地。
- 10%から20%(v/v)の量でウシ胎児血清(FBS)が存在することを特徴とする、請求項1から11のいずれか一項に記載のヒト間葉系細胞のための培養培地。
- 10%から40%(v/v)の量でヒト血漿に由来する細胞培養添加物(CCS)が存在することを特徴とする、請求項1から11のいずれか一項に記載のヒト間葉系細胞のための培養培地。
