Claims (13)
- 細胞マーカーに基づいて細胞を分離し、細胞および人工組織を成長させるためのバイオリアクターシステムであって、
第1部分および第2部分を有する内側を含み、細胞マーカーによる細胞分離、細胞成長および人工組織の成長の反応のための空間を与える1つの反応チャンバーと、
前記チャンバーの内側に配置され、内側の第1部分と内側の第2部分の間を移動することができる、磁着性アジテーターと、
細胞マーカーによる細胞分離を補助し、チャンバーの特定の内側位置にアジテーターをとどまらせ、または反応チャンバーの反転にともなって内側の第1部分と内側の第2部分の間を移動させるように操作可能な、調節可能な磁場と、を少なくとも備え、
前記反応チャンバーの内側が、
細胞培地容器と流体が行き来できるように連結し、この容器から細胞培地を受け入れ、消し去ることができ、低コロイド浸透圧溶液を含む、低浸透区域である第1の区画と、
第1の区画に隣接し、さらに、細胞、アジテーター、試薬、バッファーおよび培地を受け入れ、消し去るための出入口になるように作られ、その内部にアジテーターが配置されている、反応区画である第2の区画と、
第1の区画と第2の区画の間に配置され、第1の区画から第2の区画へと細胞培地を選択的に流すことができるように作られた、第1の膜と、
第2の区画に隣接し、第3の区画が、バッファー容器からバッファーを受け入れることができるようにバッファー容器と流体が行き来できるように連結し、さらに、廃棄物が第3の容器から廃棄物容器へと流れることができるように廃棄物容器と流体が行き来できるように連結するように作られた、高浸透区画である第3の区画と、
第2の区画と第3の区画との間に配置され、培地および廃棄物を第2の区画から第3の区画まで選択的に流すことができるように作られた、第2の膜と、
気体透過性材料で作られた反応チャンバーの側壁の少なくとも一部、pHを維持するための試薬を含む培地の1構成要素、またはチャンバーと接続した気体透過性システム、またはこれら3つの任意の組み合わせと、
バイオリアクターのカセットおよび任意のホルダーに配置された反応チャンバーの少なくとも一部と、を備える、バイオリアクターシステム。 - 複数のビーズ、または複数の開口部を有する平面部材を備える磁着性アジテーターが、
(1)全体的に剛性磁着性材料で作られ、コーティングされていないか、または細胞の接続および細胞損傷の低減を含む任意の目的のために、不活性で剛性または硬質の材料でコーティングされるか、または
(2)少なくとも部分的に、磁場の中に置かれると磁性を帯びるが、磁場が消えるか、または反応領域からはずされると、磁性ではなくなるか、または磁性が非常に弱くなるような磁着性材料で作られ、且つ
(3)前記ビーズの直径が1ミリメートルより大きく、前記反応チャンバーの体積より小さい、請求項1に記載のバイオリアクターシステム。 - 前記磁着性アジテーターは、接着性の細胞培養物中で細胞を接着させ、成長させるため、また、細胞マーカーによる細胞分離中、磁着性材料で標識された抗体または他の生体分子と結合した細胞を接続するために細胞補助材として機能し、生体分子の反応および結合ならびに磁場によって制御された細胞の捕捉および放出を含む、請求項2に記載のバイオリアクターシステム。
- 前記磁着性アジテーターを用い、反応チャンバー内の任意の2つの部分の間を移動することによって培地をゆっくりと機械撹拌し、(1)周囲にある磁場の引力、(2)アジテーターが反応チャンバー中の培地よりも密度が小さい場合には浮遊性、または(3)ビーズが反応チャンバー中の培地よりも密度が大きい場合には重力といった機構によって液体成長培地中に細胞を懸濁させたままにする、請求項2に記載のバイオリアクターシステム。
- 前記磁着性アジテーターを使用し、細胞成長のための微細環境を作り出し、該微細環境は、前記機構によってそれぞれを動かした後に磁着性アジテーターが沈殿するにつれて、アジテーターの間に空間を含み、細胞成長を妨害し、幹細胞の非特異的な分化を減らし得る任意の過剰な混み合いまたは集塊を防ぐことができる、請求項4に記載のバイオリアクターシステム。
- 前記調節可能な磁場を、
(1)電流および/または電圧を変えることによって、または電磁石の位置および向きを変えることによって、磁場を調節する電磁石によって作ることができるか、
(2)永久磁石の位置および向きを変えることによって磁場を変える永久磁石によって作ることができるか、または

- 前記細胞が、造血幹細胞、間葉幹細胞、線維芽細胞、ハイブリドーマ細胞、リンパ球、樹状細胞、昆虫細胞、胚幹細胞、種々の組織細胞、癌細胞並びに細胞株、形質変換された細胞および細胞株から選択される、請求項1に記載のバイオリアクターシステム。
- 細胞を分離し、細胞および人工組織を成長させるためのバイオリアクターシステムであって、請求項6において調節可能な磁場によって制御されるか、または請求項7において制御システムによって制御される、反応チャンバーの反転およびアジテーターの機械攪拌を組み合わせることによって、細胞およびアジテーターを懸濁状態で保持することができる反応チャンバーおよびアジテーターを備える、請求項6又は7に記載のバイオリアクターシステム。
- 細胞懸濁のために反応チャンバーの反転と細胞の機械攪拌の組み合わせを含む、請求項1に記載のバイオリアクターシステム。
- 細胞および人工組織を成長させることは、低分子を含む灌流溶液が、低浸透区画から反応区画へ、さらに高浸透区画へと単一方向に膜を介して通り抜けるような保護灌流チャンバー中で行われる、請求項1に記載のバイオリアクターシステム。
- 調節可能な磁場を、細胞マーカーによる細胞分離、細胞成長および人工組織の成長の間に、制御システムによって磁場の強度、方向または位置を変えることによって調節することができる、請求項1に記載のバイオリアクターシステム。
- 細胞マーカーによる細胞分離が、
特異的な磁着性抗体または他の生体分子を標的細胞に結合させることと、
磁着性アジテーターまたはチャンバー壁のいずれかによって、または磁着性アジテーターおよびチャンバー壁の両方によって磁気的に標識された細胞を捕捉することと、
非標識細胞を捨てるか、または洗浄することと、
標的細胞を放出させることと、を含む、請求項1に記載のバイオリアクターシステム。
