Description
前記ブロッキングのために使用される物質はカルボキシル基やアルデヒド基と反応するものであれば特に制限されず使用可能であるが、好ましくはアミン類を用いることができる。このようなブロッキング過程は下記反応式2で示すことができる。
Pは水溶性蛋白質を示し、
m、n、及びlの総合計は50乃至10,000の整数であり、nとlの合計は5乃至5,000の整数であり、lは1乃至100の整数である。
前記投与は好ましくは経皮投与であり得、前記対象は好ましくは哺乳類であり得る。
実施例1.HA−hGHコンジュゲートの製造
ヒアルロン酸(HA)(MW=100kDa)を10mg/mlの濃度で水に溶かした後、過ヨウ素酸ナトリウム(Sodium periodate、NaIO4)をヒアルロン酸単位体当り1モル(mole)倍で添加してそれぞれ3時間、6時間、12時間光を遮断した状態で反応させた後、分画分子量(Molecular weight cutoff)が7000ダルトンである透析膜を用いて蒸溜水に対して3日間透析して精製した後に3日間凍結乾燥させて置換率の異なるHA−アルデヒド誘導体を得た。
反応させた溶液をpH7.4のリン酸緩衝食塩水(Phosphate buffered saline(PBS))に対して透析した後、−70℃に保管した。
ヒアルロン酸(HA)(MW=215kDa)を10mg/mlの濃度で水に溶かした後、過ヨウ素酸ナトリウム(Sodium periodate、NaIO4)をヒアルロン酸単位体当り2モル(mole)倍で添加して3時間光を遮断した状態で反応させた後、分画分子量(Molecular weight cutoff)が7000ダルトンである透析膜を用いて蒸溜水に対して3日間透析して精製した後、3日間凍結乾燥させてHA−アルデヒド誘導体を得た。
下記のような条件でGPC分析を実施してHA−OVAを反応しないOVA、シアノ水素化ホウ素ナトリウム、エチルカルバゼートなどから分離精製した後、分離精製されたHA−OVAを遠心分離フィルターを用いて5000gで濃縮し、濃縮された溶液を−70℃に保管した。

1.ヒアルロン酸アルデヒド誘導体の置換率分析
1)分析方法
実施例1のHA−hGHコンジュゲート製造過程と実施例2のHA−OVAコンジュゲート製造過程中に製造されたHA−アルデヒド誘導体の置換率を分析するために、製造されたHA−アルデヒド誘導体を5mg/mlの濃度で100mM、pH5.2のアセテートバッファーに溶かした後、ヒアルロン酸単位体当りそれぞれ5モル倍のtert−ブチルカルバゼート(Tert butyl carbazate、TBC)とシアノ水素化ホウ素ナトリウム(Sodium cyanoborohydride、NaCNBH3)を添加し24時間反応させて、HA−アルデヒド誘導体内に含まれているアルデヒド基にTBCが結合されたHA−アルデヒド−TBCを製造した後、MWCO7000透析膜を用いて蒸溜水に対して3日間透析した後、3日間凍結乾燥して1H−NMR(DPX300、Bruker、Germany)でアルデヒドの置換率を分析した。
図2に示されているように、前記1)で製造されたHA−アルデヒド−TBC誘導体の1H−NMRスペクトルではヒアルロン酸のピーク以外にも、δ=1.2〜1.4ppmに9個の水素を示すTBCの3個のメチル(methyl)ピークが現れた。定量分析のためにδ=1.85〜1.95ppmのヒアルロン酸のアセトアミド官能基のメチル共鳴(methyl resonance of acetamido moiety of HA)を内部標準(internal standard)に決めた。
実施例1、実施例2によるHA−アルデヒドの置換率はδ=1.85〜1.95ppmのピーク面積とδ=1.2〜1.4ppmのピーク面積を比較して求めた。このような1H−NMR分析結果を通じてHA−アルデヒドの置換率を計算してみた結果、過ヨウ素酸ナトリウムとの反応時間を調節して10〜50モル%の置換率を調節して得ることができるのを確認した。即ち、HA−アルデヒドの置換率は過ヨウ素酸ナトリウムとの反応時間を長くするほど高く示され、より具体的に実施例1の場合、反応時間を3時間、6時間、及び12時間とした場合にそれぞれ10モル%、20モル%、及び50モル%の置換率を示し、実施例2の場合、反応時間を3時間とした場合に15モル%の置換率を示した。
1)分析方法
実施例1によって製造されたヒアルロン酸−ヒト成長ホルモン(HA−hGH)コンジュゲート(NaIO4と3時間反応させて約10モル%の置換率を有するHA−ald誘導体にヒト成長ホルモンをHA(100kDa)1分子(chain)当り6モル倍で入れて反応させた接合体:反応率95モル%以上)と実施例2によって製造されたヒアルロン酸−オボアルブミン(HA−OVA)コンジュゲート(NaIO4と3時間反応させて約15モル%の置換率を有するHA−ald誘導体にオボアルブミンをHA(215kDa)1分子(chain)当り3モル倍で入れて反応させた接合体:反応率80モル%以上)のGPC(Gel Permeation Chromatography)分析を通じてヒアルロン酸−蛋白質コンジュゲートの形成を確認した。HPLC(High performance liquid chromatography)を用いてヒアルロン酸−蛋白質コンジュゲートをGPC分析した。分析条件は下記の通りである。

HA−hGHのGPC分析結果、図3aに示されているように、280nmの波長で高分子量のヒアルロン酸の滞留時間(retention time)である14〜15分付近でピークが現れ、ヒアルロン酸(HA)に蛋白質がコンジュゲーションされたことを確認した。また、HA−OVAのGPC分析結果、図3bに示されているように、210nmでOVAのピークが高分子量側にシフトされ31〜32分台で現れ、蛋白質がコンジュゲーションされたことを確認した。
1)分析方法
前記実施例1及び実施例2によって製造されたHA−蛋白質コンジュゲート内の蛋白質含量は前記実験例2のGPC分析結果でピークの下の面積を測定して求めた。
HA−hGHの場合、1mg/mL濃度でhGH原液を準備した後、蒸溜水に希釈してhGH標準溶液を準備した。hGH標準溶液を前記実験例2のGPC分析条件で分析してhGH濃度によるGPCピークの下の面積の標準曲線(standard curve)を求めた。前記実施例1によって製造されたHA−hGHコンジュゲート(NaIO4と3時間反応させて約10モル%の置換率を有するHA−ald誘導体にヒト成長ホルモンをHA(100kDa)1分子(chain)当り6モル倍で入れて反応させた接合体:反応率95モル%以上)を同じ条件で分析して得られたGPCピークの下の面積を標準曲線に代入してhGH含量を求めた。
実施例1によって製造されたHA−hGHコンジュゲート内の蛋白質(hGH)含量はフィード(feed)内でHA一分子当り反応させた蛋白質の分子数によって増加することが分かった。HA一分子当り反応させた蛋白質の分子数を1、4、6、8個に変化させた時、HA一分子当り結合した蛋白質の平均分子数は1、4、6、8個に調節可能であった。即ち、蛋白質の生接合効率(Bioconjugation efficiency)(%)は分子数に関係なく95%以上と示された(図4)。実施例2によって製造されたHA−OVAコンジュゲート内の生接合効率(Bioconjugation efficiency)(%)は分子数3である時に80%内外、分子数6である時に65%内外と示された。
1)分析方法
hGH濃度を基準にして(0.25mg/ml)hGH溶液とHA−hGHコンジュゲート溶液を用いて円偏光二色性分光分析(Circular Dichroism)をした。分析条件は下記の通りである。

図5に示されているように、hGHのスペクトルとHA−hGHコンジュゲートのCDのピーク(peak)が一致することと示され、ヒト成長ホルモンの2次構造がヒアルロン酸と接合された状態でも維持されることが分かった。
1)分析方法
1mg/mlのヒト成長ホルモン溶液の原液をそれぞれ0.5、1、5、10、20、30、50ug/mlの濃度に希釈してヒト成長ホルモン標準溶液を準備し、ブラッドフォードアッセイ(Bradford assay)を用いて標準曲線を求めた。即ち、ヒト成長ホルモン標準溶液とクマシーブルー試薬(Coomassie(登録商標) Protein Assay Reagent)を1:1の体積比率で混合した後、常温で10分間反応(incubation)させた後に595nmで吸光度を測定して標準曲線を求めた。
ELISA/ブラッドフォードアッセイ(Bradford assay)の比率を通じてヒアルロン酸−ヒト成長ホルモンコンジュゲートの活性度を分析した。
図6aに示されているように、ヒアルロン酸−ヒト成長ホルモンコンジュゲートのELISA/ブラッドフォードアッセイ(Bradford assay)の比率が70%以上と示されることが分かった。
具体的に、ブラッドフォードアッセイ(Bradford assay)は蛋白質のリシン(lysine)を測定するアッセイ方法であり、ELISAはhGHと結合する抗体との相互作用を通じて定量するアッセイ方法であって、この比率を通じてヒアルロン酸−ヒト成長ホルモンコンジュゲートの活性度を測定することができ、図6aに示されているようにコンジュゲート内に存在する蛋白質中の70%以上が活性を有していることが分かった。
1)分析方法
1mg/mlのオボアルブミン溶液の原液をそれぞれ0.6、1.2、2.5、5、10、20ug/mlの濃度に希釈して製造したオボアルブミン標準溶液を使用したこと以外は前記実験例5と同様な方法で標準曲線を求めた。
ELISA/ブラッドフォードアッセイ(Bradford assay)の比率を通じてヒアルロン酸−オボアルブミンコンジュゲートの活性度を分析した。
図6bに示されているように、ヒアルロン酸−オボアルブミンコンジュゲートのELISA/ブラッドフォードアッセイ(Bradford assay)の比率が80%以上と示されることが分かった。ブラッドフォードアッセイ(Bradford assay)とELISAについては前記実験例5で言及した通りであり、したがって図6bに示されているように、ヒアルロン酸−オボアルブミンコンジュゲート内に存在する蛋白質中の80%以上が活性を有していることが分かった。
1)分析方法
製造されたヒアルロン酸−蛋白質コンジュゲートの血清安定性を確認してみるためにヒアルロン酸に結合されないhGHと、次のサンプルを使用した。
−HA−hGHコンジュゲート(20/6:アルデヒド基置換率が20モル%でありHA一分子(chain)当り6個のhGH分子が結合したもの)
図7に示されているように、ヒト成長ホルモン(hGH)よりHA−hGHコンジュゲートがヒト血清内でさらに長時間安定するのを確認することができ、特にHAに対するhGH分子の結合量が増加するほど安定性がさらに高まることが分かった。
1)分析方法
IM−9細胞はヒトBリンパ芽球細胞(human B lymphoblast)であって、hGH受容体を有しておりhGHが受容体に結合するとJAK2(Janus kinase2、ヤヌスキナーゼ2)がリン酸化される特性があるので、これを用いてHA−hGH接合体の生物学的活性度を確認した。
図8A乃至8Cに示されているように、ヒト成長ホルモンを500ng/mlの濃度で処理した時に細胞信号伝達が最もよく起こるのを確認することができ(図8のA)、同じ濃度のhGHを含むHA−hGHコンジュゲートを処理した時も細胞信号伝達が起こるのを確認することができた(図8のB)。図8Cは8A及び8Bのウエスタンブロット結果として示されたバンド強度(band intensity)を測定して定量的に示すグラフである。したがって、hGHがHAに接合された後にも生物学的活性を有するのを確認することができた。
1)分析方法
正常ヒト皮膚線維芽細胞(human skin normal fibroblast)であるDetroit551(ATCC)を10%FBSと25mM HEPESが含まれているDMEM(GIBCO)で培養した。全ての細胞は標準組織培養皿で培養し、培養皿は5%CO2(g)と相対湿度が維持される37℃培養器に保管した。Detroit551細胞にhGHとHA−hGHコンジュゲート(NaIO4と3時間反応させて約10モル%の置換率を有するHA−ald誘導体にヒト成長ホルモンをHA(100kDa)1分子(chain)当り6モル倍で入れて反応させた接合体:反応率95モル%以上)を濃度別に処理して細胞信号伝達(cell signaling)によるJAK2のリン酸化程度を実験例7のような方法でウエスタンブロットを通じて確認した。
JAK2のリン酸化は前述のようにhGHがhGH受容体に結合する時に起こる現象である。
図9Aに示されているように、Detroit551細胞にhGHを処理した時にJAK2がリン酸化されhGH受容体による信号伝達が起こるのを確認することができ、図9Bに示されているようにHA−hGH接合体を処理した時もJAK2がリン酸化されHA受容体による信号伝達が起こるのを確認することができた。図9C、9Dは濃度による信号伝達物質の濃度を図9Aと9Bでウエスタンブロット結果として示されたバンド強度(band intensity)を定量化して示すグラフである。
このようにHA−hGHコンジュゲートはヒト皮膚細胞のHA受容体及び/またはhGH受容体に結合して細胞信号伝達効果を奏することが分かった。
1)分析方法
ヒト新生児の表皮角化細胞(human neonatal epidermal keratinocyte)であるHEKn細胞(Invitrogen)を1%HKGS(human keratinocyte growth supplement)が含まれているEpiLife培地(Invitrogen)で培養した。Detroit551(ATCC)は10%FBSと25mM HEPESが含まれているDMEM(GIBCO)で培養した。全ての細胞は標準組織培養皿で培養し、培養皿は5%CO2(g)と相対湿度が維持される37℃培養器に保管した。96−ウェルプレート(well plate)の各ウェルに3*103個の細胞(A:HEKn B:Detroit551)を分注(seeding)した後、24時間経過した後にそれぞれ異なる濃度のヒアルロン酸とhGHを含むHA−hGHコンジュゲート(NaIO4と3時間反応させて約10モル%の置換率を有するHA−ald誘導体にヒト成長ホルモンをHA(100kDa)1分子(chain)当り6モル倍で入れて反応させた接合体:反応率95モル%以上)を処理した。その次に、細胞増殖程度を確認するために48時間以後にWTS−1(TaKaRa)を細胞に処理した。生きている細胞でミトコンドリア(mitochondria)の脱水素酵素(dehydrogenase)によってWTS−1(tetrazolium salt)がホルマザン(formazan)に変換されることによって培地溶液に対する吸光度の変化が起こるようになるので、WST−1を処理した96ウェルプレートを37℃で1時間放置した後、450nm波長に対する吸光度を測定した。
図10に示されているように、HA−hGHコンジュゲートの濃度変化によって細胞増殖が起こるのを確認することができた。
HAを10ug/mlの高濃度に処理した時、HEKn細胞の場合は20%の増殖効果を(図10のA)、Detroit551細胞の場合は60%の増殖効果をそれぞれ示したが(図10のB)、このようにHA濃度が高い場合には接合されたhGHの濃度が過度に高くなりhGHの信号伝達を妨害することがあるので、HA−hGHコンジュゲートの場合、HAとhGHの効果を適切に見るための低濃度条件で実験を行なった。
1)分析方法
SDラット(雌、6週齢、200g)の尾静脈を通じてPBS、hGH、HA−hGHコンジュゲートをhGH基準に1mg/ml溶液を準備して300ul容量をそれぞれ投与した後、定められた時間になった時、尾静脈から血液を採取し、各サンプルの血中濃度をhGH ELISAキット(roche)を用いて製造社の方法によって測定した。
図11に示されているように、hGH血中濃度は24時間以前に基準線(baseline)に落ちたが、HA−hGHコンジュゲートの形態に提供される場合には血中濃度が高く維持され、特にアルデヒド置換率が高い20%のアルデヒドが導入されているHA−hGHコンジュゲート(20/6)の血中濃度は3日目にも基準線に落ちなく、本発明の一実施例によるコンジュゲート形態に蛋白質薬物が提供される場合、薬物の体内半減期が長くなり薬効が長続きするのを確認することができた。
1)分析方法
毛がない(Hairless)Balb/cマウス(雌、6週齢、20g)とPBS、FITC(fluorescein isothio−cyanate)、hGH−FITC、HA−FITC、HA−hGH−FITCコンジュゲートを準備した。
FITCがタギングされたhGH−FITC及びHA−hGH−FITCコンジュゲートは先ずFITCを2mg/mlの濃度にpH9リン酸塩バッファーに溶かした後、FITCが蛋白質(hGH)の5モル倍になるようにhGH溶液と混合した後、3時間反応させた。その後、PD10カラムを用いて反応しないFITCを精製して準備した。
図12(A:control、B:FITC、C:hGH−FITC、D:HA−FITC、E:HA−hGH−FITC)に示されているように、HAと結合しないFITC(B)、hGH−FITC(C)の場合、皮膚組織層を通過せず皮膚組織の外側部分のみで蛍光が観察されるが、HAと結合を形成したHA−FITC(D)、HA−hGH−FITC(E)の場合、皮膚組織内部にも均等に蛍光が分布することと示されHAの経皮伝達特性を確認することができた。また、このような結果を通じて親水性蛋白質であるヒト成長ホルモンが接合されたHAの誘導体(E)の場合にもHAと同様に優れた経皮伝達特性を有することを確認した。
1)分析方法
毛がない(Hairless)Balb/cマウス(雌、6週齢、20g)とPBS、FITC(fluorescein isothio−cyanate)、OVA−FITC、HA−FITC、HA−OVA−FITCコンジュゲートを準備した。
FITCがタギングされたOVA−FITC及びHA−OVA−FITCコンジュゲートは先ずFITCを2mg/mlの濃度にpH9リン酸塩バッファーに溶かした後、FITCが蛋白質(OVA)の10モル倍になるようにOVA溶液と混合した後、3時間反応させた。OVA溶液と混合した後、pH9の炭酸ナトリウム(sodium carbonate)緩衝溶液で12時間反応させた。その後、過量の水に透析して反応しないFITCを精製して準備した。
図13(A:PBS、B:FITC、C:OVA−FITC、D:HA−FITC、E:HA−OVA−FITC)に示されているように、HAと結合しないFITC(B)、OVA−FITC(C)の場合、皮膚組織層を通過せず皮膚組織の外側部分のみで蛍光が観察されるが、HAと結合を形成したHA−FITC(D)、HA−OVA−FITC(E)の場合、皮膚組織内部にも均等に蛍光が分布することと示されHAの経皮伝達特性を確認することができた。また、このような結果を通じて親水性蛋白質であるオボアルブミンが接合されたHAの誘導体(E)の場合にもHAと同様に優れた経皮伝達特性を有するのを確認した。
1)分析方法
SDラット(雌、6週齢、200g)の背中皮膚の毛を刈った後、3cm×3cmの面積にPBS、hGH、HA−hGHコンジュゲートをそれぞれhGH濃度基準に1.5mg/ml溶液を300ul塗った後、定められた時間になった時に尾静脈から血液を採取して各サンプルの血中濃度をhGH ELISAキット(roche)を用いて測定した。また、経皮伝達効率を皮下注射時と比較するために、HA−hGHコンジュゲートを上記のような容量で皮下注射形態に投入した後、同様な方法で時間別血中濃度を測定した。
図14に示されているように、HAと結合されないhGHの場合、皮膚組織層を通過せず血中濃度が基準線(baseline)と示されたが、経皮伝達されたHA−hGHコンジュゲート(10/6)の場合、血中濃度がCmax:20ng/ml程度であることと示され血液でhGHが検出されたのを確認することができ、したがって、HA−hGHの形態に経皮伝達する場合、hGHが皮膚組織層を通過して血中に効果的に伝達されることが分かった。
1)分析方法
Balb/cマウス(雄、8週齢、20g)背中皮膚の毛を刈った後、1cm×1cmの面積にPBS、OVA、HA−OVAコンジュゲートをそれぞれOVA濃度基準に10mg/ml溶液を50ul塗った。経皮伝達後、それぞれ2週と4週後に血液サンプルを採取してAnti−OVA IgG抗体濃度をELISAで定量した。
図15に示されているように、同じ容量、同じ濃度のOVAとHA−OVAを経皮伝達した時、HA−OVAコンジュゲート形態に経皮伝達された場合、OVAのみを経皮伝達した場合に比べて2週後には5倍、4週後には15倍程度高い濃度の抗体を生成するようにすることが分かった。このような結果はHA−OVAコンジュゲートがOVAに比べて表皮と真皮に効果的に伝達されたためであると考えられる。皮膚組織の奥深くに透過したHA−OVAコンジュゲートは皮膚組織内免疫細胞であるランゲルハンス細胞(Langerhans cell)と樹枝状細胞(dendritic cell)を刺激して効果的に免疫反応を起こす。このような結果に基づいてHA−OVAコンジュゲートをはじめとするHA−ワクチンコンジュゲートは優れた経皮伝達特性によって抗体生成率を顕著に高めることができるのを確認することができ、したがって患者便宜性と共にワクチン効果を顕著に向上させることができる優れたワクチン伝達体として有用に用いられると期待される。
Claims (6)
- 下記化学式2のヒアルロン酸−蛋白質コンジュゲートを含む、蛋白質経皮伝達用組成物:
Pは水溶性蛋白質を示し、
mとn及びlの総合計は50乃至10,000の整数であり、nとlの合計は5乃至5,000の整数であり、lは1乃至100の整数であり、
前記ヒアルロン酸−蛋白質コンジュゲートが、ヒアルロン酸−アルデヒド1分子あたり1乃至10分子の蛋白質が結合されていて、
前記ヒアルロン酸−蛋白質コンジュゲート中のヒアルロン酸は、ヒアルロン酸またはヒアルロン酸の塩であることを特徴とする組成物。 - 前記水溶性蛋白質は、ヒト成長ホルモン、インターフェロンアルファ、エリスロポエチン(erythropoietin)、トレイル(Tumor necrosis factor−related apoptosis−inducing ligand、TRAIL)、オボアルブミンまたはインシュリンである、請求項1に記載の組成物。
- 前記ヒアルロン酸、またはヒアルロン酸の塩は分子量が10,000乃至3,000,000ダルトン(Da)である、請求項1に記載の組成物。
- 請求項1に記載の組成物を含む薬物。
- 請求項1に記載の組成物を含む化粧料。
- 請求項1に記載の組成物を含む経皮伝達用ワクチン。
