Description
本願は、2010年3月23日に出願された米国特許仮出願第61/316,747号および2010年8月12日に出願された米国特許仮出願第61/373,031号に対する優先権および便益を主張するものであり、これらの仮出願の内容は、参照によってその全体が本明細書に組み込まれる。
本発明は、国立衛生研究所(NIH)が裁定する契約第HHSN266200600014C号に基づき米国政府の支援をもって実施された。米国政府は本発明に一定の権利を有する。
さらに、製剤原料の新たな多形型が、さまざまな融点、吸湿性、安定性、溶解度および/または溶解速度、結晶化度、結晶特性、生体利用効率、毒性、ならびに製剤取扱特性を示す可能性があり、これらが有効に投与し得る薬剤を調合する際に考慮する必要のある多くの性質の一部だということをかんがえると。また、監督官庁は、薬剤の投与形態が固形である場合の有効成分の多形型に関する明確な知識、特性評価、および管理を要求している。そのため、当技術分野では、ST−246の新たな多形型を結晶化して特性評価することが必要とされている。
この詳細な説明によれば、以下の略称および定義が適用される。本明細書で用いる場合、単数形の「a、an」および「the」は複数の指示対象を含む。ただし、文脈が明らかに別の旨を指示している場合を除く。
X線粉末回折(XRPD)
XYZステージ、位置調整のためのレーザー・ビデオ顕微鏡、およびHiStar面検出器で構成されたBruker D8 Discovery回折系を用いて、回折パターンを収集した。収集時間は、名目上60秒であった。Cu Ka線1.5406管を40kVで操作し、40mAを用いてサンプルを照射した。X線光学は、Gobel鏡と0.5mmピンホールコリメータで構成される。Theta−theta連続読み取りを サンプル検出器距離15cmで採用したが、これによって4〜40°の有効な2θ測定範囲が得られる。サンプルを低バックグラウンド石英プレートに取付けた。一部の実験用には、可変温度ホットステージを用いてサンプルを操作した。
Harrick Splitpea(商標)全反射減衰装置を装備したNicolet 510 M-O フーリエ変換赤外分光計を用いて、赤外スペクトルを得た。スペクトルは、4cm−1の解像度で4000〜400cm−1から取得し、各分析につき128走査を収集した。
本発明は、以下のステップを含むST−246の多形型Iの産生方法を提供する:
a)少なくとも1の有機溶媒および溶液を作るための水にST−246を溶解し;
b)前記ST−246多形型Iの優先晶出を引き起こす温度まで前記溶液を冷却し;そして

本発明の一態様では、ST−246の結晶型が開示され、ST−246の型I、または簡単に「型I」と命名される。
型Iは、3421cm−1に単一の大きなピークを持ち、また約3300〜2600cm−1にこれらのピークの基礎となる幅広い吸光度を持つ。また、ほかにも3008および2956cm−1に2つのピークがあり、C−Hストレッチが原因であると考えられる。型Iは、1791、1717、および1671cm−1にピークを持つ。3つの型はいずれも、約1560cm−1にピークを持つ。
本発明の別の態様では、ST−246の結晶形が開示され、ST−246の型II、または簡単に「型II」と命名される。
本発明の別の態様では、発明は、ST−246の型IIIと命名されるST−246の結晶形に関わる。
型IIIは、3452および3397cm−1に分裂ピークを持つ。また、〜3008および2956cm−1にもピークがあり、C−Hストレッチによるものと思われる。約3300〜2600cm−1にもピークがある。
型IIIは、1792、1713、および1662cm−1に一連のピークがある。そのいずれも、C=Oストレッチによるものと思われる。また、1560cm−1にもピークがあり、暫定的にN−H変形に割り当てた。
155〜400cm−1では、それほど重要ではないさまざまなピークがある。
型IVのXRPDの例、単結晶X線データを、図4に示す。
本発明のさらに別の態様では、発明は、ST−246の型V、または簡単に「型V」と命名されるST−246の結晶形に関わる。
型Vは、3464および3402cm−1に分裂ピークを持ち、また3238および3206cm−1にも第2の幅広い分裂ピークを持つ。これらのピークは、OHおよびNGストレッチによるものと考えられ、3つの型の分化を可能にするように思われる。また型Vは、〜3008および2956cm−1にもピークがあり、C−Hストレッチによるものと考えられる。約3300〜2600cm−1にもさらにピークがある。
型Vはこの領域に、ST−246の他の結晶形と比較して有意に異なるスペクトル特性を持っていて、3個ではなく5個のピークを示し、これらは1791、1733、1721、1681、および1667cm−1にある。このいずれもが、C=Oストレッチによるものと思われる。3つの型はいずれも、約1560cm−1のところにピークがあり、暫定的にN−H変形に割り当てた。相Vは1519および1497cm−1にピークがある。
1500〜400cm−1では、3つの型の赤外線スペクトルはわずかな差を示すだけであり、この領域は、ここに考察するST−246の3つの型を分化させるのには役立たないのであろう。
本発明の一態様では、発明はST−246の結晶型に関わり、これはST−246の型VI、または簡単に「型VI」と命名される。
ST−246の多形体型の製剤は、製剤学の技術分野で公知の工程により調製することができる。以下の例(下記)は、当業者が本発明をいっそう明確に理解し、実施することができるように記載するものである。本発明の範囲を限定するものではなく、単にその例示および典型として考えなければならない。
さらに具体的には、ST−246の一水和物、型Iを調製するには、シクロへプタトリエンをトルエンの存在下で無水マレイン酸と反応させて、主産物である核異性体を得る。トルエン/へプタンからの結晶化によって、さらに核外異性体が約7%〜0.6%得られる。さらに、無水和物または水和物の形のヒドラジンをイソプロパノールの存在下でMethyl 4-(trifluoromethyl) benzoateと反応させて、(4-(trifluoromethyl)-benzhydrazideを得る。生成物を、次にイソプロパノールから結晶化させる。
型IおよびVの出発物質の再結晶化によって、エチルアセテート溶媒およびクロロホルム溶媒から型IIの標準を調製した。ある量のST−246型Iまたは型Vをエチルアセテートまたはクロロホルムのいずれかに溶解させ、0.45□
mの膜フィルターでろ過した。ろ過した溶液をさらに高温で過熱させて個体がすべて溶解したことを確認し、次に温度を下げて、窒素置換下で乾燥するまで蒸発させた(〜2psi)。
型IIIは、無水和のST−246を水中で再スラリー化することによって作る。型IIIを生じさせるのに用いる溶媒のさらなる例を、上の表1〜9にまとめた
型Iの出発物質を1−ブタノール溶媒から再結晶化させることによって、型IVの標準を作った。固体の出発物質を1−ブタノールに溶解させ、0.45um膜フィルターでろ過した。ろ過溶液は、固体がすべて溶解したことを確実にするためいっそう高い温度で過熱してから温度を下げ、窒素置換下で乾燥するまで蒸発させた(〜2psi)。型IVにとって好ましい結晶化条件を、表5にまとめた。型IVの結晶化のさらなる例を、表1〜3にまとめた。
型V(半水和物)はST−246の早期のGMP合成の間に作られ、国際公開第2008/079159号に開示されている。この多形体のデメリットは完全に水和されていないことである。この型は、湿潤環境に置かれるとすぐに水分を吸収し、また競争的なスラリー実験では、型Iに転化することが示されている。
型Vの出発物質をニトロメタン溶媒から再結晶化させることによって、型VIの標準を作った。固体の出発物質は、ニトロメタンに溶解させ、0.45um膜フィルターでろ過した。固体がすべて溶解したことを確実にするために温度を上げてろ過した溶液を過熱し、次に温度を下げて窒素置換下で乾燥するまで蒸発させた(〜2psi)。型VIは、いろいろな溶媒および溶媒の組み合わせからST−246を結晶化させることによって調製することができる。下記の表5に結晶化条件をまとめた。好ましい結晶化条件を、下記の表6にまとめた。型VIの結晶化のさらなる例を、表1〜3にまとめた。
ST−246は、3つの重要な物理的形状(型I、III、およびV)で存在しうることが明らかにされた。データは、さまざまな固形が製品の品質に影響を及ぼすかどうかを判定するために、多形体の関連する物理的/化学的性質および安定性について得た。このようなデータには、多形体の結晶学的性質および多形体の物理的/化学的性質(例えば、溶解度、溶解、融解範囲)の加速安定性データが含まれる。
競争的および非競争的なスラリー実験を実施してST−246の最も安定な型を決定した。スラリー実験は、ST−246の1以上の型の過剰材料を少量の水に暴露し、結果として生じた懸濁液を数日間、周囲温度および/または45℃で撹拌することによって実施した。また類似の実験も、30℃で60分間、異なるpH値で行った。スラリーをろ過し、粉末XRPDで固体を分析した。単離後に起こり得る脱溶媒和または物理的変化を回避するために、サンプルは粉末X線分析の前に乾燥させなかった。競争的および非競争的な水中スラリー実験から、型Vおよび型IIIが水中で型Iに転化し、型Iは未変化のままであることが明らかである。このスラリーデータを下記の表7にまとめた:
ST−246は、生理学的に関連のある溶媒中でその溶解度が低いため、BCSのクラスIIに属する。第1相臨床試験の試験材料は、粒径分布50%4.8μmおよび粒径分布90%12μmの粒径の微粉化された型Vを用いて作った。そのため、型IおよびIIIとも、国際公開第02/00196号に記載されたようなエアジェット摩砕機を使用し、400gmのスケールで微粉化した。どちらの型も摩砕してすぐに、(XRPDに基づく)物理的形状の変化を全く経ることなく所望の粒径を生じた。
微粉化されたものと微粉化されないものをどちらも含めた製剤原料の型Iおよび型IIIに対し、応力条件下の短期安定性評価を実施した。短期試験は完了しており、40℃/相対湿度75%で得られたデータを表8および9に示す。
型I、III、およびVに対して様々な湿度条件で吸湿性試験を実施し、収着/脱着特性を理解した。各型を約1gずつ乳鉢と乳棒ですりつぶした。TGAにより、含水率を判定した。各型を約100mgずつ、相対湿度11および97%の静止湿度室に約25℃で10日間まで配置した。第0日目から重量損失の変化を示したのは、97%の相対湿度で貯蔵した型Vのサンプルであった。このデータを下記の表11にまとめた:
薬剤の粒径がその溶解プロフィルに及ぼす影響を広範囲にわたって検討し(Fincherら、1968を参照されたい)、やや溶けにくい薬剤の粒径が小さくなると、溶媒に暴露する薬剤の表面積が増大するため、溶解速度の上昇をもたらすとの仮定が行われた。
ST−246型Iのワクシニアウイルス阻害能力は、以下の実験手順に従って確立する:
低多重度で感染したベロ細胞にワクシニアウイルス(NYCBH)のウイルス株を作り(細胞1個あたり0.01プラーク形成単位(PFU))、細胞変性効果が完全になった時点で(4+CPE)集菌する。サンプルを凍結し、解凍し、超音波で分解して、細胞結合ウイルスを開放する。細胞残屑を低速遠心分離で除去し、結果として生じたウイルス懸濁液を−80℃の1mLアリコートに貯蔵する。ウイルス懸濁液のPFU/mLを、標準のプラークアッセイでベロ細胞およびBSC−40細胞に関して定量化する。
3日間で完全なCPEを生じさせるのに必要なワクシニアウイルス株の量を決定するために、96ウェルプレート上にベロ細胞単層を播種し、ワクシニアウイルス株の2倍段階希釈液で感染させる。感染後3日目に、培養物を5%のグルタルアルデヒドで固定し、0.1%のクリスタルバイオレットで着色する。ウイルスが誘導したCPEを、OD570で分光光度的に定量化する。この分析から、HTSアッセイでの使用には、ワクシニアウイルス株の1:800希釈液を選択する。この量のワクシニアウイルスは、感染の多重度が1細胞当たり約0.1PFUであることを表わす。
ST−246型Iに対し、ワクシニアウイルスCPEアッセイでの試験を実施する。型IをDMSOに溶解し、培養液で希釈して、各ウェルの最終濃度を5pM化合物および0.5%DMSOとする。Biomek.RTMを用いたロボット制御で、型Iを培地に添加する。
試験デザイン
試験は、カニクイザル(Macaca fascicularis)を対象とするST−246型I経口薬の無作為化プラセボ対照平行群間比較縦断試験として設計される。この一連の実験のために、MPXのザイール79株5x107PFUの静脈内注射によって15のNHPを感染させ、それぞれ3NHPずつの治療群5群に無作為に割り付ける。ビヒクルまたはST−246型Iを、3mg/kg、10mg/kg、30mg/kg、および100mg/kgの用量で1日1回経口投与し、続いて、水和して均質なモンキービスケットの30%懸濁液を5ml/kgずつ与える。
飼料を与えた状態のNHPに対し、10、20、および30mg/kgのST−246を経口強制飼養により投与する。投与日には、用量投与の直前に、どの動物にも霊長動物用のビスケットスラリーを投与する。ビスケットスラリーには、Ensure1缶(8oz)または他の流動食、液体および電解質置換調製粉乳(electrolyte replacement formula)(Prang)約36g、特殊調製粉乳約94g、ろ過した果物1ジャー(2.5oz)、サル用固形ビスケット5個、ならびに水2ozが含まれ、これを混合して硬度を均一にしてある。ビスケットスラリーは、10ml/kgの用量体積のものを強制飼養で経口投与する。被験物質の各用量を投与した後、そして強制飼養管を除去する前に、10mlの水道水で管を洗い流した。投与調合物を磁石の撹拌棒と撹拌皿で撹拌する。個別の用量は、ごく最近の体重に基づく。20−mg/kg用量群を除いて、それぞれの用量群にはNHPのオス3匹とメス3匹を含め、20−mg/kg用量群にはオス4匹とメス4匹を含める。10−および30−mg/kg用量群はST−246型Iを連続14日間摂取し、20−mg/kg群のNHPはST−246の単回投与を受けた。大腿動脈/静脈予備試験(0時間)で(投与前)、ならびに第1日目、7日目、および14日目の投与後0.5、1、2、3、4、6、8、12、および24時間に血液検体を採取し、ST−246型Iの血漿中濃度を判定する。
ここに記載する試験は、MPXに静脈内感染させたNHPに対するST−246型Iの経口投与の無作為化プラセボ対照平行群間比較縦断試験である。動物は、主要組織の広範囲にわたるMPX感染を示しており、これは組織内のウイルスのレベルによって示される。ST−246型I処置を行う時点で、NHPの3分の1がウイルス病変を抱えている。ビヒクルだけの投与を受けた動物は、全匹が死亡したかまたは安楽死を必要としている。何故なら、この動物群は、33日間の試験の間瀕死の状態だったからである。これに対して、ST−246型Iを投与された動物は全匹が生き延びている。ST−246型Iの投与を受けた動物はいずれも、試験期間全体を通じて生き延びている。
試験デザイン
これは、ST−246型I(試験)とST−246型V(基準)との単回400mg(2x200mg)経口投与の薬物動態(PK;AUCの変数およびCmax)を比較し、満腹の健常志願者に対する両化合物の安全性および耐容性を評価するための第I型無作為化二重盲検クロスオーバー探索的試験であった。ふるい分けした63例の被験者(男性および妊娠していない女性、18歳〜50歳)のうち、12例の試験への参加を認め、次の順序のうちいずれかに無作為に割り付けた:型Iの後で型V、または型Vの後で型I。

- 治療的有効量の請求項1に記載の多形体を含み、さらに、担体、賦形剤、希釈剤、添加剤、充填剤、潤滑剤、および結合剤からなる群より選択される薬学的に許容し得る成分を1以上含む、オルトポックスウイルスに対する活性を有する医薬組成物。

- 請求項1に記載の4-トリフルオロメチル-N-(3,3a,4,4a,5,5a,6,6a-オクタヒドロ-1,3-ジオキソ-4,6-エテノシクロプロプ[F]イソインドール−2(1H)-イル)-ベンズアミド(ST−246)の結晶多形体型Iの生成方法であって、
a)溶媒にST−246を溶解させて溶液を作るステップと、
b)前記溶液を少なくとも15分間を超えて冷却して、前記ST−246多形体型Iの優先晶出を引き起こすステップと、そして
c)任意で、形成されたST−246の結晶を乾燥するステップと、を含み、
ここで、前記溶媒はイソプロピルアルコールまたはエチルアセテートと5%の水とを含み、前記溶液は室温に冷却される、あるいは、前記溶媒はイソプロピルアルコールまたはエチルアセテートと2%の水とを含み、前記溶液は2-5℃に冷却される、方法。 - ステップ(b)の実施中に、多形体型Iの4-トリフルオロメチル-N-(3,3a,4,4a,5,5a,6,6a-オクタヒドロ-1,3-ジオキソ-4,6-エテノシクロプロプ[F]イソインドール−2(1H)-イル)-ベンズアミド(ST−246)の種晶を添加することをさらに含む、請求項5に記載の方法。
- 前記冷却ステップが少なくとも15分間を超えて実施される、請求項5に記載の方法。
- D90%粒径が10ミクロン未満である4-トリフルオロメチル-N-(3,3a,4,4a,5,5a,6,6a-オクタヒドロ-1,3-ジオキソ-4,6-エテノシクロプロプ[F]イソインドール−2(1H)-イル)-ベンズアミド(ST−246)を200mg含む、経口投与用に製剤化された請求項2に記載の医薬組成物を含む投与単位形態。
- 4-トリフルオロメチル-N-(3,3a,4,4a,5,5a,6,6a-オクタヒドロ-1,3-ジオキソ-4,6-エテノシクロプロプ[F]イソインドール−2(1H)-イル)-ベンズアミド(ST−246)が300ミクロンまでの直径のD90%粒径を有する、請求項2に記載の医薬組成物を含む経口投与のための投与単位形態。
- 4-トリフルオロメチル-N-(3,3a,4,4a,5,5a,6,6a-オクタヒドロ-1,3-ジオキソ-4,6-エテノシクロプロプ[F]イソインドール−2(1H)-イル)-ベンズアミド(ST−246)が5ミクロンの直径のD90%粒径、16.6ミクロンの直径のD90%粒径、26.6ミクロンの直径のD90%粒径、または75ミクロンの直径のD90%粒径を有する、請求項9に記載の経口投与のための投与単位形態。
- カプセルまたは錠剤の形態で存在する、請求項10に記載の投与単位形態。
- オルトポックスウイルスに対する活性を有する経口投与のための単位投与形態であって、
(a)4-トリフルオロメチル-N-(3,3a,4,4a,5,5a,6,6a-オクタヒドロ-1,3-ジオキソ-4,6-エテノシクロプロプ[F]イソインドール−2(1H)-イル)-ベンズアミド(ST−246)が、多形体型Iである、200mgの4-トリフルオロメチル-N-(3,3a,4,4a,5,5a,6,6a-オクタヒドロ-1,3-ジオキソ-4,6-エテノシクロプロプ[F]イソインドール−2(1H)-イル)-ベンズアミド(ST−246)と、
(b)33.15mgのラクトース一水和物と、
(c)42.90mgのクロスカルメロースナトリウムと、
(d)1.95mgのコロイド状二酸化ケイ素と、
(e)13.65mgのヒドロキシプロピルメチルセルロースと、
(f)7.8mgのラウリル硫酸ナトリウムと、
(g)1.95mgのステアリン酸マグネシウムと、そして
(h)投与形態の総重量を390mgにするための、88.60mgまでの量の微結晶性セルロース、を含む、単位投与形態。
