Description
a. アシルグリセロールおよびリン不純物および少なくとも1つの追加の液体非極性溶媒および少なくとも1つの追加の液体極性溶媒を含む脂質材料を反応ゾーン内に提供する工程であって、これにより少なくとも二つの相からなる系が形成されている工程。したがって、二相系は非極性相および極性相を含む。
b. 前記相系を密閉反応ゾーン内で、150℃〜300℃の温度で攪拌下加熱する工程。温度が150℃より低い場合、不純物含有量は、不適切に高いままであり、そして、温度が300℃より上昇された場合には、例えばアシルグリセロールが分解する傾向にある。好ましくは、処理温度は、燃料成分適用のための妥当な金属およびリン不純物含有量が達成され、かつアシルグリセロールの実質的な分解が起こらない、160℃〜260℃である。処理は、前記溶媒が亜臨界状態である圧力、好ましくは選択された溶媒の蒸気圧に依存して100bar以下の圧力で、リン不純物が非極性相から除去されるまで行われる。
c. 前記相系から、アシルグリセロールを含む精製オイルを含む前記非極性相を分離し、そして回収する工程。
実施例において使用された圧力反応器は、Parr Instrumentsから入手したモデル4843であった。ヘプタンは99%純度のn−ヘプタン(J.T.Baker社)であり、エタノールは変性された99.5%純度(ETAX Ba、Altia社)であった。
菜種油(Raisio社)は、それをヘプタンを用いて1:3の比に希釈し、そして、このオイル−ヘプタン溶液を等量の水を用い、圧力反応器中でこれらの成分を攪拌することにより洗浄することによって精製した。つまり、40gの菜種油、120gのヘプタン、160gの蒸留水の混合物が、1リットルの圧力反応器中でブレードミキサーを用いて、500rpmの攪拌効率で、様々な温度で30分間混合された。続いて、相が遠心分離によって分離された。上部の非極性相が収集され、そして、精製オイル成分を回収するために、溶媒であるヘプタンが、そこからロータベーパー中にエバポレートされた。
実施例1で記載された菜種油の処理が230℃で、オイル−ヘプタン比を変化させながら行われた。処理は、1:3、1:1および3:1のオイル−ヘプタン比で行われた。オイル−ヘプタン混合物は160gの質量を有していた。使用された水の量は非極性のオイル−ヘプタン相と等量であった。試験として、オイル−ヘプタン比1:3であるが5重量%の水が使用された。結果が表2および図1に示されている。
100℃で、ヘプタンおよびエタノール(3:1)を用いて湿潤バイオマスから抽出されたナンノクロロプシスオイル(Nannochloropsis oil)を、それをヘプタンを用いて1:3の比に希釈することによって、そして、このオイル−ヘプタン溶液を、設定された温度で攪拌圧力反応器中、等量の水−エタノール(1:3)溶液で処理することによって、処理した。つまり、オイル40g、ヘプタン120g、エタノール40gおよび蒸留水120gの混合物が、1リットルの圧力反応器(500rpmでの攪拌)中、設定された温度で60分間混合された。この後、相が遠心分離によって分離された。上部の非極性相が回収され、そして、精製オイルを回収するために溶媒がロータエベーバー中にエバポレートされた。
実施例3と同じナンノクロロプシスオイルが、ヘプタン中に希釈され(オイル−ヘプタン比1:3)、230℃で処理された。実験は、極性溶媒(実施例3では水およびエタノール)が種々の実験において(1)水、(2)水−エタノール(3:1)、(3)酸性のpH(2.6)を有する水、および(4)塩基性のpH(9.5)を有する水を含むように変えられている点を除き、実施例3で示されるように行われた。表4は、元のおよび処理されたナンノクロロプシスオイルの分析結果を示している。
ヘキサンを用いて抽出されたナンノクロロプシスオイルが、ヘプタン(1:2)中に希釈され、230℃で、添加された水(1:2:1のオイル−ヘプタン−水比)によって、実施例1に開示されているように処理された。結果が表5に示されている。
160℃で、ヘプタンを用いて乾燥藻類バイオマスから抽出されたドナリエラオイル(Dunaliella oil)が、200℃および220℃で、実施例3に開示されている処理により精製された。元のおよび精製されたオイルの不純物が表6に示されている。
ロードコッカス(Rhodococcus)菌の乾燥バイオマスが100℃でヘプタンを用いて抽出された。抽出オイルは、しかしながら、リン、ナトリウム、マグネシウムおよび他の無機物を非常に多く含んでいた。
動物性脂肪(Griffin Industries Inc.)がヘプタンを用いて希釈され(1:3のオイル−ヘプタン比)、そして、200℃および240℃で極性溶媒としての水とともに処理された。処理は実施例1に開示されるように行われた。
大豆油(Control Union Argentina)がヘプタンを用いて希釈され(1:3のオイル−ヘプタン比)、そして、240℃で極性溶媒としての水とともに処理された。処理は実施例1に開示されるように行われた。
実施例1で記載された菜種油の処理が、非極性溶媒としてのヘプタンの代わりに、パーム油の水素化脱酸素反応(HDO)生成物(炭化水素混合物)を用いて、230℃で行われた。処理は、1:3のオイル−HDO生成物比で行われた。オイル−HDO生成物混合物は160gの質量を有していた。使用された水の量は非極性のオイル−HDO生成物相と等量であった。水相および固体が実施例1に記載されているように分離されたが、オイルは極性溶媒から分離されなかった。精製されたオイル−HDO生成物の分析結果が表10に示されている。
分析により84%のリン脂質、14%の中性脂質(モノ−、ジ−、トリグリセリドおよび遊離脂肪酸)ならびに2%の未同定化合物を含むことが明らかとなった、市販のレシチン(粒状レシチン(granular lecithin)、Acros Organics)を、実施例1に記載したように、ヘプタン中に希釈し(レシチン16g、ヘプタン144g)、そして水(160g)とともに200℃までおよび240℃まで加熱した。
菜種油(Raisio)の酸を用いた脱ガムから得られた菜種油ガム質が、240℃で、追加のヘプタンを用いて処理された。ガム質は、主に水(約60%)、および、水和したリン脂質(約20%)の分離に由来する多少の残留トリアシルグリセロール(約20%)を含んでいた。
種々のオイルが、極性溶媒なしで、高温で非極性溶媒中に希釈されて処理された。結果は表13に示されている。
熱処理が、種々のオイルを非極性溶媒中に希釈することなく行われた。結果は表14に示されている。
実施例5と同じナンノクロロプシスオイルが、比較のため、従来のオイル精製処理である脱ガムおよび湿式漂白によって精製された。

Claims (20)
- 生物材料起源の脂質材料を精製するための方法であって、前記方法が、
a. アシルグリセロールおよびリン不純物および少なくとも1つの追加の非極性溶媒および少なくとも1つの追加の極性溶媒を含む前記脂質材料を反応ゾーン内に提供する工程であって、これにより非極性相および極性相を含む少なくとも二つの相からなる系が形成されている工程、および
b. 前記相系を密閉反応ゾーン内で、150℃〜300℃の温度、および、前記溶媒が亜臨界状態である圧力で、リン不純物が非極性相から除去されるまで攪拌下加熱する工程、および
c. 前記相系から、アシルグリセロールを含む精製オイルを含む前記非極性相を分離し、そして回収する工程
を含むことを特徴とする方法。 - 前記リン不純物が、リン脂質の形状であることを特徴とする請求項1記載の方法。
- 前記生物材料が、植物、動物または微生物材料を含むことを特徴とする請求項1または2記載の方法。
- 前記植物材料が、種子油、植物油、果実油、およびパイン油の群より選択されることを特徴とする請求項3記載の方法。
- 前記植物材料が、菜種、キャノーラ、大豆、パーム、綿、ヒマワリ、カメリナ、ジャトロファ、トウモロコシ、大麻および使用済みの食用油から選択されることを特徴とする請求項3記載の方法。
- 前記動物材料が、ビーフ、ポーク、羊もしくは鶏肉、ラード、獣脂、バターもしくは脂肪、またはそれらの混合物から選択される精製された動物性脂肪を含むことを特徴とする請求項3記載の方法。
- 前記微生物材料が、藻類、バクテリア、菌類の群より選択されることを特徴とする請求項3記載の方法。
- 前記微生物材料が、藻類および菌類の群より選択されることを特徴とする請求項3記載の方法。
- 前記脂質材料が、オイル抽出工程由来の残渣または廃棄物を含むオイルであることを特徴とする請求項1記載の方法。
- 前記脂質材料が、糖脂質およびスフィンゴ脂質より選択される複合脂質をさらに含むことを特徴とする請求項1記載の方法。
- 前記温度が、160℃〜260℃であることを特徴とする請求項1記載の方法。
- 前記少なくとも二つの相からなる系が、固体不純物または工程の間に形成された固相残渣を含む第三の相をさらに含むことを特徴とする請求項1〜11のいずれか1項に記載の方法。
- 前記非極性溶媒が、C3〜C20の脂肪族もしくは環状アルカンまたはそれらの混合物を含むことを特徴とする請求項1〜12のいずれか1項に記載の方法。
- 前記非極性溶媒が、C 5 〜C 16 アルカンまたはそれらの混合物を含むことを特徴とする請求項1〜13のいずれか1項に記載の方法。
- 前記極性溶媒が、水を含むことを特徴とする請求項1〜14のいずれか1項に記載の方法。
- 前記極性溶媒が、水および水中に容易に溶解可能であるアルコールの混合物を含むことを特徴とする請求項1〜15のいずれか1項に記載の方法。
- 前記精製オイルが非極性溶媒中に溶解されて回収され、かつ、不純物が極性相とともにまたは固体として、非極性相から除去されることを特徴とする請求項1〜16のいずれか1項に記載の方法。
- 前記脂質材料の前記非極性溶媒に対する比が、10:1未満であることを特徴とする請求項1〜17のいずれか1項に記載の方法。
- 前記脂質材料の前記非極性溶媒に対する比が、1:1未満であることを特徴とする請求項1〜18のいずれか1項に記載の方法。
- 前記脂質材料および非極性溶媒の総計量の、前記極性溶媒に対する比が、1:10より大きいことを特徴とする請求項1〜19のいずれか1項に記載の方法。
