本発明で使われる“インフルエンザAウイルス(influenza A virus)”は、オルトミクソウイルス(Orthomyxoviridae)に属するエンベロープウイルス(enveloped virus)であり、8個の分節からなるマイナスセンス1本鎖(Negative-sense、single-strand)のRNA(ribonucleic acid)を遺伝体として持ち、A、B及びCグループに分類され、主要表面タンパク質であるHA(hemaggutinin)及びNA(neuraminidase)によって再び様々なサブタイプ(subtype)に分けられる。現在まで16種のHA及び9種のNAが知られている。
以下、本発明を詳細に説明する。
特にCT149抗体は、Group1(H1,H5,H9)のみならずGroup2(H3,H7)インフルエンザウイルスに対しても中和活性を持っている。
また、前記結合分子は、配列番号39で表されるポリペプチド配列を含む軽鎖、及び配列番号40で表されるポリペプチド配列を含む重鎖で構成されることを特徴とする。
また、前記結合分子は、配列番号41で表されるポリペプチド配列を含む軽鎖、及び配列番号42で表されるポリペプチド配列を含む重鎖で構成されることを特徴とする。
同時に、前記結合分子は、配列番号43で表されるポリペプチド配列を含む軽鎖、及び配列番号44で表されるポリペプチド配列を含む重鎖で構成されることを特徴とする。
また、前記結合分子は、配列番号47で表されるポリヌクレオチド配列を含む軽鎖及び配列番号48で表されるポリヌクレオチド配列を含む重鎖で構成されることを特徴とする。
また、前記結合分子は、配列番号49で表されるポリヌクレオチド配列を含む軽鎖及び配列番号50で表されるポリヌクレオチド配列を含む重鎖で構成されることを特徴とする。
さらに、前記結合分子は、配列番号51で表されるポリヌクレオチド配列を含む軽鎖及び配列番号52で表されるポリヌクレオチド配列を含む重鎖で構成されることを特徴とする。
前記結合分子は、インフルエンザAウイルスH1、H3、H5、H7及びH9サブタイプからなる群から選択されたいずれか一つに対して中和活性を持つことを特徴とする。また、前記インフルエンザAウイルスは、H3サブタイプはH3N2であることを特徴とするが、これに限定されるものではない。
本発明の核酸分子は、本発明で提供する抗体のアミノ酸配列が、周知の如く、ポリヌクレオチド配列にて翻訳された核酸分子をいずれも含む。したがって、ORF(open reading frame)による多様なポリヌクレオチド配列が製造され、これもいずれも本発明の核酸分子に含まれる。
また、本発明は、前記発現ベクターが形質感染されたインフルエンザAウイルスに中和活性を持つ結合分子生産細胞株を提供する。
本発明の組成物は、本発明のインフルエンザAウイルスに中和活性を持つ結合分子以外に、薬剤学的に許容可能な賦形剤を含む。薬剤学的に許容可能な賦形剤は、公知のものである。
また、本発明は、前記結合分子を含むインフルエンザAウイルス来由の疾患予防及び治療用組成物を提供する。
また本発明の予防及び治療用組成物は、少なくとも5個の異なる治療剤を含む。本発明の予防及び治療用組成物は、インフルエンザAウイルスHA及びその断片に結合する数種の単一クローン抗体を含み、これを通じて中和活性に相乗作用を示す。
前記経口形態は、錠剤、トローチ剤、薬用ドロップス剤、水性または油性懸濁液剤、酸剤または分散顆粒剤、エマルジョン剤、剛性カプセル剤、軟性ゼラチンカプセル剤、シロップ剤またはエリクサー剤、ピール剤、糖衣錠、液剤、ゲル剤またはスラリー剤として製剤化される。これら剤形は、不活性希釈剤、顆粒化または崩解剤、結合剤、光沢剤、保存剤、着色剤、風味剤または甘味剤、植物性油またはミネラル油、湿潤剤及び増粘剤を含む薬剤学的賦形剤を含むが、これらに限定されるものではない。
また、本発明は、インフルエンザAウイルスに中和活性を持つ結合分子に標識物質を接合したコンジュゲートを含むインフルエンザAウイルス診断用組成物を提供する。
また、本発明は、下記のような段階を含むインフルエンザAウイルス来由の疾患治療方法を提供する。インフルエンザAウイルスによって発生する疾患を持つ対象に、本発明のインフルエンザAウイルスに中和活性を持つ結合分子を治療学的に有効な量に投与する段階。
本発明の治療方法において、当業者に周知の治療剤を共に投与する。
本発明の治療方法において、インフルエンザAウイルス来由の疾患は、新種インフルエンザ、大流行性インフルエンザ及び季節性インフルエンザからなる群から選択されたいずれか一つであるが、これらに限定されるものではない。
また、本発明は、下記のような段階を含むインフルエンザAウイルス来由の疾患予防方法を提供する。対象に、本発明のインフルエンザAウイルスに中和活性を持つ結合分子を治療学的に有効な量に投与する段階。
本発明の予防方法において、インフルエンザAウイルスに中和活性を持つ結合分子投与量は、最適の反応を提供するように調節される。投与量の範囲は、例えば0.01〜200mg/kgであり、望ましくは0.1〜150mg/kgであり、さらに望ましくは1〜100mg/kgであるが、これらに限定されるものではない。投与回数は、様々な時間にかけて分けて投与され、投与量は、治療的状況の緊急事態によって示されるところによって比例的に減少または増加し、これは、本発明で限定するものではなく、担当医者の権限によって変化する。
また、本発明は、下記のような段階を含む患者のインフルエンザAウイルス感染有無の診断方法を提供する。1)サンプルと、本発明のインフルエンザAウイルスに中和活性を持つ結合分子とを接触させる段階と、2)前記結合分子とサンプルとの反応を検出する段階、または1)サンプルと本発明の診断用組成物とを接触させる段階と、2)前記診断用組成物とサンプルとの反応を検出する段階。
本発明の診断方法において、本発明の結合分子は、診断検出のために必要に応じて標識物質を接合させ、これは公知のものである。
本発明の診断方法において、前記サンプルは、対象の痰、唾、血液、汗、肺細胞、肺組織の粘液、呼吸器組織及び唾液からなる群から選択されたいずれか一つであることが望ましいが、これらに限定されず、当業者に周知の通常的な方法でサンプル準備が可能である。
また、本発明は、1)本発明のインフルエンザAウイルスに中和活性を持つ結合分子と、2)容器を含むインフルエンザAウイルス診断用キットとを提供する。
さらに本発明は、1)本発明のインフルエンザAウイルス診断用組成物と、2)容器を含むインフルエンザAウイルス診断用キットとを提供する。
本発明の診断用キットにおいて、前記2)の容器には、固体担体が含まれる。本発明の結合分子は、固体担体に取り付けられ、このような固体担体は、多孔性または非多孔性、平面または非平面である。
<実施例1:インフルエンザ回復患者の血液からのPBMC分離準備>
回復患者群は、新種インフルエンザ確診後2〜4週となった患者であり、血液内に新種インフルエンザウイルス(H1N1)が存在していないと確認され、かつ新種インフルエンザウイルスに対する抗体を保有している志願者で構成され、これは、臨床試験審査委員会(IRB)の承認をもらって行われた。これら患者群の特徴は、次の通りである。(1)季節インフルエンザを対するワクチンを接種されていない。(2)他の伝染性ウイルス、すなわち、HBsAgに対して陰性反応を、anti−HCV抗体及びanti−HIV抗体に対して陰性反応を示す。(3)血漿(plasma)で、インフルエンザAウイルスH1N1サブタイプのRT−PCRに陰性反応を示す。(4)血清で、インフルエンザAウイルスH1N1サブタイプの単量体(monomeric)HA(H1N1)に対するELISAで1:160以上の力価(titer)を示す。志願者から約100mlの全血を収得し、lymphoprep(商標)(Axis-Shield,Norway,1114545)方法を使ってPBMC(peripheral blood mononuclear cell)を分離した。分離されたPBMCは、燐酸緩衝溶液で3回洗浄した後、KM banker II(Cosmobio,Japan,KOJ-16092010)冷凍培地(freezing medium)で2×107cells/ml濃度に合わせ、液体窒素タンク(Liquid Nitrogen Tank)に保管された。
抗原特異抗体を分泌するB細胞の選別方法は、Jinら(Jin A.et al.,2009.Nat Med.15,1088-1092)によって説明された方法を用いた。簡略に、前記実施例1で分離されたPBMCを、用意されたマイクロアレイチップ上のそれぞれのウェルに1つずつ添加した。単一細胞から分泌された抗体は、予めコーティングされた抗ヒトIgG抗体によって確認された。これを通じて選別された抗体分泌細胞(antibody secreting cell)を対象として標識されたHA抗原を用いて、HAに結合する抗体の分泌有無を確認した。確認された個別抗体分泌細胞から、逆転写重合酵素連鎖反応(RT−PCR)方法を用いて、抗体の重鎖及び軽鎖遺伝子の全体配列を確保した。確保された重鎖及び軽鎖DNAをpcDNA3.1(+)発現ベクター(in vitrogen,USA,V790-20)に挿入し、抗体の軽鎖及び重鎖をそれぞれ生産する発現ベクターを製造した後、前記製造された軽鎖及び重鎖生産発現ベクターをCHO細胞に形質感染させた。次いで、形質感染されたCHO細胞で生産された抗体を用いて、下記の実施例3に記載のHA−ELISA方法を通じてHAに結合される抗体82個を1次的に選別した。この時には、抗体サンプルを連続して希釈せず、HAとの反応を示すすべての抗体を1次的に選別した。
1次的に選別された82個の抗体から、H3N2インフルエンザウイルスのHAへの結合力が高い単一クローン抗体を2次的に選別するために、単量体HAのサブユニット(HA1)及び三量体HAを確保してHA−ELISAを進めた。インフルエンザAウイルスの再組み合わせHA1サブユニット(11056−V08H1)は、Sino Biological Inc.(China)から購入した。購入されたHA1サブユニットは、C−末端にヒスチジン(polyhistidine)残基を含むHAのN−末端分節(Met1−Arg345)で構成されており、形質感染されたヒト細胞から生産された。再組み合わせ三量体HA(FR−61)は、IRR(Influenza Reagent Resource,USA)から提供した。前記三量体HAは、C−末端ペプチドにトロンビン切断部位、三量体形成誘導ドメイン(trimerizing domain;foldon)及び6個のヒスチジン残基を含む構造であり、バキュロウイルスシステムを用いて生産された。
HA−ELISAを進めた6個の抗体を用いて多様なインフルエンザウイルスに対する生体の外中和活性を確認するために、MN(microneutralization)分析を行った。
<実施例4−1:MDCK細胞株培養及びウイルス濃度の決定>
MDCK(Madin-Darby canine kidney)は、London line(MDCK−L)を使った。MDCK細胞株は、10%のFBS(Atlas Biologicals,USA,F0500A)、1Xペニシリン/ストレプトマイシン(Gibco,USA,15140)、25mMのHEPES(Gibco,USA,15630)及び2mMLのグルタミン(Gibco,USA,25030)が含まれたDMEM(Gibco,USA,11965)培地を用いて、37℃の5%CO2湿潤(humidified)細胞培養器で培養された。
各抗体は、ウイルス希釈剤を用いて1次的に10μg/ml濃度で合わせた。この濃度を初期濃度とし、再びウイルス希釈剤を用いて連続して2倍ずつ希釈し、96ウェルプレートのウェル当り50μlずつ添加し、100 TCID50に該当する濃度のウイルスをウェル当り50μlずつ添加して、37℃の5% CO2湿潤細胞培養器で1時間反応させた。次いで、3〜4μg/ml濃度のTPCK−トリプシン(Sigma,USA,T1426)を各ウェルに入れ、かつ100μlずつの処理されたMDCK細胞株を入れた後、37℃の5% CO2湿潤細胞培養器で20時間反応させた。20時間反応させた後の過程は、実施例4−1に言及されたウイルス定量化に使われた方法と同じ方法でMN分析を進め、OD490数値を測定した。細胞のみ入れたウェルのOD490数値より高い数値を示すウェルは、ウイルスに感染されたと判断した。各抗体別にウイルス抗原が検出されていない様々なOD490数値のうち抗体の最も低い濃度(μg/ml)を表1に示し、この抗体の濃度が低ければ低いほど、ウイルスの中和活性が高いことを意味する。
v−bottom 96−ウェルプレート上で抗体を2倍ずつ連続して希釈し、4倍のHAユニットのウイルスを添加して混合させた。混合したプレートを常温で30分間反応させた後、それぞれのウェルに1% avian red blood cellを添加した。血球凝集抑制エンドポイント(end point)は、凝集反応が観察されない最も低い抗体濃度と定めた。
<実施例6−1:マウスを通じる抗体のインフルエンザインフルエンザウイルスの予防及び治療効果の調査>
CT149抗体がマウスにおいて、H3N2ウイルスの予防及び治療効果を持つかどうかを調べるために、次のように実験を進めた。5匹で構成されたマウスの各グループに、鼻腔を通じて10LD50のA/Hong Kong/68ウイルスを感染させた。CT149抗体は、kg当たり10または20mgの量をウイルス感染24時間前、またはウイルス感染後24時間や48時間後に腹腔内注射を通じてマウスに投与された。
ヒトと類似した感受性及び症状を示してインフルエンザ研究に多く使われているフェレットを用いて、CT149抗体がH3N2及びH5N1ウイルスに対して治療効果を持つかどうかを調査するために、次のように実験を進めた。
各試験群は、9匹で構成され、H3N2(A/Hong Kong/68)インフルエンザウイルスの場合に1×106 EID50/mlを、H5N1(A/Vietnam/1203/04)インフルエンザウイルスの場合に1×102 EID50/mlを鼻腔及び機関に接種した。ウイルス接種1日後にインフルエンザと関係ない陰性対照群CT−P6抗体30mg/kgあるいはCT149抗体15mg/kgや30mg/kgを単回単独静脈注射するか、またはCT149抗体30mg/kgを毎日3日間単独静脈注射した。

カバット方法によって、配列番号1で表されるCDR1領域、配列番号2で表されるCDR2領域及び配列番号3で表されるCDR3領域を含む軽鎖及び配列番号4で表されるCDR1領域、配列番号5で表されるCDR2領域及び配列番号6で表されるCDR3領域を含む重鎖で構成される抗体または抗体断片と、
カバット方法によって、配列番号7で表されるCDR1領域、配列番号8で表されるCDR2領域及び配列番号9で表されるCDR3領域を含む軽鎖及び配列番号10で表されるCDR1領域、配列番号11で表されるCDR2領域及び配列番号12で表されるCDR3領域を含む重鎖で構成される抗体または抗体断片と、
カバット方法によって、配列番号13で表されるCDR1領域、配列番号8で表されるCDR2領域及び配列番号9で表されるCDR3領域を含む軽鎖及び配列番号10で表されるCDR1領域、配列番号14で表されるCDR2領域及び配列番号6で表されるCDR3領域を含む重鎖で構成される抗体または抗体断片と、
カバット方法によって、配列番号15で表されるCDR1領域、配列番号16で表されるCDR2領域、及び配列番号9で表されるCDR3領域を含む軽鎖及び配列番号10で表されるCDR1領域、配列番号17で表されるCDR2領域及び配列番号12で表されるCDR3領域を含む重鎖で構成される抗体または抗体断片。 - 前記抗体または抗体断片は、配列番号37で表されるポリペプチド配列を含む軽鎖、及び配列番号38で表されるポリペプチド配列を含む重鎖で構成されることを特徴とする、請求項1に記載の抗体または抗体断片。
- 前記抗体または抗体断片は、配列番号39で表されるポリペプチド配列を含む軽鎖、及び配列番号40で表されるポリペプチド配列を含む重鎖で構成されることを特徴とする、請求項1に記載の抗体または抗体断片。
- 前記抗体または抗体断片は、配列番号41で表されるポリペプチド配列を含む軽鎖、及び配列番号42で表されるポリペプチド配列を含む重鎖で構成されることを特徴とする、請求項1に記載の抗体または抗体断片。
- 前記抗体または抗体断片は、配列番号43で表されるポリペプチド配列を含む軽鎖及び配列番号44で表されるポリペプチド配列を含む重鎖で構成されることを特徴とする、請求項1に記載の抗体または抗体断片。
- 前記抗体または抗体断片は、インフルエンザAウイルスH1、H3、H5、H7及びH9サブタイプからなる群から選択されたいずれか一つに対して中和活性を持つことを特徴とする、請求項1に記載の抗体または抗体断片。

- 前記抗体または抗体断片は、Fab切片、Fv切片、ディアボディ(diabody)、キメラ抗体、ヒト化抗体またはヒト抗体であることを特徴とする、請求項1または7に記載の抗体または抗体断片。
- 請求項1または7に記載のインフルエンザAウイルスに中和活性を持つ抗体または抗体断片をコードする、単離された核酸分子。
- 請求項14に記載の単離された核酸分子が挿入された発現ベクター。
- 宿主細胞に、請求項15に記載の発現ベクターが形質感染されたインフルエンザAウイルスに中和活性を持つ抗体または抗体断片生産細胞株。
- 宿主細胞は、CHO細胞、F2N細胞、BHK細胞、SP2/0細胞、NS0細胞及びHEK293細胞からなる群から選択されたいずれか一つであることを特徴とする請求項16に記載のインフルエンザAウイルスに中和活性を持つ抗体または抗体断片生産細胞株。
- 請求項1または7に記載のインフルエンザAウイルスに中和活性を持つ抗体または抗体断片を含む組成物。
- 請求項1または7に記載のインフルエンザAウイルスに中和活性を持つ抗体または抗体断片を含むインフルエンザAウイルス由来の疾患の予防及び治療用組成物。
- 請求項1または7に記載のインフルエンザAウイルスに中和活性を持つ抗体または抗体断片に標識物質を接合したコンジュゲートを含むインフルエンザAウイルス診断用組成物。
- 前記標識物質は、酵素、ルシフェラーゼ及び放射性同位元素、毒素からなる群から選択されたいずれか一つであることを特徴とする、請求項20に記載のインフルエンザAウイルス診断用組成物。
- 請求項18〜21のいずれか一項に記載の組成物の製造における、請求項1または7に記載のインフルエンザAウイルスに中和活性を持つ抗体または抗体断片の使用。
- 前記インフルエンザAウイルスは、H1、H3、H5、H7及びH9サブタイプであることを特徴とする、請求項22に記載の使用。
- 1)請求項1または7に記載のインフルエンザAウイルスに中和活性を持つ抗体または抗体断片と、
2)容器と、を含むインフルエンザAウイルス診断用キット。 - 1)請求項20に記載のインフルエンザAウイルス診断用組成物と、
2)容器と、を含むインフルエンザAウイルス診断用キット。 - 前記インフルエンザAウイルスは、H1、H3、H5、H7及びH9サブタイプであることを特徴とする、請求項24または25に記載のインフルエンザAウイルス診断キット。
