「キメラペプチド」とは、天然には相互に関連せず、融合タンパクとしてまたは化学結合によって相互に連結された2個の成分ペプチドを有するペプチドを意味する。
本発明は、Srcとの相互作用に関与するND2のコア領域を、ND2の残基289〜321内、より具体的にはND2の残基310〜321内に特定することに、部分的に基づいている。この領域を含む、この領域に重なる、またはこの領域内からのペプチドは、ND2のSrcとの相互作用を阻害するために使用することができる。この相互作用を阻害することは、神経疾患および神経障害、疼痛ならびに癌の治療および予防に有用である。

細胞膜伝達ペプチドまたは細胞貫通ペプチドとしても知られる内在化ペプチドは、よく知られた、多くの細胞タンパクまたはウイルスタンパクが膜を横断することを可能にする比較的短い(たとえば、5〜30、7〜20または9〜15個のアミノ酸)ペプチドのクラスである。これらのペプチドは、一般に、アルギニンおよび/またはリジン残基の通常以上の表記から(一般のタンパクと比較して)正電荷を有し、それらの膜横断を容易にすると考えられている。これらのペプチドのいくつかは、少なくとも5,6,7または8個のアルギニンおよび/またはリジン残基を有する。その例には、アンテナペディアタンパク(Bonfanti, Cancer Res. 57, 1442−6 (1997))(および、その変異型)、ヒト免疫不全ウイルスのTatタンパク、タンパクVP22、単純ヘルペスウイルスタイプ1のUL49遺伝子の産物、Penetratin、SynB1およびSynB3,Transportan、Amphipathic、gp41NLS、polyArg、ならびに、リシン、モデシン、ジフテリア毒素、コレラ毒素、炭疽毒素、易熱性毒素および緑膿菌外毒素A(ETA)などのいくつかの植物毒素およびバクテリア毒素がある。他の例は、次の参考資料に記載されている(Temsamani, Drug Discovery Today, 9(23):1012−1019, 2004; De Coupade, Biochem J., 390:407−418, 2005; Saalik Bioconjugate Chem. 15: 1246−1253, 2004; Zhao, Medicinal Research Reviews 24(1):1−12, 2004; Deshayes, Cellular and Molecular Life Sciences 62:1839−49, 2005)(すべて参照によって取り込まれる)。
本発明の薬剤は、神経疾患または神経障害、疼痛および癌の治療および効果的な治療に有用である。これらの部類は相互に排他的ではない。たとえば、脳腫瘍は、これら3部類すべてに該当する。
a)治療の方法
随意的に内在化ペプチドに結合した薬剤が、治療的に有効な投与計画において、上述の疾患もしくは障害の兆候および/または症状を有する患者に投与される。この投与計画は、治療中の疾患または症状を患っている患者(または動物モデル)の集団における疾患の少なくとも1つの兆候または症状の、本発明の薬剤での治療を受けていない、その疾患または症状を患っている患者(または動物モデル)の対照集団に対する、有効な治癒、軽減またはさらなる悪化の阻止に有効な量、頻度および投与経路を意味する。この投与計画はまた、個々の治療患者が、本発明の方法により治療されない比較患者の対照集団における平均の結果よりも、良好な結果を達成する場合に、治療上有効であると認められる。投与の回数は、治療する疾患または障害に依存する。脳卒中、外傷性損傷、不安神経症、急性疼痛またはてんかんなどの急性または一過性の症状については、多くの場合、少なくとも1回の発症当たり単回の投与で十分である。神経変性疾患、たとえばアルツハイマー病またはパーキンソン病、癌、慢性疼痛などの慢性の症状については、複数回投与および、時に生涯にわたる治療が指示される。
本発明は、疾患の危険がある対象者における疾患の予防のための方法および組成物を提供する。通常このような対象者は、対照集団と比べて、疾患(たとえば、症状、病気、疾患または障害)の進行する可能性が高まっている。たとえば対照集団は、その疾患と診断されたことがなく、その疾患の家族歴を有しない、一般集団から無作為に選択された1人以上の個人を含む(たとえば、年齢、性別、人種および/または民族性で整合させる)。対象者は、疾患に関連する危険因子がその対象者に関連していることが判った場合、その疾患について危険性があると認められる。危険因子には、たとえば、その対象者の集団の統計的または疫学的研究を通じてその疾患に関連する、あらゆる活動、形質、出来事または性質を含まれ得る。対象者は、したがって、根本的な危険因子を特定する研究が具体的にその対象者を含まなかった場合でも、疾患についての危険性があると分類され得る。たとえば、心臓手術を受ける対象者は、心臓手術を受けたことのある対象者の集団における一過性脳虚血発作の頻度が、心臓手術を受けたことのない対象者の集団と比べて高いので、脳卒中の危険性がある。
本発明の薬剤、随意的に内在化ペプチドに結合された薬剤は、医薬組成物の形態で投与することが可能である。医薬組成物は、一般に、GMP条件下で製造される。医薬組成物は、単位容量の形態(つまり、単回投与のための投与量)で提供することが可能である。医薬組成物は、従来の、混合、溶解、粒状化、糖衣化、粉末化、乳化、カプセル化、封入または凍結乾燥の処理によって製造することが可能である。たとえば、凍結乾燥された薬剤は、以下に述べる剤形および組成で使用することができる。
1.スクリーニングされる薬剤
薬剤は、所望の結合または阻害活性について、まずインビトロでスクリーニングすることが可能である。薬剤には、上述したように、ND2ペプチドまたはそのペプチド模倣体が含まれ得る。スクリーニングされる薬剤は、海洋微生物、藻類、植物、菌類などの天然源から、または合成ペプチドもしくは他の化合物のライブラリーから、得ることが可能である。段階的に合成することが可能な多種類の化合物について、組み合わせライブラリーを創生することも可能である。このような化合物には、ポリペプチド、ベータ−ターン模倣体、多糖、リン脂質、ホルモン、プロスタグランジン、ステロイド、芳香族化合物、複素環化合物、ベンゾジアゼピン、N置換グリシンオリゴマーおよびオリゴカルバメートが含まれる。Affymax,国際公開公報WO95/12608号、 Affymax,国際公開公報WO93/06121号、 Columbia University,国際公開公報WO94/08051号、 Pharmacopeia,国際公開公報WO95/35503号、および、Scripps,国際公開公報WO95/30642号(これらは各々、あらゆる目的のために、参照によって取り込まれる)に記載されているコード化合成ライブラリー(ESL)法によって、化合物の大きな組み合わせライブラリーを構築することが可能である。ペプチドライブラリーは、また、ファージディスプレイ法によって創生することも可能である。たとえば、Devlin,国際公開公報WO91/18980号を参照。
薬剤は、まず、所望の活性について、たとえば、SrcまたはSrcの残基40〜49を含むSrcペプチドへの結合する能力について、スクリーニングすることが可能である。これに代えてまたは加えて、薬剤は、Srcまたはその残基40〜49もしくは残基40〜58を含むペプチドへの結合能において、ND2または上記のND2ペプチド(たとえば、ND2の残基310〜321から成るペプチド)と競合する能力について、スクリーニングすることが可能である。結合は、ELISA、蛍光偏光、またはウェスタンブロットなどの方法によって、評価することができる。いくつかのフォーマットにおいて、それらの結合試験の1つの要素は固定される。実施例において説明するように、いくつかのフォーマットが可能である。Srcに結合するおよび/またはND2もしくはND2ペプチドのSrcへの結合を阻害する薬剤の能力は、その薬剤が、神経疾患、疼痛または癌の治療において有用な薬理活性を有することの指標となる。薬剤は、次いで、以下に説明するように、様々な動物モデルにおいてさらに試験される。
薬剤は、脳卒中の種々の動物モデルで試験することが可能である。それらのモデルの1つにおいて、雄の成体Sprague−Dawleyラットを、管腔内縫合法により一過性の中大脳動脈閉塞(MCAO)に90分間付す。動物を一夜絶食させ、硫酸アトロピン(0.5mg/kg、腹腔内)を注射する。10分後、麻酔が誘導される。ラットに経口で挿管して、機械的に換気し、臭化パンクロニウム(0.6mg/kg、静脈内)で麻痺させる。体温は、加温ランプで、36.5〜37.5℃に維持する。大腿動静脈内でポリエチレンのカテーテルを使用して、血圧を連続して測定し、ガスおよびpH測定のために血液を採取する。一過性MCAOは、ポリ−L−リジン被覆の3−0モノフィラメントナイロン縫合糸(Harvard Apparatus)を、内頸動脈経由でウィリス輪に挿入して、中大脳動脈を効果的に閉塞することにより、90分間行う。これは、大脳皮質および脳幹神経節を包含する広範な梗塞を生じる。動物を、試験対象の薬剤または陰性もしくは陽性対照のいずれかで処理する。処理は、虚血の誘導前または誘導後1時間までのどちらかに、可能である。陰性対照は媒体であり得る。陽性対照は、有効であることが既に示されている、Tat−NR2B9cペプチド、YGRKKRRQRRRKLSSIESDV(配列番号6)であり得る。動物への薬剤の投与後、梗塞ボリュームおよび/または障害指数を決定する。梗塞ボリュームは通常、処理後24時に測定されるが、3,7,14または60日などのより遅い時に測定することも可能である。障害指数は、たとえば、処理後2時、処理後24時、処理後1週または1月の期間にわたって、監視することができる。梗塞ボリュームおよび/または障害指数において、薬剤で処理しない対照動物と比べて統計的に有意な減少を示す薬剤は、本発明の方法を実施するために有用な活性を有すると、確認される。
哺乳動物(たとえば、齧歯動物)における疼痛の侵害受容試験には、テールフリック(脊髄媒介侵害受容反射)試験(D’Amour et al. (1941), J. Pharmacol. Exp. Ther. 72: 74−79)、ホットプレート試験、Randall−Selitto試験(Swingle et al. (1971), Proc. Soc. exp. Biol. Med. 137: 536−538)、およびテールピンチ試験がある。これらの試験は、熱に対する閾値(テールフリック試験、ホットプレート試験、肢引き込みのHargreaves試験、および熱水への尾または後肢の短時間の浸漬)などの異種の有害刺激に対する侵害受容閾値;または、たとえば、異痛試験についてのVon Frey試験、J Neurosci法,1999 Mar 1;87(2):185−93による刺す刺激に対する触知性閾値を評価するために使用することが可能である。動的異痛は、後肢の裏の表皮を綿棒で軽く打ち、打ち始めてから8秒以内に動物が綿の刺激に反応する場合に動的異痛が存在すると判断することによって評価することが可能である。有害な化学剤に対する疼痛応答は、たとえば、希酢酸の腹腔内注射後の腹部ライジングを監視することによって、および、飲み水にカプサイシンを加えることによる嫌悪水飲み試験(これは、三叉神経侵害を評価するのに使用することができる)によって、測定することが可能である。
異なるてんかん状態の広範な動物モデルが、よく特徴づけられている。SeizuresおよびEpilepsyモデル(ed. Pitkanen et al., ISBN: 978−0−12−088554−1; Elsevier Inc., 2006)を参照。この全体が、参照によって取り込まれる。動物は、ショウジョウバエから霊長類までにわたり、ここで、てんかんは、化学物質の投与、または発作および/もしくはてんかんを自発的に発現する検体の遺伝子スクリーニングを含む様々な方法によって、引き起こされる。動物モデルの例には、発熱発作に似たラットの高熱誘導発作、ならびに、トッタラー、スターゲイザー、嗜眠性、および短時間行動停止(つまり、注視または凝視)などのヒト欠神てんかんに類似する特徴を共有する徐波てんかん(SWE)のマウスなどの、マウスの変異が含まれる。
不安神経症は、ラットなどの動物を、なじみのない環境に置いて、反応(たとえば、格子線を横断する、または開いたもしくは閉じたチューブを選択する)を観察することによって誘導される。たとえば、覚醒の状態および新しい環境に慣れる能力の双方を明らかにするために、ラットをオープンフィールドアリーナにおいて試験して、格子線を横断することから評価することができる。横断の減少は不安感の低下を示す。ラットを迷路で試験して、ラットにおける不安感/情動性を評価することもできる。好適な迷路は、中央のプラットホームから延びて床からの高さが1.5mの4本のアーム(2本は開き、2本は閉じている:幅15cmで長さ60cm)を有する。ラットを迷路の中央に置き、どのアームに入るかを自由に選択できるようにする;運用上、入るとは、頭と前肢とをアーム内に入れることと規定する。開いたアームおよび閉じたアームそれぞれでの経過時間を、2方向(頭上および水平)から同時に撮影したビデオ記録から記録し、採点する。ラットは開いた空間を避けようとする生来の性向があるので、開いたアームでの経過時間が長いことは、不安感の低下を示す。
癌に対する薬剤の活性は、免疫不全のまたはヒトの腫瘍を移植したマウスまたはラットで試験することが可能である。使用可能なマウスの免疫不全系統の例には、CD−1ヌード、Nu/Nu、Balb/cヌード、NIH−III(NIH−bg−nu−xid BR)などのヌードマウス;Fox Chase SCID(C.B−17 SCID), Fox Chase outbred SCIDおよびSCID Beigeなどのscidマウス;RAG酵素欠乏のマウス;のほか、ヌードラットがある。実験は、たとえば、Kim et al., Nature 362:841 (1992)に記載されているようにして実施する。完全DMEM培地で培養したヒト腫瘍細胞は、一般に、HBSSに採取される。雌の免疫不全の、たとえば、胸腺欠損ヌードマウス(4〜6週齢)に、一般には0.2mlのHBSS中の5×106細胞を、背側領域に注射する。腫瘍サイズが50〜100mm3に達すると、ラットを無作為にグループ分けして、薬剤の適切な投与計画を、その薬剤を欠いた対照投与計画と並行して、実施する。腫瘍サイズは、一般に週に2回、2方向(長さ(a)および幅(b))について測定することによって、判定する。腫瘍サイズは、V=ab2/2に従って計算し、平均腫瘍サイズ±SEMで表す。薬剤の効果は、時間経過による腫瘍の成長、マウスの生存期間の延長、または、一定の時点もしくは不定時に生存しているマウスの割合の増加、によって測定する。対照群と比較する統計解析は、たとえばスチューデントのt検定を使用して行ってよい。
ペプチドまたは他の薬剤は、動物における内在化または輸送活性について試験することが可能である。薬剤(Tatペプチドなど)は、たとえば標識化して、マウスなどの動物に注射することが可能である。たとえば、腹腔内または静脈内注射が好適である。注射後約1時間で、マウスを殺処分し、固定液(3%パラホルムアルデヒド、0.25%グルタルアルデヒド、10%スクロース、生理食塩中10U/mLヘパリン)で灌流する。次いで、脳を取り出して、凍結し薄片にする。切片を、共焦点顕微鏡を用いて、蛍光について分析する。内在化活性を、蛍光から、随意的に陰性対照および陽性対照と比較して、決定する。適切な陽性対照は、標準的なTatペプチドを含む薬剤である。適切な陰性対照は、Tatを欠く蛍光標識した活性薬物である。非標識媒体もまた、陰性対照として使用することができる。
GST融合タンパクを、ND2の異なるフラグメントを用いて設計し、標準的プロトコルを用いて発現および精製した。これらの精製タンパクを、ビオチニル化Src40〜58ペプチドまたはスクランブル化対照ペプチド(sSrc40〜58)で調べるために、膜にスポットした。一般に、1〜10μgのペプチドまたは組み換えタンパクを、ニトロセルロースにスポットして、一夜乾燥した。膜を5%ミルクで室温にて1時間ブロックし、次いで、ビオチニル化ペプチド(〜15μg/ml)と共に2時間インキュベートして、標準の洗浄緩衝液で洗浄した。結合プローブを、ホースラディッシュペルオキシダーゼ共役ストレプトアビジン(SA−HRP)および標準検出試薬、主に化学発光キット、での短時間インキュベーションを用いて検出した。図1Bは、構築物の第1のセットを示しており、図1Cは、ND2の全長がSrc40〜58に結合し得ること、および、ND2.1(AA239−321)と呼ばれるサブフラグメントもSrc40〜58に結合し得ることを示すドットブロットである。コアSrc結合領域を狭めるために、さらなるGST構築物を作成した(図2A)。これらの構築物でドットブロットを作成し、ビオチニル化Src40〜58を捕捉するそれらの能力について試験した(図2B)。ND2.1.4(AA289〜321)が、Src40〜58への結合に最も有効なサブフラグメントであり、一方、試験したどのフラグメントもスクランブル化陰性対照(B−sSRC40〜58)に結合せず、これは相互作用の特異性を示している。
海馬ニューロンにおけるND2,Src,NMDARサブユニットおよびPSD95の局在化を調べるために、Tat,Tat−ND2 310〜321、またはSrc40〜49−Tatの存在下における免疫細胞化学によって、一連の実験を行った。簡潔に説明すると、海馬ニューロンを、胎生期の#17/E18 Wistarラットから単離し、B27およびglutamaxを有しカバースリップを含むneurobasal培地において培養した。次いで、細胞を1μMペプチドで1時間処理し、その後、標準的な方法で固定した。特異的抗体および蛍光分子に結合した二次抗体を用いてタンパクを可視化した。Photoshopにおいて画像を融合し、総局在化蛍光クラスターをクラスターの総数で除算することにより(たとえば、Srcとの%共局在化ND2=(総局在化)/(総ND2クラスター);ND2との%共局在化Src=(総局在化)/(総Src2クラスター))、対の間の%共局在を計算することによって、蛍光の共局在を計算した。
次の一次抗体を試験において使用した:NR2Bに対するマウスmAb(Cat#:ab28373)、PSD95に対するマウスmAb(クローン7E3−1B8,Cat#:ab13552)およびSrcに対するマウスmAb(クローン327,Cat#:ab16885)をAbcam(Cambridge,MA)から;ウサギ抗NR2B(Cat#:06−600)をMillipore(Temecula,CA)から;ヤギ抗ND2(M−16,Cat#:sc−20496)およびウサギ抗GST(Z−5,Cat#:sc−459)をSanta Cruz Biotechnology(Santa Cruz,CA)から;ホスホチロシンマウスmAb(P−Tyr−100,Cat#:9411),ホスホNR2B(Tyr1472,Cat#:4208)およびウサギ抗PSD95(D27E11,Cat#:3450)をCell Signaling Technology(Danvers,MA)から得た。
カバースリップ上で培養した海馬ニューロンを、第14日に、1μmのSrc40〜49−Tat,Tat−ND2 310〜321またはTatで1時間処理した。次いで、ニューロンを、リン酸緩衝生理食塩水中4%パラフォルムアルデヒドおよび4%スクロース中で10分間、その後、室温(RT)にて20%メタノールで5分間インキュベートした。細胞を、PBS中0.25%トリトンX−100での5分間の処理、およびPBS中5%ロバ血清中での30分間の処理により、透過性にした。培養物を、様々な種を高めた一次抗体の混合物と共に、0.25%トリトンX−100中で室温にて2時間インキュベートし、洗浄して、ロバにおいて高めた、Texas RedまたはFITCフルオロフォアのいずれかに共役の種特異的二次抗体の混合物(0.25%トリトンX−100中1:200希釈)と共に、室温にて1時間インキュベートした。カバークリップをPBSで洗浄して、ProLong Gold退色防止試薬(Invitrogen)を用いてマウントした。Nikon Eclipse TE 200顕微鏡上の60×の汎蛍光対物レンズで、画像を収集した。画像は、PhotoShop 7(Adobe,San Jose,CA)で分析した。輝度およびコントラストを調整し、アンシャープマスクツールを用いて鮮明にし、色共局在化のために融合した。
共免疫沈降実験を使用して、ニューロンのNMDA受容体複合体中の選択されたタンパクの状態を調べた。これらは、3軟膜血管閉塞モデル(3PVO)を使用した脳卒中に付した海馬ニューロンとラット脳の双方において、Src−ND2相互作用の存在下および非存在下で、調べた。図12A〜図12Eは、第14日の海馬ニューロンのNMDARの状態を表している。ND2,Src,PSD95,NR2BおよびIgGに対する抗体を単独で、以下に述べるように溶解物に加えて、きれいな免疫沈降物(ブロットの上に記載した抗体)を生成するために使用した。次いで、各ブロットの下に記した抗体を用いて、各ブロットを標準的なウェスタンブロット法を用いる検出抗体によって調べた。これらの図は、免疫沈降抗体の各々がND2,Src,PSD95およびNR2Bを含有する複合体を沈降させ得ることを表している。
NMDARおよびその関連タンパクを、培養海馬(HP)ニューロンまたはラット脳から調製した。インビトロ競合実験のために、Src40〜49−Tat,Tat−ND2 310〜321もしくはTatペプチドの濃度1μMでの1時間または濃度3μMでの2時間の処理の後、2週HPニューロンを収集し、次いで、細胞を速やかに採取し均質化した。インビボ実験のために、3PVO虚血後1時間に、記載したペプチドまたは生理食塩水を、ラットに尾静脈注射で投与した。手術後2時間(化合物投与の1時間後)に、選択した大脳皮質を速やかに採取し均質化した。
下垂体アデニル酸シクラーゼ活性化ポリペプチド(PACAP)は、ラットニューロンにおけるNMDA受容体媒介反応を選択的に増加させ、このペプチドは、シナプス可塑性のみならず長期増強および長期抑圧の双方に関与していると考えられている。PACAP増強NMDA誘起電流に対するND2 310〜321およびSrc40〜49の作用を調べるために、これらのペプチドの存在下または非存在下で、単離CA1ニューロンを一連のパッチクランプ実験に付した。図14Aは、パッチピペット内でのPACAP(1nM)またはPACAP+ND2 310〜321(6mM)による規格化したピーク電流を示している。ND2 310〜321はPACAP誘導増強を防止し得る。Src選択的阻害ペプチド、Src(40〜58)(14mM)または切除ペプチド、Src(40〜49)(6mM)用いる同様の実験も行った。PACAP投与前の反応を、PACAP投与後25〜30分での応答に対して規格化した。棒グラフは、ニューロンからの記録の3群を表しており、3つの条件でのピークNMDAR電流の相対的変化を示している。ND2 310〜321はPACAP増強NMDA誘起電流を阻害するが、Src40〜49は阻害しない。したがって、ND2 310〜321は、PACAP増強NMDA誘起信号伝達を遮断するために使用可能であり、したがって、長期増強で損傷した多くの神経疾患または神経障害(たとえば、加齢、アルツハイマー、神経変性疾患、神経障害情動記憶)において使用可能である。
Tat−ND2 310〜321が痛覚過敏の軽減に有効であることを示すために、フロイントアジュバント(CFA)の皮下注射により後肢に炎症を誘導しておいたラットに、10pmol/gのTat−ND2 310〜321またはスクランブル化ND2もしくはSrc陰性対照ペプチドを尾静脈注射で投与した。痛覚過敏の測定は、ペプチドの注射後1時間または2時間に、方法の欄に記載したように、剛性の異なるフィラメントを用いて行った。Tat−ND2 310〜321で処理した動物は、スクランブル化対照ペプチドで処理した動物よりも、疼痛に対して有意に感受性が低かった。さらに、Tat−ND2 310〜321などのND2 310〜321領域を含むペプチドは、これらの実験において、Src40〜49−Tatよりも、有意に良好に作用した。この差異は、これらの条件下で、投与後1時間において、投与後2時間におけるよりも大きかった(図17)。
CFAモデル:
動物:Charles River Laboratories (St. Constant, Quebec)から入手したSprague−Dawleyラット(雄、250〜300g)で、実験を行った。ラットを、1/8”コーンコブベッドを有するプラスチックケージに収容して、12時間の明/暗サイクルに保った。ラットを対にして収容し、餌および水を自由に与えた。これらの動物の使用は、Canadian Council on Animal Careのガイドラインに従って行い、Toronto Western HospitalのAnimal Care Committeeによって承認された。
脳卒中の治療におけるND2阻害剤の有効性を、脳卒中の3軟膜血管閉塞(3PVO)モデルを用いて調べた。これは、24時間後に小さいながら確実な梗塞を引き起こす脳卒中の永続的モデルである。
体重230〜290gの雄のSprague Dawleyラット(各グループn=10)を、この研究に使用した。実験は絶食したラット(一夜、水は自由に与えたが、餌は与えなかった)で行った。永続的3軟膜血管閉塞(3PVO)は、前述のように実施した(Forder J et al.,2005,前記)。簡潔に説明すると、必要な初期用量の1/3で補完したケタミン(100mg/kg)、アセプロマジン(2mg/kg)およびキシラジン(5mg/kg)の0.5ml/kg筋肉内注射で、ラットを麻酔した。肛門温度プローブを挿入し、動物を37℃に保った加温パッドに置いた。頭蓋骨を正中切開を介して露出して、組織を擦り落とした。解剖顕微鏡および歯科用空気ドリルを使用して、頭蓋骨を貫通する長方形の孔を明け、硬膜をそのままにしながら頭蓋骨片を取り除くことによって、体性感覚皮質の上に6〜8mmの頭蓋窓(プレグマに対して、2mm後方、5mm横)を形成した。バレル皮質の周りの3軟膜血管中大脳動脈分岐を選択し、硬膜を介して電気的に焼灼した。焼灼の後、頭蓋骨を接合した。各ラットを個々のケージに戻し、ラットが完全に回復するまで、体温を保つための加温ランプの下に置いた。3PVO虚血の1時間後に、図示した薬剤(3nmol/g)または生理食塩水を、ラットに尾静脈内注射した。餌と水を与えた。手術後24時間に、脳を速やかに採取した。冠状薄片(2mm)を脳から取り、2%トリフェニルテトラゾリウムクロリド(TTC)(Sigma)中で15分、室温にてインキュベートした。画像をスキャンした(Canon 4200F)。
次に、どちらも機械的肢引き込み閾値の低下で特徴づけられるラットの末梢神経損傷(PNI)のモデルおよび完全フロイントアジュバントモデル(CFA)を用いて、神経障害性および炎症性疼痛挙動に及ぼすTat−ND2 310〜321の効果を、スクランブル化ND2陰性対照(sTat−ND2 310〜321)との対比で、試験した。機械的肢引き込み閾値へのTat−ND2 310〜321の効果を、PNI後8〜15日に評価した。静脈内に投与した場合、Tat−ND2 310〜321は、損傷神経と同側の肢引き込み閾値を有意に上昇させるが、sTat−ND2 310〜321は上昇させないことが、判明した。同様の結果がCFAモデルにおいても観察され(図17)、このモデルでは、肢引き込み閾値の上昇は、最初の45分以内に生じて、3時間の試験期間全体にわたって持続した。
慢性坐骨神経絞扼モデル:動物:末梢神経損傷(PNI)を起こすために、ラットまたはマウスの坐骨神経の周りに、イソフルラン麻酔下で、2mmのポリエチレンカフを外科的に植え込んだ。試験前の7日間、動物を順応させた。
イソフルラン麻酔化で、ラットまたはマウスの坐骨神経の周りにポリエチレンカフ(長さ2mm)を外科的に植え込むことによる、慢性坐骨神経絞扼を通じて、齧歯動物に末梢神経損傷(PNI)を誘導した。次いで、前述のvon Freyフィラメント(Liu et al, 2008)で機械的肢引き込み閾値(PWT)を、ベースラインとして手術前、および手術後8〜14日の両方に、測定した。20%酢酸中のmyr−ND2(100マイクロモル/L)を、ストック溶液として調製し、試験の前に作業濃度に希釈した。10pmolまたは100pmolのいずれかのmyr−ND2および担体を腰髄内に注射し、注射後45分、90分、135分および180分にPWT試験を行った。試験をしたmyr−ND2の濃度は、両方とも、試験をしたすべての時点にわたって疼痛に対する感受性の低下を示した(図23)。このように、myr−ND2を含めて、ND2ペプチドは、疼痛の有効な阻害剤である。
ND2ペプチドが疼痛および脳卒中後の損傷の両方の低減に有効であることを、本明細書において示してきた。Tatペプチドは、高用量で急速に投与されると血圧の低下をもたらすことが、これまで示されている。高濃度のmyr−ND2ペプチドの急速ボーラス投与の、Tat−ND2およびNA−1(Tat−NR2B9c)との対比における、有効性の試験を行った。動脈圧測定のための外科的植え込み装置を有するラットに、監視後10分に、〜25mg/kgの各ペプチドを尾静脈へのボーラス注射で投与した。Tat配列を包含する両ペプチドは、平均動脈圧を通常の半分のレベルに降下させる結果となった。これらの血圧降下は、介入なしで、約10〜15分以内に解消した。しかし、myr−ND2ペプチドは、試験をしたどの動物においても、同様の濃度で血圧の低下を全く示さなかった(図20)。図21は、血圧の最下点を示しており、さらに、myr−ND2がこのモデルにおいて血圧に影響しないことを表している。このように、myr−ND2は、Tat−ND2よりも高い用量レベルで、ヒトにおいて血圧の副作用が生じる可能性無くまたは低い可能性で、投与することが可能である。

Claims (11)
- 配列番号60の残基310〜321を含む配列番号60の残基と同一の20個以下の残基からなることを特徴とするND2ペプチドであって、
前記ペプチドは、ND2のSrcとの相互作用を阻害する、ND2ペプチド。 - 前記ND2ペプチドは、配列番号60の残基310〜321または配列番号60の残基307〜321である、請求項1に記載のND2ペプチド。
- 前記ND2ペプチドは、N末端がミリストイル化されている、請求項2に記載のND2ペプチド。
- 脂質付加されていることを特徴とする請求項1〜3のいずれか1項に記載のND2ペプチド。
- 内在化ペプチドに結合されていることを特徴とする請求項1〜3のいずれか1項に記載のND2ペプチド。
- 前記ND2ペプチドは、キメラペプチドとして、内在化ペプチドに結合しており、
前記キメラペプチドは、長さがアミノ酸50個までであることを特徴とする請求項5に記載のND2ペプチド。 - 前記キメラペプチドは、長さがアミノ酸25個までであることを特徴とする請求項6に記載のキメラペプチド。
- 神経疾患または神経障害の治療または有効な予防に用いるための、請求項1から7のいずれかに記載のND2ペプチド又はキメラペプチドを有する医薬組成物。
- 前記神経疾患または前記神経障害は、脳卒中、CNSへの外傷、てんかん、不安神経症または神経変性疾患であることを特徴とする請求項8に記載の医薬組成物。
- 疼痛の治療または有効な予防に用いるための、請求項1から7のいずれかに記載のND2ペプチド又はキメラペプチドを有する医薬組成物。
- 癌の治療または有効な予防に用いるための、請求項1から7のいずれかに記載のND2ペプチド又はキメラペプチドを有する医薬組成物。
